Cortisol and Fatty Acid Contents in Hanwoo Meat Produced by Antibiotics-free Rearing System (무항생제 사육방식으로 생산된 한우육내 코티졸과 지방산 함량)

  • This study was carried out to elucidate the effect of antibiotic-free rearing system(ARS) on cortisol level, stress hormone, and fatty acid content in the edible muscle tissues, that were of M. longissimus in Hanwoo. These cattle were reared in two different systems including antibiotic-free (ARS) and conservative system (CRS). To increase the experimental reliability, the muscle samples were purchased 3 times from 3 Korean brands of beef produced with ARS or CRS. In the muscle tissue, cortisol level was significantly lower in ARS than CRS, (p=0.0176). But the levels of total saturated- and unsaturated-fatty acids were not significantly different between ARS and CRS (p>0.05). Of total fatty acids, the total saturated fatty acid tended to be greater in CRS and the total unsaturated fatty acid tended to be greater in ARS. However, of the total unsaturated fatty acids, the level of n-6 unsaturated fatty acids was significantly higher in ARS than CRS (p=0.0040). Especially, ${\alpha}$-linolenic acid (ALA) and ${\gamma}$-linolenic acid (GLA) levels were significantly higher in ARS (p<0.01). The n-6 fatty acid content and cortisol level in muscle tissue were negatively correlated at p=0.0140. In conclusion, ARS may produce beef with higher quality which contains lower cortisol and greater n-6 fatty acids, such as ALA and GLA.

  • The arsenic resistance operon from pMH12 in Klebsiella oxytoca contains two regulatory genes. The first open reading frame for arsR extend up to 348 bp and has a translational product corresponding to a protein of 116 amino acid residue polypeptide with a molecular mass of 13 kDa. Performance Improvement of a Temperature and Humidity Measuring System for Strawberry Cultivation Greenhouse (딸기재배 온실용 온습도 계측시스템의 성능개선)

  • This study investigates the improvement in the performance of a temperature and humidity measuring system developed by Company A using the Aspirated Radiation Shield (ARS). The shield has been used in the industry and its accuracy was verified recently. The study also experimentally examines the impact of the wind speed of the ARS device on temperature and humidity. The results are summarized as follows. Before the improvement of the system, the temperature of Company A's system was up to 10.2℃ higher than that measured by the ARS device, and the measured relative humidity was approximately 20.0% lower. After improving the system, the temperature and relative humidity of nodes 1 and 2 were found to be almost identical. The temperature deviations including the maximum, mean, and minimum temperatures between those measured in node 2 and by ARS were approximately 0.2 to 0.7℃, respectively; further, the values measured by ARS tended to be slightly lower or higher. In terms of relative humidity, the ARS measurements yielded values approximately 10.0% higher immediately after sunset; otherwise, the values were approximately 1.9% lower. Moreover, when node 1 was set to minimum-middle, middle-maximum, and maximum, the deviations including the maximum, mean, and minimum temperatures of nodes 1 and 2 were 0.1 to 0.4℃, 0.0 to 0.2℃, and 0.0 to 0.5℃, respectively. The deviations including the maximum, average, and minimum temperatures of the three points of node 1 and the ARS ranged from 0.2 to 0.5℃, 0.1 to 2.2℃, and 0.1 to 1.1℃, respectively, indicating that the temperature deviation according to the wind speed was negligible. In addition, considering the results of the previous study with those from this study, the optimal wind speed to improve the temperature deviation is considered to be in the range of 1.0~2.0 m·s-1.

Feeding Value of Ammoniated Rice Straw Supplemented with Rice Bran in Sheep : I. Effects on Digestibility, Nitrogen Retention and Microbial Protein Yield

  • In vivo digestibility, nitrogen retention and microbial protein yield from diets of 100% ammonia treated rice straw (ARS) ($D_1$); 65% untreated rice straw (URS)+30% rice bran (RB)+5% SBM ($D_2$) and 85% ARS+15% RB ($D_3$) were determined using three Japanese Corriedale wethers in a $3{\times}3$ Latin Square Design. Results showed that DM consumption and organic matter digestibility were highest in $D_3$; but did not promote high protein digestibility, which RB+SBM had effected in URS based-diet. Dry matter intake and OM digestibility were the same for $D_1$ and $D_3$. Solubility of fiber bonds was increased by ammoniation, resulting in higher NDF digestibility. Nitrogen retention and microbial protein yield of rice bran supplemented groups was higher than ARS, but supplementation did not significantly increase efficiency of microbial protein synthesis from ARS which did occur when RB+SBM was added to untreated straw. The quality of ammoniated rice straw could be improved through RB supplementation because of its positive effects on DM digestibility, nitrogen retention and microbial protein yield. However, the addition of RB+SBM to URS resulted to more efficient N utilization.

