Causes of Nitrogen Loss during Animal Manure Analysis (가축분의 정량과정에서 생기는 질소 손실에 대한 여러 원인)

  • Since nitrogen(N) is a volatile compound affected by many environmental factors, determining the N content of manure tends to be difficult. Upon arrival in the laboratory, the manure should be moist and refrigerated. Manure samples will have variable N contents due to drying temperature, and the presence of soil in the sample will affect N content. Acidification of the sample prevents ammonia volatilization and should be done before drying. It is recommended that manure samples be pretreated with a strong oxidizing agent, KMnO$_4$, followed by digestion under reduced conditions (reduced Fe-$H_{2}$ $SO_{4}$ ), which achieves a complete recovery of both $NO_{3}$ -N and $NO_{2}$ -N without a low recovery of $NH_{4}$ -N, resulting in a more accurate determination of N content. Accuracy of results for N content determined by recently developed rapid analysis techniques in the field should be tested by comparison with results obtained at laboratories using approved standard methods. Most commonly, the Kjeldahl system is used to determine manure N content. More research is needed on the effects of species, breed, age and individuals on the nutrient contents of manure. The procedures for manure sampling on the farm, shipping and handling of the sample until it reaches the laboratory, and the methods of sampling of the manure at the laboratory must be studied. Development of animal agricultural laboratories where feed, manure, soil, and water are all analyzed by appropriate specialists is needed.

Anatomical, Chemical, and Topochemical Characteristics of Transgemic Poplar Down-regulated with O-methyltransferase

  • The present work was undertaken to investigate the anatomical and chemical characteristics of transgenic poplar down-regulated with antisense OMT gene. Also the distribution of lignin in transgenic poplar trees was investigated at cellular level. No visible abnormal phenotype was observed in the fibers and vessel elements of transgenic poplar. Any marked differences in the staining intensities of Wiesner and Mäule color reaction were not identified in the transgenic poplar. TEM micrographs did not show any staining intensities in the cell walls stained with KMnO4. Interestingly, the UV spectroscopy of semi-thin sections exhibited a distinct decrease of lignin absorption at 280 nm in the vessel walls, indicating transgenic poplar wood with lower amount of guaiacyl lignin in vessel elements. Chemical composition of antisense OMT poplar was almost identical to that of wild-type poplar. Klason lignin content of transgenic poplar did not show any significant difference from that of the controls. The solid state NMR spectra revealed the transgenic poplar with only slightly more syringyl lignin than the control. The present work showed that antisense OMT gene constructed in the poplar was not enough to reduce the overall content of Klason lignin, and suggested that the expression of transformation was confined to vessel walls.

Effect of Gas Absorbents on Quality Attributes and Respiration Characteristics of Mature-Green Mume (Prunus mume Sieb. et Zucc) Fruits during Storage at Ambient Temperature (가스흡착제 처리가 상온 유통 청매실의 품질 및 호흡특성에 미치는 영향)

  • During storage at $25^{\circ}C$, the effect of gas absorbents, such as carbon dioxide scavenger, ethylene absorber, and their combinations, on respiration characteristics and quality attributes of mature-green Mume fruits packaged in $30\;{\mu}m$ low density polyethylene (LDPE) film was examined. Changes in quality attributes of the fruits were observed in terms of weight loss, titratable acidity, pH, fish firmness, color, water-soluble solid, and chlorophyll contents. In the presence of ethylene absorber $(KMnO_4)$, the physiological injury was remarkably suppressed, and there was no significant injury in Mume fruits at $25^{\circ}C$ for 10 days. Yellowing and softening were also noticeably reduced by the combination of plastic film packaging and inclusion of ethylene absorber. The respiration rate was slower in fruits sealed with ethylene absorber than in those with absorbent-free packaging. Using ethylene absorber, levels of oxygen and carbon dioxide were maintained at 2-3 and 7-8%, respectively, during storage at $25^{\circ}C$ for 10 days. The addition of carbon dioxide scavenger $(Ca(OH)_2)$, negatively affected the quality attributes and respiration characteristics of the fruits. Overall results showed that ethylene removal by gas absorbent in the film packages significantly prolonged the shelf life of the fruits at ambient temperature.

