Effects os Cold Drawing Ratio on the Hardness of Inconel 718 Wire (Inconel 718 선재의 경도에 미치는 냉간신선가공의영향)

  • Effect of cold drawing ratio and aging time on the hardness of lnconel 718 wire aged at 11l6K were investi¬gated by hardness measurement and scanning & transmission electron microscopy. Hardness which was 245Hv in as¬solution treated condition increased very rapidly to 450Hv as cold drawing ratio increased to 50%. The hardness in the early stage of aging was increased by the precipitation of $\gamma^{'}$ and $\gamma^{'}$ phases and after the peak hardness, the hardness was decreased by the transformation of $\gamma^{'}$ phase to $\delta$ phase. The time to reach peak hardness during aging appeared to be reduced with the increase of cold drawing ratio, and those times were 30, 10, and 5 minutes for 0, 30 and 50% cold drawn materials, respectively. For the 50% cold drawn material. $\gamma^{'}$ and $\gamma^{'}$ were precipitated by aging for 5 minutes at 1116K. The hardness in the same material was largely decreased under the initial hardness by the recrystallization.

Differential Expression of a Chimeric nos-npt II Gene in 9 Years Old Hybrid Poplars (Populus koreana x P. nigra)

  • The expression of a chimeric transgene (nos-npt II) has been examined in 9 years old transgenic poplars (Populus koreana x P. nigra) growing in a nursery. The expression of the gene in twenty six independentely transformed plants were examined by 1) enzyme (NPT II) assay, 2) RT-PCR, and 3) resistance to kanamycin. High NPT II activities in young leaves of all the transformed plants were found even without a selection pressure for antibiotics for 9 years. However, the activity varied with the positions of leaves in the stem in that young leaves showed higher activity than did mature tissues. When leaf segments were cultured in the presence of 150 mg/l kanamycin, only those from young leaves produced vigorously growing callus. However, as in the case of NPTII assay, the leaf segments from mature leaves did not form callus well on the media. RT-PCR with nptII specific primers also showed that amplification products were observed only when RNAs from young tissues were used. The total RNA gel showed that while RNA in young leaves are relatively stable and in a large quantity, those in old leaves were mostly degraded. All the above results suggest that the gene is transcriptionally active only in young tissue even though it is attached to a constituitive promoter. Therefore, the expression of foreign gene in poplar plants seemed to be affected by the metabolic state of the cells and thus vary greatly with the developmental stages and the age of tissue.

Manual Contouring Based Volumetric Evaluation for Colorectal Cancer with Liver Limited Metastases: A Comparison with RECIST

  • Background: To compare response evaluation criteria in solid tumours (RECIST) and volumetric evaluation (VE) for colorectal cancer with liver-limited metastasis. Patients and Methods: VE of liver metastases was performed by manual contouring before and after chemotherapy on 45 pairs of computed tomography (CT) images in 36 patients who suffered from metastatic colorectal cancer (mCRC) with liver metastasis only. Cohen kappa was used to compare the agreement between VE and RECIST. Pearson correlation was performed for their comparison after cubic root transformation of the aggregate tumor volumes. Logistic regression was done to identify clinical and radiographic factors to account for the difference which may be predictive in overall response (OR). Results: There were 16 partial response (PR), 23 stable disease (SD) and 6 progressive disease (PD) cases with VE, and 14 PR, 23 SD and 8 PD with RECIST. VE demonstrated good agreement with RECIST (${\chi}$=0.779). Discordant objective responses were noted in 6 pairs of comparisons (13.3%). Pearson correlation also showed excellent correlation between VE and RECIST ($r^2$=0.966, p<0.001). Subgroup analysis showed that VE was in slightly better agreement with RECIST for enlarging lesions than for shrinking lesions ($r^2$=0.935 and $r^2$=0.780 respectively). No factor was found predictive of the difference in OR between VE and RECIST. Conclusions: VE exhibited good agreement with RECIST. It might be more useful than RECIST in evaluation shrinking lesions in cases of numerous and conglomerate liver metastases.

