The Analysis of Failure Pattern in Locally Advanced Stomach Cancer Treated with Surgery and Post-Op Chemotherapy: To Explore The Role of Post-Op Irradiation (수술과 항암요법으로 치료한 국소 진행된 위함 환자에서의 치료실패 양상분석 : 수술후 방사선 치료의 역할에 대한 연구)

  • A Retrospective study to analyze the failure pattern in locally advanced stomach cancer, treated with radical surgery and post-op chemotherapy was perfomed. Among 107 patients who underwent radical gastrectomy in Asan Medical Center between June 1989 and August 1990. there were 20 stage II(T2NO, T2N1) and 87 stage III(T3N1, T3N2) and 91 patients were eligible for study. 57 patients treated with 6 cycles of postop adjuvant chemotherapy. Among 57 patients treated with postop adjuvant chemotherapy, local failure occurred in $21\%$ and distant failure in $12\%$. Among 34 patients who were not treated with postop chemotherapy, local failure occurred in $24\%$ and distant failure in $26\%$. Among 29 failures including 13 locoregional, 9 distant metastasis and 7 locoregional and distant metastasis, 11 cases recurred in the anastomotic site, 3 in the gastric bed,7 in the regional lymph nodes and peritoneal seeding occurred in 6 cases. The true incidences of gastric bed, nodal and peritoneal failures may be higher in the longer follow-up or reoperative or autopsy series. Our data sugest that postop chemocherapy is beneficial by reducing distant failure rate. Our data suggest that postop chemocherapy is beneficial by reducing distant failure rate. Postop adjuvant locoregional radiotherapy in addition to the systemic adjuvant therapy may reduce the local failure rate and potentially benefit in at least $20\%$ of patients who developed the local failure only.

Characterization of Traditional Korean Unifloral Honey Based on the Mono-, Di-, and Trisaccharides (한국산 벌꿀의 밀원별 단당, 이당 및 삼당류의 정량 특성)

  • Sugar profiles of 45 Korean honey samples (15 acacia, 15 multi-floral, 10 chestnut, and 5 artificial honey samples), which are commercially available in the Korean markets, were analyzed using gas chromatography/mass spectrometry (GC/MS) through TMS-oxime and TMS-methoxime derivatization. The average invert sugar contents in acacia, multi-floral, chestnut, and artificial honey samples were $71.2{\pm}1.05$, $68.7{\pm}3.26$, $63.2{\pm}1.85$, and $68.0{\pm}2.10%$, respectively. Fourteen disaccharides were detected from the samples, and the average content of major disaccharides was higher in order of turanose, maltulose, maltose, trehalulose, kojibiose, isomaltose, and nigerose. The average content of total disaccharides was highest in chestnut and lowest in acacia. Seven trisaccharides were detected from the samples, and the average content of trisaccharides was the highest in artificial honeys, which had high erlose content. The total content of disaccharides and trisaccharides was highest ($16.0{\pm}2.03%$) in chestnut honey and lowest ($9.70{\pm}1.75%$) in acacia honey.

  • Menadione is known as an anti-tumor factor. Many studies have reported the potential anti-cancer role of menadione against a range of cancer cell lines. In this study, the anti-cancer effects of menadione and the underlying molecular signaling involved in apoptosis was investigated in gastric cancer cell lines. The menadione treatment decreased the cell viability of MKN45 gastric cancer cells. The decreased cell viability was attributed to the induction of apoptosis, which was confirmed by the results indicating the activation of caspase-3 and -7 and the cleavage of PARP in Western blotting. The upstream regulatory molecules involved in apoptosis were investigated further and it was discovered that menadione reduced the expression of survivin, an inhibitor of upstream apoptosis proteins. In addition, a transcription factor ${\beta}$-catenin, which is known to regulate survivin expression, was down-regulated by menadione. A previous report showed that menadione inhibited XIAP expression to induce apoptosis and induced G2/M cell cycle arrest in AGS cells. This study elucidated another inhibitory mechanism of menadione against gastric cancer cells in a different cell line. Although further studies will be needed, the inhibitory mechanism demonstrated in this study will help better understand the anti-cancer effects of menadione.

