Effect of Embryogenic Callus Conditions on Plant Regeneration in Satsuma Mandarin (Citrus unshiu Marc.)

  • The ability to form embryiod from callus in satsuma mandarin is low and unstable. In this study, the conditions of embryogenic calli induced from nucellar tissue for promotion of plant regeneration in satsuma mandarin were investigated. The calli of I, II and III line were divided into two sizes of 0.5 mm and 1.0 mm in diameter and two weight gradients of percoll at 40% and 50% though the filter mesh. The frequency of embryo formation from $\phi$ 1.0mm-40% was slightly higher than callus that from others. Adventitious embryoids developed to a globular stage were transferred to regeneration medium. In 'Miyagawa Wase', the embryos from I and II line developed into a heart stage from most of $\phi$ 0.5 mm- 40% and $\phi$1.0 mm-40% calli, but it failed in 'Sugiyama Unshu'. In the cultivar of 'Miyagawa Wase', 63% of adventitious embryos transferred to the regeneration medium developed into the heart stage from the most $\phi$ 1.0 mm-40% calli of I line, but of 'Sugiyama Unshu' failed in some calli condition. The embryoids from two callus lines developed further to shoots and plantlets, while the embriods from III line abnormal failed to regenerate in the cultivar. From these results, it is suggested that the plant regeneration from embryogenic callus in satsuma mandarin could be affected by callus conditions.

Effects of Genetic Polymorphisms of ADD1 Gene on Economic Traits in Hanwoo and Jeju Black Cattle-derived Commercial Populations in Jeju-do (제주도 한우와 제주흑우실용화축군 집단에서 ADD1 유전자의 다형성이 경제형질에 미치는 영향)

  • Genetic polymorphisms of adipocyte determination and differentiation factor 1 (ADD1) gene were screened in Hanwoo and Jeju Black cattle-derived commercial (JBC-DC) populations. The ADD1 genotypes were determined using the presence/absence of 84-bp fragment at intron 7 region. The association of ADD1 genotypes for economic traits was examined in both populations. In the Hanwoo steers, ADD1 D/- carcasses showed significantly thicker backfat levels than those from WW (p<0.05). However, the thickest level of backfat appeared in WD heterozygotes, whereas thicker backfat did not appear in DD homozygotes in the JBC-DC population (p<0.05), leading to the supposition that synergic effects of alleles W and D increase backfat deposition. On the other hand, there was no association between the ADD1 genotypes and intramuscular fat deposition measured as meat quality index and marbling score. From these results, we concluded that the bovine ADD1 affected the backfat in subcutaneous tissue, rather than intramuscular fat in muscle tissue. In addition, the DD animals showed higher levels of meat color than those from W/- (p<0.05). Interestingly, a highly significant difference was found between the genotypes and carcass weights only in the JBC-DC population, and D/- animals were heavier by more than 38 kg than those from WW (p<0.001). The results of this study reveal faster growth rate and differences in steer productivity according to genotypes of the ADD1 gene. These findings demonstrate that ADD1 genotypes may effectively function as molecular genetic markers for the improvement of Hanwoo and Jeju Black cattle-related crossbreeding systems.

Efficacy of a petroleum spray oil for the control of citrus pests, spirea aphid (Aphis citricola) and two scales (lcerya purchasi and Planococcus cryptus) in Jeju island (감귤의 조팝나무 진딧물 및 깍지벌레류에 대한 Petroleum Spray Oil의 방제효과)

  • The efficacy of petroleum spray oil (PSO; D-C Tron $Plus^{(R)}$) against spirea aphid (Aphis citricola) and two scales (lcerya purchasi and Planococcus cryptus) on citrus was tested in Jeju, Korea. The control value of 0.33% PSO against spirea aphid was over 90% at 3 and 6 days after treatment (DAT), and then decreased to about 80% at 9 and 14 DAT. Spraying of PSO (0.33, 0.66, 1%) against cottony-cushion scale showed a much lower control value (less than 50%) during the investigation period compared to that of the conventional pesticide (Methion). When used against tile scale, Planococcus cryptus, the control value of 1% PSO was 95%, but that of 0.33% and 0.66% PSO only ranged from 50 to 80%, lower than the conventional pesticide. The results of this study, as with a previous study, suggest that spraying of 0.33% and 0.5% PSO not only controls the spirea aphid, but also reduces the density of two scales (lcerya purchasi and Planococcus cryptus) on citrus tree in Jeju, without phytotoxic damage.

