단백질인산화효소 C 활성화제로 유도된 기도 뮤신 생성 및 유전자 발현과 점액 과분비 모델동물에 대한 수종(數種) 방제의 영향 (Effects of Several Oriental Medicines on Protein Kinase C Activator-Induced Production and Gene Expression of Airway Mucin and Animal Model for Airway Mucus Hypersecretion)

  • The author investigated whether Chwiyeon-tang(PC), Haengso-tang(PH), Jawanchihyo-san(PJ) and Gamisocheongryong-tang(PS) significantly affect both PMA-induced mucin production and MUC5AC gene expression in airway epithelial cells and sulfur-dioxide-induced airway goblet cell hyperplasia and mucus hypersecretion animal model using rat. Possible cytotoxicity of each herbal medicine was assessed by measuring the survival and proliferation rate of NCI-H292 cells. Confluent NCI-H292 cells were pretreated for 30 min in the presence of PC, PH, PJ and PS, respectively, and treated with PMA(10 $ng/m{\ell}$), to assess the effect of each herbal medicine on PMA-induced mucin production by enzyme-linked immunosorbent assay(ELISA). Effects of each herbal medicine on PMA-induced MUC5AC gene expression from the same cells were investigated. Also, hypersecretion of airway mucus and goblet cell hyperplasia were induced by exposure of rats to $SO_2$ during 3 weeks. Effects of orally-administered PC, PH, PJ and PS during 1 week on intraepithelial mucosubstances and hyperplasia of goblet cells were examined using histological analysis after staining the epithelial tissue with PAS-alcian blue. (1) PC, PJ, PS and PH did not show significant effects on the survival and proliferation of NCI-H292 cells ; (2) PC, PJ and PS significantly decreased PMA-induced mucin production from NCI-H292 cells ; (3) PC, PJ and PS significantly inhibit the expression levels of PMA-induced MUC5AC gene in NCI-H292 cells ; (4) Among PC, PJ, PS and PH, only PS decreased $SO_2$-induced hyperplasia of airway goblet cells and intraepithelial mucosubstances. This result suggests that PC, PJ and PS can not only affect the production of mucin but also affect the expression of mucin gene and this can explain, at least in part, the traditional use of PC, PJ and PS for controlling airway diseases showing hypersecretion of mucus in oriental medicine.

The Role of Excipients in Iontophoretic Drug Delivery: In vitro Iontophoresis of Isopropamide and Pyridostigmine through Rat Skin and Effect of Ion-pair Formation with Organic Anions

  • The iontophoretic delivery across rat skin of quaternary ammonium salts (isopropamide: ISP, pyridostigmine: PS), which are positively charged over a wide pH range, was measured ill vitro. The study showed that: (a) iontophoresis significantly enhanced delivery of ISP and PS compared to respective passive transport; (b) delivery of ISP and PS was directly proportional to the applied continuous direct current density over the range of $0-0.69\;mA/cm^2;$ (c) delivery of ISP and PS was also proportional to the drug concentration in the donor compartment over the range of $0-2{\time}l0^{-2}M:$ (d) sodium ion in the donor compartment inhibited the drug transport possibly due to decreasing the electric transference number of the drug; (e) delivery of ISP and PS increased as the pH of the donor solution increased over the pH range 2-7 suggesting permselective nature of the epidermis, and inhibition of the transference number of the drugs by hydronium ion; (f) some organic anions such as taurodeoxycholate, salicylate and benzoate which form lipophilic ion-pair complexes with ISP inhibited the delivery of ISP. The degree of inhibition by the organic anions was linearly proportional to the extraction coefficient $(K_e)$ of ISP from the partition system with each counteranion between phosphate buffer (pH 7.4) and n-octanol. For PS, however, taurodeoxycholate, but not salicylate and benzoate inhibited the iontophoretic delivery. It suggests that not only sodium ion and hydronium ion but also the counteranions which form lipophilic ion-pairs with quaternary ammonium drugs are not favorable components in formulating the donor solution of the drugs to achieve an effective iontophoretic delivery.

