Inhibition of Teeth Decalcification and Glucosyltransferase Activity by Juniperus rigida S. et Z. (두송실의 치아탈회 및 Glucosyltransferase 활성억제효과)

  • We investigated the inhibition effects of teeth decalcification and glucosyltransferase(GTase) activity on the Juniperus rigida S. et Z. Teeth decalcifications by Streptococcus mutans were respectively inhibited to 70.13, 74.93% on methanol and n hexane fractions. In the inhibition test of GTase activities by solvent fractions of J. rigida, they were respectively inhibited to 86.6, 89.5% to a similar degree. And in the result to identify GTase produced by S. mutans with SDS PAGE, the band near 65KD estimated as GTase did not show in the lanes of methanol, n hexane and chloroform fractions.

Extractives from the needles of Juniperus rigida Siebold et Zucearin and Antioxidant activity (노간주나무(Juniperus rigida Siebold et Zucearini) 잎의 추출성분 및 항산화 활성)

  • The dried neeldes(1.5 kg) of Juniperus rigida were ground, extracted with acetone-H2O(7:3, v/v), concentrated, and fractionated with a series of hexane, CH2Cl2, EtOAc and water on a separatory funnel. Each fraction was freeze dried to give dark-brown powder and EtOAc and water soluble fractions were chromatographed on a Sephadex LH-20 column using a series of aqueous methanol and ethanol-hexane mixture as eluent. Spectrometric analysis such as NMR and FAB or EI-MS including TLC were performed to characterize the structures of the isolated compounds. The needles of Juniperus rigida contained a large amount of (+)-catechin, quercetin-3-O-α-L-rhamnopyranoside and isoconiferin, in addition to a small amount of umbelliferone and quercetin-3-O-β-D-rutinoside. The antioxidative activities of each fraction and isolated compounds were tested by DPPH radical scavenging method, and EtOAc soluble fraction, (+)-catechin and quercetin-3-O-α-L-rhamnopyranoside were effective.

Fungal Endophytes Isolated from Needle Leaves of Three Coniferous Species on Mt. Seodae of Korea

  • We investigated endophytic fungi diversity of two Pinaceae species (Pinus densiflora, Pinus rigida) and one Cupressaceae species (Juniperus rigida) on Mt. Seodae, Korea. In total, 34 isolates were obtained from 19 host plants and identified according to internal transcribed spacer (ITS) region. As a result, they were placed in 13 taxa. Among them, 52.9% belonged to Leotiomycetes, 32.3% belonged to Sordariomycetes, 5.8% belonged to Agaricomycetes, and 3.0% for isolates belonged to either Eurotiomycetes, Dothideomycetes or Ustilaginomycetes. In particular, Lophodermium growing in pine would be an especially important instance of biodiversity for endophytes, suggesting that further study examining its ecological function in the environment is critical.

Inhibition Effect by Juniperus rigida S. et Z. on Organic Acids Production from Streptococcus mutans (두송실에 의한 충치균의 유기산 생성 억제효과)

  • To develop the natural antibacterial agents which don't have any toxicity against man, collected several species of medicinal plants were tested for their antibacterial activity and inhibition effect on organic acids production from Streptococcus mutans. Among the tested medicinal plants, methanol extracts and n-hexane fraction of Juniperus rigida S. et Z. had the comparatively strong activities. For example, strong antibacterial effect against S. mutans were showed in methanol extracts, n-hexane and chloroform fractions. Organic acids production were reduced to 89.3 and 90.8% of control in methanol extracts and n-hexane fraction treated concentration of 1 mg/ml, respectively.

Diversity of Endophytic Fungi Isolated from Pinus densiflora and Juniperus rigida Distributed in Mt. Baekryeonsan and Mt. Johangsan, Korea

  • We investigated the biodiversity of endophytic fungi in Pinus densiflora and Junipreus rigida from Mt. Baekryeonsan and Mt. Johangsan in Korea. In total, 59 isolates were discovered from 26 host plants and identified using sequences from the rDNA internal transcribed spacer (ITS) region. Twenty taxa of endophytic fungi were identified in this study; 62% of isolates belonged to Leotiomycetes, 34% belonged to Sordariomyetes, and 4% belonged to Agaricomycetes. Results confirmed three previously unreported endophytic fungi in Korea: Phanerochaete concrescens, Creosphaeria sassafras, and Scopuloides rimosa. Community analysis showed that the fungal diversity patterns of the samples from Mt Baekryeonsan and Mt. Johangsan are similar to those from previous studies and varied depending upon host and locality.

