Growth Promotion of Pavlova viridis by Bacteria Isolated from the Microalga (파블로바 비리디스로부터 분리한 세균에 의한 미세조류의 생장 촉진)

  • The marine microalga Pavlova viridis can grow fast and has the ability to accumulate essential nutrients for culturing marine animals, such as EPA and DHA, and it has been used as food for raring larval fish and prawn. The symbiotic relationship between the flagellate microalga Pavlova viridis and its associated bacteria was investigated. An axenic culture of P. viridis was obtained by repeated treatment of the microalga with an antibiotic cocktail. The axenic status was confirmed after sub-culturing three times in a sterile f/2 medium without an antibiotic. The axenic alga was then co-inoculated with five bacteria, arbitrarily designated as I1–I5, isolated from the alga to test the growth promotion of the algae. All bacterial strains promoted the growth of P. viridis, and bacterial isolate I3 was the most effective among the five bacteria tested. The cell number of P. viridis in the co-culture with I3 was significantly higher than that of the control culture. A sequence analysis of the 16S rRNA gene isolated from I3 revealed a 97% nucleotide sequence similarity to that of Citrobacter sp. The growth of strain I3 was also significantly enhanced by co-culturing with P. viridis, indicating a symbiotic relationship between the microalga and its associated bacterium. The association between the microalga and bacterium was confirmed by scanning electron microscopy.

Flavor Components of Mideoduck (Styela clava) (미더덕의 향기 성분)

  • The flavor extracts of Mideoduck muscle and its juices were concentrated by simultaneous distillation and extraction (SDE) and solid-phase microextraction (SPME) methods. Each component present in the extracts was identified with GC and GC-MS by the n-paraffin hydrocarbon retention index and standard MS library data system. By SDE, $371.3\;{\mu}g/g$ of hexanal, $80.1\;{\mu}g/g$ of 1-tridecanol, $72.1\;{\mu}g/g$ of (Z)-4,5-dimethylhex-2-en-4-ol with other alcohols, aldehydes and acids were present in the flavor extracts, with the alcohols having the highest composition and being the most important factor in Mideoduck muscle flavor. By SPME, 9 alcohols, 1 acid, 1 aldehyde, 1 hydrocarbon, 1 ester, 1 amine and 2 ketones were detected in the extracts, with alcohol such as 1-nonanol, 1-decanol and 1-tridecanol as the major components. In SPME, the muscle sample, consisting of $31.6\;{\mu}g/g$ of 1-nonanol, $20.3\;{\mu}g/g$ of (E)-2-butenedioic acid dibutylester, and $26.7\;{\mu}g/g$ of heptadecanoic acid made up the 62.1% of total flavor extracts of Mideoduck muscle. The results of the SPME methods were similar to the composition of the raw material flavor of the sample even at a low concentration.

Evaluation of Leather Meal and Meat and Bone Meal as the Fish Meal Replacer in Israeli Carp Diets (잉어사료에 있어서 피혁분과 육골분의 어분대체원으로서의 평가)

  • This study was conducted to evaluate the possible utilization and the replacing range of leather meal (LM) or meat and bone meal (MBM) as a fish meal replacer for the dietary animal protein source of growing israeli carp, Cyprinus carpio. Five different diets were formulated on isonitrigenous and isocaloric basis of 38.5% curde protein and 15.3 KJ/g diet. Percentage of the graded levels of replacement of white fish meal (WFM) by LM/MBM on the basis of crude protein were at follows : diet 1, 100% WFM (control) ; diet 2, 75% WFM+25% LM ; diet 3, 50% LM ; diet 4, 75% WFM+25% MBM ; diet 5, 50 WFM+50% MBM and LM or MBM, and approximately 64.5% of plant protein. After one week of conditioning period, fish averaging 10g were divided into five groups and fed one of the experimental diets for 12 weeks. In the first and the second 4 weeks of feeding trials, there were no significant differences in weight gain between two groups of fish fed diet 1 (control) and 2 (P>0.05), while fish fed diet 3, 4 and 5 had a significantly lower body weight gain than fish fed diet 1 (P<0.05). In contrast to those two periods, all diet groups were not significantly different compared to diet 1 in body weight gain during the third 4 weeks of feeding trial (P<0.05). These results indicated that LM and MBM could be used as a fishmeal replacer up to 50% in growing israeli carp.

