The relationships of body mass index, waist-to-height ratio, and body fat percentage with blood pressure and its hemodynamic determinants in Korean adolescents: a school-based study

  • Purpose: Obesity is an important risk factor for hypertension in adolescents. We investigated the relationship of obesity-related indices (body mass index [BMI], waist-to-height ratio [WHR], and body fat percentage [%BF]) with blood pressure and the hemodynamic determinants of blood pressure in Korean adolescents. Methods: In 2008, 565 adolescents, aged 12-16 years, were examined. The %BF of the participants was measured by bioelectrical impedance analysis. Echocardiography and brachial artery pulse tracing were used to estimate the stroke volume (SV), cardiac output (CO), total vascular resistance (TVR), and total arterial compliance (TAC). Results: We noted that BMI, WHR, and %BF were positively correlated with systolic blood pressure (SBP) and diastolic blood pressure (DBP). The positive correlation between BMI and blood pressure (SBP and DBP) persisted after adjustment for WHR and %BF. However, after adjustment for BMI, the positive associations between blood pressure (SBP and DBP) and WHR as well as %BF, were not noted. With regard to the hemodynamic factors, BMI, but not WHR and %BF, was an independent positive factor correlated with SV and CO. TVR had an independent negative association with BMI; however, it was not associated with WHR or %BF. Moreover, we noted that BMI, WHR, and %BF did not affect TAC. Conclusion: In Korean adolescents, BMI had an independent positive correlation with SBP and DBP, possibly because of its effects on SV, CO, and TVR. WHR and %BF are believed to indirectly affect SBP and DBP through changes in BMI.

Inhibition of NF-ĸB, Bcl-2 and COX-2 Gene Expression by an Extract of Eruca sativa Seeds during Rat Mammary Gland Carcinogenesis

  • The effect of Eruca sativa seed extract (SE) on nuclear factor kappa B (NF-${\kappa}B$), cyclooxygenase-2 (COX-2) and B-cell lymphoma-2 (Bcl-2) gene expression levels was investigated in rat mammary gland carcinogenesis induced by 7,12 dimethylbenz(${\alpha}$)anthracene (DMBA). DMBA increased NF-${\kappa}B$, COX-2 and Bcl-2 gene expression levels and lipid peroxidation (LP), while, decreased glutathione-S-transferase (GST) and superoxide dismutase (SOD) activities and total antioxidant concentration (TAC) compared to the control group. After DMBA administration, SE treatment reduced NF-${\kappa}B$, COX-2 and Bcl-2 gene expression levels and LP. Hence, SE treatment reduced inflammation and cell proliferation, while increasing apoptosis, GST and SOD activities and TAC. Analysis revealed that SE has high concentrations of total flavonoids, triterpenoids, alkaloids and polyphenolic compounds such as gallic, chlorogenic, caffeic, 3,4-dicaffeoyl quinic, 3,5-dicaffeoyl quinic, tannic, cinnamic acids, catechin and phloridzin. These findings indicate that SE may be considered a promising natural product from cruciferous vegetables against breast cancer, especially given its high antioxidant properties.

Combination between Taxol-Encapsulated Liposomes and Eruca sativa Seed Extract Suppresses Mammary Tumors in Female Rats Induced by 7,12 Dimethylbenz(α)anthracene

  • Taxol (paclitaxel) is a powerful anti-cancer drug widely used against several types of malignant tumors. Because Taxol may exert several side effects, a variety of formulations have been developed. One of these features liposomes, regarded as one of the most promising drug carriers, biocompatible and best able to reduce drug toxicity without changing efficacy against tumor cells. Eruca sativa seed extract (SE) is considered a promising natural product from cruciferous vegetables against breast cancer, increasing chemotherapeutic and eliminating harmful side effects. The effects of Taxol-encapsulated liposomes (T) alone and in combination between Eruca sativa seed extract on nuclear factor kappa B (NF-${\kappa}B$), cyclooxygenase-2 (COX-2) and B-cell lymphoma-2 (Bcl-2) gene expression levels were investigated in rat mammary gland carcinogenesis induced by 7,12 dimethylbenz(${\alpha}$) anthracene (DMBA) using qRT-PCR. The results showed that DMBA increased NF-${\kappa}B$, COX-2 and Bcl-2 gene expression levels and lipid peroxidation (LP), while decreasing glutathione-S-transferase (GST) and superoxide dismutase (SOD) activities and total antioxidant concentration (TAC) compared to the control group. T and T-SE treatment reduced NF-${\kappa}B$, COX-2 and Bcl-2 gene expression levels and LP. Hence, T and T-SE treatment appeared to reduce inflammation and cell proliferation, while increasing apoptosis, GST and SOD activities and TAC.

