Damages caused by infection with viruses in cut-flower production of Lilium $\chi$. fomolongi

  • Viral disease symptoms were investigated in the field grown Longiflorum hybrid cultivars, and the damages caused by infection with Lily mottle virus (LMoV) and Cucumber mosaic virus (CMV) were assessed by comparing growth of plants produced from seeds of Longiflorum hybrid cultivar both infected by artificial inoculation and free from infection with theses viruses. Dominant symptom caused by spotaneous infection with LMoV and CMV in the field was mottle combined with chlorotic stripe on leaves. LMoV developed brownish necrotic lesion on floral leaves. The incidence of viral disease by mixed infection with LMoV, CMV or Lily symptomless virus (LSV) in the filed grown Longiflorum hybrid cultivar, cultivated for more than 6 years, was 80 to 84 percent. In comparison with virus-free plants, plants doubly infected with CMV and LMoV by artificial inoculation decreased stem length by 14 percent and fresh weight by 38 percent. In conclusion, flower quality and the stem length of Longiflorum hybrid cultivar were affected by LMoV and CMV infection.

Discrimination of Korean ginseng (Panax ginseng Meyer) cultivar Chunpoong and American ginseng (Panax quinquefolius) using the auxin repressed protein gene

  • Background: Korean ginseng (Panax ginseng) is one of the most important medicinal plants in the Orient. Among nine cultivars of P. ginseng, Chunpoong commands a much greater market value and has been planted widely in Korea. Chunpoong has superior quality "Chunsam" ($1^{st}$ grade ginseng) when made into red ginseng. Methods: A rapid and reliable method for discriminating the Chunpoong cultivar was developed by exploiting a single nucleotide polymorphism (SNP) in the auxin repressed protein gene of nine Korean ginseng cultivars using specific primers. Results: An SNP was detected between Chunpoong and other cultivars, and modified allele-specific primers were designed from this SNP site to specifically identify the Chunpoong cultivar and P. quinquefolius via multiplex polymerase chain reaction (PCR). Conclusion: These results suggest that great impact to prevent authentication of precise Chunpoong and other cultivars using the auxin repressed protein gene. We therefore present an effective method for the authentication of the Chunpoong cultivar of P. ginseng and P. quinquefolius.

A New Early Flowering, Spray Chrysanthemum Cultivar for Cut Flower, 'Biz' with Single Type and White Petals. (조기개화성의 백색 홑꽃 절화용 스프레이국화 '비즈' 육성)

  • A new spray chrysanthemum(Dendranthema grandiflorum) cultivar, 'Biz' was developed from a cross between 'S03-117' and 'Angaesoguk' by selection of seedlings and lines at the Flower Research Institute, Gyeongnam Agricultural Research and Extension Services(ARES) from 2003 to 2008. Its characteristics were investigated three times from 2006 to 2008 under condition of forcing culture in spring and retarding culture in autumn. The natural flowering time of 'Biz' was October 26th, and year-round production was possible by day length treatment. This cultivar was white in color, single in flower type for the spray type cut flower. Its capitulum was 3.0 cm in diameter, and had 15.6 head per stem in autumn. Its ray floret was green central zone. The days to flowering under the short day treatment is about 42 days in spring season, and 'Biz' showed the vase life of 24.1 days in autumn. This cultivar was registered for a commercialization in 2008.

  • A cultivar 'Yellow Cream' of Phalaenopsis(P. Blume) was developed by cross breeding and line selection at the Flower Research Institute, Gyeongnam ARES, from 2000 to 2008. Characteristic trials for this cultivar were carried out three times from 2006 to 2008. 'Yellow Cream' was developed from a crossing between P. 'Sogo Firework' and P. 'Sogo Gold'. 'Yellow Cream' exhibits light yellow flower color with pink stripe and favorable flower shape. Individual flowers are formed with parallel arrangement and have long life with more than 55 days. 'Yellow Cream'(Grant No.3232) was registered to the Korea seed and variety Service(KSVS) for commercialization in 2010 and suitable for the cultivation under greenhouse conditions in Korea.

  • A new oak mushroom cultivar 'Hwadam' was bred from monokaryotic strains of 'GMLE36062' and 'LE15401'. The optimum temperature for mycelial growth of 'Hwadam' on potato dextrose agar was 25℃. The cultivation period of 'Hwadam' lasted for 129 days until the first harvest of the fruiting bodies, including 35 days for spawn run, 84 days for browning, and 10 days for development of fruiting bodies. The fruiting bodies of 'Hwadam' had a hemispherical, brighter pileus and thicker stipe than the control cultivar (Sanjo 701ho). The three-cycle yield of 'Hwadam' was 554 g, which was higher than that of control cultivar (405 g). In addition, 'Hwadam' contained 79.4 mg/g of beta-glucan and 0.93 mg GAEs/g of phenolic compounds, which was similar to that of the control cultivar but higher than that of 'L808' and 'Chujae 2ho'.