Isolation of an Isocarbophos-Degrading Strain of Arthrobacter sp. scl-2 and Identification of the Degradation Pathway

  • Isocarbophos is a widely used organophosphorus insecticide that has caused environmental pollution in many areas. However, degradation of isocarbophos by pure cultures has not been extensively studied, and the degradation pathway has not been determined. In this paper, a highly effective isocarbophos-degrading strain, scl-2, was isolated from isocarbophos-polluted soil. The strain scl-2 was preliminarily identified as Arthrobacter sp. based on its morphological, physiological, and biochemical properties, as well as 16S rDNA analysis. The strain scl-2 could utilize isocarbophos as its sole source of carbon and phosphorus for growth. One hundred mg/l isocarbophos could be degraded to a non detectable level in 18 h by scl-2 in cell culture, and isofenphos-methyl, profenofos, and phosmet could also be degraded. During the degradation of isocarbophos, the metabolites isopropyl salicylate, salicylate, and gentisate were detected and identified based on MS/MS analysis and their retention times in HPLC. Transformation of gentisate to pyruvate and fumarate via maleylpyruvate and fumarylpyruvate was detected by assaying for the activities of gentisate 1,2-dioxygenase (GDO) and maleylpyruvate isomerase. Therefore, we have identified the degradation pathway of isocarbophos in Arthrobacter sp. scl-2 for the first time. This study highlights an important potential use of the strain scl-2 for the cleanup of environmental contamination by isocarbophos and presents a mechanism of isocarbophos metabolism.

Enhanced Production of Itaconic Acid through Development of Transformed Fungal Strains of Aspergillus terreus

  • Metabolic engineering with a high-yielding mutant, A. terreus AN37, was performed to enhance the production of itaconic acid (IA). Reportedly, the gene cluster for IA biosynthesis is composed of four genes: reg (regulator), mtt (mitochondrial transporter), cad (cis-aconitate decarboxylase), and mfs (membrane transporter). By overexpressing each gene of the IA gene cluster in A. terreus AN37 transformed by the restriction enzyme-mediated integration method, several transformants showing high productivity of IA were successfully obtained. One of the AN37/cad transformants could produce a very high amount of IA (75 g/l) in shake-flask cultivations, showing an average of 5% higher IA titer compared with the high-yielding control strain. Notably, in the case of the mfs transformants, a maximal increase of 18.3% in IA production was observed relative to the control strain under the identical fermentation conditions. Meanwhile, the overexpression of reg and mtt genes showed no significant improvements in IA production. In summary, the overexpressed cis-aconitate decarboxylase (CAD) and putative membrane transporter (MFS) appeared to have positive influences on the enhanced IA productivity of the respective transformant. The maximal increases of 13.6~18.3% in IA productivity of the transformed strains should be noted, since the parallel mother strain used in this study is indeed a very high-performance mutant that has been obtained through intensive rational screening programs in our laboratory.

  • The effect of stress relief heat treatment temperature and duration time on the microstructure, residual stress and Vickers hardness of additively manufactured Ti-6Al-4V alloy using laser powder bed fusion process was clarified. As a result of stress relief heat treatment for 240 minutes at 823 K and 60 minutes or more at 873 K, residual stress was decreased less than 30 MPa without grain growth and phase transformation which causes dimensional distortion and deterioration of mechanical properties. In addition, hardness was increased with increasing heat treatment temperature and duration time. It was deduced that the refinement of acicular martensitic α' phase due to the increasing duration time of isothermal heat treatment at 773~873 K, which was not detected by XRD and phase map analysis using SEM-EBSD, probably increases the hardness.

  • Escherichia Coli ornithine transcarbamylase is the enzyme which catalyzes the L-citrulline biosynthesis from L-ornithine and carbamyl phosphate. To facilitate the purification of enzyme which will be used for many biochemical studies such as structure and function relationships and catalytic mechanisms, the cloning and expression of E. coli argI gene for ornithine transcarbamylase was conducted. argI was amplified from genomic DNA of E. coli strain of $DH5{\alpha}$, by polymerization chain reaction (PCR) method. The amplified argI gene was ligated to the prokaryotic expression vector pKK223-3 and used for transformation of E. coli TB2 which was deficient of ornithine transcarbamylase. The over-produced enzyme by the tnansformant was purified by ammonium sulfate fractionation, heat denaturation and affinity chromatography. The result of SDS denaturation gel electrophoresis for the purified enzyme showed a single band of about 38 kDa of ornithine transcarbamylase. Kinetic data for the expressed enzyme gave almost the s?????? values as those of the wild type enzyme. The $k_{cat}$, of the enzyme was $1.0{\times}10^5min^{-1}$, and $K_ms$ for ornithine and carbamyl phosphate were 0.35 mM and 0.06 mM, respectively.

