Analysis of Genetic Relationship and Cultural Characteristics of Penicillium species Isolated from Postharvest Decay of Pear by Random Amplified Polymorphic DNA (저장 배에서 분리한 Penicillium속의 배양적 특성과 RAPD에 의한 유연관계 분석)

  • Fifteen isolates from pear, and sixteen isolates from KCTC, KCCM, and Chungnam Univ. of Penicillium spp. were investigated for the analysis of their relationships of cultural characteristics and RAPD genetic variation by RAPD. The cultural characteristics of Penicillium spp. were shown different growth rate, morphology, and color. In addition, the cultural characteristics and RAPD analysis were conducted for the pear rot pathogens and related isolates. RAPD patterns were applied to compare the taxonomic and genetic diversity of the Penicillium species between 15 groups isolated from pear fruits and 16 standard species. The genomic DNA were amplified from $0.1{\sim}2.0kb$ by five URP primer and 744 bands were detected. The cluster analysis showed four genomic DNA RAPD groups and its similarity was 47.7%. Intraspecific relationships were 87.4, 97.5 and 95.2%, in P. expansum, P. solitum, and P. crustosum, respectively. These results appeared to be that there were high similarities between isolates, and consistent with the results of cultural morphological characteristics analysis.

Effects of Chitosan on Red Pepper (Capsicum annuum L.) Cultivation for Eco-friendly Agriculture (고추 친환경 재배를 위한 Chitosan 처리 효과)

  • The present study evaluated the effect of chitosan on the changes of soil chemical properties, soil microbial population, and yield of red pepper (Capsicum annuum L.) for eco-friendly agriculture in an upland field. We utilized four treatment groups, control, foliar spray, soil drench, and foliar spray + soil drench with chitosan, and analyzed these variations throughout the seven, fourteen, and twenty one days interval. The pH values, organic matter, and available phosphate in the upland soil at the harvesting stage decreased in the chitosan treatment. The populations of bacteria, actinomyces, and fungi in the upland field were increased in plots treated with chitosan. The chlorophyll content of leaves was no significant differences between the control and the chitosan treatments, while calcium content of leaves was significantly higher in the chitosan treatments than in the control. In addition, the nitrogen content of leaves was no significant differences between the foliar spray and the soil drench. The yield of red pepper was significantly higher in the control ($383kg\;10a^{-1}$) than the chitosan treatments and the yield of soil drench with chitosan reached up to 95% of control.

Yield Loss Assessment and Determination of Control Thresholds for Powdery Mildew of Chili pepper (Capsicum annuum L) (시설 고추에 발생하는 흰가루병의 경제적 방제수준에 따른 고추수확량 변화 예측)

  • This study was carried out to develop the economic thresholds for powdery mildew on pepper. To investigate the relationship between powdery mildew incidence degree and yield, experimental plots with ten treatments as initial disease degree were established. Disease intensity exhibited negative and significant correlation with fruit characters like fruit length, fruit diameter, fruit weight. The adverse effect of the disease on these characteristics was low yield, exhibiting significant negative correlation with disease intensity. There existed close correlation between rate of infected leaf area and yields in the plastic house (Chonhatongil: Y = -3.44X+291.09 $R^2=0.73$, Buchon: Y = -2.14X+327.9 $R^2=0.78$). There existed close correlation between rate of infected leaf area and yield loss in the plastic house (Chonhatongil: Y = 2.14X+15.45 $R^2=0.76$ $r=0.87^{**}$, Buchon: Y = 3.44X+114.21 $R^2=0.73$ $r=0.85^{**}$). Control thresholds diseased rate on powdery mildew of pepper was below 3.2 to 7.3% rate of infected leaf area per plant in the plastic house. The economic thresholds for powdery mildew of pepper was below 3.8 to 6.2% rate of infected leaf area per plant in the plastic house.

