Characterization of Two Mannitol-Producing Leuconostoc Strains from Pa-Kimchi and Their Application for Juice and Yogurt Fermentation

  • Two mannitol producing lactic acid bacteria were isolated from pa (green onion)- kimchi, identified and named as Leuconostoc mesenteroides SKP 88 and Leuconostoc citreum SKP 92, respectively. Both isolates grew well at 25-30℃, initial pH 6-8, and 3% and lower NaCl concentration. Both isolates converted fructose into mannitol efficiently when grown on MRS broth containing fructose and glucose. Glucose was used as a carbon source and fructose was used as a precursor for mannitol. Mannitol yields were the highest in MRS broth with 3% fructose and 2% glucose. Shine muscat juice fermentation was done using each isolate as a starter. As fermentation progressed, decrease in pH and increases in titratable acidity and viable counts were observed. L. mesenteroides SKP 88 showed better mannitol conversion ability than L. citreum SKP 92, and shine muscat juice fermented with L. mesenteroides SKP 88 showed the mannitol production of 41.6 g/l at 48 h, and juice fermented with L. citreum SKP 92 showed 23.4 g/l at the same time. Yogurt fermentations showed similar patterns, and yogurt fermented with L. mesenteroides SKP 88 showed the mannitol production of 15.13 g/l. These results showed that both strains are useful as starters for healthy fermented foods with reduced fructose contents.

Quality and Lactic Acid Bacteria Diversity of Pork Salami Containing Kimchi Powder

  • We investigated the quality properties and bacteria diversity of pork salami containing homemade kimchi powder (KP). Pork salamis were manufactured with commercial starter culture (control), and 1% KP (KP1), 3% KP (KP3), and 5% KP (KP5). The salami was fermented and aged for 2 days and 56 days, respectively. The pH and $A_W$ values of salamis with KP were significantly lower than that of the control (p<0.05). The 2-thiobarbituric acid reactive substance values of all salamis with KP increased but were below 0.2 mg MDA/kg. Salamis with KP, decreased the $L^*$ value, but increased the $a^*$ and $b^*$ values (p<0.05). The Lactobacillus count was significantly higher in the salamis with KP than in the control by day 14 (p<0.05). The number of Leuconostoc was higher in salamis with KP than in the control and was the highest in salamis in KP1. The KP1 protected lipid oxidation and showed low TBARS value of pork salami compared to the control. This study demonstrates that KP can improve pork salami properties and can serve as a potential natural compounds for fermented meat prodcuts like Metzgerei.

Isolation of 2 Bacillus Strains with Strong Fibrinolytic Activities from Kimchi

  • Two Bacillus strains, K3 and K208, both demonstrating strong fibrinolytic activities were isolated from Kimchi, a traditional Korean preparation of fermented vegetables. Isolates were subjected to various molecular biology based identification methods including RAPD-PCR and identified as B. subtilis and B. velezensis, respectively. Tryptic soy broth (TSB) was found to best maintain both the growth and the fibrinolytic activity of these strains. Culture supernatants were analyzed by SDS-PAGE and fibrin zymography, and the results indicate that a 40 and 27 kDa band seem to be responsible for the fibrinolytic activities of these two isolates and the 27 kDa band was subsequently identified as the mature form of AprE, the major fibrinolytic enzyme. Thus the aprE genes were cloned and the translated amino acid sequences demonstrated 99.3% identity with each other, and 86.5% identity with BsfA, a fibrinolytic enzyme from B. subtilis ZA400 also isolated from Kimchi, and AprE2, a fibrinolytic enzyme from B. subtilis CH3-5 isolated from Cheonggukjang, a traditional Korean fermented soy. Given this B. subtilis K3 and B. velezensis K208 may be promising starter cultures in the production of fermented foods.

Properties of a Novel Glutamate Decarboxylase (GAD) from Levilactobacillus brevis B737 Isolated from Cabbage Kimchi

  • γ-Aminobutyric acid (GABA) is a multi-functional compound with broad applications for food industry. GABA producing bacteria were isolated from cabbage kimchi. Among them, B737 was the best GABA producer when culture supernatants were analyzed by TLC. B737 was identified as Levilactobacillus brevis by 16S rRNA gene sequencing. Its glutamate decarboxylase (GAD) gene was cloned by PCR and the nucleotide sequence determined. B737 GAD consisting of 485 amino acids is the largest in size among GADs reported from LAB so far. gadB from L. brevis B737 was overexpressed in Escherichia. coli BL21(DE3) using pET26b(+).pET26b(+). The recombinant GAD was purified and its size was 55 kDa by SDS-PAGE. Maximum GAD activity was observed at pH 5 and 40℃ and the activity was dependent on pyridoxal 5'-phosphate. Km and Vmax of recombinant GAD were 6.2 ± 0.06 mM and 0.34 ± 0.002 mM/min, respectively. L. brevis B737 can be used as a starter for fermented foods with high GABA contents.

