세포내 수송 기구는 세포의 작용과 생존에 필수적이다. 이러한 세포내 수송은 긴 미세소관을 따라서 운반체를 운반하는 미세소관 의존 분자 모터 단백질인 kinesin과 cytoplasmic dynein에 의하여 이루어진다. Kinesin은 ATP 의존적으로 미세소관의 plus-end방향으로 이동하는 모터 단백질로 세포내 소기관, 분비소포, RNA 복합체, 단백질 복합체들을 수송한다. Kinesins에 의한 다양한 운반체의 수송의 이상은 세포의 기능 이상과 연관된다. Kinesins에 의한 운반체 수송의 기본 단계는: 운반체 혹은 adaptor 단백질과의 결합, kinesin 기능 활성화와 미세소관을 따라서 이동, 그리고 올바른 위치에서 운반체와의 분리 단계로 나뉘어 진다. 최근의 연구결과들에서 kinesin 모터 기능 활성화, 운반체와의 결합, 운반체와의 해리 기전이 확인되고 있으며 세포내 운반체 수송은 kinesin과 운반체를 연결하는 adaptor 단백질에 의하여서도 조절된다. 단백질 인산화 효소, 탈 인산화 효소를 포함하는 kinesin 모터 활성 조절 단백질들은 kinesin의 인산화 혹은 탈 인산화를 통하여 직접적으로 세포내 수송을 조절하거나, c-Jun NH-terminal kinase-interacting proteins (JIPs)와 같은 adaptor 단백질들과 미세소관의 간접적 수식을 통하여 세포내 수송을 조절하기도 한다. 이러한 연구결과들은 세포의 기능과 형태 유지에 관여하는 kinesin에 의한 다양한 세포내 수송 조절 기전을 이해하는데 기초적인 토대가 된다. 또한 각각의 kinesin에 대한 조절 기전을 밝히는 것은 세포생물학과 신경생리학을 이해하는데 중요하므로 본 종설에서는 kinesin에 의한 세포내 수송을 조절하는 단백질과 kinesin과 수송체와의 결합이 어떻게 조절되는지를 고찰하고자 한다.

In eukaryotes, chromosomes undergo a series of complex and coordinated movements during cell division. The kinesin motor proteins, such as the chicken Chromokinesin, are known to bind DNA and transport chromosomes on spindle microtubles. We previously cloned a family of retrograde C-terminus kinesins in Caenorhabditis elegans that mediate chromosomal movement during embryonic development. Here we report the cloning of a C. elegans klp-12 cDNA, encoding an ortholog of chicken Chromokinesin and mouse KIF4. The KLP-12 protein contains 1609 amino acid and harbors two leucine zipper motifs. The insitu RNA hybridization in embryonic stages shows that the klp-12 gene is expressed during the entire embryonic development. The RNA interference assay reveals that, similar to the role of Chromokinesin, klp-12 functions in chromosome segregation. These results support the notion that during mitosis both types, the anterograde N-terminus kinesins such as KLP-12 and the retrograde C-terminus kinesins, such as KLP-3, KLP-15, KLP-16, and KLP-17, may coordinate chromosome assembly at the metaphase plate and chromosomal segregation towards the spindle poles in C. elegans.