A Study on the Diatomaceous Earth Filtration of Settling Basin Effluent (정수장 침전지 유출수의 규조토 여과에 관한 연구)

  • The objective of this investigation was to evaluate applicability of precoat filtration that can be substituted for rapid sand filter of conventional water treatment system(CWTS). Precoat filter used in this experiment are candle filter. Element disk of candle are pore size $10{\mu}m(R),\;20{\mu}m(B)$ And diatomaceous earth are cake pore size $3.5{\mu}m$(Standard Super- Cel; A), $7{\mu}m$(Hyflo Super-Cel; B) and $17{\mu}m$(Celite 545RV; C). $2kg/m^2$ diatomaceous earth is used for precoating, it coated candle in $5{\sim}6mm$ thickness. 1. Al adsorption dosages by diatomaceous earth used in experimental we Hyflo Super-Cel 0.843mg/g, Standard Super-Cel 0.782 mg/g and Celite 545RV 0.766 mg/g. 2. Filtrate of precoat filter during 60min are R-C combination 20.7($m^3/m^2$)>B-C 18.3($m^3/m^2$)>B-B 15.0($m^3/m^2$)> R-B 12.9($m^3/m^2$)> R-A 11,093($l/m^2$). 3. Water quality of precoat filter effluent are thus. $KMnO_4$ consumption are $1.10{\sim}2.20mg/l$, removal rate are $30.9{\sim}65.6\%$. They are R-A 1.10(mg/l)(removal rate $65.6\%$). R-C(2.20 mg/l)(removal rate $30.9\%$). 4. $Al^{3+}$ are not detected with all combination, removal rate $100\%$. 5. Considering water quality and flux, continued running time of R-A combination is 7 hr. Accumulated filtrate are $74.4 m^3/m^2$, average flux is $177.2 l/m^2{\cdot}min$. And filtrate per diatomaceous earth 1g are 37.2 l. 6. R-A effluent's water quality are $KMnO_4$ Consumption 1.10(mg/l), DOC 1.161 mg/1, Al 0.0 mg/1, $UV_{254}$ 0.016/cm, Turbidity 0.1(NTU). R-A combination is suitable to precoat filtration for the settling basin effluent treatment.

Determining the Safer Thickness of the Epoxy Coating on Wooden Utensils (식품 안전성 확보를 위한 목재 식기용 에폭시 코팅의 두께 결정)

  • Overall migration through epoxy layer coated wood was investigated to estimate the coating thickness satisfying the regulatory limit. As an index of overall migration, KMnO$_4$ oxidizable extractives by the food simulant water solution was used. Migration pattern in interest range could described by a simple diffusion model and the temperature dependence of the permeability index could be explained by Arrhenius equation. The thickness of epoxy coating greater than 0.004 mm was analyzed to be required for satisfying the regulatory guideline.

A Studyd on Adsorptive Properties of Activated Carbons Produced from Rice-Straw

  • Activated carbons from rice-straw can be used as an adsorbents for the purification of water were prepared and evaluated. The adsorptive capacities of activated carbons were measured by iodine, potassium permangante, phenol and metals. It was observed by electron microscope (SEM) and IR spectrum that organic components in the rice-straw and its carbonization product were disappeared. Slit-shaped and porousstructures were formed by activation. There was no relationship between temperature and adsorption of iodine but adsorption of potassium permanganate increased as temperature rose. The adsorption of the phenol was greater than 99%. The adsorption data of phenol at $25^\circ{C}$ obeyed the Freundlich's isotherm. Various metals except sodium were not removed by activated carbon.