Influences of Seeding Dates and Pinching Height on Tender Shoot Productivity of Moloheiya (Corchorus olitorius L.) (모로헤이야 파종시기와 적심높이에 따른 신초의 생산성 비교)

  • This study was conducted to seek optimum sowing time and pinching height for tender shoots production of moloheiya (Corchorus olitorius L.), which is a subtropical leaf vegetables, under an unheated greenhouse in the south of Korea. Seeds of moloheiya did not germinate at all at $10^{\circ}C$, but germinated well at $18{\sim}30^{\circ}C$ with 95.5~98.5% germination rate. Germination rate was 75.0% at $15^{\circ}C$, but germination speed was very low. The pods of moloheiya sown before April 28 was matured after late-August, but those sown after May 7 was not matured in that year. In tender shoot production and harvest frequency, treatment sown on April 14 was most plentiful among sowing time treatments, and showed marketable yield increase by 14~42% compared to other sowing times. Mean-while, pinching at 100cm high from soil surface was more effective than other heights for increment of branches and tender shoot. All above suggest that it was suitable to sow seed on mid-April and to pinch at 100cm high from soil surface for increment of tender shoots in the south of Korea.

  • Previous studies on genetic transformation of chrysanthemum using cold regulated gene (BN115) have been conducted and the PCR and Real-Time PCR based method to determine the presence of the transferred cold regulated gene in the chrysanthemum was established. To check whether over-expression of BN115 gene in transgenic chrysanthemum will enhance their tolerance to cold stress, the transgenic chrysanthemum were grown under low temperature condition and several cold signalling including growth characteristics, stoma size and shape, SPAD value and ion leakage test were investigated. The transgenic chrysanthemum in the low temperature growth chamber grow much faster in term of the height, number and size of the leaves than those of wild-type plants and damage of transgenic plant caused by the low temperature was much less than that of wild-type plants. The stoma type and size of transgenic plant leaves grown at $5^{\circ}C$ were much similar to of wild-type plant cultured on $25^{\circ}C$ It has been found that SPAD value of transgenic plants was much higher than those of wild-type, but the EC density being lower under low temperature condition.

Condition of mycelial culture and inoculum volume of spawn on cultivation of Agrocybe cylindracea (버들송이의 균사배양조건 및 최적 접종량 설정)

  • Studies were made to optimize the media composition and cultural condition for mycelial growth of Agrocybe cylindracea. Sawdust spawn of media composition for optimal growth was found to be pine sawdust combination of 30% wheat bran and poplar sawdust combination of 20% corn bran were the best of the optimal combination. The optimal concentration of white sugar was 1.0~1.5%. The nitrogen sources was found to be yeast extract and soybean powder. Also, optimal concentration were $0.7g/{\ell}$ and $0.1g/{\ell}$, respectively. The mineral sources of optimal medium compositions were $MgSO_4{\cdot}7H_2O\;0.3g/{\ell}$, $KH_2PO_4\;0.5g/{\ell}$ and $K_2HPO\;1.2g/{\ell}$. Optimal amount of inoculum for cultivation of A. cylindracea were $20{\sim}25g/850m{\ell}$ and $25m{\ell}/850m{\ell}$ in the sawdust spawn and liquid spawn, respectively.

Determination of Optimal Collecting Date and Exogenous Auxin Dipping Treatments in Cutting Transplants of 'Seolhyang' Strawberry (Fragaria × ananassa Duch.) ('설향' 딸기 삽목묘의 최적 삽수 채취시기와 오옥신 처리 구명)

  • The aim of this study was to investigate the effects of timing of collecting date and concentration of IBA and NAA, in order to enhance initial activity and seedling quality of domestic strawberry. Strawberry cuttings were separately taken twice, in June 7 and in July 5, and IBA and NAA were treated with the concentrations of 0.025, 0.05 and 0.1% at cutting date, respectively. The seedlings were evaluated for the percentage of survival during 18 days at 6 times after tunnel cultivation. The NAA treatment was inappropriate for strawberry cutting due to the high rate of seedling mortality, regardless of the collecting date. The vitality of the seedlings was highest at IBA 0.1% in June collecting and at IBA 0.05% in July collecting. The seedlings from June collecting had a higher quantum yield at IBA 0.1% and the seedlings from July collecting at IBA 0.05%. Therefore, IBA could be more effectively applied than NAA to promote the vitality and quality with the appropriate concentration of 0.1% at June collecting and 0.05% at July collecting, respectively.

