사람 단핵구에서 결핵균에 의해 유도되는 CCL3 및 CCL4 발현에 대한 Phospholipase-Protein Kinase C-MEK-ERK 경로의 역할 분석 (The Phospholipase-Protein Kinase C-MEK-ERK Pathway is Essential in Mycobacteria-induced CCL3 and CCL4 Expression in Human Monocytes)

  • Background: Little information is available on the identification and characterization of the upstream regulators of the signal transduction cascades for Mycobacterium tuberculosis (M. tbc)-induced ERK 1/2 activation and chemokine expression. We investigated the signaling mechanisms involved in expression of CCL3 /MIP-1 and CCL4/MIP-1 in human primary monocytes infected with M. tbc. Methods: MAP kinase phosphorylation was determined using western blot analysis with specific primary antibodies (ERK 1/2, and phospho-ERK1/2), and the upstream signaling pathways were further investigated using specific inhibitors. Results: An avirulent strain, M. tbc H37Ra, induced greater and more sustained ERK 1/2 phosphorylation, and higher CCL3 and CCL4 production, than did M. tbc H37Rv. Specific inhibitors for mitogen-activated protein kinase (MAPK) kinase (MEK; U0126 and PD98059) significantly inhibited the expression of CCL3 and CCL4 in human monocytes. Mycobactetia-mediated expression of CCL3 and CCL4 was not inhibited by the Ras inhibitor manumycin A or the Raf-1 inhibitor GW 5074. On the other hand, phospholipase C (PLC) inhibitor (U73122) and protein kinase C (PKC)specific inhibitors ($G\ddot{o}6976$ and Ro31-8220) significantly reduced M. tbc-induced activation of ERK 1/2 and chemokine synthesis. Conclusion: These results are the first to demonstrate that the PLC-PKC-MEK-ERK, not the Ras-Raf-MEK-ERK, pathway is the major signaling pathway inducing M. tbc-mediated CCL3 and CCL4 expression in human primary monocytes.

Proteomic Analysis and the Antimetastatic Effect of N-(4methyl)phenyl-O-(4-methoxy) phenyl-thionocarbamate-Induced Apoptosis in Human Melanoma SK-MEL-28 cells

  • We employed human SK-MEL-28 cells as a model system to identify cellular proteins that accompany N-(4-methyl)phenyl-O-(4-methoxy)phenyl-thionocarbamate (MMTC)-induced apoptosis based on a proteomic approach. Cell viability tests revealed that SK-MEL-28 skin cancer cells underwent more cell death than normal HaCaT cells in a dose-dependent manner after treatment with MMTC. Two-dimensional electrophoresis in conjunction with matrixassisted laser desorption/ionization-time of flight (MALDI-TOF) mass spectrometry analysis or computer matching with a protein database further revealed that the MMTC-induced apoptosis is accompanied by increased levels of caspase-1, checkpoint suppressor-1, caspase-4, NF-kB inhibitor, AP-2, c-Jun-N-terminal kinase, melanoma inhibitor, granzyme K, G1/S specific cyclin D3, cystein rich protein, Ras-related protein Rab-37 or Ras-related protein Rab-13, and reduced levels of EMS (oncogene), ATP synthase, tyrosine-phosphatase, Cdc25c, 14-3-3 protein or specific structure of nuclear receptor. The migration suppressing effect of MMTC on SK-MEL-28 cell was tested. MMTC suppressed the metastasis of SK-MEL-8 cells. It was also identified that MMTC had little angiogenic effect because it did not suppress the proliferation of HUVEC cell line. These results suggest that MMTC is a novel chemotherapeutic and metastatic agents against the SK-MEL-28 human melanoma cell line.

홍수위 해석을 위한 미측정 하천 단면 추정 (Estimating Ungauged River Section for Flood Stage Analysis)

  • The objective of this study was to develop the simple method to estimate ungauged river section for flood stage analysis. Damage prediction should be prioritized using hydrological modeling to reduce flood risk. Mostly, the geographical data using hydrological modeling depends on national river cross-section survey. However because of the lack of measured data, it is difficult to apply to many local streams or small watersheds. For this reason, this study suggest the method to estimate unguaged river cross-section. Simple regression equations were derived and used to estimate river cross-section by analyzing the correlation between the river cross-sectional characteristics (width, height and area). The estimated cross-sections were used to simulate flood level by HEC-RAS (Hydrologic Engineering Center's River Analysis System). The applicability of this method was verified by comparing simulated flood level between measured and estimated cross-section. The water surface elevation of the flood stage analysis was 6.56-7.24 m, 5.33-5.95 m and 6.12-6.75 m for measured cross section, for estimated cross section and for estimated cross section based on DEM elevation, respectively. Further study should consider other factors for more accurate flood stage analysis. This study might be used one of the guidelines to estimate ungauged river section for flood stage analysis.