Effect of polishing methods on color change by water absorption in several composite resins (여러 복합레진에서 수분 흡수에 의한 색변화에 연마가 미치는 영향)

  • Purpose: The aim of this study was to evaluate the influence of polishing methods on the color stability of composite resins. Materials and Methods: Two bulk-fill and four conventional resin composites were filled in cylindrical molds (6 mm diameter, 4 mm height) and light-cured. The specimens were stored in $34^{\circ}C$ distilled water for 24 h. Spectrophotometer was used to determine the color value according to the CIE $L^*a^*b^*$ color space. Each group was divided into three groups according to polishing methods (n = 5). Group 1 was control group (Mylar strip group), group 2 was polished with PoGo, and group 3 was polished with Sof-Lex Spiral wheels. Color evaluation was performed weekly for 4 weeks after immersion in $34^{\circ}C$ distilled water. The results were analyzed by generalized least squares method (P < 0.05). Results: Generalized least squares analysis revealed that Sof-Lex Spiral wheels group showed the significantly lower ${\Delta}E$ values compared to PoGo and control group (P < 0.05). The ${\Delta}E$ values of polished group showed the significantly lower than the ${\Delta}E$ values of unpolished group (P < 0.05). Regarding color changes of composite resins, there was no significant difference between the ${\Delta}E$ values of Filtek Z250 and Filtek Z350 XT Universal restorative in all time intervals (P < 0.05). Tetric N-Ceram Bulk Fill showed the significantly lower ${\Delta}E$ values compared to other composite resins in 1, 2, 3 weeks (P < 0.05). Conclusion: Within the limitations of this study, polishing methods influence the color stabilities of composite resins. The group polished with Sof-Lex Spiral Wheels showed more resistance to discoloration than group polished with PoGo.

Characteristics of New Ever-bearing Strawberry 'Bokha' Bred for Tropical and Subtropical zone Cultivation (열대 및 아열대 지역 재배용 사계성 딸기 '복하' 육성)

  • 'Bokha' is a new strawberry (Fragaria x ananassa Duch.) cultivar, which was released by the Highland Agriculture Research Institute in 2016. The 'Bokha' cultivar originated from a cross between 'Goha' and 'Saebong No. 3' that showed excellent ever-bearing characteristics, including continuous flowering habit and high soluble-solids contents under long-day and high temperature conditions in 2011. This cultivar was initially named 'Saebong No. 8' after examining its characteristics and productivity in summer culture from 2013 to 2015. After regional adaptability tests in 2016, 'Bokha' was selected from Saebong No. 8 as an elite cultivar. The general characteristics of 'Bokha' include semispreading type, elliptical leaves, and moderately vigorous growth. The fruits are conical in shape, and red in color. 'Bokha' plants have 32.7 leaves, 9.9 more than 'Goha' plants. The soluble-solids content of 'Bokha' was 9.2%, which was 0.2% higher than that of 'Goha'. The average fruit weight of 'Bokha' was about 9.5g and the marketable yield was $27,701kg{\cdot}ha^{-1}$, 72% higher than 'Goha'. 'Bokha' is suitable for tropical and subtropical zone cultivation as a high soluble solids contents cultivar, because it shows continuous flowering habit under long-day and high temperature conditions.

International and domestic research trends in longitudinal connectivity evaluations of aquatic ecosystems, and the applicability analysis of fish-based models (수생태계 종적 연결성 평가를 위한 국내외 연구 현황 및 어류기반 종적 연속성 평가모델 적용성 분석)

