Inhibitory Effect of a Decoction Combined with Ostericum koreanum Maxim. and Aralia continentalis Kitagawa on Collagen II-induced Arthritis Mice (Type II Collagen으로 유도된 관절염에 대한 강활(羌活), 독활(獨活) 배합약물의 억제 효과)

  • Objectives : To clarify the anti-arthritic activity of Ostericum koreanum Maxim. (OS) plus Aralia continentalis Kitagawa (AC) in vivo. Methods : All mice were immunized with bovine type II collagen. After a second collagen immunization, mice were treated with OS plus AC once a day for 7 weeks. Oral administration of OS plus AC (200 or 50 mg/kg) significantly suppressed the progression of CIA, which extend is comparable to that of methotrexate (MTX, 0.3 mg/kg), a positive control. The severity of arthritis within the knee joints was evaluated by histological assessment of cartilage destruction and pannus formation. Results : Administration of OS plus AC significantly suppressed the progression of CIA and inhibited the production of TNF-${\alpha}$ and IL-6 in serum. The erosion of cartilage was dramatically reduced in mouse knees after treatment with OS plus AC. In conclusion, our results demonstrates that OS plus AC significantly suppressed the progression of CIA and that this action was characterized by the decreased production of IL-6, IFN-${\gamma}$ and collagen II specific antibody in serum, CD3+CD69+ T cells, MHC class II+/CD11c+ (in DLN), CD11b+Gr-1+ cells (in PBMC), CD11b +Gr-1+ cells, B220+/CD23+ (in paw joint). Conclusions : The the levels of IFN-${\gamma}$ in the culture supernatant of splenocytes stimulated with CD3/CD28 or collagen were dramatically decreased, while those of IL-4 was increased. In the serum of OS and AC-treated mice, the levels of IgM RA factor were decreased.

Seed Germination Rate and Growth Characteristics according to Ripening Stages in Angelica acutiloba Kitagawa (일당귀의 등숙에 따른 종자 발아 및 생육 특성)

  • Background: Angelica acutiloba Kitagawa belongs to the flowering plant family Apiaceae. Its young leaves are consumed as a vegetable, and its roots have medicinal properties. This study was conducted to analyze the seed germination characteristics of A. acutiloba according to its ripening stages. Methods and Results: The seeds were harvested from a research farm managed by the Department of Herbal Crop Research, Rural Development Administration in 2018 and were divided into six groups according to their specific gravities. In particular, we studied the effect of harvesting periods and umbel orders on seed characteristics. The results showed that the rates of germination, emergence, and early growth improved as the specific gravity of the seeds increased. In addition, the germination rates of the seeds harvested in mid July and early August were significantly higher than those harvested in the other seasons, and the seeds obtained from the first floret had the greatest mass and weight. Conclusions: In this study, we demonstrated that it is possible to improve the germination rate by appropriate selection of seeds and harvesting period both of which are closely related to seed maturity.

Effect of GA3 Treatment on Seed Germination, Seedling Growth and Useful Component Content of Angelica acutiloba (Siebold & Zucc.) Kitagawa (GA3 처리 왜당귀의 종자 발아, 유묘 생육 및 유용성분 특성 분석)

  • Germination, growth characteristics, and content of useful components were analyzed through GA3 treatment of Angelica acutiloba (Siebold & Zucc.) Kitagawa seeds, to provide basic data for cultivation technology. The moisture absorption rate of the seeds was 62.07% after 2 hours of soaking, and the highest value was 122.15% after 48 hours. The appropriate seed germination temperature for the species ranged from 10℃ to 25℃. It was confirmed that the growth of seedlings increased as the GA3 concentration increased, but there was no significant difference in the content of useful components according to the GA3 treatment.

