The amelioration of plasma lipids by Korean traditional confectionery in middle-aged women: A cross-over study with western cookie

  • BACKGROUND/OBJECTIVES: The purpose of this study was to examine whether plasma lipid profiles are affected differently by snack kinds with equal calorific values. SUBJECTS/METHODS: We compared a Korean traditional confectionery (dasik) with Western confectionery (cookie) in this regard. Controlled cross-over study consisted of two 3-week snack intake phases and for separating, a 2-week washout period (3-2-3) was carried out with 30 healthy women aged between 40-59 years old. Brown rice based Korean traditional confectionery and wheat flour based Western confectionery were used. The participants consumed either dasik or cookie every day for 3 weeks, providing 93 kcal a day. RESULTS: The total cholesterol (TC) in the dasik group had decreased significantly after 3 weeks (P < 0.05). Furthermore, in the dasik group, reduction in TC and low-density lipoprotein-cholesterol were greater than those in the cookie group (P < 0.05). CONCLUSIONS: Prioritizing functional snacks like dasik improves plasma lipid profiles; this may be useful information for individuals who cannot refrain from snacking.

Molecular Cloning, Purification, and Characterization of a Cold-Adapted Esterase from Photobacterium sp. MA1-3

  • The gene encoding an esterase from Photobacterium sp. MA1-3 was cloned in Escherichia coli using the shotgun method. The amino acid sequence deduced from the nucleotide sequence (948 bp) corresponded to a protein of 315 amino acid residues with a molecular weight of 35 kDa and a pI of 6.06. The deduced protein showed 74% and 68% amino acid sequence identities with the putative esterases from Photobacterium profundum SS9 and Photobacterium damselae, respectively. Absence of a signal peptide indicated that it was a cell-bound protein. Sequence analysis showed that the protein contained the signature G-X-S-X-G included in most serine-esterases and lipases. The MA1-3 esterase was produced in both soluble and insoluble forms when E. coli cells harboring the gene were cultured at $18^{\circ}C$. The enzyme was a serine-esterase and was active against $C_2$, $C_4$, $C_8$ and $C_{10}$ p-nitrophenyl esters. The optimum pH and temperature for enzyme activity were pH 8.0 and $30^{\circ}C$, respectively. Relative activity remained up to 45% even at $5^{\circ}C$ with an activation energy of 7.69 kcal/mol, which indicated that it was a cold-adapted enzyme. Enzyme activity was inhibited by $Cd^{2+}$, $Cu^{2+}$, $Zn^{2+}$, and $Hg^{2+}$ ions.

Tuna by-product meal as a dietary protein source replacing fishmeal in juvenile Korean rockfish Sebastes schlegeli

  • This study was conducted to evaluate the use of tuna by-product meal (TBM) as a substitute for fishmeal in juvenile Korean rockfish (Sebastes schlegeli) diets. Five isonitrogenous (52% crude protein) and isocaloric (4.9 kcal/g) diets were prepared. Control diet (Con) was formulated to contain brown fishmeal, whereas 25, 50, 75, and 100% of fishmeal was substituted with the same percentage of TBM in the TBM25, TBM50, TBM75, and TBM100 diets, respectively. Three replicate groups of fish (initial weight, $29.5{\pm}0.6g$) were fed one of the five diets for 12 weeks. The mean weight gain of fish fed the Con and TBM25 diets was significantly higher than that of fish fed the TBM100 diet (P < 0.05), but it was not significantly different from the weight gain of fish fed the TBM50 and TBM75 diets. The feed efficiency and protein efficiency ratios of fish fed the Con diet were significantly higher than those of fish fed the TBM100 diet (P < 0.05), but were not significantly different from those fed the TBM25, TBM50, and TBM75 diets. The TBM25, TBM50, TBM75, and TBM100 diets had significantly (P < 0.05) lower incidence cost and higher profit index than the Con diet. These results suggest that the tuna by-product meal used in this study could replace up to 75% of fishmeal protein in the diet without reducing growth and feed utilization in juvenile Korean rockfish weighing 29-53 g. By considering feed cost, the TBM 75 diet is recommendable for Korean rockfish farming.

Effects of Feeding Single or Multiple Probiotics on Performance and Intestinal Microflora of Broiler Chicks (단일 및 혼합 생균제의 급여가 육계의 생산성 및 장내 미생물에 미치는 영향)