  • A new spray chrysanthemum (Dendranthema grandiflorum) 'Blue Hope' was bred by the Flower Research Institute, Gyeongsangnam-do Agricultural Research & Extension Services in 2009. The cultivar 'Blue Hope' was initially derived from the cross in 2005 between 'Ford', a spray chrysanthemum cultivar, with white anemone type, and 'Chopin', a spray chrysanthemum cultivar with white anemone type. The cultivar has anemone type with white petals. After the evaluation of the characteristics under shade culture in summer and retarding culture in spring and consecutive selection from 2007 to 2009, 'Blue Hope' was selected finality. The natural flowering time of 'Blue Hope' was October 24th, and year-round flowering is possible by shading or lighting treatment. The growth of plant was very vigorous and response time 6.5 weeks. The diameter of flower was 4.9 cm. Number of flowers per stem was 19.1 in autumn. Days to flowering under the short day treatment was about 45 in spring and its vase life was 23.8 days in the autumn season. This cultivar was resistance white rust and consumer's preference of new spray is high level than the control.

Polymorphism and Expression of Isoflavone Synthase Genes from Soybean Cultivars

  • Isoflavones are synthesized by isoflavone synthases via the phenylpropanoid pathway in legumes. We have cloned two isoflavone synthase genes, IFS1 and IFS2, from a total of 18 soybean cultivars. The amino acid residues of the proteins that differed between cultivars were dispersed over the entire coding region. However, amino acid sequence variation did not occur in conserved domains such as the ERR triad region, except that one conserved amino acid was changed in the IFS2 protein of the GS12 cultivar ($R_{374}G$) and the IFS1 proteins of the 99M06 and Soja99s65 cultivars ($A_{109}T$, $F_{105}I$). In three cultivars (99M06, 99M116, and Simheukpi), most of amino acid changes were such that the difference between the amino acid sequences of IFS1 and IFS2 was reduced. The expression profiles of three enzymes that convert naringenin to the isoflavone, genistein, chalcone isomerase (CHI), isoflavone synthase (IFS) and flavanone 3-hydroxylase (F3H) were examined. In general, IFS mRNA was more abundant in etiolated seedlings than mature plants whereas the levels of CHI and F3H mRNAs were similar in the two stages. During seed development, IFS was expressed a little later than CHI and F3H but expression of these three genes was barely detectable, if at all, during later seed hardening. In addition, we found that the levels of CHI, F3H, and IFS mRNAs were under circadian control. We also showed that IFS was induced by wounding and by application of methyl jasmonate to etiolated soybean seedlings.

The natural flowering time of 'Magic' was October 26th, and year-round production was possible by day length treatment. Its capitulum was 6.1 cm in diameter, and had 31.9 ray florets and 11.7 head per stem in autumn. Its ray floret was white-purple with yellow green central zone. Days to flower of 'Magic' was about 43 days in spring, and 'Magic' showed the vase life of 20.6 days in autumn. This cultivar was registered for a commercialization in 2007.

'Kowon', a New Korean Ginseng Cultivars with High Yield and Alternaria Blight Resistance

  • Recently, there has been increased attention to the development of new plant cultivars with enhanced resistance to biotic and abiotic stress. To develop ginseng cultivars with such traits, systematic breeding programs and comprehensive field studies are prerequisites. In this study, we applied a pure-line selection method to identify a ginseng cultivar with enhanced stress resistance. Phenotypic and agronomic characteristics, seed yield, and physiological responses to biotic and abiotic stresses were investigated according to the guidelines of the International Union for the Protection of New Varieties of Plants (UPOV). In the newly developed 'Kowon' cultivar, the time of emergence, flowering, and berry maturity were intermediate between those of the controls, 'Yunpoong' and 'Chunpoong'. The stem length of 'Kowon' was intermediate, whereas the root length was shorter and the main root diameter was greater than those of 'Chunpoong'. In local adaptability tests conducted in three regions, the yield of 'Kowon' was $666kg{\cdot}10a^{-1}$; 27% and 4% higher than that of 'Chunpoong' and 'Yunpoong'. Diseases such as Alternaria blight, Phytophthora blight, mulberry mealybug, and nematode infestation did not occur in 'Kowon'; and it also exhibited moderate resistance to damping-off and anthracnose. In these cases, yellow spots occurred on aerial parts and the rusty skin of the root, and it exhibited moderate resistance at high temperatures. Our study demonstrates that 'Kowon', which has a high root weight and enhanced biotic/abiotic stress resistance, is a superior cultivar that could increase farmers' income.

  • We characterized the resistances such as to waterlogging, Phytophythora and viruses in hybrids between Italian and Korean mother lines and screened them for complex resistances and agronomic traits to select elite multi-resistant lines for hybrid breeding. Resistance to waterlogging was selectable due to diversity of the resistance. Phytophythora resistance introduced from Italian lines could also be combined with resistance to other diseases and restoration abilities from cytoplasmic male sterility that has been maintained in Korean varieties.
