Bacillus subtilis JK-1 showed degradation activity against crude oil, gasoline, kerosene, and light oil, and this strain was used as a crude biosurfactant producing microorganism in this study. To optimize the culture medium for production of crude biosurfactant, the influences of various carbon, nitrogen and mineral sources were assessed. The highest biosurfactant production by B. subtilis JK-1 was observed after 96 h cultivation, containing 1.0% (w/v) soluble starch as a carbon source and 0.5% (w/v) skim milk as a nitrogen source, and carbon to nitrogen concentraion (C/N) ratio was 2.0. For the biosurfactant production 0.1% (w/v) of $KNO_3$ was the most effective mineral source. Comparison of biosurfactant production indicates that B. subtilis JK-1 produces more biosurfactant in the optimum medium established in this study than LB and TSB. Under the optimum medium, the surface tension of culture broth of B. subtilis JK-1 was decreased from 47.3 dyne/cm to 24.0 dyne/cm after cultivation of 48 h.

A turbocharger is an engine supercharger that is driven by exhaust gas. It improves the output and fuel efficiency by increasing the charging efficiency of the mixture gas, which is achieved by changing the rotatory power of the turbine connected to the exhaust passage. It is important to control the supercharging for this purpose. A nozzle slide joint is one of the core parts. Austenitic stainless steel is currently used as the material for this part, and its excellent mechanical properties include high heat resistance and corrosion resistance. However, because of its poor machinability, there are many difficulties in producing products with complicated shapes. Machining is used in the production of nozzle slide joints for high dimensional accuracy after metal powder injection molding. As design variables in this study, we investigated the sintering temperature, product stress, deformation rate, radius of curvature of the punch, and angle of the chamfer punch, which are related to the strain and shapes. The goal is to suggest a forming process using Nitronic 60 that does not require machining to manufacture a nozzle slide joint for a turbocharger. Accordingly, we determined the best process environment using finite-element analysis, the signal-noise ratio, and the Taguchi method for experiment design. The relative density and hydrostatic pressure of the final product were in accordance with the results of the finite element analysis. Therefore, we conclude that the Taguchi method can be applied to the design process of metal powder injection molding.

This study was done to determine the effect of antioxidants added perilla oil diet on the content of cholesterol, vitamin E, and lipid peroxide in serum and tissue of rats. Four groups of experimental diets, such none added perilla oil diet, ascorbic acid added perilla oil diet, vitamin E added perilla oil diet, EDTA added perilla oil diet were fed ad libitum to the 4 weeks white male rats of Sprague-Dawley strain. The results obtained are summarized as follow: 1) The body weight gain in all experimental diet groups was higher than the control gorup and EDTA added diet group was lower than the other experimental diet group, while food intake in vitamin E added diet group was the highest and vitamin C added diet group was the lowest in the control group. 2) Total cholesterol levels in serum of all experimental diet groups were lower than that of the control group and especially the level of total cholesterol in none added diet group and vitamin C added diet group were significantly lower than that of control group. 3) HDL-cholesterol levels of all experimental diet groups were lower than that of the control group and especially none added diet group was significantly lower than that of control group. 4) The activities of glutamic oxaloacetic transaminase (GOT) in serum of all experimental diet group except EDTA added diet group were higher than that of the control group and especially none added diet group was significanly higher than that of the control group. The activites of glutamic pyruvic transaminase (GPT) in serum of all experimental diet groups except vitamin C added group were higher than that of control group. 5) Vitamin E levels in serum of none added diet group and vitamin C added diet group were lower than that of the control group and vitamin E added diet group and EDTA added diet group were higher than that of the control group. 6) Vitamin E levels in liver of all experimental diet groups were higher than that of control group and especially none added diet group and vitamin E added diet group were significantly higher than that of the control group. 7) Lipid peroxide in serum of all experimental diet group were lower than that of control group and especially EDTA added diet group. 8) Lipid peroxide in liver and spleen of all experimental diet groups were higher than that of the control group and lipid peroxide in kidney of all experimental diet groups except EDTA added diet group were higher than that of the control group. Four these results, as vitamin C, vitamin E and EDTA added diets have an effect to lipid peroxide by antioxidants, it could be suggested that perilla oil diet has required to add antioxidant because it has not sufficient vitamin E for antioxidant and intake and overtake level of perilla oil diet should be studied to go ahead.