  • Two experiments were conducted to investigate the effents of feeding single or combined probiltics on performance and intestinal micreflora of broiler chicks for five weeks. Diets based on corn and soybean meal contained 21.50, 19% CP and 3,100, 3,150kcal/kg ME for starting and finishing period, respectively. Lactobacillus salvarius isolated from chicks intestine(LSC), Lactobacillus salvarius isolated from piglet(LSP), Bacillus polyfermenticus(BP) were fed with alone and mixed ones at the level of 0.21 and 0.1% in experiment 1 and 2, respectively. Three hundred eighty four chicks were randomly assigned to eight treatments with four replicates of 12 chicks each per treatment. Weight gain, feed consumption, feed conversion ratio(FCR) were weekly measured for 5 weeks. The number of intestinal micreflora was examined at the end of experiment. In both experiments, weight gain of chicks fed probiltics tended to be similar or higher than control, but was not in LSC+LSP treatment. Feed intake was not consistent among treatments. In experiment 1, FCR of chicks fed LSC alone was the lowest of all treatments, whereas it was significantly higher in LSC+LSP treatments than control(P〈0.05). In experiment 2, LSP supplemental groups tended to increase FCR compared to the control. The number of ileal E. coli was the lowest in LSP alone treatment of experiment 1, whereas cecal E. coli was higher concentration in probiotics supplemental groups than control. Total Lactobacillus of chicks fed probiotics was decreased in ileum, but was no consestency in cecum. In experiment 2, ileal total yeast tended to be higher in probiotics supplemental groups except LSP alone supplement than control. Total Lactobacillus of chicks fed LSC or LSP alone treatments was significantly higher than control(P〈0.05).

Influence of Dietary Supplemental Bio-Plus on Performance and Intestinal Microflora of Broiler Chicks (바이오 플러스의 급여가 육계의 생산성과 장내 미생물에 미치는 영향)

  • Two experiments were conducted to evaluate the dietary supplemental fruit extract(Bio-plus) on performance and intestinal microflora of broiler chicks. Seven treatments were allocated into control, 0.05, 0.10, 0.20, 0.10, 0.20, 0.40, 0.80, 1.60% with four replications for five weeks, in both experiments. Diets contained 21.5% CP, 3, 100 kcal/kg ME for the rest two weeks. Weight gain, feed intake, feed conversion ratio(FCR) were weekly measured. Birds were sacrificed to examine the E. coli and Salmonella from cecum and ileum at the end of experiment. In experiment 1, Weight gain of Bio plus treatments tended to increase compared to that of control, but was not consistency between them. Chicks fed Bio-plus showed significantly lower feed conversion ratio(FCR) than control(P〈0.05). In experiment 2, Weight gain of Bio plus treatments was significantly different from control(P〈0.05) for starting period and linearly increased upto 0.80% for the rest of two weeks, but was not significantly different. Feed intake of chicks fed Bio-plus tended to be higher than control. Bio-plus treatments was not statistically different FCR from control. ND titer showed no significance, but tended to be higher in Bio-plus treatments than control. The number of ileum salmonella was significantly decreased in Bio-plus treatments compared to that of control in expt 2 (P〈0.05). Total yeast and Lactobacillus spp. of illeum tended to increase in expt 1 and were significantly higher in expt 2 than control(P〈0.05). The results of these experiment indicated that dietary Bio-plus supplements increased weight gain, the number of ileum yeast and Lactobacillus spp. of broiler chicks.

A Comparison of Feeding Lactobacillus and Virginiamycin Influence on Performance and Intestinal Microflora of Broiler Chicks (유산균과 버지니아마이신의 급여가 육계의 생산성 및 장내 미생물에 미치는 영향)

  • This experiment was conducted to investigate the effect of feeding two strains of Lactobacillus and virginiamycin on performance, nutrients digestibility and intestinal microflora of broiler chicks(Abor acres$\times$Abor Acres) were randomly allocated into six treatments with four replications for five weeks. Control(no supplement), 0.05% virginiamycin(VM), Lactobacillus crispatus avibro1(LC), Lactobacillus reuteri avibro2(LR), LC+0.05% VM(LC+VM), LR+0.05% VM(LR+VM) were supplemented into basal diets, which contained ME 3,100kcal/kg and CP 22.0, 20.0% for starting and finishing period, respectively. Weight gain, feed intake and feed conversion(FC) were weekly measured. Nutrients digestibility, intestinal microflora and fecal noxious gas were examined at the end of experiment. Weight gains of chicks fed Lactobacillus or VM was significantly higher than control(P〈0.05). Feed intake increased significantly in those supplemental groups(P〈0.05). FC of chicks fed Lactobacillus or VM significantly lower than control(P〈0.05). Degestibility of crude protein, calcium, and phosphorus improved significantly in alone or combined Lactobacillus treatments(P〈0.05). Whereas DM, crude fat and ash digestibility were not statistically different. Feeding Lactobacilli tended to increase the total Lactobacillus spp. in ileum at one and three weeks of age(WOA) and showed significantly higher in cecum than control at 5 WOA. Total yeast were not shown difference at 1 and 3 WOA, but significantly increased at 5 WOA(P〈0.05). The ileal and cecal anaerobes were started to increase from the first WOA. Fecal NH$_3$gas tended to decrease in Lactobacillus treatments compared to that of other treatments.

