  • The objectives of this investigation were to produce artificially an antitumor constituent by submerged culture of the mycelium of Coriolus versicolor (Fr.) Quel., to characterize the influence of various modifications of the nutrient and culture conditions with respect to the pro­duction, to determine chemical composition of the antitumor constituent, and to examine effects of the constituent on the immune response of mice. Submerged agitation of the mycelium in flasks containing a nutrient solution showed its adequate growth. Especially the mycelial growth in the medium containing glucose and yeast extract was abundant. The addition of cotton seed flour or ginseng waste to the medium increased the yield of mycelial growth and the production of the antitumor constituent. The replacement of glucose with starch also yielded the adequate growth. The antitumor constituent extracted from the mycelium and isolated from the culture filtrate was a protein-bound polysaccharide. The analyses of this constituent by GLC and amino acid autoanalysis showed that it contained four monosaccharides and fifteen amino acids. The protein-free polysaccharide of the constituent was also found to exert greater antitumor activity against sarcoma-180 in mice than the entire constituent. The antitumor constituent was found to potentiate the immune response of mice against sheep red blood cell. The protein-bound polysaccharide exerted more favorable influence on the immunity than the protein-free moiety.

  • A sensitive and efficient assay method was applied to determine the level of glutamic acid (GA) and ${\gamma}-aminobutyric$ acid (GABA) in frontal cortex and hippocampus of rat administrated with ethanol and drugs. The compounds were derivatized with ο-phthalaldehyde (OPA) and 2-mercaptoethanof for precolunm analysis. The condition for the simultaneous determination of GA, GABA and $\beta-aminobutyric$ acid (BABA) by high performance liquid chromatography with electrochemical detection was reverse phase $C_{18}$ column as stationary phase, 0.1 M phosphate buffer containing 0.1 mM $Na_4EDTA$ : methanol = 55:45 (v+v) pH 3.8 as mobile phase and 0.7V electrode voltage. The stability of reaction product of GA, GABA and BABA with OPA could be increased by adding the same volume of polyethylene glycol 400 to reaction mixture. The GABA level in frotal cortex of rat was significantly decreased by the administration of picrotoxin and diazepam, but it was significantly increased by the administration of red ginseng total saponin, N-methyl-D-glucamine and (-)-deprenyl.

Potential Effects of Microglial Activation Induced by Ginsenoside Rg3 in Rat Primary Culture: Enhancement of Type A Macrophage Scavenger Receptor Expression

  • Brain microglia are phagocytic cells that are the major inflammatory response cells of the central nervous system and widely held to play important pathophysiologic roles in Alzheimer's disease (AD) in both potentially neurotoxic responses and potentially beneficial phagocytic responses. In the study, we examined whether ginsonoside Rg3, a by-product of red ginseng, enhances the microglial phagocytosis of $A{\beta}$. We found that Rg3 promoted $A{\beta}$ uptake, internalization, and digestion. Increased maximal $A{\beta}$ uptake was observed at 4 and 8 h after Rg3 pretreatment (25 ${\mu}g/mL$), and the internalized $A{\beta}$ was almost completely digested from cells within 36 h when pretreated with Rg3 comparing with single non-Rg3-treated groups. The expression of MSRA (type A MSR) was also up-regulated by Rg3 treatment in a dose- and time-dependent manner which was coincidently identified in western blots for MSRA proteins in cytosol. These results indicate that microglial phagocytosis of $A{\beta}$ may be enhanced by Rg3 and the effect of Rg3 on promoting clearance of $A{\beta}$ may be related to the MSRA-associated action of Rg3. Thus, stimulation of the MSRA might contribute to the therapeutic potentials of Rg3 in microglial phagocytosis and digestion in the treatment of AD.

  • As a basic study to elucidate nutritional physiology and composition of synthetic medium of red rotting bacteria, Erwinia carotovora, of ginseng, the effects of hydrogen ion concentration, various kinds of carbon sources, nitrogen source, micrometallic salts and it's concentration on the gorwth of the bacteria were investigated and the results were as follows. Optimal pH in the basal medium for the growth of the bacteria was 6.5. After incubation the pH in culture media was neutralized. Among the various kinds of carbon sources, sucrose, glucose mannitol, but organic acids were not utilized effectively as nutrients. After incubation the pH turned acidic. Alanine as organic nitrogen sources nad ammonium sulfate as inorganic nitrogen promoted the growth, but L-valine and sodium nitrite were the least effective. Ferric chloride 1.0mg/dl and ferrous sulfate 100mg/dl were the most effective as micrometallic sources. Control and boric acid were the least effective. New synthetic medium based on the above results was follows ; Alanine 1.0g, $KH_2PO_4$ 1.0g, sucrose 30.0g, $MgSo_4$ $7H_2$O 0.5g, $FeCl_36H_2$O 1.0mg thiamine 200.gamma.g, and distilled water 1000ml, pH6.5.