Growht and Size Variation of (Brachionus plicatilis) Reared at Different Conditions (배양조건에 따른 Rotifer (Brachionus plicatilis)의 성장과 크기변화)

  • The growth and size variation of rotifer (Brachionus plicatils) were examined at the various water temepratures ranging form 18 to $36^{\circ}C$, and under continuous light condition. Either Chlorella ellipsoidea or $\omega$-yeast were fed to rotifer as a food organism. Compared to large-type rotifer fed of $\omega$-yeast, those fed on C. ellipsoildea generally showed higher growth at 7th after starting culture under the water temperaure of less than $30^{\circ}C$. Above such temperature, the large-type rotifer fed on $\omega$-yeast showed higher growth rather than those fed on C. ellipsoidea and the maximum cell number reached 1,420 ind./m$\ell$ at $36^{\circ}C$, whereas when the small-type rotifer was fed on $\omega$-teast, its maximum cell number was to be 1,669 ind./m$\ell$at $34^{\circ}C$. The striking decrease in size was found in large-type rotifer which was to be $112.1\mu m$m(initial : 202.0${\mu}{\textrm}{m}$) when they were fed on C. ellipsoidea at $34^{\circ}C$ for 5 days after starting culture. A great increase in size from the initial of 157.0 to $201.3\mu m$mwas show in small-type rotifer fed on $\omega$-yeast at $20^{\circ}C$,for 5 days. The present results demonstrated that size variation of rotifer could be controlled by changing culture temperature.

  • Growth of triploid abalone, Haliotis discus hannai induced by cody (3$^{\circ}C$) shock and its feed efficiency were investigated from larva to adult for 51 months. After 51 months from triploidy induction, the triploid abalones have outgrown to diploid abalones in shell length and total weight. Triploid abalones with inhibition of extrusion of first polar body (3n-1pb) were outgrown to diploid abalones, however, triploid abalones with inhibition of extrusion of second polar body (3n-2pb) were not significantly different from diploid controls in shell length and total weight through the whole rearing period (P<0.05), because of their heterozygosity differences. Daily feeding rates and feed conversion rates decreased with the growth of abalones and both rates had no differnce between two experimental groups. After 51 months from inducing triploid, conditin index of triploid abalone (64.1%) was higher than that of diploid control (59.4%) (P<0.05). Survival rate was 63.0% in triploid group (3n-1pb 62.0%, 3n-2pb 64.0%) and 62.0% in diploid group during the experimental period.

Effects of Fish Meal Analogue on the Growth of Israeli Strain of Common Carp, Cyprinus carpio (이스라엘 잉어(Cyprinus carpio)에 대한 어분 대체품의 성장 효과)

  • This study was conducted to investgate the effect of fish meal analogue (FMA) on the growth of Israeli carp, Cyprinus carpio averageing 43g. Leather meal, meat and bone meal, feather meal, squid liver powder, poultry by-product and blood meal were used for ingredient of FMA. Four experimantal diets were formulated to contain 36% crude protein and 15.4 kJ available enery g-1. The percentage of follows ; Diet 1, 100% 3, 60% WFM + 40% FMA (40% FMA) ; Diet 4, 40% WFM + 60% FMA (60% FMA). In this expriment, fingerling Israeli carp averaging 43 g were assigned to each diet treatment. When the experiment was terminated, weigth gain (WG), feed conversion (FC), specific growth rate (SGR) and protein efficiency ratio (PER) were measured respectively. In the experiment, WG, FC, SGR and PER of Diet 2 were not significantly different from those of the control diet (P0.05). And WG, FC and SGR of Diet 3 were not significantly of the control Diet. In Diet 4, WG, FC and PER were not significantly different from those of the control Diet, but SGR was lower than that fo the control Diet. According to the results from this study, it might be possible to replace fish meal protein by FMA up to 60% in fingerling Israeli carp diets.

  • Successive management of prawn farm is strongly dependent upon monitoring of pond seawater quality which is generally influenced by an excessive food supplied sediment type and phytoplankton composition in the pond. For good condition of seawater quality it must need exchangning of fresh seawater by tidal current. Two distinct shrimp ponds Galha and Yunho which were different in seawater exchanging system and sediment type were selected to understand how some factors affected to seawater and sediment qualities in the pond. Prawn growth was also determined. Galha pond characterized by sand bottom with water exchanging by turn of the tidal current accumulated 1.8 mgS/g-dry as sulfide in sediment while Yunho pond mud- bottomed with seawater exchanging of pumping system showed 4.7mgS/g-dry when it was highest, Ammonia-N and hydrogen sulfide measured in the seawater were 0.31mg/${\ell}$ and 21.2 ${\mu}$${\ell}$/${\ell}$in Yunho and 0.10mg/${\ell}$and 10.8${\mu}$${\ell}$/${\ell}$in Galha pond respectively. Dissolved oxygen remained below 6.0mg/${\ell}$ in Galha and 5.0mg/${\ell}$in Yunho pond from June through August. Less growth of prawn was found in Yunho pond than in Galha pond. Prawn growth expressed as body length and weight were 138.3mm 22.9g in Yunho pond while they were length 158.2mm and 28.9g in Galha pond respectively when they were harvested in October. These results indicate that higher levels of ammonia-N and hydrogen sulfide and lower dissolved oxygen in bottom seawater of Yunho pond might affect the growth of cultured prawn.