Antioxidative and Nitrite Scavenging Abilities of Medicinal Plant Extracts (한약재 추출물의 항산화작용 및 아질산염 소거작용)

  • The purpose of this study was to investigate antioxidative and nitrite scavenging abilities of extracts of medicinal plants. Three kinds of medicinal plants, Angelica acutiloba Kitagawa (AA), Akebia quinate Decaisne (AQ), and Caragana chamlagu (CC), were extracted with water and $70\%$ ethanol. The yields of water extracts were increased in the order of Akebia quinate Decaisne ($35.63\%$, Caragana chamlagu ($31.36\%$ and Angelica acutiloba ($8.98\%$). Those of ethanol extracts were Akebia quinate Decaisne ($31.82\%$, Angelica acutiloba Kitagawa ($14.15\%$, and Caragana chamlagu ($5.20\%$). The highest electron donating ability (EDA) at 300-1,000 ppm of water extract was Angelica acutiloba Kitagawa, ranged from $62\%$ to $88\%$ and that of ethanol extract was Akebia quinate Decaisne, ranged from $72\%$ to $87\%$. Nitrite scavenging ability (NSA) of extracts measured at various pH (1.2, 3.0, 4.2, 6.0) showed highest ability in all extracts at pH 1.2 and decreased with increasing pH. The highest NSA of water extracts of 1,000 ppm at pH 1.2 showed $35-40\%$, in Caragana chamlagu and that of ethanol extract was $33-42\%$, in Akebia quinate Decaisne. From these results, Akebia quinate Decaisne was found as a potential raw material baring functionality in the respects of extract yield, EDA and NSA.

Plant Regeneration from Somatic Embryo with Structural Diversity from Leaf Explant Culture of Ostericum koreanum Kitagawa (강활(Ostericum koreanum Kitagawa)의 잎절편체 배양으로 부터 발생된 여러가지 형태의 체세포배를 통한 식물체 재생)

  • This study was carried out in order to establish plant regeneration via somatic embryogenesis from leaf explant of Ostericum koreanum Kitagawa and to elucidate the effects of NAA and cytokinins (kinetin, BA) on the abnormalities of somatic embryo and the relationship between thecotyledon numberand germinability. Calli were formed on leaf explants cultured on MS agar medium supplemented with various concentrations (0, 0.1, 0.5, 1, 2 mg/L) of NAA and cytokinins. The calli were white, watery and soft, became browning during cultures. Somatic embryos were formed from pale yellowish calli derived browning calli. High frequency somatic embryos were observed on MS medium containing 1 mg/L NAA and 0.1 mg/L BA after 60 days of culture. The mature somatic embryos germinated into plantlets without subculture after 2 weeks. The frequency of normal somatic embryo with two cotyledons was 39.8%. On the other hand, cotyledonary abnormalities of somatic embryos were observed at considerable frequency: 33.6% of somatic embryo with one cotyledon, 15.3% cotyledons with three, 8.2% four cotyledons and 3.1% jar shaped cotyledon. Germination frequency of somatic embryos with two cotyledons was 97.4%, and that of the embryos with abnormal cotyledon was almost similar to that of embryos with two cotyledons, except jar shaped somatic embryos (33.3%).

Studies on Germination Rates and Early Seedling Growth Characteristics by Different Storing Temperatures, Durations, and Methods in Aralia cordata var. continentalis (Kitagawa) Y.C.Chu (독활 종자의 저장온도, 기간 및 방법에 따른 발아율과 유묘 생육특성 연구)

  • This study was conducted to improve the managing and storing methods of the seeds of Aralia cordata var. continentalis (Kitagawa) Y.C.Chu, to examine the viability and the germination ability of seeds with different storing conditions and methods, and to develop new ways to propagate and have better healthy seedling. Therefore, the germination rate, days required for germinating seeds, and early growth responses of Aralia cordata var. continentalis (Kitagawa) Y.C.Chu were investigated with different storing temperatures, durations and methods. The germination rate was higher in stratified storage than that in dry storage condition. The highest germination rate was with outdoor temperature at 30 days after stratified storage. The days required for germinating seeds were less than 10 days with the treatment of $25^{\circ}C$ and outdoor temperature in stratified storage. In dry condition, they were shorter with $4^{\circ}C$ and $25^{\circ}C$ than those with $-20^{\circ}C$ and outdoor temperature. Leaf number of seedling was higher in stratified storage compared to that in dry condition, while it was not clearly different according to storage temperatures and durations. Leaf length and leaf width of seedling was not difference among the treatment of storage methods, temperatures, and durations. Stem length of seedling was higher in stratified storage than those in dry condition, while root length was not clearly different among the treatments. It would be assumed that temperatures, methods and durations of storage could affect much to the germination rate and the early seedling growth response.

  • An objective and one-step detection procedure of multiple outliers is suggested by using the gaps of the order statistics. The detection procedure can be used as a routine outlier detection method of a statistical analysis computer program. The procedure is applied to some examples including the data selected by Kitagawa.