Comparison of Growth Characteristics and Compounds of Ginseng Cultivated by Paddy and Upland Cultivation (논 . 밭재배에 따른 인삼의 생육 및 성분 특성 비교)

  • This study was carried out to investigate the difference of growth characteristics and the content of root chemical components in four years old ginseng by paddy and upland cultivation at farmers' field in Korea. Proportions of silt, clay, liquid phase and porosity were higher in paddy soil than upland soil. The range of liquid phase was $17.5{\sim}19.5%$ in paddy and $7.0{\sim}12.8%$ in upland during growth period. EC and the other contents of OM, $NO_3^-,\;K_2O$, and Mg in paddy soil were higher than those of upland soil, while the contents of $P_2O_5$ and Ca were less than those of upland soil. The levels of chemical components of tested soil exceeded recommended range in EC, $NO_3^-$ and Ca of paddy soil, and in $P_2O_5$ and Ca of upland soil. Stem length, fresh root weight and total dry weight per plant in paddy were greater than those of upland. Root weight in paddy-ginseng showed a great increase on September, while it was not increased in upland because of early defoliation. Net assimilation rate and crop growth rate by paddy and upland cultivation showed distinct differences on May and September, and those of paddy-ginseng were higher than those of upland-ginseng. Yield and ratio of red-colored root showed no significant difference by paddy and upland cultivation, while significant differences were observed in diameter and length of primary root, contents of crude saponin and 50% ethanol extracts of primary root, and water content of root. Hardness of primary root showed no significant difference by paddy and upland cultivation until August, but it showed distinct difference on September, at which the hardness in upland cultivation was drastically decreased.

  • In order to improve the physical properties on moisture sorption of spray dried red ginseng extract powder (SD-RGEP), the various additives and coating agents were treated, and solubility, sedimentation rate and storage stability of RGEPs treated were investigated. For the moisture-proofing, additive itself was effective in the order casein>dextrin>starch>avicell, but RGEPs treated with additives were effective in the order cord oil+tween-40>starch>casein. But there was no significant difference between RGEP treated less than 1% additive and the moisture-proofing. The coating effect of AEA and CAP on RGEP could not be recognized for the moisture-proofing, whereas that of HPC, corn oil, lecithin and HPC+corn oil was proved to be very effective for the moisture-proofing of RGEP. Also it is required to control the initial moisture content of RGEP. The sedimentation rate of RGEP nontreated was 65.1 seconds, whereas that of RGEPs coated with 1% HPC, 1% corn oil, 1% lecithin and 1% HPC+0.5% corn oil was in the range of 96.2 to 114.3 seconds. The sedimentation rate of RGEPs coated was 1.5 to 1.8 times higher than that of RGEP nontreated, and there was significant difference between solubility and sedimentation rate. Therefore it was no matter for the keeping quality. The sorption rates of RGEPs coated with lecithin $(0.5{\sim}1.0%$), corn oil(0.8%) and cellulose acetate (0.8%) were ranged 54 to 56%, 51 to 55% and 52 to 54%, respectively, and it is found that the moisture-proofing effect of RGEPs coated was about 2 times higher than that of RGEP nontreated. From the result of storage experiment during 3 months under the maltreated condition of $48{\pm}2^{\circ}C$, 75% RH, the moisture sorption of RGEPs coated with corn oil$(0.5{\sim}1.0%$), cellulose acetate(DES, $0.5{\sim}0.8%$) and lecithin (DES, 1.0%) was not at all observed. Therefore it is considered that the circulating period of RGEPs coated with these coating agents could to give more than 3 years.

Evaluation on the Muscular Strength Activity of Medicinal Herb Hot-Water Extracts (생약 열수추출물의 근력향상 효능 평가)

