강황(薑黃)이 MIA 유도 골관절염 모델에 미치는 영향 (Effects of Curcuma longa Rhizoma on MIA-induced Osteoarthritis in Rat Model)

  • Objectives: The aim of this study was to investigate the anti-inflammatory effects of Curcuma longa rhizoma extract in an experimental rat model of osteoarthritis. Methods: Osteoarthritis was induced in rats by injecting monosodium iodoacetate (MIA) into the knee joint cavity of rats. The rats were divided into 5 groups (Normal, Control, positive comparison, low (CL) and high (CH) concentration groups). Rats in the low concentration (CL) group had MIA-induced osteoarthritis; they were treated with Curcuma longa rhizoma extract at a dose of 50mg/kg body weight. Rats in the high concentration (CH) group had MIA-induced osteoarthritis; they were treated with Curcuma longa rhizoma extract at a dose of 100mg/kg body weight. Hind paw weight distribution and ROS levels were measured. At the end of all treatments, changes in alanine aminotransferase (ALT), aspartate aminotransferase (AST), blood urea nitrogen (BUN), and creatinine levels were analyzed. In addition, inflammatory protein levels were evaluated by western blot analysis. Results: In this study, hind paw weight distribution significantly improved in the CL and CH groups, while. Reactive oxygen species (ROS) production significantly decreased in both. The levels of ALT, AST, BUN, and creatinine did not significantly change in either group. The production of nicotinamide adenine dinucleotide phosphate oxidase 4 (NOX4), $p47^{phox}$, and Ras-related C3 botulinum toxin substrate 1 (RAC1) decreased in both. Catalase, heme oxygenase-1 (HO-1) and superoxide dismutase (SOD) significantly increased in the CL and CH groups, respectively. Nuclear factor erythroid 2 (Nrf2) increased, but there were no significant differences between the experimental and control groups. Inflammatory cytokines, including nuclear factor-kappa Bp65 (NF-${\kappa}Bp65$), interleukin-1beta (IL-$1{\beta}$), and tumor necrosis factor-alpha (TNF-${\alpha}$), decreased significantly in both the CL and CH groups. Conclusions: Our results showed that Curcuma longa rhizoma extract has anti-inflammatory effects. Anti-inflammatory activity is regulated by the inhibition of inflammatory cytokines and mediators, such as NF-${\kappa}B$, therefore, it suppresses cartilage damage as well.

괴화산(槐花散)이 Dextran Sulfate Sodium으로 유도된 생쥐의 궤양성 대장염에 미치는 영향 (Effects of Goihwa-san on Ulcerative Colitis Induced by Dextran Sulfate Sodium in Mice)

  • Objectives : This study was carried out to investigate the effects of Goihwa-san(GHS) against ulcerative colitis induced by DSS (dextran sulfate sodium). Method : The sample group was divided into three. The control group consisted of mice that were not inflammation-induced. The pathological group was composed of untreated colitis elicited mice. The experimental group was administered GHS after colitis elicitation. The effects on ulcerative colitis were evaluated by the morphological change of colonic mucosa, the anti-oxidant effect, HSP 70, NF-${\kappa}$B, COX-1, COX-2 and iNOS. Results : In terms of immunohistochemical changes, the distribution of COX-1 in mice treated with GHS increased noticeably more than that in the pathological group. The distributions of HSP70, NF-${\kappa}$B, COX-2, iNOS in mice treated with GHS more decreased than those in the pathological group. Regeneration of surface epithelial cell and goblet cell in mucosa was observed by optical microscope. The colonic lengths in GHS-treated mice were more elongated than those of DSS only treated mice. Conclusion : GHS is a candidate treatment for ulcerative colitis.