Antioxidant and Anti-Inflammatory Effects of Various Cultivars of Kiwi Berry (Actinidia arguta) on Lipopolysaccharide-Stimulated RAW 264.7 Cells

  • The present study evaluated the total phenolic and flavonoid contents as well as total antioxidant capacity (TAC) of three cultivars of Actinidia arguta Planch. kiwi berries; cv. Mansoo (Mansoo), cv. Chiak (Chiak), and cv. Haeyeon (Haeyeon). In addition, the anti-inflammatory effects of the three cultivars of kiwi berries were investigated using a lipopolysaccharide (LPS)-stimulated RAW 264.7 murine macrophage cell line. Mansoo had the highest total phenolic content and TAC among the three cultivars, whereas Chiak had the highest total flavonoid content. The total antioxidant capacities of the kiwi berry extracts were more strongly correlated with total phenolic content than with total flavonoid content. The kiwi berry extracts suppressed the secretion of pro-inflammatory cytokines, including interleukin-6 and tumor necrosis factor-α, from LPS-stimulated RAW 264.7 cells. The release of nitrite, an indirect indicator of nitric oxide, was also ameliorated by pre-treatment with the kiwi berry extracts in a dose-dependent manner. Cellular-based measurements of antioxidant capacity exhibited that the kiwi berry extracts had cellular antioxidant capacities. Such cellular antioxidant effects are possibly attributed to their direct antioxidant capacity or to the inhibition of reactive oxygen species generation via anti-inflammatory effects. Our findings suggest that kiwi berries are potential antioxidant and anti-inflammatory agents.

동해안 대게 자망어업의 어획 성능 및 어획 능력 평가 (Assessment of fishing power and fishing capacity of the snow crab (Chionoecetes opilio) gillnet fishery in the East Sea)

  • Currently, environment-friendly and sustainable fisheries are one of the major issues among fisheries authorities. A variety of alternative management policies and projects are going on to enhance fisheries management systems and fishery resources such as a TAC management program and a VDS (Vessel Day Scheme) management scheme for distant water fishing nations in Western and Central Pacific Fisheries Commission (WCPFC) convention area. These kinds of efforts among fisheries management authorities are aimed at limiting fishing capacity or fishing power. In terms of fisheries management, Fishing capacity and fishing power have increased the importance of the impact on a fishery and level of the resources. Increased fishing capacity and fishing power have caused not only depletion of fisheries resource, but also additional fishing cost. therefore, there is a clear need for authorities to manage fishing capacity and fishing power. It is also help ensure the conservation and sustainability of the fishery resources. Because of lack of data, absolute fishing power is difficulty to measure. The notion of relative fishing power is frequently used. In this study, relative fishing power was assessed using Mastuda (1991) method for fishing power index. The raw data for assessment was based on fishermen's logbook data from sampled fishing vessels in coastal snow crab gillnet fishery. Data Envelopment Analysis (DEA) method was used to assess fishing capacity. DEA is a linear programming methodology to measure the efficiency of a set of entities called Decision-Making Units (DMUs). It was recommended by FAO for assessing capacity in fisheries.