Kinesin is an ATP-driven microtubule motor protein that plays important roles in control of microtubule dynamics, intracellular transport, cell division and signal transduction. The kinesin superfamily is composed of numerous members that are classified into 14 subfamilies. Animal kinesins have been well characterized. In contrast, plant kinesins have not yet to be characterized adequately. Here, a novel plant-specific kinesin gene, GhKCH2, has been cloned from cotton (Gossypium hirsutum) fibers and biochemically identified by prokaryotic expression, affinity purification, ATPase activity assay and microtubule-binding analysis. The putative motor domain of GhKCH2, $M_{396-734}$ corresponding to amino acids Q396-N734 was fused with 6$\times$His-tag, soluble-expressed in E. coli and affinity-purified in a large amount. The biochemical analysis demonstrated that the basal ATPase activity of $M_{396-734}$ is not activated by $Ca^{2+}$, but stimulated 30-fold max by microtubules. The enzymatic activation is microtubule-concentration-dependent, and the concentration of microtubules that corresponds to half-maximum activation was about 11 ${\mu}M$, much higher than that of other kinesins reported. The cosedimentation assay indicated that $M_{396-734}$ could bind to microtubules in vitro whenever the nucleotide AMP-PNP is present or absent. As a plant-specific microtubule-dependent kinesin with a lower microtubule-affinity and a nucleotide-independent microtubule-binding ability, cotton GhKCH2 might be involved in the function of microtubules during the deposition of cellulose microfibrils in fibers or the formation of cell wall.
Kinesins, as a kind of microtubule-based motor proteins, have a conserved microtubule-binding site in their motor domain. Here we report that two homologous kinesins in Arabidopsis thaliana, KatB and KatC, contain a second microtubule-binding site in their tail domains. The prokaryotic-expressed N-terminal tail domain of the KatC heavy chain can bind to microtubules in an ATP-insensitive manner. To identify the precise region responsible for the binding, a serious of truncated KatC cDNAs encoding KatC N-terminal regions in different lengths, KatC1-128, KatC1-86, KatC1-73 and KatC1-63, fused to Histidine-tags, were expressed in E. coli and affinity-purified. Microtubule cosedimentation assays show that the site at amino acid residues 74-86 in KatC is important for microtubule-binding. By similarity, we obtained three different lengths of KatB N-terminal regions, KatB1-384, KatB1-77, and KatB1-63, and analyzed their microtubule-binding ability. Cosedimentation assays indicate that the KatB tail domain can also bind to microtubules at the same site as and in a similar manner to KatC. Fluorescence microscopic observations show that the microtubule-binding site at the tail domain of KatB or KatC can induce microtubules bundling only when the stalk domain is present. Through pull-down assays, we show that KatB1-385 and KatC1-394 are able to interact specifically with themselves and with each other in vitro. These findings are significant for identifying a previously uncharacterized microtubule-binding site in the two kinesin proteins, KatB and KatC, and the functional relations between them.
Secretory proteins, including plasma membrane proteins, are generally known to be transported to the plasma membrane through the endoplasmic reticulum-to-Golgi pathway. However, recent studies have revealed that several plasma membrane proteins and cytosolic proteins lacking a signal peptide are released via an unconventional protein secretion (UcPS) route, bypassing the Golgi during their journey to the cell surface. For instance, transmembrane proteins such as the misfolded cystic fibrosis transmembrane conductance regulator (CFTR) protein and the Spike protein of coronaviruses have been observed to reach the cell surface through a UcPS pathway under cell stress conditions. Nevertheless, the precise mechanisms of the UcPS pathway, particularly the molecular machineries involving cytosolic motor proteins, remain largely unknown. In this study, we identified specific kinesins, namely KIF1A and KIF5A, along with cytoplasmic dynein, as critical players in the unconventional trafficking of CFTR and the SARS-CoV-2 Spike protein. Gene silencing results demonstrated that knockdown of KIF1A, KIF5A, and the KIF-associated adaptor protein SKIP, FYCO1 significantly reduced the UcPS of △F508-CFTR. Moreover, gene silencing of these motor proteins impeded the UcPS of the SARS-CoV-2 Spike protein. However, the same gene silencing did not affect the conventional Golgi-mediated cell surface trafficking of wild-type CFTR and Spike protein. These findings suggest that specific motor proteins, distinct from those involved in conventional trafficking, are implicated in the stress-induced UcPS of transmembrane proteins.