토끼에 있어서의 Physostigmine의 혈압상승작용 (Pressor Action of Physostigmine in the Rabbit)

  • 흰쥐, 개, 고양이의 뇌내의 머스커린수용체에 작용함이 알려져 있는 physostigmine(PS)의 동맥혈압에 미치는 효과를 urethane마취토끼에서 조사하였다. 정맥내 (iv) PS $25{\sim}250{\mu}g/kg$은 혈압변동을 일으키지 않았다. 그러나 토끼를 chlorisondamine(CS), hexamethonium, 뇌실내 (icv) clonidine, icv xylazine, icv reserpine으로 처리후 또는 척수이단후에는 승압반응을 일으켰다. CS처리토끼의 iv PS승압반응은 prazosin또는 pirenzepine처리후에는 현저히 약화되었다. Iv PS는 CS처리나 척수이단토끼에서 일어나는 McN-A-343의 승압효과를 억제하였고 또 McN-A-343주입시에는 iv PS는 승압을 일으키지 않았다. DMPP의 승압효과는 iv PS의 영향을 받지 않았다. Icv PS $12{\sim}100{\mu}g/kg$은 승압반응을 일으켰고 이는 CS처리로 강화되었다. 이 승압효과는 완전치는 않으나 prazosin 또는 pirenzepine으로 억제되었다. Angiotensin II 길항약인 $(Sar^{1},\;Ala^{8})-angiotensin$ II와 prazosin또는 pirenzepine으로 토끼를 처리할때는 icv PS승압효과는 거의 볼 수 없었다. 그러나 이 angiotensin II 길항약은 prazosin, pirenzepine의 iv PS승압반응에 대한 억제효과는 항진시키지 않았다. Icv pirenzepine은 icv PS승압반응은 차단하였으나, iv PS승압효과에는 영향을 미치지 않았다. 본실험성적은 CS처리 및 척수이단토끼에서 볼 수 있는 iv PS승압은 교감신경절의 머스커린수용체의 흥분으로 일어나고, icv PS승압은 뇌내의 머스커린수용체의 흥분으로 교감신경계 및 angiotensin계의 활성도가 높아져서 일어남을 가리키고 있다. 또한 토끼에서는 교감신경절니코틴수용체차단, 교감신경절에 미치는 중추 교감신경의 지배력의 감소 또는 척수이단등으로 교감신경절 머스커린수응체의 감수성이 바꾸어지지 않은한 iv PS는 승압반응을 일으키지 못함을 시사하고 있다.

중금속 검지를 위한 디티존 기능화된 폴리스티렌 제조 (Preparation of Dithizone Functionalized Polystyrene for Detecting Heavy Metal Ion)

  • 색센서는 액상 내 존재하는 특정 물질에 대하여 선택적인 색상변화를 통하여 대상물질을 검지할 수 있다. 기존 유기염료를 이용한 색센서는 단일종의 중금속 이온만을 검지한다는 단점이 있지만, 디티존(dithizone, DTZ)은 최소 5개 이상의 중금속 이온을 서로 다른 색상 변화로 검지해 낼 수 있다는 장점이 있다. 즉 다중검지용 색센서 물질로 사용할 수 있다. 그러나 디티존은 액상에서 주로 사용되어서 종이나 분말형태로 제조된 사례가 없어서 활용범위가 제한적이다. 이에 본 연구에서는 폴리스티렌(polystyrene, PS)과 디티존을 결합시켜 DTZ/PS 펠렛을 제조하여 10가지 중금속 이온에 대한 선택적인 색상변화를 관찰하였다. 제조한 DTZ/PS 펠렛에 대하여 10 ppm의 중금속 이온의 노출 결과, 수은과 코발트가 뚜렷한 색상 변화를 보였으며 선택적인 중금속 이온 검지에 활용할 수 있음을 확인하였다.

Effect of Porcine Serum as Macromolecule on the Meiotic Maturation and Embryonic Development of Porcine Oocytes

  • This study was conducted to establish an in vitro maturation (IVM) system by selection of efficient macromolecule in the porcine in vitro production (IVP) technology. To choose the efficient macromolecules in the development of porcine embryos, the effects of 3 kinds of macromolecules (porcine serum; PS, porcine follicular fluid; pFF, and polyvinyl alcohol; PVA) supplemented in IVM media on the maturation, cleavage, and development rates to blastocyst of parthenogenetic activation (PA) and in vitro fertilization (IVF) embryos were examined. The maturation rates of porcine oocytes in media supplemented with PS were significantly higher than those with pFF and PVA (92.4% vs. 85.4%, 77.1%; p<0.05). In the cleavage and development to blastocyst rates, supplement with PS or pFF in the IVM media was more effective than PA. However, there were no significant differences in cleavage and development to blastocyst between PS and pFF group. From the results of this study, it was demonstrated that PS was optimal macromolecule in the porcine IVM media.

