Improvement of the Biodegradability of Polyvinyl Alcohol by Radiation Treatment (방사선 처리에 의한 폴리비닐 알콜의 생분해도 개선)

  • Radiation treatment with gamma-rays and electron-beams was used to remove polyvinyl alcohol(PVA), one of the main components of dyeing wastewater. PVA was effectively decomposed by radiation treatment, thus the removal was near 100 % at an initial PVA concentration of 44 mg/L. However, total organic carbon(TOC) removal was less than 5 % due to lower transformation of PVA to $CO_2$. This directly indicates the radiation treatment alone is not appropriate for the complete decomposition of PVA. In this sense, the improvement of biodegradability($BOD_5/COD$) of PVA by radiation treatment was studied. Both gamma-ray and electron-beam treatments significantly increased the biodegradability of PVA by transforming non-biodegradable PVA to biodegradable by-products. This suggests radiation treatment, especially electron-beam treatment that showed better improvement of biodegradability, can be used as a pre-treatment of biological degradation process of PVA.

Effect of Temperature on Persistence of Recombinant Plasmid pCU103 in Different Waters

  • The recombinant plasmid of pCU103 constructed by cloning pcbCD genes in pBluescript SK(+) was studied for the effect of temperature on its persistence in different waters by the methods of electrophoresis, Southern hybridization, quantification, and transformation. The plasmid was very rapidly degraded out in non-sterile FW water without regards to water temperature, probably due to the effect of biochemical factor such as nucleases. The pCU103 was most persistent at 4$^{\circ}C$ in any water environments, moderately persistant at 15$^{\circ}C$ but least stable at 3$0^{\circ}C$ such results could be explained by the facts that hydrogen bonds in double-stranded plasmid DNAs become unstable and that nucleases are activated by increasing temperature. The intact structure of pCU1-3 was generally observed by gel electrophoresis under the conditions which the plasmid should be 2.0 ng/$\mu\textrm{l}$ or higher in concentration and that about 10$^2$ CFU/ml or more transformant cells should be recovered.

  • The transformational conditions of plasmids $pPL-\lambda$ and pAS 1 are as follows in E. coli, Ts-mutant M 5248 strain at $30^{\circ}C$. When the culture time was 2.5 hours of mid logarithemic phase, the cell concentration was $4.5\times10^7\;cells/ml$, the optical density was equal to 0.45 at 590 nm wave length, the transformational frequencies of plasmid$pPL-\lambda$ and pAS 1 had the highest values as $2\times10^{-6}\;and\;1.5\times10^{-6}\;and\;1.5\times10^{-6}$ and respectively. For $9\times10^6$ competent cells in $200{\mu}l$, the transformational frequency was as high as $4.4\times10^{-6}$ at 510 ng plasmid concentration. The competent cells treated with the mixture of calcium chloride and thymidine twice rates of transformation than those treated with calcium chloride. The ampicillin resistance of transformants was expressed in LB broth after 2 hours at $30^{\circ}C$.

Mineralization of Cattle Manure Compost at Various Soil Moisture Content (우분퇴비 시용후 토양수분 조절에 따른 질소 및 탄소의 전환)