A Sphingosine Kinase-1 Inhibitor, SKI-II, Induces Growth Inhibition and Apoptosis in Human Gastric Cancer Cells

  • SKI-II has been reported as an inhibitor of sphingosine kinase 1 and has been extensively used to prove the involvement of sphingosine kinase and sphingosine-1-phosphate (Sphk1) in cellular processes. In the current study, we investigated the effects of SKI-II and its potential mechanisms in human gastric cancer SGC7901 cells. After treatment with SKI-II, cell growth, cell cycle distribution, apoptosis, expression of Sphk1, NF-${\kappa}B$, Bcl-2, Bax and p27 were assessed by MTT assay, flow cytometry, electron microscopy, immunocytochemistry and Western-blot assay, respectively. Our results showed that SKI-II markedly inhibited SGC7901 cell survival in a dose-dependent manner, reduced cell proliferation with accumulation of cells in the G0/G1 phase and induced apoptosis in the tumor cells. Furthermore, Western blotting and immunocytochemistry showed that the expression of p27 and Bax was increased significantly, but the expression of NF-${\kappa}B$, Bcl-2 and Sphk1 decreased by different degrees. These results indicate that SKI-II induced cell growth arrest and apoptosis. The increased apoptotic sensitivity of SGC7901 was correlated with NF-${\kappa}B$ or Bcl-2/Bax activation.

사상체질(四象體質)의 유전성(遺傳性) 검토(檢討)에 대한 통계적(統計的) 연구(硏究) (Statistical Study on Heredity in SaSang Constitutional Medicine)

  • Purpose : Sasang Constitutional Medicine explains the pathology peculiar to constitution and suggests treatment for each constitution. In Sasang Constitutional Medicine hwnan beings are classified on four groups; Taeyangin, Taeumin, 5oyangin, Soeumin. These four constitution has their's own symptoms and treatments. In treatment, control of mind inclination, that is to say, moderation takes a very important role. But the study on heredity in Sasang Constitutional Medicine has not done not biological study but also statistical study. So we used several statistical methods and analyzed 163 samples. Methods : We implemented Fishers exact test for adjusting chi-squared test, kappa coefficient to estimate agreement of parent's and children's constitutions, and finally plotted bi-plot using correspondence analysis. Results : From Fisher's exact test result, we could know that parent's and children'S constitution's distribution had significant difference. In kappa coefficient, mother and daughter's estimated value produced highest result. In correspondence analysis we only plotted the case of mother and son for easy interpretation. Conclusion : In the study of heredity of SaSang constitution, we cannot know exactly the heredity of constitution in terms of biology or genetics. But this research can be helpful for further analysis, that is, a study of biological or genetical aspects. And we could conclude that in statistical aspects the heredity in SaSang constitution is meaningful.

2차원과 3차원 아음속 공동 유동 특성에 대한 수치적 연구 (NUMERICAL ANALYSIS OF TWO- AND THREE-DIMENSIONAL SUBSONIC TURBULENT CAVITY FLOWS)

  • The flight vehicles have cavities such as wheel wells and bomb bays. The flow around a cavity is characterized as unsteady flow because of the formation and dissipation of vortices due to the interaction between the freestream shear layer and cavity internal flow, the generation of shock and expansion waves. Resonance phenomena can damage the structures around the cavity and negatively affect aerodynamic performance and stability. In the present study, numerical analysis was performed for cavity flows by the unsteady compressible three dimensional Reynolds-Averaged Navier-Stokes (RANS) equations with Wilcox's ${\kappa}\;-\;{\omega}$ turbulence model. The cavity has the aspect ratios of 2.5, 3.5 and 4.5 for two-dimensional case, same aspect ratios with the W/D ratio of 2 for three-dimensional case. The Mach and Reynolds numbers are 0.53 and 1,600,000 respectively. The flow field is observed to oscillate in the "shear layer mode" with a feedback mechanism. Based on the SPL(Sound Pressure Level) analysis of the pressure variation at the cavity trailing edge, the dominant frequency was analyzed and compared with the results of Rossiter's formula. The MPI(Message Passing Interface) parallelized code was used for calculations by PC-cluster.

후방계단유동에 대한 저레이놀즈 수 난류모형의 예측성능에 관한 연구 (The study of predictive performance of low Reynolds number turbulence model in the backward-facing step flow)

  • Incompressible flow over a backward-facing step is computed by low Reynolds number turbulence models in order to compare with direct simulation results. In this study, selected low Reynolds number 1st and 2nd (Algebraic Stress Model : ASM) moment closure turbulence models are adopted and compared with each other. Each turbulence model predicts different flow characteristics, different re-attachment point, velocity profiles and Reynolds stress distribution etc. Results by .kappa.-.epsilon. turbulence models indicate that predicted re-attachment lengths are shorter than those by standard model. Turbulent intensity and eddy viscosity by low Reynolds number .kappa.-.epsilon. models are still greater than DNS results. The results by algebraic stress model (ASM) are more reasonable than those by .kappa.-.epsilon. models. The convective scheme is QUICK (Quadratic Upstream Interpolation for Convective Kinematics) and SIMPLE algorithm is adopted. Reynolds number based on step height and inlet free stream velocity is 5100.

