Association analysis of polymorphisms in six keratin genes with wool traits in sheep

  • Objective: The purpose of this study was to investigate the genetic effects of six keratin (KRT) genes on the wool traits of 418 Chinese Merino (Xinjiang type) (CMXT) individuals. Methods: To explore the effects and association of six KRT genes on sheep wool traits, The polymerase chain reaction-based single-strand conformation polymorphism (PCR-SSCP), DNA sequencing, and the gene pyramiding effect methods were used. Results: We report 20 mutation sites (single-nucleotide polymorphisms) within the six KRT genes, in which twelve induced silent mutations; five induced missense mutations and resulted in $Ile{\rightarrow}Thr$, $Glu{\rightarrow}Asp$, $Gly{\rightarrow}Ala$, $Ala{\rightarrow}Ser$, $Se{\rightarrow}His$; two were nonsense mutations and one was a same-sense mutation. Association analysis showed that two genotypes of the KRT31 gene were significantly associated with fiber diameter (p<0.05); three genotypes of the KRT36 gene were significantly associated with wool fineness score and fiber diameter (p<0.05), three genotypes of the KRT38 gene were significantly associated with the number of crimps (p<0.05); and three genotypes of the KRT85 gene were significantly associated with wool crimps score, body size, and fiber diameter (p<0.05). Analysis of the gene pyramiding effect between the different genotypes of the gene loci KRT36, KRT38, and KRT85, each genotype in a gene locus was combined with all the genotypes of another two gene loci and formed the different three loci combinations, indicated a total of 26 types of possible combined genotypes in the analyzed population. Compared with the other combined genotypes, the combinations CC-GG-II, CC-HH-IJ, CC-HH-JJ, DD-HH-JJ, CC-GH-IJ, and CC-GH-JJ at gene loci KRT36, KRT38, and KRT85, respectively, had a greater effect on wool traits (p<0.05). Conclusion: Our results indicate that the mutation loci of KRT31, KRT36, KRT38, and KRT85 genes, as well as the combinations at gene loci KRT36, KRT38, and KRT85 in CMXT have significant effects on wool traits, suggesting that these genes are important candidate genes for wool traits, which will contribute to sheep breeding and provide a molecular basis for improved wool quality in sheep.

Adsorption Characteristics of Pb(II), Cu(II), Cr(III), and Zn(II) Ions by Domestic Loess Minerals (국내산 황토를 이용한 수용액중의 Pb(II), Cu(II), Cr(III) 및 Zn(II) 이온의 흡착 특성)

  • Removal of Pb(Ⅱ), Cu(Ⅱ), Cr(Ⅲ), and Zn(Ⅱ) ions from aqueous solutions using the adsorption process on domestic loess minerals has been investigated. Variations of contact time, pH, adsorption isotherms and selectivity of coexisting ions and leachate were experimental parameters. YDI, YPT and KRT samples diluted in 1% aquous solution which was adjusted pH 10.8, 8.0 and 6.50, respectively. The result of XRD measurement, Quartz was mainly observed in all samples. In the case of KRT sample, Kaolinite, Feldspar, Chlorite consisting of clay minerals shows almost same pattern with YPT samples. Different properties showed from the YDI sample containing Iillite, remarkably. For all the metals, maximum adsorption was observed at 30min∼60min. Adsorption of metal ions on loess minerals were reached an equilibrium by shaking the solution for about 30min. Removal efficiency of Pb(Ⅱ) ion for KRT, YPT and YDI were 84.7%, 92% and 100%, respectively. The Cu(Ⅱ) and Zn(Ⅱ) adsorptivity on KRT showed the low in various pH solution However, those on YPT and YDI were high than 90% except for the pH 2 solution. The orders of adsorptivities for domestic loess minerals showed as following : YPT>KRT>YDI. The adsorption isotherms of Cu(Ⅱ) and Zn(Ⅱ) ions on clay minerals were fitted to a Freundlich's. Freundlich constants(1/n) of KRT and YPT domestic loess minerals were 0.63, 0.97 and 0.36, 0.25, respectively.