  • Recently, stream longitudinal connectivity has been a topic of investigation due to the frequent disconnections and the impact of aquatic ecosystems caused by the construction of small and medium-sized weirs and various artificial structures (fishways) directly influencing the stream ecosystem health. In this study, the international and domestic research trends of the longitudinal connectivity in aquatic ecosystems were evaluated and the applicability of fish-based longitudinal connectivity models used in developed countries was analyzed. For these purposes, we analyzed the current status of research on longitudinal connectivity and structural problems, fish monitoring methodology, monitoring approaches, longitudinal disconnectivity of fish movement, and biodiversity. In addition, we analyzed the current status and some technical limitations of physical habitat suitability evaluation, ecology-based water flow, eco-hydrological modeling for fish habitat connectivity, and the s/w program development for agent-based model. Numerous references, data, and various reports were examined to identify worldwide longitudinal stream connectivity evaluation models in European and non-European countries. The international approaches to longitudinal connectivity evaluations were categorized into five phases including 1) an approach integrating fish community and artificial structure surveys (two types input variables), 2) field monitoring approaches, 3) a stream geomorphological approach, 4) an artificial structure-based DB analytical approach, and 5) other approaches. the overall evaluation of survey methodologies and applicability for longitudinal stream connectivity suggested that the ICE model (Information sur la Continuite Ecologique) and the ICF model (Index de Connectivitat Fluvial), widely used in European countries, were appropriate for the application of longitudinal connectivity evaluations in Korean streams.

LC-MS/MS analysis and anti-inflammatory effects of crude extract from Coptidis Rhizoma (황련 추출물의 LC-MS/MS 분석 및 항염증 효과)

  • Objectives : The main aim of this study was to examine the LC-MS/MS used to identify phenolic compounds of CRE(Coptidis Rhizoma 70% EtOH Extract). Also, we investigated antioxidative activities and Anti-inflammatory activities. Methods : LC-MS/MS Analysis HPLC and LC-MS/MS were performed on a 1260 series HPLC system (Agilent Technologies, Inc., California, USA) and 3200 QTrap tandem mass system (Sciex LLC) operated in positive ion mode (spray voltage set at -4.5 kV). The solvent used was DW and Acetonitrile containing 0.1% formic acid, a gradient system was used at a flow rate of 0.5 mL/min for analysis, and a Prontosil C18 column (length, 250 mm; inner diameter, 4.6 mm; particle size, 5 ㎛; Phenomenex Co., Ltd., California, USA, Biochoff Chromatography) was used. The solvent conditions used in the mobile phases were 0-10 min at 10-15% B, 10-20 min at 20% B, 20-30 min at 25%, 30-40 min at 40%, 40-50 min at 70%, 50-60 min at 95%, and 60-70 min at 95%. The analysis was performed at a wavelength of 284 nm and a temperature of 35℃. The cell viability was measured using a 3-(4,5-dimethyethiazol-2-yl)-2,5-diphenyltetrazolium bromide (MTT) assay and 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activity. We examined the effects of CRE on the lipopolysaccharide (LPS)-induced production of nitric oxide (NO) in a RAW 264.7 cells Results : The chemical analysis CRE by Liquid chromatography-tandem mass spectrometry (LC-MS/MS) confirmed that Rosmarinic acid, Ferrulic acid, 3-O-feruloylquinic acid, and 5-O-feruloylquinic acid as phenolic components. DPPH radical scavenging activity was the inhibitory activity of CRE showed at 200 ㎍/mL a statistically significant level. MTT assay demonstrated that the CRE did not have a cytotoxic effect in RAW 264.7 and LPS-induced RAW264.7 cells. Also, CRE reduced NO production in RAW 264.7 cells stimulated with LPS. Conclusions : Based on these findings, The chemical analysis 4 major components CRE such as Rosmarinic acid, Ferrulic acid, 3-O-feruloylquinic acid, and 5-O-feruloylquinic acid. Moreover, we confirmed that CRE has effects antioxidant and anti-inflammatory. The results demonstrate that CRE can be used as an antioxidant and a powerful chemopreventive ingredient against inflammatory diseases.