익위탕(益胃湯)의 Indomethacin유발 위점막 손상에 대한 효과 (The Effects of Ikwitang Extract on Indomethacin-induced Gastric Mucosal Lesions)

  • This study was carried out to investigate the effects of Ikwitang extract on indomethacin-induced gastric mucosal lesions in mice. Experimental groups were classified into a non-treated group(NM group), a non-administered group(CON group), the Misoprostol-administered group(MI group) and the Ikwitang-extract-administered group(IW group). This study examined the morphological change, distribution of mast cells, mucous surface cells, acid mucose secreted cells, and apoptic cells, BrdU, COX-1, Hsp70, $NF-{\kappa}B\;p50$, PKC, COX-2 and IL-12B of gastric mucosa. Results : The results of this study were as follows: 1. In the Misoprostol-administered group and the Ikwitang-extract-administered group, the hemorrhagic erosion of gastric mucous and infiltrated mast cells decreased. 2. Mucous surface cells and acid mucose secreted cells abserved in the Misoprostol-administered group and the Ikwitang-extract administered group. 3. The distribution of apoptic cells, Hsp70, $NF-{\kappa}B\;p50$, PKC, COX-2, and IL-12B increased in the Control group, but decreased in the Misoprostol-administered group and Ikwitang-extract-administered group. 4. Cells proliferation of gastric mucosa and the COX-l positive cells decreased in the Control group, but increased in the Misoprostol-administered group and the Ikwitang-extract -administered group. The above results suggest the lkwitang extract had beneficial effects on indomethacin induced gastric mucosal lesions.

Holstein 종(種) 유우(乳牛)의 유단백질(乳蛋白質)의 유전적다형(遺傳的多型)과 유조성분간(乳組成分間)의 연관성 (Association between Genetic Polymorphisms of Milk Proteins and Milk Compositions in Holstein Cows)

  • 본(本) 연구(硏究)는 유단백질(乳蛋白質)의 유전적표식(遺傳的標識)(genetic marker)를 유우(乳牛)의 유전적(遺傳的) 개량(改良)을 위한 유우등록 및 선발(選拔) 자료로 활용하고자 1992년 국립 종축원에서 사육증인 Holstein종 159두에서 각각 유즙 시료를 채취하여 Polyacrylarnide gel(PAGE)에 의한 전기영동방법에 의하여 ${\alpha}S1$-casein(${\alpha}S1$-CN) 및 ${\beta}$-casein(${\beta}$-CN), ${\kappa}$-casein(${\kappa}$-CN) 및 ${\beta}$-lactoglobulin(${\beta}$-LG)의 유전적(遺傳的) 다형(多型)을 분석(分析)하여 유전적(遺傳的) 변이체의 표현형(表現型), 유전자형(遺傳子型) 및 유전자빈도(遺傳子頻度)를 추정하고 이들 단백질의 유전적(遺傳的) 다형(genetic polyrnorphisms)과 유조성분과의 연관성을 분석(分析) 검토(檢討)하였다. 유단백질(乳蛋白質)인 ${\alpha}S1$-CN, ${\beta}$-CN, ${\kappa}$-CN 및 ${\beta}$-LG 유전좌위(遺傳座位)의 유전적(遺傳的) 변이체의 표현형(表現型) 분포의 관찰치와 기대치간에 유의적(有意的)인 차이가 인정 되지않아 이들 유전자(遺傳子)들이 유전적(遺傳的) 평형을 이루고 있는 것으로 사료되었다. 유단백질(乳蛋白質) 좌위(座位)의 유전자형(遺傳子型)을 분석(分析)한 결과(結果) ${\alpha}S1$-CN BB유전자형(遺傳子型), ${\beta}$-CN AA유전자형(遺傳子型), ${\kappa}$-CN AA유전자형(遺傳子型) 및 ${\beta}$-LG AB 유전자형(遺傳子型)의 출현빈도(出現頻度)가 각각 79.87%, 84.28%, 71.70% 및 49.10%로 높게 나타났다. 유단백질(乳蛋白質)의 유전자빈도(遺傳子頻度)에 있어서는 ${\alpha}S1-CN^B$${\alpha}S1-CN^C$가 각각 0.899 및 0.101이었고, ${\beta}-CN^A$${\beta}-CN^B$은 각각 0.921 및 0.079이었으며, ${\kappa}-CN^A$${\kappa}-CN^B$은 각각 0.837 및 0.163이었고, ${\beta}-LG^A$${\beta}-LG^B$은 각각 0.378 및 0.622이었다. 분산분석(分散分析) 결과(結果) 유단백질(乳蛋白質)의 유전자형(遺傳子型)들은 유조성분인 유지율, 단백질 함량 및 총 고형분 함량에서 유의적(有意的)인 연관성이 인정되었다. 유조성분 함량에 있어서는 ${\kappa}$-CN좌위(座位)의 BB유전자형(遺傳子型)은 유지율 및 유단백질(乳蛋白質) 함량에서 ${\kappa}$-CN좌위(座位)의 AA 및 AB 유전자형(遺傳子型)보다 유의적(有意的)으로 높게 나타났으며, ${\beta}$-LG좌위(座位)의 AA유전자형(遺傳子型)은 유지율에서 ${\beta}$-LG좌위(座位)의 AB 및 BB 유전자형(遺傳子型)보다 유의적(有意的)으로 높게 추정되었다. 이상의 연구결과(硏究結果)를 종합하여 볼때 유조 성분인 유지율 및 단백질함량을 높히기 위하여서는 ${\kappa}$-CN좌위(座位)의 BB 유전자형(遺傳子型)과 ${\beta}$-LG좌위(座位)의 AA 유전자형(遺傳子型)을 선발(選拔)하는것이 보다 유리할 것으로 생각되었다.