KRT13, FAIM2 and CYP2W1 mRNA Expression in Oral Squamous Cell Carcinoma Patients with Risk Habits

  • Background: Expression of KRT13, FAIM2 and CYP2W1 appears to be influenced by risk habits, thus exploring the associations of these genes in oral squamous cell cancer (OSCC) with risk habits, clinico-pathological parameters and patient survival may be beneficial in identifying relevant biomarkers with different oncogenic pathways. Materials and Methods: cDNAs from 41 OSCC samples with and without risk habits were included in this study. Quantitative real-time PCR was used to analyze KRT13, FAIM2 and CYP2W1 in OSCC. The housekeeping gene (GAPDH) was used as an endogenous control. Results: Of the 41 OSCC samples, KRT13 was down-regulated in 40 samples (97.6%), while FAIM2 and CYP2W1 were down-regulated in 61.0% and 48.8%, respectively. Overall, there were no associations between KRT13, FAIM2 and CYP2W1 expression with risk habits, selected socio-demographic and clinico-pathological parameters and patient survival. Conclusions: Although this study was unable to show significance, there were some tendencies in the associations of KRT13, FAIM2 and CYP2W1 expression in OSCC with selected clinic-pathological parameters and survival.

Anti-aging Effect of Artocarpin in UVA-irradiated Normal Human Epidermal Keratinocytes (자외선 조사에 의해 노화된 인간각질형성세포에서 Artocarpin의 항노화 효능)

  • The aim of this study was to investigate the epidermal moisturizing effects of artocarpin on normal human epidermal keratinocytes (NHEKs). To investigate the effects of artocarpin on NHEKs, cell viability and the expression of mRNAs related to skin hydration were measured. In addition, hyaluronic acid (HA)-ELISA assay was performed. Here, the effects of artocarpin on AQP3, HAS2, KRT1, and KRT10 mRNA expression, and on HA production, following UVA treatment were reported. The Quantitative real-time PCR results demonstrate that artocarpin increased AQP3, HAS2, KRT1, and KRT10 mRNA levels. The HA-ELISA assay revealed that artocarpin also increased HA production in NHEKs. Through these experiments, the epidermal moisturizing effects of artocarpin have been elucidated, providing evidence that artocarpin may be a potent cosmetic ingredient in skin anti-aging and moisturizing products. Based on these results, I anticipate that further research on the mechanisms of action of artocarpin may allow the development of not only cosmetics, but also medicines and healthcare foods.

Luciferase Assay to Screen Tumour-specific Promoters in Lung Cancer

  • Objective: Specific promoters could improve efficiency and ensure the safety of gene therapy. The aim of our study was to screen examples for lung cancer. Methods: The firefly luciferase gene was used as a reporter, and promoters based on serum markers of lung cancer were cloned. The activity and specificity of seven promoters, comprising CEACAM5 (carcinoembryonic antigen, CEA), GRP (Gastrin-Releasing Peptide), KRT19 (cytokeratin 19, KRT), SFTPB (surfactant protein B, SP-B), SERPINB3 (Squamous Cell Carcinoma Antigen, SCCA), SELP (Selectin P, Granule Membrane Protein 140kDa, Antigen CD62, GMP) and DKK1 (Dickkopf-1) promoters were compared in lung cancer cells to obtain cancer-specific examples with strong activity. Results: The CEACAM5, DKK1, GRP, SELP, KRT19, SERPINB3 and SFTPB promoters were cloned. Furthermore, we successfully constructed recombinant vector pGL-CEACAM5 (DKK1, GRP, SELP, KRT19, SERPINB3 and SFTPB) contained the target gene. After cells were transfectedwith recombinant plasmids, we found that the order of promoter activity from high to low was SERPINB3, DKK1, SFTPB, KRT19, CEACAM5, SELP and GRP and the order for promoters regarding specificity and high potential were SERPINB3, DKK1, SELP, SFTPB, CEACAM5, KRT19 and GRP. Conclusion: The approach adopted is feasible to screen for new tumour specific promoters with biomarkers. In addition, the screened lung-specific promoters might have potential for use in lung cancer targeted gene therapy research.