  • To investigate the transformation characteristics of nitrogen and carbon from cow manure compost amended in soil under different moisture conditions, dynamics of nitrogen and carbon were determined periodically for 15 weeks of aerobic incubation at room temperature during July${\sim}$November, 1996. Cow manure compost matured with mixing saw dust was amended with the 4 ratios (0, 2, 4, 6%(wt/wt)) in Ap horizon soil, which collected from green house in Yesan, Chungnam. Moisture was controlled with 0.2, 0.3, 0.4, and 0.5 of mass water conte nt (${\theta}$m) to air dried soil, and water loss was compensated at every sampling. During incubation, soil pH was decreased continuously, that was caused by hydrogen generated from nitrification of ammonium nitrogen. And pH became higher with inclining cow manure compost amendment and water treatment, that meaned the increase of mineralization of organic-N to $NH_4\;^+-N$. Total nitrogen was reduced with increasing water content, but total carbon showed the contrast tendency with that of nitrogen. Therefore, C/N ratio slightly decreased in the low water condition (${\theta}$m 0.2) during incubation, but increased continuously in high water condition over ${\theta}$m 0.4. As a result, it was assumed that soil fertility is able to be reduced in the high water content over available water content. Nitrate transformation rate increased lasting in the low water content less than ${\theta}$m 0.3. Itdropped significantly in the first $2{\sim}3$ weeks of incubation over ${\theta}$m 0.4. In particular, nitrate was not detected in ${\theta}$m 0.5 of water content after the first $2{\sim}3$ weeks. In contrast, ammonium transformation was inclined with increasing water treatment. Nitrogen mineralization rate, which calculated with percentage ratio of (the sum of ex.$NH_4\;^+-N$ and $NO_3\;^--N$)/total nitrogen, was continuously increased in the low water content of ${\theta}$m 0.2 and 0.3. But it saw the different patterns in high water content over ${\theta}$m 0.4 that was drastically declined in the initial stage and then gradually inclined . From the above results, nitrogen transformation patterns differentiated decisively in water content between ${\theta}$m 0.3 and 0.4 in soil. Thus, it is very important for the maintain of suitable soil water content to enhance fertility of soil amended with manure compost. However, excess treatment of manure compost might enhance the possibility of contamination of small watershed and ground water around agricultural area.

Development of a Guided Wave Technique for the Inspection of a Feeder Pipe in a Pressurized Heavy Water Reactor

  • One of the recent safety issues in the pressurized heavy water reactor (PHWR) is the cracking of the feeder pipe. Because of the limited accessibility to the cracked region and a high dose of radiation exposure, it is difficult to inspect all the pipes with the conventional ultrasonic method. In order to solve this problem, a long-range guided wave technique has been developed. A computer program to calculate the dispersion curves in the pipe was developed and the dispersion curves for the feeder pipes in PHWR plants were determined. Several longitudinal and/or flexural modes were selected from the review of the dispersion curves and an actual experiment has been carried out with the specific alignment of the piezoelectric ultrasonic transducers. They were confirmed as L(0,1)) and/or flexural modes(F(m,2)) by the short time Fourier transformation(STFT) and were sensitive to the circumferential cracks, but not to the axial cracks in the pipe. An electromagnetic acoustic transducers(EMAT) was designed and fabricated for the generation and reception of the torsional guided wave. The axial cracks were detected by a torsional mode(T(0,1)) generated by the EMAT.

Development of character recognition system for the billet images in the steel plant

  • In the steel production line, the molten metal of a furnace is transformed into billet and then moves to the heating furnace of the hot rolling mill. This paper describes about the realtime billet characters recognition system in the steel production line. Normally, the billets are mixed at yard so that their identifications are very difficult and very important processing. The character recognition algorithm used in this paper is base on the subspace method by K-L transformation. With this method, we need no special feature extraction steps, which are usually error prone. So the gray character images are directly used as input vectors of the classifier. To train the classifier, we have extracted eigen vectors of each character used in the billet numbers, which consists of 10 arabia numbers and 26 alphabet aharacters, which are gathered from billet images of the production line. We have developed billet characters recognition system using this algorithm and tested this system in the steel production line during the 8-days. The recognition rate of our system in the field test has turned out to be 94.1% (98.6% if the corrupted characters are excluded). In the results, we confirmed that our recognition system has a good performance in the poor environments and ill-conditioned marking system like as steel production plant.

Estimation of Superoxide Dismutase and Glutathione Peroxidase in Oral Submucous Fibrosis, Oral Leukoplakia and Oral Cancer - A Comparative Study

  • Present study was undertaken to estimate and compare erythrocyte superoxide dismutase (E-SOD) and Glutathione peroxidase (GPx) levels in oral submucous fibrosis, oral leukoplakia and oral cancer patients and age/sex matched healthy subjects, 25 in each group. Statistically significant (P<0.001) decrease in E-SOD and GPx levels were observed in OSF, oral leukoplakia and oral cancer groups as compared to the control group. Oral leukoplakia group showed lower levels in comparison with OSF (P>0.05). Oral cancer group had the lowest levels amongst the study groups. Imbalance in antioxidant enzyme status may be considered as one of the factors responsible for the pathogenesis of cancer and may serve as a potential biomarker and therapeutic target to reduce the malignant transformation in oral premalignant lesions/conditions.