  • Effects of medicinal herbs on the improvement of muscular strength along with the related fatigue factors were evaluated in SD rats. Nine types of treatment groups were fed diets supplemented with 1% herb hot-water extracts (Liriope spicata (LS), Schizandra chinensis (SC), Acanthopanax sessiliflorus (AS), Saliconia herbacea (SH), Eucommia ulmoides (EU), Lycium chinensis (LC), Panax ginseng (red ginseng) (PG), Polygonum multifloum (PM), Glycyrrhizae uralensis (GU)) for 4 weeks. The muscular strength of all groups were measured weekly for 4 weeks. After 4 weeks serum was collected and liver dissected out for the analysis of glycogen and fatigue factors. The liver glycogen contents of all treatment groups ($21.0{\sim}25.9mg/g$) were higher than that of control group (18.1 mg/g). A significantly increase of muscular strength in the fourth week were found in the AS (282.5 gf), SH (277.4 gf), PG (287.2 gf) groups (p<0.05). LS (7.88 mg/dL), SC (7.85 mg/dL), AS (7.64 mg/dL), EU (7.54 mg/dL), LC (7.81 mg/dL) and PG (7.75 mg/dL) groups were significantly reduced in serum inorganic phosphorus concentration measured after 4 weeks (p<0.05). Serum lactate levels of treatment groups ($8.61{\sim}12.18{\mu}g/dL$) were significantly lower than that of control group ($17.45{\mu}g/dL$). These results suggest that medicinal herbs enhanced muscular strength of rats by delaying accumulation of muscular fatigue factor.

A study of consumers' perceptions and prediction of consumption patterns for generic health functional foods

  • The Korea Food and Drug Administration (KFDA) revised the Health Functional Food Act in 2008 and extended the form of health functional foods to general food types. Therefore, this study was performed to investigate consumers' perceptions of the expanded form of health functional food and to predict consumption patterns. For this study, 1,006 male and female adults aged 19 years and older were selected nationwide by multi-stage stratified random sampling and were surveyed in 1:1 interviews. The questionnaire survey was conducted by Korea Gallup. The subjects consisted of 497 (49.4%) males and 509 (50.6%) females. About 57.9% of the subjects recognized the KFDA's permission procedures for health functional foods. Regarding the health functional foods that the subjects had consumed, red ginseng products were the highest (45.3%), followed by nutritional supplements (34.9%), ginseng products (27.9%), lactobacillus-containing products (21.0%), aloe products (20.3%), and Japanese apricot extract products (18.4%). Opinions on expanding the form of health functional foods to general food types scored 4.7 points on a 7-point scale, showing positive responses. In terms of the effects of medicine-type health functional foods versus generic health functional foods, the highest response was 'same effects if the same ingredients are contained' at a rate of 34.7%. For intake frequency by food type, the response of 'daily consistent intake' was 31.7% for capsules, tablets, and pills, and 21.7% for extracts. For general food types, 'daily consistent intake' was 44.5% for rice and 22.8% for beverages, which were higher rates than those for medicine types. From the above results, consumers had positive opinions of the expansion of health functional foods to generic forms but are not expected to maintain accurate intake frequencies or amounts. Thus, continuous promotion and education are needed for proper intake of generic health functional foods.

The Use of Complementary and Alternative Medicine in Patients with Type 2 Diabetes Mellitus: Community Based Survey (지역사회 기반 당뇨병 환자의 보완대체요법 이용 실태조사)

  • Objectives : To investigate the prevalence and utilization pattern of complementary and alternative medicine (CAM) in individuals with type 2 diabetes. Methods : We conducted a nationwide survey and a total of 535 individuals with type 2 diabetes were asked about their demographic, diabetes-related characteristics and CAM use. We used multiple logistic regression modeling to determine independent predictors of CAM use. Results : Of the 535 subjects, 417 (77.9%) used CAM in last 12 months. The most commonly used modality was red ginseng (17.9%). CAM use was independently associated with being married, northern area residence, higher education and diabetes duration of 3-5 years. More than half(58.3%) of the CAM users used it expecting a better outcome in addition to conventional therapy. Information sources about CAM were mostly friends (36.0%) and family (31.9%), only 28.1% of diabetic patients seek counsel from medical doctors or oriental medical doctors about CAM. Many people perceived CAM as effective (62.7%) and reported side effects only rarely (1.2%). Conclusions : Many diabetic patients used CAM and found it effective, which urges clinicians to pay attention to CAM use of their diabetic patients. Also, further studies are required regarding efficacy and safety about CAM.

Postbiotics Enhance NK Cell Activation in Stress-Induced Mice through Gut Microbiome Regulation

  • Recent studies have revealed that probiotics and their metabolites are present under various conditions; however, the role of probiotic metabolites (i.e., postbiotics in pathological states) is controversial. Natural killer (NK) cells play a key role in innate and adaptive immunity. In this study, we examined NK cell activation influenced by a postbiotics mixture in response to gut microbiome modulation in stress-induced mice. In vivo activation of NK cells increased in the postbiotics mixture treatment group in accordance with Th1/Th2 expression level. Meanwhile, the Red Ginseng treatment group, a reference group, showed very little expression of NK cell activation. Moreover, the postbiotics mixture treatment group in particular changed the gut microbiome composition. Although the exact role of the postbiotics mixture in regulating the immune system of stress-induced mice remains unclear, the postbiotics mixture-induced NK cell activation might have affected gut microbiome modulation.