Identification of Antioxidant Activities and Stimulation of Human Keratinocytes Differentiation Effects of Syzygium claviflorum Extract (Syzygium claviflorum 추출물의 항산화 활성 및 각질형성세포 분화유도 효과)

  • We validated the physiological activity of Syzygium claviflorum (Roxb.) Wall. ex A.M. Cowan & Cowan (S. claviflorum) extracts (leaves, stems, fruits, and flowers) as a cosmetic ingredient. Firstly, S. claviflorum extracts removed over 80% of free radicals at various concentrations in antioxidant experiments using the DPPH and ABTS assay. In cytotoxicity experiments using human epidermal keratinocytes, S. claviflorum extracts showed low cytotoxicity. In addition, S. claviflorum extracts significantly increased the expression of keratin (KRT)1, KRT2, KRT9, KRT10, which are differentiation markers of keratinocytes, as well as genes involved in the maintenance of skin barrier function, including involucrin (IVL), loricrin (LOR), filaggrin (FLG), and claudin1 (CLDN1). In particular, the expression of FLG protein, inhibited by interleukin (IL)-4/IL-13 in atopic dermatitis, was restored by S. claviflorum extracts in in vitro experiments. Therefore, S. claviflorum extracts with excellent antioxidant efficacy and skin barrier improvement function will be useful materials for the development of future atopic dermatitis treatments and cosmetics.

Vacuolar H(+)-ATPase is not restricted to clear cells of the epididymal epithelium in cattle

  • Communication among epididymal epithelial cells creates the best luminal condition where spermatozoa mature, transport and are stored. Vacuolar ATPase (V-ATPase) and cytokeratin 5 (KRT5) have been used as signal indicators for clear and basal cells of the epididymal epithelium, respectively, in mice, rats, bats, and pigs; however, these two markers have not yet been described in the epididymis of bulls. Here, we examined the presence and distribution of the B1 subunit of V-ATPase (B1-VATPase) and KRT5 in the distinct regions of adult bovine epididymides, specifically, the caput, corpus, and cauda. Immunofluorescence staining and confocal microscopy showed that narrow shaped-clear cells were placed in the caput and corpus regions of the bovine epididymis; however, they were absent in the cauda epididymis. In addition, B1-VATPase was highly expressed in the cauda spermatozoa; however, it was rarely detected in the caput spermatozoa. On the other hand, KRT5-positive cells, basal cells, were maintained beneath the basal lamina and they had the traditional form with a dome-shaped morphology from the caput to cauda region of the bovine epididymis. The co-expression of B1-VATPase and KRT5 was confined to basal cells placed in the basal region of the epithelium. In summary, 1) clear cells were present with region-specific localization, 2) B1-VATPase was present in the corpus and cauda spermatozoa but absent in the caput, 3) co-expressed cells with B1-VATPase and KRT5 were present in the adult bovine epididymis, and 4) B1-VATPase was not a specific marker for clear cells in the bovine epididymis. Therefore, the perfect epididymal luminal condition created by the specific expression and localization patterns of B1-VATPase might be necessary to obtain fertilizing capacity of spermatozoa in the bovine epididymis.

The Synthesis and Hydraulic properties of Calcium Sulfo Aluminate(CSA) derived from Secondary Refining Slag. (제강 2차 정련 슬래그를 재활용한 칼슘설포알루미네이트(CSA) 합성 및 수화 특성)

  • The synthesis and hydration of Calcium Sulfo Aluminate[$3CaO{\cdot}3Al_2O_3{\cdot}CaSO_4(C_4A_3{\overline{S}})$, CSA cement utilizing secondary steelmaking refining slags is studied for recycling the discarded steel plant wastes to meet the environmental requrations imposed on the steel industry. Raw materials of secondary refining slag, lime sludge, gypsum and bauxite were prepared to be sintered at $1,250^{\circ}C$. The sintered samples were hydrated for 1, 3 and 7 days to evaluate the mineralogical and physico-mechanical properties. The hydration products evaluated with the aid of SEM and XRD analyses confirmed the formation and the continuing growth of ettringite phase with the further hydration times, which plays a role in developing the early strength and the expansion properties of cements. The physico-mechanical properties of hydrated CSA products employing the recycled steelmaking refining slags determined in terms of compressive strength and linear expansion of hydrated products are found to be superior to those of the Ordinary Portland Cement(OPC) or the other commercial CSA cements.

  • In collaboration with SAMPYO KRT, VOSSLOH COGIFER has designed and supplied all turnouts for the first Korean high speed project.. The track is laid on ballast and the turnouts were designed accordingly and as per the SNCF standard for turnouts at commercial speed of 300 km/h and design speed of 350 km/h. KNR has now decided to implement a ballastless technology for the extension of the line between (South line). To the best of our knowledge, it has also been decided that the system to be used is ballastless Concrete bearer. Thus, the following presentation has been done with a technology for ballastless track with the same system.