  • Many nucleoside diphosphate (NDP) kinases are ubiquitous enzymes responsible for the exchange of ${\gamma}$-phosphates between tri- and diphosphonucleosides. The catalytic Many nucleoside diphosphate (NDP) kinases are ubiquitous enzymes responsible for the exchange of ${\gamma}$-phosphates between tri- and diphosphonucleosides. The catalytic reaction follows a ping-pong mechanism in which the enzyme is transiently phosphorylated on a histidine residue conserved in all nucleoside diphosphate kinases. Beside their role in nucleotide synthesis, these enzymes present additional functions, possibly independent of catalysis, in processes such as differentiation, cell growth, tumor progression, metastasis and development. To clone murine nm23-M5, several expressed sequence tags (ESTs) of the GenBank data base, selected according to their homology to nm23-H5 cDNA, reconstituted a complete open reading frame (GenBank AF222750). To test whether murine NDPKs (1, 2, 3, 4, 5, and 6) can inhibit Bax-mediated toxicity in yeast, co-transformation was performed respectively. The yeast S.cerevisiae was transformed with a copy expression plasmid containing the histidine selection marker and expressing murine Bax under the control of a galactose-inducible promoter. Several clones were selected and found to be growth inhibited when Bax expression was induced with galactose. A representative clone was transformed again with a copy expression plasmid containing the tryptophane selection marker and expressing either murine Bcl-xL or NDPK under the control of a galactose-inducible promoter. Several subclones of the double-transformants were selected and characterized. The ability of Bcl-xL and NDPKs to suppress Bax-mediated toxicity was determined by growing yeast cells overnight in galactose media and spot-testing on galactose plates starting with an equal number of yeast cells as determined by taking the OD$_{600}$. Ten-fold serial dilutions were used in the spot-test. Plates were grown at 3$0^{\circ}C$ for 2-3 days. All murine NDPKs suppressed Bax dependent apoptosis. Futher study will be peformed whether Bax-toxicity inhibition was caused by NDP kinase activity or additional function.n.

Basic and Mechanical Properties by Film Type to Minimize the Sound Pressure Level of PTFE Laminated Vapor-permeable Water-repellent Fabrics (PTFE(Polytetrafluoroethylene) 라미네이팅 투습발수직물의 총음압 최소화를 위한 필름 타입 별 기본 특성과 역학 특성)

  • This study investigates the sound properties of fabric frictional sound (SPL, ${\Delta}L$, ${\Delta}f$) according to the film type of PTFE laminated vapor-permeable water-repellent fabrics in order to understand the relationship between SPL and the basic properties of fabrics such as layer, yarn type, and thickness of fiber. This study accesses their mechanical properties and determines how to control them to minimize SPL. Eight PTFE laminated water-repellent fabrics, composed of four different film types (A, B, C, D) and with two different fabrics, were used as test specimens. Frictional sounds generated at 1.21m/s were recorded by using a fabric sound generator and SPLs were analyzed through Fast Fourier Transformation (FFT). The mechanical properties of fabrics were measured by KES-FB. The SPL value was lowest at 74.4dB in film type A and highest as 85.5dB in type D. Based on ANOVA and post-hoc test, specimens were classified into less Loud Group (A, B) and Loud Group (C, D). It was shown that SPL was lower when 2 layer (instead of 3 layer), filament yarn than staple, and thin fiber than thick were used. In Group I, shearing properties (G, 2HG5), geometrical roughness (SMD), compressional properties (LC, RC) and weight (W) showed high correlation with SPL however, elongation (EM) and shear stiffness (G) did with SPL in Group II.

  • A fuzzy-ART(adaptive resonance theory) network for the PVC(premature ventricular contraction) classification using wavelet coefficient is designed. This network consists of the feature extraction and learning of the fuzzy-ART network. In the first step, we have detected the QRS from the ECG signal in order to set the threshold range for feature extraction and the detected QRS was divided into several frequency bands by wavelet transformation using Haar wavelet. Among the low-frequency bands, only the 6th coefficient(D6) are selected as the input feature. After that, the fuzzy-ART network for classification of the PVC is learned by using input feature which comprises of binary data converted by applying threshold to D6. The MIT/BIH database including the PVC is used for the evaluation. The designed fuzzy-ART network showed the PVC classification ratio of 96.52%.