Isolation and Characterization of Transcriptional Elements from Corynebacterium glutamicum

  • A promoter-probe shuttle vector pSK1Cat was constructed for the isolation of transcriptional signal sequences from Corynebacterium glutamicum. Besides conferring resistance to kanamycin in Escherichia coli and C. glutamicum, the vector carried a promoterless cat gene to confer resistance to chloramphenicol upon insertion of the appropriate transcriptional signals in the multiple cloning site. By utilizing the vector, a series of transcriptionally active fragments were isolated from the genome of C. glutamicum. The clones, ranging from 200 bp to 1 kb in size, were grouped into 3 classes of strong, medium, and weak, based on the chloramphenicol acetyltransferase (CAT) activity and sensitivity to the chloramphenicol of the clone-carrying C. glutamicum cells. C. glutamicum cells carrying the $P_{19}$ clone, a representative in the strong class, were able to grow on minimal agar plates containing over $40 mg/mell$ chloramphenicol, and showed CAT activity of 10 m㏖/mgㆍmin, performing slightly better than the cells carrying $P_{tac}$ , a strong E. coli promoter. Subcloning analysis of the $P_{19}$ clone identified a 180 bp intergenic fragment ($P_{180}$), which was located upstream of a gene encoding a hypothetical membrane protein. The expression conferred by $P_{180}$ was not affected by either the kinds of carbon sources or changes in temperature. These properties make the $P_{180}$ clone useful for the deregulated expression of biosynthetic genes in C. glutamicum during amino acid fermentation.

청국장 메탄올 추출물이 T98G 세포와 허혈성 뇌졸중 백서에 미치는 영향 (The Effects of Methanol Extract from Cheonggukjang in T98G Cells and Early Stage of Focal Ischemia Rodent Models)

  • This study was conducted to evaluate the neuroprotective effects of Cheonggukjang extract in in-vitro and in-vivo models. T98G-human glioblastoma cells were pretreated with various concentrations (1~10 mg/mL) of Cheonggukjang extract for 24 h and then exposed to $H_2O_2$ (1 mM) for 3 h. The neuroprotective effects of Cheonggukjang extract were measured using a CCK-8 kit assay, total antioxidant capacity (TAC) assay, reactive oxygen species (ROS) assay, and lactate dehydrogenase (LDH) release assay. The early stage focal ischemia rodent model was used as the in-vivo neurotoxicity model. Various concentrations (10~200 mg) of Cheonggukjang extract were administered to the animal models for 1 week. Peripheral blood was analyzed for glutathione peroxidase (GPx) expression by ELISA, and infarct volume reduction was analyzed by TTC staining. Cheonggukjang extract significantly (p<0.05) increased cell viability in T98G cells against $H_2O_2$ as well as against the induced neurotoxicity. Indeed, treatment with the Cheonggukjang extract induced a decrease in ROS and LDH expression and increased TAC significantly (p<0.05). However, Cheonggukjang extract did not induce a decrease in infarct volume or an increase in GPx expression in the in-vivo model. Despite the limitation in neuroprotection, Cheonggukjang extract may be useful for treating ROS injury.

플로팅 해상호텔의 부하계산에 관한 연구 (A Study on Load Estimation of Floating Hotel)

  • 경제적 여유와 함께 사람들의 요구가 다양화 되면서 해양 레저에 대한 관심이 증가하고 있다. 삼면이 바다인 우리나라는 바다를 이용한 해양 레저의 잠재력이 매우 높기 때문에 해양 리조트 개발이 증가할 것으로 예상된다. 그러나 이미 포화상태에 이르렀으며 또한 환경문제를 내포하고 있는 매립에 의한 개발은 상당한 어려움이 따를 것으로 예상된다. 이에 대한 대안으로 플로팅 건축에 의한 개발이 확대될 것으로 기대되지만 플로팅에 의한 해상호텔에 대한 연구는 아직 초보적인 단계에 있으며 이용객의 쾌적성에 직접적인 영향을 미치는 공조 설비 용량을 선정하기 위한 부하계산에 대한 연구는 전무하다고 할 수 있다. 본 연구에서는 육상의 일반적인 호텔과 선박의 공조부하 계산법을 비교 분석함으로써 해상호텔의 공조부하계산방법에 대해 검토한다.