비닐 케톤계 고분자와 폴리스티렌 블렌드의 광분해성 (Photodegradability of Blends of Polystyrene and Vinyl Ketone Polymers)

  • 카보닐기를 측쇄에 가지는 폴리페닐비닐케톤(PPVK)과 폴리스티렌(PS)을 블렌드한 후 이 블렌드에 자외선이 조사되었을 때 자외선 조사시간에 따른 광분해성 변화에 대해 연구하였다. 또한 PPVK의 카보닐기중 일부가 수소화된 수소화 폴리페닐비닐케톤(PPVK(H))도 폴리스티렌과 블렌드한 후 광분해성을 조사하여 광분해에 있어서의 카보닐기의 역할에 대해 연구하였다. PPVK의 페닐기가 메틸기로 대치된 폴리메틸비닐케톤(PMVK)도 폴리스티렌과 블렌드된 후 광분해성이 조사되었고 스티렌과 메틸비닐케톤(MVK)을 공중합한 폴리스티렌메틸비닐케톤(SMVK)에 대해서도 광분해성을 측정하여 PS/PMVK 블렌드와 비교하여 보았다. PPVK 또는 PPVK(H)를 PS와 블렌드한 경우의 광분해성은 PPVK를 블렌드한 경우가 PPVK(H)를 블렌드한 경우보다 더 높은 것으로 나타났다. 순수한 PPVK가 PPVK(H)보다 광분해성이 높게 나타난 것으로부터 이는 자연스러운 결과라고 할 수 있겠으며 또한 PPVK(H)는 PPVK의 카보닐기의 일부가 수소화된 고분자라는 점을 감안할 때 이 실험 결과로부터 카보닐기가 광분해에 참여한다는 것을 확인할 수 있었다. 또한 PMVK를 PS에 블렌드 하였을때의 광분해성은 PPVK를 PS에 블렌드한 경우보다 높게 나타났으며 따라서 PS의 광분해를 위하여 비닐케톤 폴리머를 PS에 블렌드할 경우에는 PMVK가 가장 효과적이고 다음으로 PPVK, PPVK(H)의 순서인 것으로 관찰되었다. 또한 스티렌과 메틸비닐케톤을 공중합시킨 SMVK가 PS와 PMVK 블렌드보다 더 높은 광분해성을 보여주어 비닐케톤 폴리머를 PS와 블렌드 시키는 것 보다는 스티렌과 직접 공중합 시키는 것이 더 효과적인 것으로 나타났다.

Comparison of Blue Luminescence Between Spark-processed Photoluminescian Silicon and Ambient Air Aged Anodically Etched Porous Silicon

  • Ambient air aged anodically etched porous silicon (PS) and spark-processed silicon (sp-Si) show interesting similarities and dissimilarities in some of their luminescence-related properties. Among these similarties are: (1) the photoluminescence (PL) peak maximum in the blue/violet (410 nm);(2) the blue/violet PL peak positions are essentially unchanged with temperature ; (3) PL decay times in the nanosecond region which are independent of the detection wavelength, which is much faster in decay times compared to that of observed decay time in $SiO_2$.Among the dissimilarities are: (1) the PL intensity of blue/violet luminescence, namely, the PL intensity of sp-Si is at least 2 orders of magnitude larger than that of an ambient air aged PS; (2) the blue/violet PL intensity of sp-Si is more stable than that of ambient air aged PS under UV illuminations; (3) FTIR spectra of sp-Si favor those modes, which involve silicon -oxygen bonds in $SiO_2$ stoichiometry, whereas ambient air aged PS can be considered as a nonstoichiometric oxide judging from the observed vibrational spectra.