Sporulation in the fission yeast Schizosaccharomyces pombe has been regarded as an important model of cellular development and differentiation. S. pombe cells proliferate by mitosis and binary fission on growth medium. Deprivation of nutrients especially nitrogen sources, causes the cessation of mitosis and initiates sexual reproduction by malting between two sexually compatible cell types. Meiosis is then followed in a diploid cell in the absence of nitrogen source. DNA fragment complemented with the mutations of sporulation gene was isolated from the S. pombe gene library constructed in the vector, pDB 248' and designated as pDB (spo 5)1. We futher analyzed six recombinant plasmids, pDB (spo 5)2, pDB(spo 5)3, pDB(spo 5)4, pDB(spo 5)5, pDB(spo 5)6, pDB(spo 5)7, and found each plasmids is able to rescue the spo 5-2, spo 5-3, spo 5-4, spo 5-5, spo 5-6, spo 5-7, mutations, respectively. Mapping of the integrated plasmid into the homologous site of the S. pombe chromosomes demonstrated that pDB (spo 5)1, and pDB (spo 5)R1 contained the spo 5 gene. Transcipts of spo 5 gene were analyzed by Northern hybridization. Two transcripts of 3.2 kb and 25 kb were detected with 5 kb Hind III fragment containing a part of the spo 5 gene as a probe. The small mRNA (2.5 kb) appeared only when a wild-type strain was cultured in the absence of nitrogen source in which condition the large mRNA (3.2 kb) was produced constitutively. Appearance of a 2.5 kb spo 5-mRNA depends upon the function of the mei1, mei2 and mei3 genes.
The transformational conditions of plasmids $pPL-\lambda$ and pAS 1 are as follows in E. coli, Ts-mutant M 5248 strain at $30^{\circ}C$. When the culture time was 2.5 hours of mid logarithemic phase, the cell concentration was $4.5\times10^7\;cells/ml$, the optical density was equal to 0.45 at 590 nm wave length, the transformational frequencies of plasmid$pPL-\lambda$ and pAS 1 had the highest values as $2\times10^{-6}\;and\;1.5\times10^{-6}\;and\;1.5\times10^{-6}$ and respectively. For $9\times10^6$ competent cells in $200{\mu}l$, the transformational frequency was as high as $4.4\times10^{-6}$ at 510 ng plasmid concentration. The competent cells treated with the mixture of calcium chloride and thymidine twice rates of transformation than those treated with calcium chloride. The ampicillin resistance of transformants was expressed in LB broth after 2 hours at $30^{\circ}C$.
One bacterium with high proteinase production and spore-forming ability was isolated from korean traditional soybean paste(doenjang). The isolated strain was identified as Bacillus subtilis, based on gram-staining, biochemical properties and l6S rRNA gene sequencing, and designated as B. subtilis DJI. Its growth rate was very fast, and it reached its stationary phase within 9 h, and then started to form spores. Bacterial-koji and doenjang were prepared using B. subtilis DJI. Chemical components of the doenjang were determined after 2 months of aging period: amino nitrogen 507 mg%, crude protein 14.3%, crude fat 4.8% and water 54.9%. The composition of total and free amino acids and their ratios of doenjang were changed during the aging period. Among total amino acids in DJI doenjang, glutamic acid, aspartic acid, leucine and arginine were the major amino acids. The fibrinolytic activities of DJI doenjang and traditional doenjangs were 909.7 units/ml and $363.3{\sim}618.6\;units/ml$, respectively. Flavor compounds of DJI doenjang and traditional doenjang were extracted by SDE(simultaneous steam distillation and extraction), and analyzed by GC/MS; DJI doenjang possessed the typically favorable flavor compounds in traditional korean doenjang, with reduced off-flavor compounds.
One bacterium, which showed strong antagonistic activity against H. pylori KCCM 41756, was isolated from kimchi. The strain NO1 was designated as Lactobacillus plantarum NO1 based on Gram staining, biochemical properties, and 16S rRNA gene sequencing. The culture medium $(2{\sim}4{\mu}g/ml)$ of Lb. plantarum NO1 reduced $(40{\sim}60%)$ the urease activity of H. pylori KCCM 41756. Lb. plantarum NO1 inhibited the binding of H. pylori to human gastric cancer cell line, AGS cells, by more than 33%. Lb. plantarum NO1 exhibited high viability (maintained initial viable cell count of $10^9CFU/ml$) in 0.05 M sodium phosphate buffer (pH 3.0) for 2 h, in artificial gastricjuice for 2 h and in 0.3%, 0.5% oxgall for 24 h. Hemolysis phenomena did not observed when Lb. plantarum NO1 was incubated in the blood agar media. We concluded that Lb. plantarum NO1 can be a good candidate as a probiotic, harboring anti-H. pylori activity.