삼치어종의 동태적 최적어업관리 (The Dynamic Optimal Fisheries Management for Spanish Mackerel)

  • 본 연구의 목적은 다수어업에 의해 어획되는 삼치어종에 대해 잉여생산모형을 이용하여 최대지속적어획량(YMSY) 및 최대지속적어획노력량(FMSY)을 추정한 후 현재 가치 해밀토니안 기법을 적용하여 삼치어종을 어획하는 다수어업의 이윤 극대화를 위한 최적어업관리 수준과 민감도 분석을 통한 삼치어종의 제도적 접근 방안을 제공함에 있다. 분석 내용으로, 우선, 다수어업의 어획노력량을 Gavaris의 일반선형모형을 적용하여 하나의 단위로 표준화한 후, 다양한 잉여생산모형 중 CYP 모형을 채택하여 정태적 자원평가를 시도하였다. 다음으로, 생물·기술적 계수와 경제적 매개변수를 가지고 현재가치 해밀토니안 기법을 이용하여 어업 이윤을 극대화하는 동태적 최적 수준을 추정하였다. 분석 결과, 우선, MSY 수준과 이윤이 극대화되는 수준에서 우리나라 삼치자원은 남획되고 있는 것으로 분석되었다. 다음으로, 경제적 주요 변수인 생산가격과 단위당 어업비용 변화에 따른 민감도 분석을 시도해본 결과, 생산가격 및 어업비용이 변화할 때 삼치어종의 자원량이 민감하게 반응하는 것으로 나타났다. 특히, 실제 삼치어종에 대한 어획노력량이 과도하게 투입되고 있어, 동 자원의 남획으로 인해 삼치어종을 어획하는 쌍끌이대형저인망어업, 대형선망어업, 대형트롤어업의 경영에도 비효율성을 야기하고 있는 것으로 분석되었다. 결론적으로, 본 연구에서는 우리나라 연근해 수산자원의 회복과 TAC 총량규제의 정부 정책의 확대에 맞추어 삼치어종도 TAC 대상 품종에 포함하여 체계적으로 자원을 관리해 나가길 제안하였다.

천마 침출약용주의 만성적인 섭취가 뇌 조직에서 항산화력에 미치는 영향 (Effects of Chronic Consumption of Liquor prepared with Gastrodiae rhizoma on Antioxidant Activity in Brain Tissue of Rats)

  • This experiment was conducted to verify whether or not chronic consumption of liquor prepared with Gastrodiae rhizoma exhibits antioxidant activity in brain tissue of rats. In the experiment, male Sprague-Dawley rats were given a liquid diet containing 6% (GA6), 18% (GA18), and 36% (GA36) of total calories as liquor for 6 weeks. The control rats (GAC6, GAC18, GAC36) received an isocaloric diet containing Soju instead of liquor. Caloric intakes and body weight gains of the 36% groups were significantly lower than those of the 6% and 18% groups (p<0.05), whereas alcohol intakes were significantly high in the 36% group (p<0.05). MDA contents of plasma and brain showed a positive correlation with alcohol intakes of the liquor and Soju consuming groups. Especially, contents of MDA were significantly higher in the 18% and 36% groups than the 6% group (p<0.05). SOD activities of serum and brain were significantly highest in the 18% group, and they tended to be higher in the liquor consuming groups compared with the control group. Catalase activity of serum showed no significant changes in the three groups consuming liquor, and catalase activity of brain tissue significantly increased in the GA18 group compared with the control group (p<0.05). In the liquor consuming groups, TAC level of serum tended to be higher in the GA36 group compared with the control group (p<0.05). However, TAC level of brain tissue showed no significant changes between the liquor consuming and control groups. These results show that as alcohol intake increased by chronic administration of liquor and Soju, brain tissue of rats was damaged more due to oxidative stress. However, a liquid diet containing 18% of total calories as liquor instead of Soju is expected to have a protective effect on brain damage induced by oxidative stress.