Anti-lipase and Lipolytic Activities of EtOH Extract from Juniperus rigida (노간주나무 에탄올추출물의 지방 흡수 억제 및 지방분해 효능)

  • Obesity is an important risk factor that significantly increases mortality and disease rates in the cardiovascular disease, diabetes, and various diseases. So far, the most powerful way to inhibit fat absorption is pancreatic lipase inhibitors. In this study, we investigated the anti-obesity effect of the extract of Juniperus rigida. Juniperus rigida extract (JRE) had a inhibitory effect on pancreatic lipase activity ($IC_{50}$=8.63 ${\mu}g$/ml). In in vivo oil-emulsion loading test, this extract also inhibited the intestinal fat absorption. In addition, we measured inhibitory effects of JRE on activity of phosphodiesterase (PDE) and hormone sensitive lipase (HSL) among the important enzymes associated with lipolysis. JRE strongly inhibited PDE activity ($IC_{50}$=4.56 ${\mu}g$/ml), whereas inhibitory effect on HSL activity was very weak compared with orlistat. As a result, JRE inhibited the absorption of fat by inhibiting the activity of pancreatic lipase and induced lipolysis through inhibition of PDE activity. Therefore, we suggest that Juniperus rigida may be a potential therapeutic agent improving obesity.

Karyotype Analysis of Juniperus rigida Sieb. et Zucc. of Two Different Provenances in Korea (한국산(韓國産) Juniperus rigida의 두 산지(産地)의 핵형분석(核型分析))

  • Karyotypes are described for Juniperus rigida Sieb. et zucc, in two provenances of Gyeong-nam and Choong-puk. Chromosome numbers of two provenances, are 2n=22. The most common feature of mitotic chromosomes was shown at the chromosome 7, which has secondary constriction on the short arm. And the most differential chromosome was shown at chromosome 9 from Gyeong-nam and chromosome 5 from Choong-puk provenance which bore secondary constriction. The karyotype formulae are as follows; Gyeong-nam, Jinyang provenance race is $$K(2n)=22=2A^m+2B^m+2C^m+2D^{sm}+2E^{st}+2F^m+2^{sc}G^m+2H^m+2^{sc}I^t+2J^{st}+2K^m$$ Choong-puk, Jechun provenance race is $$K(2n)=22=2A^m+2B^m+2C^m+2D^{st}+2^{sc}E^{sm}+2F^m+2^{sc}G^m+2H^m+2I^m+2J^{st}+2K^{sm}$$.

A Study on the Flora in Tanyang Limestone Area (단양 석회암지역의 식물상에 관한 연구)

  • Biomass and soil properties of plant communites was investigated in the limestone ares, Tanyang, Ch'ungbuk Province in Korea. Plants was classified in calcicoles and calcifuges within plant communities occurring in the limestone area. Biomass of plant populatins in the limestone area was measured that Stellaria aquatica was the lowest(0.58g/plant), Chrysanthemum boreale was the highest(8.87g/plant) and that Pennisetum alopecuroides, Miscanthus sinensis was 6.67g/plant, 5.76g/plant, respectively. Diameter of breast height among the tree layer communities was investigated that Pinus densiflora was the widest(20cm), Juniperus rigida was the narrowest(6cm) and that Quercus aliena, Populus ${\times}$ tomentiglandulosa was 10cm, 12cm, respectively. The high of density in the tree layer was found out Populus ${\times}$ tomentiglandulosa, Juniperus rigida, Quercus aliena, Pinus densiflora in this order. Dominant species in the study area were Pinus densiflora, Quercus aliena, Juniperus rigida, Quercus dentata in the tree layer, Lespedeza bicolor, Buxus microphylla, Rhus chinensis in the shrub layer, Humulus Japonicus, Erigeron canadensis, Xanthium strumarium, Oenothera odorata Persicaria hypropiper on the riverside, Pennisetum alopecuroides, Vicia amoena, Chrysanthemum boreale on the roadside. Soil properties of the limestone area was investigated around the protrusion of rock in the upper region, around the cave and underlayer accumulated by corrosion in the study area. Lime-chlorosis was observed in the upper region, underlayer and around the cave. It was observed that biota of kum-gul and dungbong-gul was Rhinolophus ferrumequinum korai, Epanerchodus kimi, Antrokoreana gracilipes, Diestrammena japonical. Dominant species were Humulus japonicus, Stellaria aquatica, Hydrocotyle maritima, Phragmites communis, Miscanthus sinensis around the inlet of a kum-gul. It was classified that plants in the study area was 34 order, 53 family, 135 species in all. Calcicoles were 11 order, 16 family, 18 species and calcufuges were 6 order, 8 family, 11 species of them all.

  • Human skin is constantly exposed to ultraviolet (UV) radiation, polluted air, and chemical products. UV rays, in particular, will affect the skin in a variety of ways, including causing wrinkles, fine lines, rough skin, and xeroderma, thereby resulting in skin aging. This study aimed to investigate the whitening effects of Juniperus rigida Sieb., which is a cedar tree that is found throughout the world. The whitening efficacy that was measured by tyrosinase inhibition revealed 49.4% efficacy in water extract and 80.0% efficacy in ethanol extract. Among the B16F10 black cells, the effect of the ethanol extract was higher than the effect of the water extract in the restrain creation of melanin pigment, tyrosinase, microphthalmia-associated transcription factor (MITF), tyrosinase related protein-1 (TRP-1), and tyrosinase related protein-2 (TRP-2). Thus, the results of these studies demonstrated that the ethanol extract had greater efficacy than the water extract and Juniperus rigida Sieb. Ethanol extracts could be utilized as materials for functional cosmetics, such as whitening products.
