Optimization of KOH pretreatment conditions from Miscanthus using high temperature and extrusion system (고온 압출식 반응시스템을 이용한 억새 바이오매스의 KOH 전처리조건 최적화)

  • The purpose of this study is to investigate the optimum conditions of biomass pretreatment with potassium hydroxide (KOH) for efficient utilization of cellulose, hemicellulose and lignin from Miscanthus. The optimization of variables was performed by response surface methodology (RSM). The variation ranges of the parameters for the RSM were potassium hydroxide 0.2~0.8 M, reaction temperature 110~190℃ and reaction time 10~90 min. The optimum conditions of alkali pretreatment from Miscanthus were determined as follows: concentration of KOH 0.47 M, reaction temperature 134℃ and reaction time 65 min. At the optimum conditions, the yield of cellulose from the solid fraction after pretreatment was predicted to be 95% by model prediction. Finally, 66.1 ± 1.1% of cellulose were obtained by verification experiment under the optimum conditions. The order contents of solid extraction were hemicellulose 26.4 ± 0.4%, lignin 3.7 ± 0.1% and ash 0.5 ± 0.04%. The yield of ethanol concentration of 96% was obtained using separated saccharification and fermentation.

  • The steam explosion-chemical pretreatment is a more effective wood pretreatment technique than the conventional physical pretreatment by accelerating reactions during the pretreatment process. In this paper, two-stage pretreatment processes of hardwood were investigated for its enzymatic hydrolysis and the succinic acid yield from the pretreated solid. The first stage pretreatment was performed under conditions of low severity to optimize the amount of solid recovery. In the second stage pretreatment washed solid material from the first stage pretreatment step was impregnated again with chemical (alkaline or chlorine-based chemicals) to remove a portion of the lignin, and to make the cellulose more accessible to enzymatic attack. The effects of pretreatment were assessed by enzymatic hydrolysis and fermentation, after the two stage pretreatments. Maximum succinic acid yield (16.1 g $L^{-1}$ and 77.5%) was obtained when the two stage pretreatments were performed at steam explosion -3% KOH.

Preparation of Polyester Using Waste Ethylene Glycol Generated from the Wastepaper Pretreatment Process (Ethylene glycol을 사용한 폐지의 전처리공정에서 발생된 폐액으로부터 polyester 제조)

  • This research was to investigate the polyester preparation using waste ethylene glycol (EG) generated from the wastepaper pretreatment process. Waste EG was obtained from using EG five times repeatedly in the pretreatment of wastepaper. The hydroxyl value of the waste EG was 441 mg KOH/g and its composition was 0.68% cellulose, 6.5% hemicellulose, 6.1% lignin, and 86.7% EG. Maleic acid was used as carboxylic acid. The effect of reaction temperature and time except carboxyl group/hydroxyl group ratio on the crosslinkage of the prepared polyester was marginal. Citric acid, lithium hydroxide and dicumyl peroxide were used as additive or catalyst to enhance the crosslinkage of polyester. Among them, 10% of citric acid was found to be most effective. The crosslinkage was 86% when the polyester was prepared at an optimum condition such as $130^{\circ}C$ and 15 minutes, 1.5 of C/H ratio, and 10% of citric acid, and its insoluble percentage in boiling water for 6 hours was 47%. The weight loss of the prepared polyester was approximately 40% when it was buried in damp soil for 5 months, indicating that it is readily biodegradable. This results can provide some information for future development of wastepaper pretreatment by organic solvent.

  • Effects of phenobarbital pretreatment on the pharmacokinetics of enterohepatic recirculating griseofulvin were investigated by comparing normal to bile duct cannulated rats and also the effects of enhanced endogeneous bile flow on the absorption of griseofulvin were studied by means of in situ recirculation method in phenobarbital-pretreated rats. Phenobarbital was administered orally for five days at the dose of 75 mg/kg/day. The influence of phenobarbital pretreatment on the absorption rate constant, area under the plasma concentration-time curve, maximum plasma concentration of orally administered griseofulvin was not found in bile duct cannulated rats. Decreased absorption clearance and apparent partition coefficient of griseofulvin in accordance with the amount of endogeneous bile juice seemed to be due to the decrease of thermodynamic activity of griseofulvin as bile forms the micelle with griseofulvin.