Protein-protein Interaction Analysis of Bradykinin Receptor B2 with Bradykinin and Kallidin

  • Bradykinin receptor B2 (B2R) is a GPCR protein which binds with the inflammatory mediator hormone bradkynin. Kallidin, a decapeptide, also signals through this receptor. B2R is crucial in the cross-talk between renin-angiotensin system (RAS) and the kinin-kallikrein system (KKS) and in many processes including vasodilation, edema, smooth muscle spasm and pain fiber stimulation. Thus the structural study of the receptor becomes important. We have predicted the peptide structures of Bradykinin and Kallidin from their amino acid sequences and the structures were docked with the receptor structure. The results obtained from protein-protein docking could be helpful in studying the B2R structural features and in the pathophysiology in various diseases related to it.

  • Interaction between wood-destroying basidiomycetes and blue stain fungi were studied during conjoint cultivation on 2% malt extract agar. The ability of basidial cultures to decolourise dark pigments of ophiostomatoid fungi was the main characteristics estimated during this investigation. Antrodiella hoehnelii(S28/91), Bjerkandera fumosa(137), Gleophyllum odoratum(124), Trametes versicolor (B18/91) cultures were found to be the most active in decreasing dark colour of blue stain fungi colonies. The cultures were recommended for further development as agents of biopulping and control of blue stain fungi in wood chips.

  • Artificial neural networks (ANN) has been proven to be able to predict the compressive strength and elastic modulus of recycled aggregate concrete (RAC) made with recycled aggregates (RAs) from different sources. However, ANN is itself like a black box and the output from the model cannot generate an exact mathematical model that can be used for detailed analysis. So in this study, sensitivity analysis is conducted to further examine the influence of each selected factor on the output value of the models. This is not only conducive to the determination and selection of the more important factors affecting the results, but also can provide guidance for researchers in adjusting mix proportions appropriately when designing RAC based on the variation of these factors.

위성사진을 이용한 도시하천 홍수범람도 작성에 관한 연구 (A Study of Make Inundation Map Using Satellite Photograph at Urban River)

  • This study used to the satellite photograph and drew up a flood inundation map. To past used digital map and showed reflect the inundation map and change of the city was difficult. The acquisition of the satellite photograph is easy from internet Site of Korea and is updated quickly. So, used the satellite photograph from flood inundation drawing up of existing and reflected the change of the city. Drawing up method of the inundation map in compliance with the research which sees the reflection of the land use which is actual is possible. Results of HEC-RAS with depth and an inundation location to be show easily. But there is to a combination method of the satellite photographs and GIS data and more objective and the subject the development of the technique which has reached fixed quantity must be researched continuously judges.

  • 2,3,7,8-Tetrachlorodibenzo-p-dioxin(TCDD) is known to induce cytochrome p450 1A1 and to activate c-Src kinase and p21 Ras. This study examined the molecular interactions of adaptor proteins including Shc, Grb2, and Sos in rat primary hepatocytes and their relationship to the induction of CYP1A1 by TCDD. TCDD induced CYP1A1 level and EROD activity in a dose-dependent mode. Sos/Grb2 association isincreased by TCDDㅑㅜ a dose dependent mode. Tyrosine phosphorylated Shc, mainly p152, onloads to Grb2/Sos complex upon TCDD stimulation. The electrophoretic mobility shift of Sos is showed by TCDD. These results indicate that TCDD modulated the molecular interaction features of adaptor compoes proteins including Shc, Grb2, and Cnl in early signaling pathway of TCDD-mediated CYP 1A1 induction of rat primary hepatocyte.

Theoretical Structure Prediction of Bradykinin Receptor B2 Using Comparative Modeling

  • Bradykinin receptor B2, a GPCR protein, binds with the inflammatory mediator hormone bradkynin. Also, it is involved in many processes including vasodilation, edema, smooth muscle spasm and pain fiber stimulation. Hence, studuying the structural features of the receptor becomes important. But the unavailability of the three dimensional structure of the protein makes the analysis difficult. Hence we have performed the homology modelling of Bradykinin receptor B2 with 5 different templates. 25 different homology models were constructed. Two best models were selected based on the model validation. The developed models could be helpful in analysing the structural features of Bradykinin receptor B2 and in pathophysiology of various disorders related to them.

부등류해석을 이용한 QUAL2E 모형의 개선 (Improvement of QUAL2E Model using Nonuniform Flow Analysis)

  • Recently, as water pollution accidents in rivers have increased, there is an increased interest in water quality forecast with accurate simulation. QUAL2E model, widely used for water quality analysis, uses the same hydraulic characteristics, such as depth and velocity, in a reach. The flow of the river is changed by various hydraulic constructions or by topography in a real river channel. In this study, a hydraulic connection module is developed to consider flow variations of river channels in QUAL2E model. The module uses the simulations results of non-uniform flow of a 1-D hydraulic model such as DWOPER or HEC-RAS. The improved QUAL2E model with this module was applied to a downstream section of Paldang Dam on the Han River. The results show the variation of water quality very well in a reach where flowing vary abruptly, like the Jamsil submerged weir.