  • Population growth and increasing consumption of resources in the process of the industrial development has caused environmental pollution, climate change, and resource exhaustion. Therefore 'sustainable development' has become the important issue for the future. The sustainable development aims at effective resource use, less environmental impacts, and higher social security. Generally the rural area including agricultural fields and forest has various and plentiful natural resources which could make future development sustainable. To develop potential rural resources, the values for energy, environment and economy should be assessed considering the life-cycle of resources. The purposes of this study are to suggest the E3 (Energy, Environment, and Economy) assessment model for rural biomass considering life-cycle of resource and to apply the model to rice, the major agricultural product. As the results of this study, it turned out through E3 assessment that economic gain of rice cultivation is 578,374 won/10a, carbon absorption is $1,530kgCO_2/10a$, carbon emission is $926.65kgCO_2/10a$, and bio-energy potential of by-product is 394,028 kcal/10a. When E3 assessment was applied to by province, the results varied by regions because of the amount of input during cultivation. These results would be useful to realize the rural biomass and design regional resources plan in integrated E3 perspective.

Body Composition Variations in the Paretic and Nonparetic Regions of Patients with Strokes Caused by Cerebral Hemorrhage or Cerebral Infarction

  • Indicators to quantitatively evaluate the body function may help to optimize the effectiveness of rehabilitation therapy for stroke patients. In this study, we analyzed the body composition in the paretic and nonparetic regions of stroke patients with hemiplegia caused by cerebral hemorrhage (7 cases) and cerebral infarction (13 cases) using multifrequency bioelectrical impedance. Specifically, we considered fat mass (FM), fat-free mass (FFM), FFMI index (FFMI), FM/FFM relation, body cell mass (BCM), basal metabolic rate (BMR), and BMR/FFM relation to evaluate the bodily function in the paretic and nonparetic regions. These values showed considerable differences according to grades determined by the stroke causes and the paralysis status. In the paretic regions, the FFM, FFMI, BCM, and BMR were low and the FM was high. In contrast, the nonparetic regions showed a high FFM and low FM. Furthermore, the paretic and nonparetic regions of all patients suitably fit a linear relation (slope: 22.17 kcal/day/kg) between BMR and FFM. Therefore, bio-electrical impedance measurements can be very useful to quantitatively assess paretic and nonparetic regions in hemiplegic stroke patients.

Effects of Taeumjowe-tang-gagambang on the Glycometabolism and Lipidmetabolism in the Liver Tissue of Diet-induced Obesity Mice (비만 유도 흰쥐 간조직에서 태음조위탕(太陰調胃湯) 가감방(加減方)이 당과 지질대사에 미치는 영향)

  • The aim of this study was to investigate the effect of Taeeumjowuitanggagam-bang (TJV) on the mRNA expression of Sterol regulatory element binding proteins (SREBPs), Tumor necrosis factor-$\alpha$ (TNF-$\alpha$) and Interlukin-6 (IL-6) that are considered to play an important role in lipid and glucose metabolism. For diet-induced obese studies, we split mice into 2 groups. The low fat diet group (LFD, n=8) were supplied with general diet for 10 weeks and the high fat diet group (HFD, n=18) were supplied with 60 kcal% fat diet for 10 weeks. And then The HFD group, the diet-induced obese group, were divided into 3 groups ; a group supplied with normal saline, a group treated with TJV 200 mg/kg and a group treated with TJV 500 mg/kg. They were treated orally with TJV and measured their body weight every day during 10 weeks. After that, we measured mRNA expressions of TNF-$\alpha$, IL-6 and SREBP-1c in liver, and blood concentrations of glucose, total cholesterol and triglyceride too. The results are as follows. The TJV reduced glucose and total cholesterol of blood concentration. The TJV reduced the mRNA expressions of TNF-$\alpha$ and SREBPs in liver. However, We couldn't find the TJV effects on the mRNA expression of IL-6, triglyceride blood concentration, and body weight among groups. The TJV stained liver tissue less red than control group. These results suggest that TJV may be effective for regulation of lipid and glucose metabolism in liver.

Single Oral Dose Toxicity Test of Blue Honeysuckle Concentrate in Mice

  • The objective of this study was to obtain single oral dose toxicity information for concentrated and lyophilized powder of blue honeysuckle (Lonicera caerulea L., Caprifoliaceae; BHcL) in female and male ICR mice to aid in the process of developing natural origin medicinal ingredients or foods following proximate analysis and phytochemical profile measurement. The proximate analysis revealed that BHcL had an energy value of 3.80 kcal/g and contained 0.93 g/g of carbohydrate, 0.41 g/g of sugar, 0.02 g/g of protein, and 0.20 mg/g of sodium. BHcL did not contain lipids, including saturated lipids, trans fats, or cholesterols. Further, BHcL contained 4.54% of betaine, 210.63 mg/g of total phenols, 159.30 mg/g of total flavonoids, and 133.57 mg/g of total anthocyanins. Following administration of a single oral BHcL treatment, there were no treatment-related mortalities, changes in body weight (bw) or organ weight, clinical signs, necropsy or histopathological findings up to 2,000 mg/kg bw, the limited dosage for rodents of both sexes. We concluded that BHcL is a practically non-toxic material in toxicity potency.