김치제조시 사용되는 천일염이 김치의 장기저장에 미치는 영향 (Effect of Solar Salt on Kimchi Fermentation during Long-term Storage)

  • 천일염이 김치의 품질에 미치는 직접적인 영향을 규명하기 위해 발효조건(김치 원 부재료, 발효미생물, 발효온도 및 기간 등)을 동일하게 하여주고, L. citreum GJ7을 종균으로 사용하여 김치내 미생물 제어를 통한 원 부재료내의 토착미생물의 변수를 최소화한 환경하에서 사용된 소금에 따른 차이가 김치의 저장기간 중 김치품질특성에 미치는 영향을 조사하였다. 사용소금의 성분분석결과 NaCl 함량은 1년 숙성 천일염보다 4년 숙성 천일염에서 더 높았고, 수분과 $SO_4$ 및 무기양이온(Mg, K, Ca)은 4년 숙성 천일염보다 1년 숙성 천일염이 더 높게 나타나, 소금의 숙성에 따라 간수가 제거됨으로써 이러한 현상이 나타나는 것으로 보였다. 천일염 2종(4년 숙성염, 1년 숙성염) 및 정제염을 사용하여 제조된 김치는 발효하여 산도가 0.5-0.6%에 이르렀을 때 $-1^{\circ}C$에 보관하면서 5개월 동안 김치품질 특성을 조사하였다. 막 담근 김치에서 알맞게 김치가 익는데(산도 0.5-0.6%) 소요되는 시간은 정제염김치가 천일염김치보다 더 오래 걸리지만, 알맞게 익은 김치를 장기저장시(5개월)에는 정제염김치가 가장 빨리 그리고 제일 많이 시어지고, 4년 숙성염김치가 같은 기간내에 가장 덜 시어진다는 것을 알 수 있었다. 김치내 총균수 및 유산균수는 4년 숙성염김치가 가장 높게 나타났으며, 그 다음으로는 1년 숙성염김치 그리고 정제염김치순으로 나타났다. 김치 저장 5개월에 사용종균을 포함한 이종발효유산균의 점유율은 4년 숙성염김치가 77.2%, 1년 숙성염김치는 65.6%, 정제염김치는 53.1%로 4년 숙성염김치가 가장 높게 나타났으며, 김치연부를 일으키는 효모는 4년 숙성염김치에서 가장 낮게(1.3 log CFU/mL), 정제염김치에서 (5.1 log CFU/mL)로 가장 높게 나타났다. 즉, 김치의 품질특성에 바람직한 영향을 미치는 종균을 포함한 이종발효유산균 균총의 검출율과 그렇지 못한 동종발효유산균과 효모의 검출률 비율이 김치의 저장기간이 장기화될수록 사용소금간 김치에서 확연이 차이가 벌어짐을 확인할 수 있었다. 김치의 색도변화에서는 황색도(b값)가 저장기간이 경과함에 가장 유의적으로 증가하는 경향을 나타내었으며, 정제염김치의 증가폭이 가장 크게 나타났다. 저장기간에 따른 경도변화에 있어서 4년 숙성염김치가 가장 견고하였으며 정제염김치의 경도가 가장 낮은 것으로 나타났다. 또한 2개월, 5개월 동안 저장한 김치의 관능검사결과 4년 숙성염김치가 가장 높게 평가되었으며 정제염김치가 가장 낮게 평가되었다. 이와 같은 소금에 따른 김치품질특성 차이는 1개월 이내의 단기저장에서보다 5개월 이상의 장기저장에 따라 점점 더 확연한 차이를 나타냄으로서 4년 숙성염이 발효 김치의 장기저장에 가장 적합한 것으로 나타났다.

종균이 첨가된 저염김치의 발효온도 및 소금 농도에 따른 발효 특성 (Fermentation Characteristics of Low-Salt Kimchi with Starters on Fermentation Temperature and Salt Concentration)

  • 본 연구에서는 김치의 소금 함량을 낮추기 위해 유기산과 젖산 그리고 종균을 첨가하고 과일을 첨가하여 김치의 저염화와 동시에 미각증진 효과를 가져올 수 있는 저염김치를 개발하였다. 그리고 소금 농도와 발효온도를 달리하였을 때 저염김치의 품질특성에 미치는 영향을 조사하기 위하여 저염김치에 대한 이화학분석, 미생물학 분석, 그리고 관능적 특성에 대한 분석을 하였다. 저염김치의 pH 및 산도 변화는 저장기간이 경과함에 따라 온도가 높을수록 변화가 빠르게 나타났고, pH는 점차 감소하였으며, 산도는 증가하였는데, 특히 소금 농도가 낮을수록 pH가 낮았고, 산도가 가장 높게 나타났다. 저염김치의 염도 변화는 발효가 진행됨에 따라 작은 변화를 나타내었으나, 유의성 있는 증감을 나타내지 않았다. 환원당의 경우, 모든 처리구에서 비슷한 비율로 감소하는 경향을 나타내었다. 조직강도의 경우 저장기간이 길어짐에 따라 종균 첨가 여부 및 종류에 따라서 차이가 났음을 알 수 있었다. 또한 같은 소금 농도에서 저장온도에 따른 조직감 차이는 저장기간이 길어짐에 따라 현저한 차이를 나타내었다. 또한 저염김치의 미생물 결과 총균수의 경우, 저장기간 동안 농도와 균 첨가와 관계없이 총균이 증식함을 알 수 있었다. 저염김치의 관능평가 결과, 냄새, 짠맛과 전반적인 기호도에서 이형발효 종균을 첨가한 김치의 관능결과가 모두 우수한 것으로 나타났다. 종균과 유기산을 첨가하여 소금 함량을 낮춘 저염김치를 개발하였고, 저장온도에 따라 발효하는 동안 이화학, 미생물 그리고 관능적인 변화를 통해 적숙기를 파악하였다. 이 연구는 건강한 일반인, 당뇨와 고혈압 질환이 있는 환자들의 식이요법에 도움이 될 것이다.