Breeding of New Ever-bearing Strawberry 'Miha' with High Hardness (고경도 사계성 딸기 '미하' 육성)

  • 'Miha' is a new strawberry (Fragaria x ananassa Duch.) cultivar, which was released by the Highland Agriculture Research Institute in 2019. The 'Miha' cultivar originates from a 2014 cross between 'Monterey' and 'Saebong No. 3', both of which exhibited excellent ever-bearing characteristics, including continuous flowering and large fruits under long-day and high temperature conditions. This new cultivar was initially named 'Saebong No. 12' after examining its characteristics and productivity during summer cultivation between 2015 and 2019. After regional adaptability tests, 'Miha' was selected from 'Saebong No. 12' as an elite cultivar. The general characteristics of 'Miha' include intermediate, elliptic leaves, and strong growth. The fruits are conical and of a dark-red color. The number of leaves of 'Miha' was 21.9, which was 6.2 fewer than that of the control cultivar, 'Goha' with 28.1. The number of flower clusters of 'Miha' was similar to that of 'Goha'. The average fruit weight of 'Miha' was 13.4 g, which was 4.3 g heavier than that of 'Goha'. The fruit hardness of 'Miha' was 36.2 g·mm-2, which was 10.1 g·mm-2 harder than that of 'Goha'. The marketable yield of 'Miha' was 37,393 kg·ha-1, 156% more than that of 'Goha' with 23,970 kg·ha-1. Therefore, the new cultivar of ever-bearing strawberry 'Miha' is expected to be very popular in the export or bakery market because it is hard.

A Survey on Pesticide Residues and Risk Assessment for Agricultural Products Marketed in the Northern Area of Seoul from 2022 to 2023 (서울 북부지역 유통 농산물의 잔류농약 실태 및 위해성 평가 (2022-2023))

  • In this study, we investigated the concentrations of pesticide residues in agricultural products marketed in northern Seoul and assessed their potential health risks. A total of 1,737 samples were collected using the QuEChERS method, followed by LC-MS/MS and GC-MS/MS. Pesticide residues below the MRLs were detected in 560 samples of 72 items (32.2%), but 38 samples of 22 items had pesticide residues above the MRLs. Residual pesticides were detected in 53.8% of fruits, 33.0% of vegetables, 28.6% of herbs, 15.4% of beans, and 10.5% of rice samples. Most of the samples that exceeded the MRLs were vegetables, especially leafy, stalk, stem, and root vegetables; herbs and mushrooms also exceeded the permitted MRLs. Of the 105 pesticides investigated, dinotefuran, fluxametamide, chlorfenapyr, azoxystrobin, and carbendazim were the most frequently detected, whereas 23 pesticide residues, including terbufos, carbendazim, and fluxametamide, were detected above the MRL values. The hazard indices were calculated as 0.00003-1.31406%, which suggests that the investigated pesticide residues in the samples were within safe levels, but continuous monitoring of pesticides in agricultural products is needed to ensure the safety of consumers.

Breeding of New Ever-bearing Strawberry 'Jinha' with High Soluble Solid Content (당도가 높은 사계성 딸기 '진하' 육성)

This new cultivar was initially named 'Saebong No. 11' after examining its characteristics and productivity during summer cultivation between 2012 and 2016. After regional adaptability tests, 'Jinha' was selected from 'Saebong No. 11' as an elite cultivar. The general characteristics of 'Jinha' include intermediate, elliptic leaves, and medium growth. The fruits are conical and of a red color. The plant height of 'Jinha' is simiar to that of the control variety, 'Flamenco', but it has a lot of number of leaves. The cluster length of 'Jinha' was 35.5 cm, 10.8 cm longer than 24.7 cm of the control variety. The number of flower clusters of 'Jinha' appeared 14.4, which was 4.1 more than that of 'Flamenco'. The average fruit weight of 'Jinha' was 10.1 g, which was 0.8 g heavier than that of 'Flamenco'. The soluble solid content of 'Jinha' was 10.2 °Brix, which was 2.0 °Brix higher than that of 'Flamenco'. The marketable yield of 'Jinha' was 25,931 kg·ha-1, 440% more than that of 'Flamenco' with 5,900 kg·ha-1. Therefore, the new cultivar of ever-bearing strawberry 'Jinha' is expected to be very popular in the export or bakery market because it is high soluble solid content and good shape.