Polymorphisms and Allele Distribution of Novel Indel Markers in Jeju Black Cattle, Hanwoo and Imported Cattle Breeds (제주흑우, 한우 및 수입 소 품종에서 새로운 indel 마커의 다형성과 대립인자 분포)

  • The aim of this study was to screen the polymorphisms and distribution of each genotype of insertion/ deletion (indel) markers which were found in a preliminary comparative study of bovine genomic sequence databases. Comparative bioinformatic analyses were first performed between the nucleotide sequences of Bovine Genome Project and those of expressed sequence tag (EST) database, and a total of fifty-one species of indel markers were screened. Of these, forty-two indel markers were evaluated, and nine informative indel markers were ultimately selected for population analysis. Nucleotide sequences of each marker were re-sequenced and their polymorphic patterns were typed in six cattle breeds: Holstein, Angus, Charolais, Hereford, and two Korean native cattle breeds (Hanwoo and Jeju Black cattle). Cattle breeds tested in this study showed polymorphic patterns in eight indel markers but not in the Indel-15 marker in Charolais and Holstein. The results of analysis for Jeju Black cattle (JBC) population indicated an observed heterozygosity (Ho) that was highest in HW_G1 (0.600) and the lowest in Indel_29 (0.274). The PIC value was the highest in HW_G4 (0.373) and lowest in Indel_6 (0.305). These polymorphic indel markers will be useful in supplying genetic information for parentage tests and traceability and to develop a molecular breeding system for improvement of animal production in cattle breeds as well as in the JBC population.

Analysis of Melanocortin Receptor 1 (MC1R) Genotype in Korean Brindle Cattle and Korean Cattle with Dark Muzzle (칡소와 비경흑색 한우의 Melanocortin Receptor 1 (MC1R) 유전자형 분석)

  • PCR-RFLP analysis was carried out to investigate the genotype of Melanocortin receptor 1 (MC1R) gene in Korean Brindle Cattle and Korean Cattle with dark muzzle, which are coat color and muzzle pigmentation variants of Korean Cattle, respectively. Allelic variants of MC1R in cattle were analyzed by digestion with BsrFⅠ, AciⅠ. Among six genotypes, $E^D/E^D,\;E^D/E^+,\;E^D/e,\;E^+/E^+,\;E^+$/e and e/e, detected in cattle, only two genotypes, $E^+/E^+\;and\;E^+$/e, were observed in Korean Brindle Cattle, probably reflecting the necessary of $E^+$ allele for the expression of black brindle coat color. As in Korean Cattle with light muzzle, the $E^+$/e and e/e genotypes were detected in Korean Cattle with dark muzzle. The $E^+$ and e alleles frequencies in two populations of Korean Cattle with dark muzzle and with light muzzle were 0.37, 0.63 and 0,11, 0.89, respectively. Although the frequency of $E^+$ allele in Korean Cattle with dark muzzle was higher than in Korean Cattle with light muzzle, the $E^+$ allele was not completely associated with dark muzzle pigmentation. The results of this experiment indicate that the difference of MC1R genotype and frequency may be useful for fixation of coat color in Korean Cattle as well as Korean Brindle Cattle.