The Effect of Phenobarbital Pretreatment on the Pharmacokinetics of Diltiazem in Rats (랫트에 있어서 페노바르비탈 전처리가 딜티아젬의 생체내 동태에 미치는 영향)

  • The influence of phenobarbital (PB) pretreatment (75 mg/kg/day, i.p. for 4 days) on the pharmacokinetics of diltiazem (DTZ) and its metabolite, desacetyldiltiazem (DAD), was investigated in rats. DTZ was injected via femoral (3 mg/kg) or portal (10 mg/kg) vein to the control and PB-pretreated rats. DAD was also injected separately via femoral (3 mg/kg) vein to both groups of rats. The intrinsic hepatic plasma clearance of DTZ was found to be significantly increased (6.8-fold) by the PB pretreatment. However, the fraction of an intravenous DTZ dose converted to DAD $(F_mi)$ was only slightly (6%) increased and calculated metabolic rate constant of DTZ to DAD was not affected by the pretreatment. On the other hand, plasma free fraction of DTZ was increased (1.8-fold) from $4.24{\pm}0.25%$ to $7.45{\pm}0.54%$ by the pretreatment. However, the l.8-fold increase in the free fraction of DTZ would not explain the 6.8-fold increase in the hepatic intrinsic clearance of DTZ. Therefore, the increase in either the hepatic blood flow or the metabolism other than to DAD was expected as the probable mechanism(s) of the increased hepatic clearance of DTZ. Sequential metabolism of DAD to further metabolites, however, would be a more potential cause of the apparently unchanged metabolism of DTZ to DAD by the PB-pretreatment.

Microwave Assisted Energy Efficient Biodiesel Production from Crude Pongamia pinnata (L.) Oil Using Homogeneous Catalyst

  • Microwave assisted biodiesel production from crude Pongamia pinnata oil using homogeneous base catalyst (KOH) was unsuccessful because of considerable soap formation. Therefore, a two step process of biodiesel production from high free fatty acid (FFA) oil was investigated. In first step, crude P. pinnata oil was acid catalyzed using $H_2SO_4$ and acid value of oil was reduced to less than 4 mg KOH/g. Effect of sulfuric acid concentration, alcohol-oil molar ratio and microwave irradiation time on acid value of oil was studied. Result suggested that 1.5% $H_2SO_4$ (w/w), 6:1 methanol oil molar ratio and 3 min microwave irradiation time was sufficient to reduce the acid value of oil from 12 and 22 mg KOH/g to 2.9 and 3.9 mg/KOH/g, respectively. Oil obtained after pretreatment was subsequently used for microwave assisted alkali catalyzed transesterification. A higher biodiesel yield (99.0%) was achieved by adopting two step processes. Microwave energy efficiency during alkali catalyzed transesterification was also investigated. The results suggested a significant energy saving because of reduced reaction time under microwave heating.

Effect of Phenobarbital on the Nonlinear Pharmacokinetics of Naproxen (나프록센의 비선형 체내동태에 미치는 페노바르비탈의 영향)

  • In order to elucidate the effect of phenobarbital (PB) on the nonlinear pharmacokinetic behavior of naproxen (NAP), we compared the dose dependent hepatic intrinsic clearance, biliary excretion and protein binding of NAP in control rats to those in the PB-pretreated rats which were intraperitoneally pretreated with PB sodium (75 mg/kg) once a day for four days. NAP was injected via femoral (1.5 mg/kg) and portal(0.25, 0.5, 1.5, 15 and 30 mg/kg) vein to the control and PB-pretreated rats, respectively. And also, we measured the plasma free fraction of NAP with the equilibrium dialysis method and the biliary excreted total amounts of NAP in both rats. Plasma free fraction of NAP was decreased in lower concentration than $150\;{\mu}g/ml$ of NAP due to PB pretreatment. In higher concentration, however, plasma free fraction was increased. These in vitro results suggest that the total protein concentration was increased but the total binding capacity of NAP to protein was decreased by PB-pretreatment. The total plasma clearance and the hepatic intrinsic clearance of NAP had similar values in both groups, respectively. And, both clearances of NAP were significantly increased by PB-pretreatment. Even though the plasma free fractions of NAP in both groups were constantly remained within the concentration range according to the increase of administration dose, the hepatic intrinsic clearances of NAP were significantly increased in both groups with the increased dose. And, the biliary excreted total amounts of NAP were significantly increased by PB-pretreatment at the lower dose, but decreased at the higher dose. These in vivo results suggest that NAP represents the uncommon nonlinear pharmacokinetic behavior that the hepatic intrinsic clearance was enhanced with the increased dose, and that PB enhances further the hepatic intrinsic clearance of NAP with the increased dose due to its enzyme induction effect.