Effects of Bisroot in the Diet on Growth, Body Composition, Immume responses of Nile Tilapia, Oreochromis niloticus (사료 중 비스루트가 나일틸라피아, Oreochromis niloticus의 성장, 체조성 및 면역 방응에 미치는 영향)

  • The present study investigated the effects of Bisroot, that contains live bacteria (Bacillus polyfermenticus, Bacillus mesentericus, Streptococcus faecalis, & Bifidobacterium breve) and digestive enzymes (protease, lipase), on the growth, body composition and immune response of Nile tilapia fingerlings. One percent of the Bisroot was added to the experimenta feed. All exprimental fish were fed for 60 days. The weigh gains among the experimental fish were not significntly different (P>0.05). Hematocrit value, hemoglobin, total protein, glucose, GOT, and GPT were unaffected by Bisroot treatment. However, it was observed that glucose, GOT, and GPT value in the fish that were fed Bisroot, were lower than the control. The complement activity ($CH_50$) tended to be significantly increased by Bisroot treatment, but not lysozyme activity. Phagocytosis and respiratory burst activities of macrophages in the head kidney were enhanced by Bisroot. Therefore, the Bisroot diet enhances the cellular immune activities were enhanced by Bisroot. Therefore, the Bisroot diet enhances the cellular immune activities of non-specific immune responses. When fish were challenged with a virulent strain of Edwardsiella tarda, the Bisroot treated fish were more resistant than the control. The present results suggest that the introduction of Bisroot into the diet of Nile tilapia could increase their resistance against bacterial infection, reduce fish mortality, and offers economic benefits.

  • The influence of starvation on survival, growth and feeding incidence of larval tiger puffer Takifugu rubripes was investigated for 16 days after let them hatched at the temperature of 16.5~18.5${\circ}C$. The effect by delayed initial feeding on larvace was also studied. Almost all larvae began to be fed from the stage of yolk absorption about 5 days after hatching, in which rearing trials revealed that their survival and growth were influenced by food availability. Survival of unfed larvae, which stated to decrease drastically since the oil globule was exhausted about 9 days after hatching, dropped to 50% and 0% respectively on the 11.5th and 16th day. the difference of growth between fed and unfed larvae came into sight on about 5th day after hatching when the initial feeding was started, and had reduced in proportion to the length of starvation period. The condition factors for the unfed larvae were lower than thost of the fed. Of the larvae which had starved less than 5 days, more than 80% could consume food upon feeding. No fish had the ability in having food after 10 days. It appeared that the turning point of recover for the starving larvae came on 7th or 8th day. Their taking-food ability seemed to be affected by the change in swimming activity which depended on the duration of starvation. For the seed production of tiger puffer, therefore, feeding should be initiated as soon as possible, so as to enhance the production and survival of healthy seeds.

Comparison of Acute Toxicity of Ammonia in Juvenile Rockfish and Red Sea Bream (조피볼락과 참돔 유어에 대한 암모니아 독성 비교)

  • This study compared acute ammonia toxicity in juvenile rockfish and red sea bream. Oxygen concentration affected ammonia toxicity to aquatic animals. Without aeration, the 96-hr $LC_50$ values of $NH_4^+$and un-ionhized ammonia for juvenil rockfish(3.6g, 6.6cm) were 2.61 and 0.09 mg/l, respectively. The 3-hr $LC_50$ values of $NH_4^+$ for the size of 1.9g (5.2cm) and 3.6g (6.6cm) rockfish, and 1.0g (3.8cm) and 2.0g (5.0cm) red sea bream without aeration were 3.96, 3.94, 4.35 and 3.75 mg/l, respectively. When oxygen level was low, larger fish were more susceptible to ammonia toxicity than smaller one because of stress resulted from low oxygen. However, with aeration, the 96-hr $LC_50$ values of $NH_4^+$ for the size of 1.2g (3.9cm) and 2.3g (5.1cm) red sea bream with aeration were 3.84 and 3.90mg/l, respectively. The 6-hr $LC_50$values of $NH_4^+$ for the size of 3.1g (5.9cm) and 6.2g (7.0cm) rockfish with aeration were 3.83 and 3.94 mg/l, respectively. When oxygen level was high, larger rockfish and red sea bream were less susceptible to ammonia toxicity than smaller ones. The 6 hr-or 96 hr-$LC_50$ values of $NH_4^+$for rockfish with reduced ammonia toxicity. In comparing 96 hr-$LC_50$ values of $NH_4^+$ and un-ionized ammonia for juvenil rockfish with those for juvenil red sea bream, the values for rockfish were lower than for red sea bream. This indicates that juvenile rockfish is more susceptible to ammonia toxicity than juvenile red sea bream.