Experimental investigation of organic fouling mitigation in membrane filtration and removal by magnetic iron oxide particles

  • Here magnetic iron oxide particles (MIOPs) were synthesized under atmospheric air and which size was controlled by regulating the flow rate of alkali addition and used for efficient removal of bovine serum albumin (BSA) from water. The MIOPs were characterized using field-emission scanning electron microscopy (FE-SEM), Fourier transformation-Infrared spectroscopy (FT-IR) and vibrating sample magnetometer (VSM). The results revealed a successful preparation of the MIOPs. The removal efficiency for BSA using MIOPs was found to be about 100% at lower concentrations (≥ 10 mg/L). The maximum adsorption of 64.7 mg/g for BSA was achieved as per the Langmuir adsorption model. In addition, microfiltration membrane for removal of BSA as model protein organic foulant is also studied. The effect of various MIOPs adsorbent sizes of 210, 680 and 1130 nm on the absorption capacity of BSA was investigated. Water permeability of the BSA integrated with the smallest size MIOPs membrane was increased by approximately 22% compared by the neat BSA membrane during dead-end filtration. Furthermore, the presence of small size MIOPs were also effective in increasing the permeate flux.

Modification of amylose content of sweetpotato starch by RNAi technique

  • In the storage roots of sweetpotato (Ipomoea batatas (L.) Lam. cv. Kokei 14), 10 to 20% of starch is essentially unbranched linear amylose and the other major component is branched amylopectin. Amylose is produced by the enzyme GBSSI (granule bound starch synthase I), whereas amylopectin is produced by a concerted action of soluble starch synthase and starch branching enzymes (SBEI and SBEII). We constructed double-stranded RNA (dsRNA) interference vectors of GBSSI and IbSBEII and introduced them into sweetpotato genome via Agrobacterium-mediated gene transformation. The endogenous GBSSI expression was inhibited by dsRNA of GBSSI in 73 % of transgenic plants giving rise to the storage tubers containing amylopectin but not amylose. On the other hand, all sweetpotato plants transformed with dsRNA of IbSBEII contained a larger amount of amylose than the non-transgenic control (up to 25% compared to 10% in the controls). The RNA interference (RNAi) is effectively inhibited the gene expression in thestarch metabolic pathway and modified the characteristics of starch in sweetpotato.

RECK Gene Promoter rs10814325 Polymorphism in Egyptian Patients with Hepatocellular Carcinoma on Top of Chronic Hepatitis C Viral Infection

  • Background: The reversion-inducing-cysteine-rich protein with Kazal motifs (RECK) gene is a novel transformation suppressor gene linked to several malignancies. Objective: To analyze any association between RECK gene rs10814325 single-nucleotide polymorphism (SNP) and HCC susceptibility with various clinicopathological and laboratory data. Materials and Methods: RECK gene rs10814325 SNP was estimated, using real-time PCR, in 30 HCC patients on top of HCV infection, 30 HCV related cirrhotic patients and 30 healthy controls. Results: No special pattern of association could be detected on comparing the RECK gene rs10814325 genotypes(P=0.5), or alleles(P=0.49) among the studied groups. HCC patients with TT genotype had younger age (mean of $54.1{\pm}6.0$ years vs $60.6{\pm}10.2$ years for TC/CC genotypes, P=0.035). Abdominal distension was significantly greater in TT genotype patients (75% vs 30%for TC/CC genotypes, P=0.045). The TT genotype was present in 75% of patients with lymph node metastasis. Serum GGT levels were higher in TT genotype patients [80 (48.5-134.8) IU/L vs 40 (33-87.5) IU/L for TC/CCgenotypes], and lower limb edema was observed in 60% for TT vs 20% for TC/CCgenotypes, but both just failed to reach significance (p=0.05 and p=0.06 respectively). Conclusions: RECK gene rs10814325 T>C could not be considered a risk factor for HCC development on top of HCV, but may be related to the disease progression and metastasis.
