Anti-Inflammatory Effects of N1-Benzyl-4-Methylbenzene-1,2-Diamine (JSH-21) Analogs on Nitric Oxide Production and Nuclear Factor-kappa B Transcriptional Activity in Lipopolysaccharide-Stimulated Macrophages RAW 264.7

  • $N^1$-Benzyl-4-methylbenzene-1,2-diamine (JSH-21) and its analogs were chemically synthesized and their anti-inflammatory potentials investigated. JSH-21 inhibited nitric oxide (NO) production in lipopolysaccharide (LPS)-stimulated macrophages RAW 264.7 in a dose-dependent manner, with an $IC_{50}$ value of 9.2 ${\mu}M$, where pyrrolidine dithiocarbamate and parthenolide as positive controls exhibited $IC_{50}$ values of 29.3 and 3.6 ${\mu}M$, respectively. The inhibitory effect of JSH-21 on the NO production was attributable to its down-regulatory action on LPS-inducible NO synthase (iNOS), which was documented by iNOS promoter activity. In the mechanism of the anti-inflammatory action, JSH-21 exhibited inhibitory effects on LPS-induced DNA binding activity and transcriptional activity of nuclear factor-kappa B (NF-$_KB$). Structural analogs of JSH-21 also inhibited both the LPS-induced NO production and NF-$_KB$). transcriptional activity, where diamine substitution at positions 1 and 2 of JSH-21 seems to play an important role in the anti-inflammatory activity.

드론센싱자료와 식생지수를 활용한 환경피해범위 산출 정확도 평가 (Accuracy Assessment of Environmental Damage Range Calculation Using Drone Sensing Data and Vegetation Index)

  • 본 연구는 사고현장에서 화학물질로 인해 발생한 사고지점의 피해면적을 식생지수로 산출하는 방안을 연구하였다. 자료수집은 두 종류의 드론을 활용하였으며, 사진측량기법을 적용한 3차원 점군자료에서 피해면적을 산출하였다. 다중분광센서의 분광대역 정보를 활용하여 제작한 정사영상을 토대로 식생지수 영상을 제작하였고, 임계값에 따른 피해면적의 결과로 사고현장에 대한 통계를 분석하였다. 근적외선 밴드 기반의 식생지수 Kappa 값은 0.79, 녹색 밴드 기반의 식생지수는 0.76으로 화학물질사고 조사현장에서 식생지수를 활용한 피해면적 분석 방법을 활용할 수 있을 것으로 판단된다.

위암의 임상적 병기 설정을 위한 전산화단층촬영 및 초음파 내시경의 진단력 평가 (Analysis of Diagnostic Performance of CT and EUS for Clinical TN Staging of Gastric Cancer)

  • 목적: 위암의 수술 전 병기 설정은 환자의 예후를 결정하고 치료 방침을 결정하기 위해 매우 중요하다. 이에 저자들은 위암의 TN병기 설정을 위해 주요한 진단 도구인 전산화단층촬영과 초음파 내시경의 진단력에 대해 분석하였다. 대상 및 방법: 2003년 5월부터 2007년 12월까지 분당서울대병원에서 위암으로 위절제술을 시행 받은 1,206명의 환자 중, 본원에서 시행한 검사가 아니거나, 내시경적 점막절제술 후 위절제술을 시행한 환자의 경우를 제외한 1,174명을 대상으로 후향적인 연구를 시행하였다. 두 검사의 수술 전 병기와 술 후 조직병리학적 병기가 일치하는 정도를 보기 위해서 코헨의 카파값(kappa value)과 ROC곡선하면적(Area under the ROC curve, AUC)을 이용하였다. 결과: 1,174명의 환자의 평균 나이는 $59.31{\pm}11.98$세였고, 조기위암이 638예, 진행 위암이 536예였다. 1,117명의 환자에서 전산화단층촬영이 시행되었으며, 초음파 내시경은 839예에서 시행되었다. T병기를 진단하는데 있어 전산화단층촬영이나 초음파 내시경의 카파값은 각각 0.4039 (P=0.021), 0.4201 (P=0.026)로 중등도의 일치도(moderate agreement)를 보였으나, 전체 T병기를 진단하는데 있어서 통계적으로 유의한 차이는 없었다. 이외에도 조기위암에서의 점막 병변과 점막하 병변을 구분하는 초음파 내시경의 진단력은 정확도 58.92%, 카파값 0.206으로 fair agreement를 보여 예상보다 진단력이 낮음을 알 수 있었다. 이외에도 T2 이상의 병변과 T2 이하의 병변을 감별하기 위해 두 검사를 동시에 시행한 환자군에서 두 검사 중 높은 병기를 진단한 검사를 채택하였을 때 AUC가 0.84로 한 개의 검사를 시행했을 때보다 진단력이 증가하였다. N병기를 진단하는 데 있어서는 전산화단층촬영의 진단력이 초음파 내시경보다 더 높음을 알 수 있었다. 결론: 위암의 T병기를 진단하기 위해 전산화단층촬영과 초음파 내시경 중 어느 한 검사가 우월하다고 할 수 없었으나, T2 이상의 병변과 T2 이하의 병변을 구별하기 위해서는 두 검사를 모두 시행하여 높은 병기를 채택한 경우 진단력이 높았다. 또한, 림프절 전이 여부를 알기 위해서는 전산화단층촬영이 우월하였다. 점막 병변과 점막하 병변을 구별하기 위한 초음파 내시경의 진단력은 낮았다.