Ab Initio Dispersion Polymerization of Styrene in the Presence of the Poly(methacrylic acid) Macro-RAFT Agent

  • Stable, spherical, polystyrene particles were synthesized in ab initio dispersion polymerization by using the poly(methacrylic acid)[PMAA] macro-RAFT agent. The presence of the PMAA macro-RAFT agent on the polystyrene (PS) particles was confirmed by NMR and FTIR spectroscopy. The PS particle size was influenced by the concentration of the RAFT agent and monomer due to the initial nucleation. When the concentration of the PMAA macro-RAFT agent was increased from 2 to 10 wt% relative to the monomer, the average particle size decreased from 2.31 to 1.36 ${\mu}m$, the conversion decreased from 93.3 to 88.9%, the weight-average molecular weight increased from 46,300 to 150,200 g $mol^{-1}$ and the PDI decreased from 2.79 to 1.94, respectively. In particular, the incorporation of 10 wt% of PMAA macro-RAFT agent produced monodisperse PS spheres of 1.36 ${\mu}m$ with a coefficient of variation (CV) of 6.44%. Thus, the PMAA macro-RAFT agent worked as a reactive steric stabilizer providing monodisperse, micron-sized, PS particles.

Influence of Supplementing Dairy Cows Grazing on Pasture with Feeds Rich in Linoleic Acid on Milk Fat Conjugated Linoleic Acid (CLA) Content

  • Three experiments were conducted to investigate the hypothesis that cows grazing on pasture produce the highest proportion of c-9 t-11 CLA in milk fat and no further increase can be achieved through supplementation of diets rich in linoleic acid, such as full-fat extruded soybeans or soybean oil. In experiment 1, 18 lactating Holstein cows were used in a randomized complete block design with measurements made from wk 4 to 6 of the experiment. In experiment 2, three cannulated lactating Holstein cows were used in a $3{\times}3$ Latin square design. Each period was 4 wk with measurements made in the final wk of each period. Cows in both experiments were assigned at random to treatments: a, conventional total mixed ration (TMR); b, pasture (PS); or c, PS supplemented with 2.5 kg/cow per day of full-fat extruded soybeans (PES). In both experiments, feed intake, milk yield, milk composition, and fatty acid profile of milk and blood serum were measured, along with fatty acid composition of bacteria harvested from rumen digesta in experiment 2. In experiment 3, 10 cows which had continuously grazed a pasture for six weeks were assigned to two groups, with one group (n = 5) on pasture diet alone (PS) and the other group (n = 5) supplemented with 452 g of soy oil/cow per day for 7 d (OIL). In experiment 1, cows in PS treatment produced 350% more c-9, t-11 CLA compared with cows in TMR treatment (1.70 vs. 0.5% of fat), with no further increase for cows in PES treatment (1.50% of fat). Serum c-9, t-11 CLA increased by 233% in PS treatment compared with TMR treatment (0.21 vs. 0.09% of fat) with no further increase for cows in PES treatment (0.18% of fat). In experiment 2, cows in PS treatment produced 300% more c-9 t-11 CLA in their milk fat compared with cows in TMR treatment (1.77 vs. 0.59% of fat), but no further increase for cows in PES treatment (1.84% of fat) was observed. Serum c-9, t-11 CLA increased by 250% for cows in PS treatment compared with cows in TMR treatment (0.27 vs. 0.11% of fat), with no further increase for cows in PES treatment (0.31% of fat). The c-9, t-11 CLA content of ruminal bacteria for cows in PS treatment was 200% or more of TMR treatment, but no further increase in bacterial c-9, t-11 CLA for cows in PES treatment was observed. Supplementation of soy oil in experiment 3 also did not increase the c-9 t-11 CLA content of milk fat compared with cows fed a full pasture diet (1.60 vs. 1.54% of fat). Based on these findings, it was concluded that supplementing with feeds rich in linoleic acid, such as full-fat extruded soybeans or an equivalent amount of soy oil, to cows grazing perennial ryegrass pasture may not increase milk fat c-9 t-11 CLA contents.

Controlled Synthesis of Spherical Polystyrene Beads and Their Template-Assisted Manual Assembly

  • Polystyrene beads (PS beads) with narrow size distribution were synthesized, and their diameter was controlled from 1.2 to $5{\mu}m$ by varying the injection rate of a styrene solution containing initiator and the concentration of reactant, such as initiator and capping material. The diameter of the PS beads increased with increasing in the injection rates and the initiator concentration or decreasing the capping material concentration. Then, we used the PS beads as building block, and organized them into a hexagonally close-packed monolayer on substrate with template-assisted manual assembly. We showed perfect hexagonally close-packed organization of the PS beads with various sizes in large-scale area. And we demonstrated the superiority of the dry manual assembly over the wet self-assembly in terms of simplicity, speed, perfect ordering, and large scale.