개정 Prand시 이론을 이용한 유사 농도 분포식 (A Sediment Concentration Distribution Based on a Revised Prandtl Mixing Theory)

  • 멱 유속 분포를 구하기 위해 개정 Prandtl 혼합 거리 이론이 이용되었으며, 여기에서 사용된 지수값의 범위는 1/4~1/7이었다. 이 개정된 유속분포를 이용하여 간단한 부유사 농도 분포식을 개발하였다. 미국 지질조사국이 리오그란데강에서 실측한 자료와 명목값인 $\beta$=1.0, $textsc{k}$=0.4, 그리고 가시관에 의해 얻어진 침강속도를 이용한 개정 농도식 계산결과와 실측지와의 비교는 양호한 편이었으며, 지수에 임의로 두배를 해주었을 경우에는 좋은 결과를 보였다. 적당한 $\beta$, $textsc{k}$, 그리고 침강속도를 선택하기 위해 더 많은 연구가 필요하지만 이러한 연구가 대규모 난류와 이차류 영향에 대한 설명을 할 수는 없을 것이다. 하지만 실용적인 측면에서 보면 어느 관측지점에 대한 매우 자세하고 특별하게 측정된 자료는 그 지점에 맞는 지수나 계수를 찾아낼 수도 있다.

디퓨저에서 벽면으로의 방출유로에서의 난류유동에 관한 수치 해석적 연구 (A Numerical Study on the Turbulent Flow in the Discharge Flow Path from a Diffuser to a Wall)

  • A numerical study was made to choose the better turbulence model for the flow in the discharge flow path from a diffuser to a wall. In this study standard $\kappa-\epsilon$ model(SKE), RNG $\kappa-\epsilon$ model(RNG), and Reynolds stress model(RSM) were applied. In case of the flow with relatively high Reynolds number at a diffuser inlet, the pressure loss coefficients by RNG have a tendency to be near to those by SKE at small ratio(below about 0.35) of $h/D_o$, but to those by RSM at large ratio(above about 0.35). At large ratio RNG begins to enlarge the effects of rapid strain and streamline curvature. RNG & RSM are recommended as the appropriate turbulence models for this case. But it is noticeable that the velocity gradient pattern in RNG is same as in SKE, and also that the total pressure distribution in RNG is same as in RSM only at swirling flow area, same as in SKE only at main flow area.