LPS를 처리한 RAW 264.7 세포에서 털여뀌와 양지꽃 추출물의 NF-κB 활성화 및 Nitric Oxide 생성 저해 (Persicaria orientalis and Potentilla fragarioides Extracts Inhibit NF-κB Translocation and Nitric Oxide Production in LPS-stimulated RAW 264.7 Cells)

  • Persicaria orientalis (L.) Spach (Po)와 Potentilla fragarioides var. major Maxim (Pf)의 추출물로 reactive oxygen species (ROS)와 같은 free radical의 억제를 통한 항염증 활성을 살펴보았다. 또한, Po와 Pf로 LPS를 처리한 murine macrophage RAW 264.7 세포에서 발생한 nitric oxide의 저해와 NF-${\kappa}B$의 핵으로 translocation의 저해를 살펴보았다. 3-morpholinosydnonimine hydrochloride (SIN-1) $50{\mu}M$에 의해 유도된 ROS의 Po에 의한 50% 저해값 ($IC_{50}$)은 $23.35{\pm}1.27mg/mL$, Pf에 의한 $IC_{50}$$8.46{\pm}1.22mg/mL$이었다. 또한, SIN-1 $50{\mu}M$에 의해 유도된 peroxynitrite의 Po에 의한 $IC_{50}$$2.19{\pm}0.04mg/mL$, Pf에 의한 $IC_{50}$$0.80{\pm}0.02mg/mL$이었다. LPS 1 mg/mL을 처리한 RAW 264.7 세포에서 Nitric oxide는 증가하였으나 Po와 Pf 추출물을 처리한 그룹에서 농도의존적, 유의적으로 감소하였다. Po 추출물을 처리한 그룹의 Nitric oxide 생성량은 $13.34{\pm}0.67{\mu}M$, Pf 추출물을 처리한 그룹의 Nitric oxide 생성량은 $11.45{\pm}0.57{\mu}M$이었다. 또한, Po와 Pf 추출물은 LPS를 처리한 RAW 264.7 세포에서 NF-${\kappa}B$의 핵으로의 translocation을 저해하였다. 그러므로, Po와 Pf는 항염증 소재로서의 가능성이 충분하다고 사료된다.

Fungal Taxol Extracted from Cladosporium oxysporum Induces Apoptosis in T47D Human Breast Cancer Cell Line

  • Purpose: The present study concerns molecular mechanisms involved in induction of apoptosis by a fungal taxol extracted from the fungus Cladosporium oxysporum in T47D human breast cancer cells. Materials and Methods: Apoptosis-induced by the fungal taxol was assessed by MTT assay, nuclear staining, DNA fragmentation, flow cytometry and pro- as well as anti-apoptotic protein expression by Western blotting. Results: Our results showed inhibition of T47D cell proliferation with an $IC_{50}$ value of $2.5{\mu}M/ml$ after 24 h incubation. It was suggested that the extract may exert its anti-proliferative effect on human breast cancer cell line by suppressing growth, arresting through the cell cycle, increase in DNA fragmentation as well as down-regulation of the expression of NF-${\kappa}B$, Bcl-2 and Bcl-XL and up-regulation of pro-apoptotic proteins like Bax, cyt-C and caspase-3. Conclusions: We propose that the fungal taxol contributes to growth inhibition in the human breast cancer cell through apoptosis induction via a mitochondrial mediated pathway, with possible potential as an anticancer therapeutic agent.

사각 단면을 갖는 철도차량 주위의 3차원 유동해석 (Three-Dimensional Flow Analysis around Rolling Stock with Square Cross Section Using Low Re ${\kappa}-{\epsilon}$)

  • Three-dimensional numerical study is performed for the flow analysis around the rolling stock with square cross section (Mugungwha train model). The height (H) of rolling stock is considered as the characteristic length and the total length of rolling stock is 40 which correspond to 1/2 unit of rolling stock. The gap between the surface and rolling stock is 0.17H which is average value. The relative velocity between the surface and rolling stock is assumed to be zero and Re=10,000 based on the characteristic length. Low Re ${\kappa}-{\epsilon}$[15] is employed for the calculation of turbulence which resolve all the way to the solid surface (laminar sub-layer). Large flow separation occurred at the front head of train and a pair of vortex is generated on both top and side of rolling stock. The behavior of vortices on the top of the rolling stock is believed to affect the performance of the pantograph which should be intensively investigated. The difference between the high pressure in the front stagnation region of train and the low pressure in the rear separated region causes a large pressure drag. A large pair or vortex are generated in the rear of train and the size of vortex is increased more than the size of cross section of train.