조지 포크가 경험한 1884년 조선의 음식문화 (Joseon Dynasty's Food Culture Experienced by George C. Foulk in 1884)

  • This research is the first known to introduce and analyze food-related content among the records left by George Clayton Foulk (1856-1893), a naval attache dispatched as part of a U.S. delegation to Korea during the Joseon dynasty in the 19th century. Sketches and memos by Foulk provide important clues in understanding the food culture during the late Joseon dynasty. By analyzing the types of foods, table composition, and intervals between serving the nine rounds of food from the rural government office, which Foulk ate during his local trip, he was able to confirm that there was a starter ('preliminary table' or 'hors d'œuvre') before the main table and that it was served before the main dish.

냉면용 동치미액 제조를 위한 항균 활성이 높은 젖산균의 선발 (Screening of high Antibacterial Lactic Acid bacteria for the Preparation of Dongchimi-Juice for Naengmyon)

  • In order to screen lactic acid bacterial starter having high antibacterial activity and giving good flavor in Dongchmi for Naengmyon antibacterial activity and acid producing ability of 24 strains of Kimchi lactic acid bacteria were tested. Among 24 strains tested Leuconostoc mesenteroides subsp. mesenteroides C16, Leu. paramesenteroides C18 Lactobacillus bavaricus B01, B06, C19, C32 Lac. homohiochii B21 and B22 showed high antibacterial activity and their antibacterial activities were more active against Listeria monocytogenes and Staphylococcus aureus than against Escherichia coli and Salmonella typhimurium. The odors of Dongchimi-juices fermented with Leu. mesenteroides or Leu. paramesenteroides were more favorable than those with Lac. bavaricus or Lac. homohiochii. But the odors of Naengmyon-broths which were made with each Dongchimi-juice were not significantly different each other. In consideration of antibacterial ac-tivity and flavor of Dongchimi-juice two strains of lactic acid bacteria Lac. homohiochii B21 and Leu. mes-enteroides subsp. mesenteroides C16 were finally screened out as Dongchimi starters.

Probiotic Property and Anti-Obesity Effect of Lactiplantibacillus plantarum KC3

  • Lactic acid bacteria are representative probiotics that have beneficial effects on humans. Nineteen strains among the 167 single strains from kimchi was selected and their physiological features were investigated. The selection of a strain was based on strong enzyme (lipase, α-amylase, and α-glucosidase) inhibitory activities and anti-obesity effects in the adipocytes. For the final selection, the strain Lactiplantibacillus plantarum KC3 was tested for its potential as a starter. To assess its functionality, a freeze-dried culture of L. plantarum KC3 was administered to a diet-induced obese mouse model receiving a high-fat diet. The animal group administered with L. plantarum KC3 showed significant body weight loss during the 12-week feeding period compared to the high-fat control group. This study investigated the physiological characteristics of selected strain and evaluated its potential as an anti-obesity probiotic in mice.

Development of Cabbage Juice Medium for Industrial Production of Leuconostoc mesenteroides Starter

  • Leuconostoc mesenteroides is used as a starter to produce high-quality kimchi products. In this study, an efficient and economical cabbage juice medium (CJM) was developed by process optimization of cabbage extraction and pasteurization and by compositional supplementation of various lacking nutrients. The pasteurized cabbage juice was determined to be a good medium candidate to cultivate L. mesenteroides, showing maximal cell numbers ($9.85{\times}10^8CFU/ml$) after 24 h. Addition of sucrose and yeast extract with soy peptone resulted in increment of bacterial cell counts in CJM, showing the supplementing effect of the lacking nutrients. Furthermore, addition of shell powder gave a protective effect on bacterial cells by preventing pH decline and organic acid accumulation in CJM, resulting in a 2-fold increase of bacterial counts. The optimized composition of CJM was 70% cabbage juice diluted with water, 0.5% (w/v) sucrose, 1% (w/v) yeast extract, 1% (w/v) soy peptone, and 1.5% (w/v) ark shell powder. The CJM developed in this study was able to yield a comparable level of bacterial counts with MRS medium and reduced the cost by almost 10-fold.