Biogenic amines such as histamine, tyramine, cadaverine, and putrescine may have detrimental effects on consumers' health; therefore, they must be considered hazardous substances in foods. In recent years, the application of microorganisms that can degrade biogenic amines has become an emerging method for reducing the amount of these amines in foods. Primarily, Bacillus sp. BCNU 9171 was isolated from Doenjang, a Korean traditional fermented soybean paste. The inhibitory effects of the cell-free supernatant (CFS) of Bacillus sp. BCNU 9171 on biogenic amine production by 4 amine-positive food pathogenic bacteria were investigated using high performance liquid chromatography (HPLC). Our results showed that 4 different CFS concentrations-10% [1 ml CFS + 9 ml histidine decarboxylase broth (HDB)], 25% (2.5 ml CFS + 7.5 ml HDB), 50% (5 ml CFS + 5 ml HDB), and 75% (7.5 ml CFS + 2.5 ml HDB)-reduced the biogenic amine production up to 87% compared with that of the control without CFS. These results suggested that it is advisable to use Bacillus sp. strain BCNU 9171 as a starter organism for the manufacture of fermented foods and to ensure food safety since it prevents the accumulation of high amounts of biogenic amines in fermented foods by amine-positive bacteria.
Fiftysix male weanling rats of the Sprague-Dawley strain weighing $52.6{\pm} 0.9g$ were fed with 77% starch-15% casein diet by ad libitum for four days to get them adapted and divided into eight groups. For three weeks, the body weight gain of rats was controlled in three different ranges. After the period of food restriction, the rats were recovered by being fed by ad libitum for seventeen days. During the experimental period, the amount of food intake and body weight were measured. And the experimental groups were compared when they reached at the same age and at the same body weight. Anterior Tibialis, Extensor Digitorum Longus, Soleus, Plantaris, Gastrocnemius were used as analytic items of skeletal muscle. Wet weight of muscle, muscle protein, water content were measured from each of five skeletal muscles. Sodium, potassium, magnesium content were measured from Anterior Tibialis, Extensor Digitorum Longus, Soleus ana Plantaris. Phosphorus was measured only from Gastrocnemius. The whole carcass protein except the skeletal muscles was also measured. During the period of food restriction, food intake was $311.7{\pm}19.5g$ for the control group. $130.2{\pm}1.5g$ for the second group and $161.7{\pm}2.1g$ for the third group. During the period of food restriction, body weight gain was $106.8{\pm}12.7g$ for the control group, $3.6{\pm}2.1g$ for the second group and $18.9{\pm}3.3g$ for the third group. Comparing the body weight when they reached at the age 66 days, the control group showed higher value than the other groups. In the concentration of electrolytes of skeletal muscles sodium and magnesium tend to increase and potassium and phosphorus tend to decrease by food restriction. But after their recovery, there was no significant difference between the groups. By the degree of food restriction, there was time difference in reaching at the same body weight. But when they reached at the same body weight, there was no significant difference in the value of electrolytes concentration. Regarding all the results of this study, the ages of rats which reached the same body weight were different by food restriction level during weanling. Once food·restricted rats reached the same body weight by recovery, the concentration of electrolytes and protein tend to become almost the same.
In order to demonstrate the usefulness of Deodeok. the functions of Deodeok and the qualities of Cheongkukjang were investigated. DPPH (1.1-diphenyl-2-picryl-hydrazyl) radical scavenging activity of Deodeok extracted with 70% ethanol was higher than that of water extract. The direct antimutagenic effect of ethanol extract of Deodeok was examined by Ames test using Salmonella typimurium TA 98. The inhibition rates on ethanol extract at concentrations of 200, 1.000, 2.000, 3.000 and 4.000 ${\mu}g$/plate were 5.75, 31.38, 34.75, 53.50 and 83.75%, respectively. The inhibition rates of ethanol extract was higher over 2 times than that of water extract. The qualities including physicochemical and sensory properties of Deodeok Cheongkukjang were investigated over the following range of Deodeok levels; 5, 10, 15 and 20% (w/w). The strain used in Cheongkukjang manufacture was Bacillus sp. B-3 with the highest enzyme activities such as amylase and protease. During fermentation at $40^{\circ}C$ for 6 days. amino-type nitrogen content of Cheongkukjang containing 10% (w/w) Deodeok was more than others. When Deodeok content exceeded 15% (w/w), higher contents resulted in lesser amino-type nitrogen production. The results showed that Deodeok had influenced the growth of Bacillus sp. B-3. The L-value of Deodeok Cheongkukjang was decreased according to increasing the Deodeok contents. Sensory evaluation showed that Deodeok Cheongkukjang containing 10% (w/w) Deodeok was superior to other tested.