Food nutritional characteristics of fruit of Cudrania tricuspidata in its various maturation stages (꾸지뽕나무 열매의 숙기별 식품학적 특성)

  • This study was conducted to investigate the food value of Cudrania tricuspidata at its various maturation stages. The pH, total acid and reducing sugar contents of its fruit juice were determined to have been 4.2~5.1, 1.4~2.0% and 5.4~8.6%, respectively. The general chemical components of its fruit were observed as 76~80% moisture, 2.2~3.5% crude protein, 1.7~2.9% crude fat, 0.8~1.2% ash and 14.5~16.4% carbohydrate. Its free sugar, glucose and fructose contents were determined. The fructose contents of both its ripened and over-ripened fruits were higher than their glucose contents. Organic acids such as oxalic acid, citric acid, tartaric acid, malic acid and succinic acid were detected, and the concentration of the malic acid and the succinic acid were found to have been most abundant. The K content was higher than the amounts of other minerals, such as Ca, Fe, K, Mg, Na and P. Its vitamin C and the total amount of its dietary fiber were 127.5~149.2 mg% and 22.7~38.7%, respectively. Its insoluble dietary fiber content was higher than its soluble dietary fiber content. Its total polyphenol and flavonoid content were 18.9~19.6 mg% and 40.9~48.2 mg%, respectively.

Set up and Running Status of School Gardening at Elementary Schools - Focus on Jeollabuk-do (초등학교 학교 텃밭의 조성현황과 운영실태 분석 - 전북지역을 중심으로)

  • This study was conducted to investigate the set up and running status of school gardening at elementary schools in Jeollabuk-do. Among 416 elementary schools in Jeollabuk-do, 164 schools (39.4%) had school gardens. Ninety-seven schools in cities and 67 in counties had school gardens. The total area and school garden size at schools in Jeollabuk-do were $45,490m^2$ and $277m^2$ per school, respectively, as well as $1.6m^2$ per students. School gardens varied in type, and percentages of outdoor and off-campus gardens were 67.2% and 17.2%, respectively. There were differences in the set up, type of garden, annual operating budget, and participants in school garden programs according to the location (city or county) of the school The installation and automation of facilities in the garden (such as greenhouse, tool shed, resource recycling facility, etc.) were poor. Most schools grew various kinds of plants, including vegetables, crops, ornamentals, and fruits. Teachers most often operated school gardens and taught students. Teachers had difficulty managing school gardens due to absence of knowledge about sustaining gardens. Most respondents reported the need for a school garden training program. Sixty-one percent of schools reported that the garden was used for academic instruction, especially during class. The majority of respondents agreed that school gardens have a positive effect and wanted to increase classes related to school gardens. Accordingly, in order to sustain school gardens and maximize their effects, systematic and customized support is needed that considers the characteristics and circumstances of the school. The facilities and features of the garden should be improved, and the school garden training program for teachers should attempt to reduce the effort required to manage the garden and increase utilization efficiency. In addition, participation of garden coordinators, parents, and community volunteers in managing gardens and implementing garden lessons is required.

Changes in the Physico-Chemical Properties of Growing Media and the Growth of Oriental Melon Seedlings(Cucumis melo L.) by Charcoal Application (활성탄 혼합 비율에 따른 상토의 이화학성 변화와 참외(Cucumis melo L.)묘의 생육)

  • To investigate the effect of charcoal on the physico-chemical characteristics in the growing media and the growth of oriental melon, six treatment, 0%, 10%, 20%, 30%, 40%, and 50% charcoal, were added into the growing media, Baroker. The value of pH in the charcoal contained growing media (CGM) was recorded higher and further increased by raising the charcoal ratio and by growing period. In 20% CGM, the range of pH was adequate to grow crop as 5.2${\sim}$5.8. Contents of Ca and K in CGM increased by raising the ratio of charcoal, while contents of Mg and Na decreased. The growth of oriental melon seedlings in 20% and 30% CGM was better than in other treatments, in terms of fresh and dry weight of shoot, plant height, leaf area, lear width and relative growth rate. Both the physico-chemical properties of growing media and the growth of oriental melon seedlings were changed by the addition of charcoal. These results suggest that charcoal can be used as mixing material with other potting media for producing the seedling of good quality.