The aim of this study was to investigate the role of kinesin superfamily member 15 (KIF15) in nasopharyngeal carcinogenesis (NPC) and explore its underlying mechanisms. We employed various assays, including the CCK-8 assay, flow cytometry, the Transwell and scratch assay, Western blotting, and nude mice transplantation tumor, to investigate the impact of KIF15 on NPC. Our findings demonstrate that KIF15 plays a critical role in the proliferation, apoptosis, migration, and invasion of NPC cells. Furthermore, we discovered that silencing KIF15 inhibits cell proliferation, migration, and invasion while promoting apoptosis, and that KIF15's effect on NPC cell growth is mediated through the PI3K/AKT and P53 signaling pathways. Additionally, we showed that KIF15 promotes nasopharyngeal cancer cell growth in vivo. Our study sheds light on the significance of KIF15 in NPC by revealing that KIF15 knockdown inhibits NPC cell growth through the regulation of AKT-related signaling pathways. These findings suggest that KIF15 represents a promising therapeutic target for the prevention and treatment of NPC.
세포내 운반체는 kinesin과 dynein과 같은 미세소관 분자 모터단백질에 의하여 운반된다. Kinesin은 분자 모터단백질의 큰 그룹을 형성하며, kinesin-1은 미세소관 위를 정방향으로 세포내 소기관, 단백질 복합체, 그리고 mRNAs을 운반한다. Kinesin-1은 kinesin superfamily protein (KIF) 5A, 5B, 그리고 5C (또 다른 명칭으로 kinesin장쇄) 그리고 kinesin 단쇄로 구성되어져 있다. Kinesin-1은 KIF5s의 carboxyl (C)-말단 부위를 통하여 다양한 단백질과 결합한다는 사실은 알려져 있지만, 결합단백질에 대하여서는 아직 충분히 밝혀지지 않았다. 본 연구에서는 KIF5A의 C-말단 특정영역과 결합하는 단백질을 효모 two-hybrid system을 사용하여 탐색한 결과, Glutamate-rich 4 (ERICH4)를 분리하였다. ERICH4는 KIF5A의 C-말단 특정영역과 결합하지만, KIF5B와 KIF3A (kinesin-2의 모터단백질)와는 결합하지 않았다. 그리고 KIF5A는 ERICH4의 다른 isoform인 ERICH1과는 결합하지 않았다. 또한 KIF5A은 GST-ERICH4, GST-ERICH4-amino (N)-말단과는 결합하지만 GST-ERICH4-C말단과 GST와는 결합하지 않았다. HEK-293T 세포에 ERICH4와 KIF5A을 발현시켰을 때 ERICH4와 KIF5A는 세포 내의 같은 부위에서 발현하며, ERICH4을 면역침강한 결과 KIF5A와 KLC은 같이 침강하였다. 이러한 결과들은 ERICH4는 kinesin-1이 운반하는 수송체와 KIF5A와의 결합에 매개단백질로의 역할의 가능성을 시사한다.

KIF5A의 C-말단 영역과 상호 작용하는 어댑터 단백질을 규명하기 위해 효모 2-하이브리드 스크리닝을 하여, cyclin-dependent kinase 2-associated protein 1 (CDK2AP1)를 확인하였다. CDK2AP1은 KIF5A의 C-말단 영역에 결합하고 KIF3A, KIF5B, KIF5C 및 kinesin light chain 1 (KLC1)과는 결합하지 않았다. CDK2AP1의 C-말단 영역은 KIF5A와의 결합에 필수적이었다. HEK-293T 세포에 CDK2AP1 및 kinesin-1은 동시 발현하여 면역침강하면 CDK2AP1 및 kinesin-1은 같이 면역침강하였다. 그리고 CDK2AP1 및 kinesin-1은 세포내에서도 같은 위치에 발현하였다. 이러한 결과들은 KIF5A-CDK2AP1결합은 kinesin-1이 cargo를 운반할 때 kinesin-1과 cargo 사이를 연결하는 어댑터 단백질 역할을 시사한다.