Development of Pretreatment Method for Analysis of Vitamin B12 in Cereal Infant Formula using Immunoaffinity Chromatography and High-Performance Liquid Chromatography

  • Vitamin B12 deficiency may lead to serious health issues in both infants and adults. A simple analytical method involving sample pretreatment with enzyme, followed by cyanide addition under acidic conditions; separation on an immunoaffinity column; and high-performance liquid chromatography (HPLC) was developed for the rapid detection and quantitation of vitamin B12 in powdered milk. Detection limit and powdered milk recovery were determined by quantitative analysis. The limits of detection and quantitation were 2.71 and 8.21 ㎍/L, respectively. Relative standard deviations of the intra-day and inter-day precisions varied in the ranges of 0.98%-5.31% and 2.16%-3.90%, respectively. Recovery of the analysis varied in the range of 83.41%-106.57%, suggesting that the values were acceptable. Additionally, vitamin B12 content and recovery in SRM 1849a were 54.10 ㎍/kg and 112.24%, respectively. Our results suggested that the analytical method, including the sample pretreatment step, was valid. This analytical method can be implemented in many laboratory-scale experiments that seek to save time and labor. Therefore, this study shows that immunoaffinity-HPLC/ultraviolet is an acceptable technique for constructing a reliable database on vitamin B12 in powdered milk containing starch as well as protein and/or fat in high amounts.

The removal of saw marks on diamond wire-sawn single crystalline silicon wafers

  • The diamond wire sawing method to produce silicon wafers for the photovoltaic application is still a new and highly investigated wafering technology. This technology, featured as the higher productivity, lower wear of the wire, and easier recycling of the coolant, is expected to become the mainstream technique for slicing the silicon crystals. However, the saw marks on the wafer surface have to be investigated and improved. This paper discusses the removal of saw marks on diamond wire-sawn single crystalline silicon wafer. With a pretreatment step using tetramethyl ammonium hydroxide ($(CH_3)_4NOH$, TMAH) and conventional texturing process with KOH solution (1 % KOH, 8 % IPA, and DI water), the saw marks on the surface of the diamond wire-sawn silicon wafers can be effectively removed and they are invisible to naked eyes completely.

  • The influence of phenobarbital (PB) pretreatment (75 mg/kg/day, i.p. for 4 days) on the metabolite kinetics of diltiazem (DTZ) was studied in rats in order to elucidate the effect of esterase induced by PB on the formation of DTZ to desacetyldiltiazem (DAD), DAD was injected via portal vein (3 mg/kg) to the control and PB-pretreated rats, The intrinsic hepatic clearance of DAD was significantly increased by PB pretreatment and the absolute bioavailability of DAD was significantly decreased in the PB-pretreated rats. According to the hepatic biotransformation model of DTZ, the fraction of systemic clearance of DTZ which forms DAD $(G_{mi})$ was different from that of DTZ which furnishes the available DAD to the systemic circulation $(F_{mi})$ in control rats. This result shows that DTZ was suspected of the sequential hepatic first-pass metabolism. On the other hand, PB pretreatment enhanced the $G_{mi}$ value of DTZ by 44%. It may be concluded that the deacetylation of DTZ to DAD in rats is increased by the esterase induced by PB but the transfer rate of DAD immediately formed from DTZ into systemic circulation is not affected by PB due to the 27% decreased absolute bioavailability of DAD resulting from PB pretreatment.