A Study on the Removal of Organics and Disinfection Effect in Sand Filter Using Nano Silver Sand (은나노 모래를 이용한 모래여과에서 유기물질 제거 및 소독 효과에 관한 연구)

  • In this study, novel nano silver sand filtration method was compared with UV treatment and normal sand filtration method through filtering treated water from sewage treatment plant. As a result, $BOD_5$ removal rate of nano silver sand filtration showed higher approximately 31% and 23%, comparing with UV treatment and sand filtration. Moreover, $KMnO_4$ removal rate of nano silver sand was about 6.6 and 2.8 times higher than other two methods. In addition, it showed better for removing SS and total coliform, comparing with others. Also, there is no bacteria on nano silver sand after experiments. Therefore, nano silver sand filtration will be effective for advanced water treatment.

Aspergillus sp. BY-54가 생산하는 Dextranase의 정제 및 특성

  • Aspergillus sp. BY-54 which produced a strong dextran hydrolyzing enzyme was isolated from soil. Using this strain, the optimal cultural conditions, enzyme purification and characterization were studied. The results are as follows : The optimal concentration of dextran as carbon source was l%. and the optimum temperature and the initial pH for enzyme production was 3$0^{\circ}C$, and 7.0, respectively. Dextranase was purified by DEAE-cellulose column chromatography with a linear gradient increase in NaCl. Km value of dextranase was 0.222%, and several glucans containing various types of glucosidic linkages such as DEAE-sephadex, CM-sephadex and sephadex G-100 were almost digested to a large extent with this dextranase. The enzyme was strongly inhibited by sodium fluoride, KMnO4 and p-CMB, while KCN caused 20% of activation.

Mineral Products and Characteristics of Phase Transformation after Hydrothermal Treatment according to the Synthetic Method and Cation Combination during Birnessite Synthesis (버네사이트 합성 시 합성 방법 및 양이온 조건에 따른 생성 광물 및 열수처리 후 상전이 특성)

  • The birnessite (7Å manganate, δ-MnO2) which is a manganese oxide and comprises manganese nodules, is a major manganese mineral on the earth surface and a precursor in the synthesis of todorokite. In this study birnessite was synthesized by three different methods: Feng et al. (2004) and Luo et al. (1998) based on redox reaction and Ma et al. (1999) based on reduction reaction. 12 birnessite samples were synthesized by different combinations of Na+ and K+ cations based on the base (OH-) and permanganate (MnO4-) reagents in the synthesis. The mineral compositions of synthesized birnessite were identified by XRD, and the two cation ratio in the mineral was measured by ICP. The products obtained after hydrothermal treatment of Mg-buserite, by the precursor of birnessite, was examined by XRD, and then phase transition to todorokite and their characteristics were compared. Our results show that the byproducts and the characteristics of phase transition by each synthetic method have different trends. Hausmannite (γ-Mn3O4) and feitknechtite (β-MnOOH) were formed by both methods in the redox reaction mechanism. By Feng et al. (2004)'s method, manganite (γ-MnOOH) phase only appeared when cation was predominantly Na+. Two birnessite samples synthesized by redox reaction mechanism showed phase transition to todorokite (10Å manganate, OMS-1) when both NaOH and KMnO4 were used together. However, single-phase birnessite was formed by Ma et al. (1999)'s method, and phase transition was confirmed only for the sample when the cation was only composed of Na+.

  • In order to develope the in vitro short term screen-ing method for carcinogen, we studied a purification method for thymine glycol in oxidaized DNA. Thymine glycol (5,6-dihydroxy-5, 6-dihydrothymine) is the major stable radiolysis poduct in thymine by chemical oxidants and ionzing radiation and it is a useful biomarker among oxidized DNA adducts, related with carcinogenests. Standard thymine glycol was prepared by oxidation of 〔$^3$H〕 thymine with KMnO$_4$ followed by purification with HPLC-LSC system and it was assayed by TLC and gas chromatography-MSD. 〔$^3$H〕 DMA adducts was isolated from E. coli (wild type ) treated with oxidative agents such as benzo(a)pyrene, adriamycin, aflatoxin B$_1$ and KBrO$_3$. These oxidative agents generated free radicals in cells by oxidative metabolism. As a result, thymine glycol was produced in cultured E. coli by four chemicals. This result shows that this methodology should be useful tool in screening oxidative carcinogen.