  • According to the 4th BPE(Basic Plan for long-term Electricity Supply and Demand), Electricity demand of Jeju island in 2012 will be reached to 682MW. Jeju island will be faced with severe shortage of electricty, also needed to find transport route to supply green energy which will be made from "Carbon free island" energy policy toward land at that time. For that reason, The plan which construct 400MW size HVDC connection line was decided for potential supply stabilization and transportation of green energy. KEPCO organized special construction office not only to observe the successful project completion which aims 2011 December but also to build up professional manpower. KEPCO is putting spur to the HVDC project these days. Construction site of converter stations has been already confirmed and contracts of cable, converter have done as per turn-key early this year. On this report, we would like to discuss about project scheme, main characteristics of system, and the furture progress plan of "Jindo~Jeju HVDC construction project expecially converter station part" which is being pushed by KEPCO.

Effect of 11-ketotestosterone (11-KT) on Gonadal Sex Reversal and Spermatogenesis of Honeycomb Grouper Epinephelus merra

  • We investigated the androgenic effects of 11-ketotestosterone (11-KT) on gonadal sex reversal and spermatogenesis in honeycomb grouper Epinephelus merra by method of gonadal biopsy. 11-KT was injected intramuscularly at a concentration of 1 and $10{\mu}g$ body weight. The proportion of cross sectional area of the gonad occupied by each germ cell type was measured and compared pre- and post-injection group. During the sex change phase, the distribution ratio of oocytes was decreased in all fish of 11-KT treatment group while the distribution ratio of spermatocytes was increased than pre-injection group. In male phase, all fish of 11-KT treatment group shown the increased distribution ratio of spermatocytes, but the distribution ratio of spermatozoa was decreased than pre-injection group. The present results suggest that 11-KT can stimulate degeneration of oocytes, proliferation of spermatocytes and spermiation in honeycomb grouper.

A Molecular Genetic Analysis of the Introduced Wild Boar Species (Sus scrofa coreanus) on Mount Halla, Jeju Island, Korea (제주도 한라산에 서식하는 도입종 야생멧돼지에 대한 분자유전학적 분석)

  • An wild boar species which has been known as an extinct species on Jeju Island, was recently observed in the surrounding areas of Mount Halla. Based on the molecular techniques, this study examines whether they are crossbred with domesticated pig breeds. Intraspecific genetic relationships with other wild boar populations and molecular sexing were examined as well. Total of four molecular markers on mitochondrial DNA(control region and ND2) and nuclear DNA(MC1R and KIT) were applied to test crossbreeding between with domesticated pig breeds, such as Landrace, Large White, Berkshire, Hampshire, and Duroc. All individuals of wild boar population had identical mtDNA control region(CR) sequences. In addition, the sequences were the same as those of some native pig breeds which are distributed in Northeast China, but different from those previously reported from the Korean Peninsula up to date. These results suggest that this population may have originated from a genetic lineage had been not previously studied and genetically related to Chinese native pig breeds. Molecular sexing results show that there are twice as many females as male. Thus the population is under expansion and its size will dynamically increase if not controlled.

Community Structure of Macrobenthic Polychaetes and its Health Status (Assessed by Two Biotic Indices) on the Adjacent Continental Shelf of Jeju Island, in Summer of 2020 (2020년 하계 제주도 인근 대륙붕 해역의 저서다모류군집 구조 및 저서생태계 건강도 평가)

  • This study was carried out to investigate the benthic polychaetous community and benthic ecosystem quality status on the adjacent continental shelf of Jeju Island, and field surveys were conducted at 31 stations in July and August, 2020. The surface sediment was generally composed of muddy sand facies and sandy mud facies, and the average particle size was medium silt (6.1±1.6∅). The benthic polychaetous community revealed a total of 73 species with a mean density of 242 ind./m2. The major dominant species were Notomastus latericeus, Ampharete arctica and Onuphis shirikishinaiensis. By the cluster analysis and nMDS results based on species composition of the benthic polychaetous community, the study area was divided into three station groups arranged from east to west by the water depth and sedimentary facies. The station group located in the west was subdivided into two station groups from south to north. From results of correlation analysis and PCA, it was found that the benthic polychaetous community in the study area had a strong correlation with the sedimentary environment and water depth. The benthic faunal community (or ecosystem) on the adjacent continental shelf of Jeju Island was assessed to be in a healthy state by biotic indices such as AMBI and BPI.