척수손상환자에서 보행 기능 평가도구인 WISCI II의 타당도와 신뢰도 (Validity and Reliability on the Walking Index for Spinal Cord Injury in the walking assessment of the SCI patient)

  • The purpose of this study was to determine the validity and the reliability of the WISCI II to ascertain its value in the walking function evaluation of spinal cord injury patient. The WISCI II consists of 20 variables with a total valus ranging from 6 to 20 score. A group of 23 spinal cord injury patient were included in this study. To determine the validity, kappa statistics between the WISCI II and SCIM II were measured. The result of this study are as follows: 1) In the validity study, the kappa statistics between the WISCI II and SCIM II were 0.79 and 0.84 for an initial total score and a discharge total score respectively, indicating a reasonable agreement between the two test. 2) In the reliability study, the Cronbach's alpha coefficient was 0.83 and 0.95 for total score indicating a good internal consistency. The finding suggest that the WISCI II demonstrated an acceptable validity and reliabilit for the evaluation of walking function capacity of spinal cord injury patient in clinical practice.

Object-oriented Classification and QuickBird Multi-spectral Imagery in Forest Density Mapping

  • Forest cover density studies using high resolution satellite data and object oriented classification are limited in India. This article focuses on the potential use of QuickBird satellite data and object oriented classification in forest density mapping. In this study, the high-resolution satellite data was classified based on NDVI/pixel based and object oriented classification methods and results were compared. The QuickBird satellite data was found to be suitable in forest density mapping. Object oriented classification was superior than the NDVI/pixel based classification. The Object oriented classification method classified all the density classes of forest (dense, open, degraded and bare soil) with higher producer and user accuracies and with more kappa statistics value compared to pixel based method. The overall classification accuracy and Kappa statistics values of the object oriented classification were 83.33% and 0.77 respectively, which were higher than the pixel based classification (68%, 0.56 respectively). According to the Z statistics, the results of these two classifications were significantly different at 95% confidence level.

Immunomodulatory effects of six Acetobacter pasteurianus strains in RAW-Blue macrophage

  • In this study, we investigated the immunological properties of six strains of Acetobacter pasteurianus through nuclear factor-kappa B/activator protein-1 (NF-κB/AP-1) transcription factor activation and nitric oxide (NO) and cytokine production in macrophages. We found that the six A. pasteurianus strains had no significant inhibitory effect on the cell viability of RAW-BlueTM cells at the concentration of (25, 50, 100 CFU/macrophage). The production of NO and cytokines (TNF-α, IL-1β, and IL-6) showed different abilities of immune activation for each strain, and it was 0.7 to 0.9 times higher than that of the LPS (100 ng/mL, v/v) positive control and 7 to 8 times superior to that of the negative control group. To explore the underlying mechanism, we evaluated the mRNA expression of pro-inflammatory genes. Consequently, we found that inducible nitric oxide synthase (iNOS) and cyclooxygenase (COX)-2 expression including genes expression of cytokines were elevated by the six A. pasteurianus treatment. These results suggested that the six strains of A. pasteurianus have an excellent industrial application value as a functional material for the purpose of enhancing immune function.

A Novel Dental Plaque Index Using Intraoral Camera Images

  • Background: This study aimed to evaluate whether a Novel Plaque Index (NPI) using intraoral camera images would be more useful for assessing the accumulated area of dental plaque compared to three other indices. Methods: This study evaluated 80 disclosed plaque tooth surface images from 40 participants, including the facial surfaces of the maxillary left first molar and mandibular left central incisor. The tooth surface was divided longitudinally into three sections, with each section further subdivided into four subsections. A score ranging from 0 to 5 was assigned to each longitudinal third, and the average score was determined as the NPI score for the tooth surface. Two examiners assessed the NPI, Patient Hygiene Performance Index, Quigley and Hein Plaque Index, and Rustogi's Modified Navy Plaque Index scores obtained using the plaque images. The assessments were repeated twice with a 2-weeks washout period. The Plaque Percent Index (PPI) score calculated via image analysis is considered the gold standard. Results: The NPI showed the highest correlation coefficient with PPI compared to the other indices (examiner 1: 0.87, examiner 2: 0.88). The intraexaminer reliability of the NPI was excellent for both examiners. The intraclass correlation coefficient (ICC) for examiner 1 and 2 was 0.95 and 0.93, respectively. The weighted kappa values were >0.85 for both examiners. The ICC of the interexaminer analysis was 0.93 and the weighted kappa showed a reliability value of 0.81. Conclusion: The NPI was found to have greater validity and reliability than the three existing indices for the quantitative scoring of dental plaque.