CARCASS CHARACTERISTICS AND BEEF PALATABILITY AS INFLUENCED BY FEEDING RICE STRAW AND ALFALFA

  • Ninety feeder steers, predominantly Herefords weighing 280 kg, were assigned at random to each of nine diets: basal (high concentrate); 25 or 50% of untreated rice straw (25- or 50 URS) or ammoniated rice straw (25- or 50 ARS); a 50:50 mixture of URS - alfalfa or ARS - alfalfa replacing a proportion of the basal at 25 and 50%. Animals were slaughtered after 154 or 161 days of feeding. The cattle fed 50% URS had the lowest (P<.05) carcass weight, dressing & external fat thickness, kidney, pelvic and heart fat, rib eye area, marbling score, quality and yield grades, followed by cattle fed 50% ARS, 50% rice straw/alfalfa, and 25% rice straw alone or 25% rice straw/alfalfa mixture. Ammoniation of rice straw improved all measured traits. A 25% substitution of basal diet with untreated or ammoniated rice straw and a 50% substitution with rice straw/alfalfa mixture did not significantly affect carcass traits compared to the basal group. Only 50% ARS and 50% URS cattle showed differences in body composition (P<.05) with lower fat and higher water and protein contents. No significant differences were found in shear value, panel tenderness, connective tissue, juiciness, flavor and overall palatability of meat from steers fed the basal, ARS or URS diets.

A Study on Integrated Control of AFS and ARS Using Fuzzy Logic Control Method (Fuzzy Logic 제어를 이용한 AFS와 ARS의 통합제어에 관한 연구)

  • An Integrated Dynamics Control system with four wheel Steering (IDCS) is proposed and analysed in this study. It integrates and controls steer angle of front and rear wheel simultaneously to enhance lateral stability and steerability. An active front steer (AFS) system and an active rear steer (ARS) system are also developed to compare their performances. The systems are evaluated during brake maneuver and several road conditions are used to test the performances. The results showed that IDCS vehicle follows the reference yaw rate and reduces side slip angle very well. AFS and ARS vehicles track the reference yaw rate but they can not reduce side slip angle. On split-${\mu}$ road, IDCS controller forces the vehicle to go straight ahead but AFS and ARS vehicles show lateral deviation from centerline.

Risk Factors for Adverse Reactions to Iodinated Contrast Media in Computed Tomography (컴퓨터 단층촬영을 위한 요오드화 조영제 사용으로 인한 부작용 발생의 위험인자 연구)

  • Objective: The increasing use of imaging examinations such as computed tomography (CT) results in increased contrast media use, which increases contrast media-induced adverse reactions (AR). This study investigated the risk factors of ARs to nonionic iodinated contrast media. Methods: This study evaluated patients who were administered iodinated contrast media during CT scanning in Yeouido St. Mary's Hospital in Seoul, Korea in 2012. Among the subjects, those with contrast media-induced ARs were classified as the AR group. The control group included individuals without ARs who were selected through simple random sampling. The effects of sex, age, contrast media type and dose, CT region, previous contrast media administration, allergy history, and comorbidity were analyzed in the AR and control groups. Results: Multivariate logistic regression analyses were performed to evaluate the identified AR risk factors in 103 subjects in the AR group and 412 subjects in the control group. The results confirmed that the risk of developing ARs was significantly higher in females [odds ratio (OR): 2.206; 95% confidence interval (CI): 1.353-3.598], in individuals administered Iohexol (OR: 9.981; 95% CI: 2.361-42.193), in individuals with an allergy history (OR: 3.982; 95% CI: 1.742-9.101), and in individuals with comorbid asthma (OR: 6.619; 95% CI: 1.377-31.826). Most of the ARs were mild and immediate. Conclusion: In patients who were administered contrast media during CT scans, female gender, Iohexol use, allergy history, and asthma were risk factors for ARs. Therefore, special care is required for patients with such risk factors to prevent ARs.

Establishment of a Selection System for the Site-Specific Incorporation of Unnatural Amino Acids into Protein (비천연 아미노산의 위치특이적 단백질 삽입을 위한 Amino Acyl-tRNA Synthetase 선별시스템 개발)

  • Site-specific incorporation of unnatural amino acids (SSIUA) into protein can be achieved in vivo by coexpression of an orthogonal pair of suppressor tRNA and engineered aminoacyl-tRNA synthetase (ARS) that specifically ligates an unnatural amino acid to the suppressor tRNA. As a step to develop the SSIUA technique in Escherichia coli, here we established a new 2-step screening system that can be used for selecting an ARS variant(s) that ligates an unnatural amino acid to a suppressor tRNA. A positive selection system consists of chloramphenicol acetyl transferase gene containing an amber mutation at the $27^{th}$ residue, and efficiently concentrated amber suppressible ARS with a maximum enrichment factor of $9.0{\times}10^5$. On the other hand, a negative selection system was constructed by adding multiple amber codons in front of a lethal gene encoding the control of cell death B toxin (ccdB) which acts as an inhibitory protein of bacterial topoisomerase II. Amber suppression of ccdB by an orthogonal pair of Saccharomyces cerevisiae tyrosyl-tRNA synthetase (TyrRS) and an amber suppressor tRNA significantly inhibits bacterial growth. This selection system was also able to efficiently remove amber suppressible ARS which could ligate natural amino acids to the suppressor tRNA. Thus, sequential combination of these two selection systems might be able to function as a powerful tool for selecting an ARS variant that specifically ligates an unnatural amino acid to the suppressor tRNA from an ARS mutant pool.
