Brevibacterium sp.에 의한 L-leucine의 생산에 관한 연구

  • Biotin 요구성 glutamic acid 생산능을 가진 Br-evibacterium sp.의 isoleucine 영양요구성 변이주가 다량의 L-leucine을 생성함을 발견하였다. 본균주에 관한 탄소원, 질소원, 무기염, vitamin, amino acid 등의 영양물질과 온도, 공기주입량, pH등에 관한 실험으로 다음과 같은 결과를 얻었다. 본균주는 탄소원, 질소원으로서 10% glucose와 4% $(NH_4)_2SO_4를$ 공합하여 $30^{\circ}C,$ 88시간 배양시킨 결과, 액중에 21 mg/ml의 L-leucine이 축적되었다. Fe$^{2+}$$Mn^{2+}$의 동시첨가는 L-leucine생성에 효과적이었으며 thiamine 역시 L-leucine 생성에 효과적이었다. 30ι jar fermentor 에서 폐당밀을 주원료로 한 실험결과, 폐당밀, $(NH_4)/SO_4,$ $KH_2PO_4,$ $MgSO_4ㆍ7H_2O로$ 구성된 액체배지에서 $30^{\circ}C,$ 36시간 배양시킨 것이, 27mg/ml의 L-leucine이 축적되었으며 이때의 대당수율은 16%이었다.

영양요구성주 및 유사체 내성 대장균 변이주에 의한 L-스레오닌 생산 (Production of L-Threonine by Auxotrophs and Analogue Resistant Mutants of Escherichia coli)

  • 대장균 W3110으로부터 NTG 및 UV를 사용하여 여러 단계의 돌연변이 실험을 거치면서 스레오닌 고생산균주인 대장균 TF427를 선별하였다. 선별된 변이주는 스레오닌 유사체인 AHV 내성, 메치오닌 및 이소루이신 요구성을 특징으로 한다. 5-L 발효조 실험에서 44시간 발효하였을 때 46.5g/l의 스레오닌이 생산되었다. 효모분석에 의하면, TF427의 아스파토키나아제 I의 활성은 스레오닌에 의해 저해받지 않았으며, 이 효소의 합성은 스레오닌과 이소루이신에 의해 억제를 받지 않았다.

Corynebacterium sp. K-199가 생산하는 단백질성 생물응집제

  • About 600 microorganisms isolated from soil, marsh, compost, etc. were examined for their flocculating ability in the kaolin suspension and swine wastewater. Among them, K-199 was the best producer of flocculant and was identified to be a species belong to the genus Corynebacterium. The maximum production of the flocculant from Corynebacterium sp. K-199 was observed in culture medium containing 2% glycerol, 0.4% peptone, 0.3% K$_{2}$HPO$_{4}$, 0.1% CaCO$_{3}$, 0.05% NaCl, 0.1% yeast extract, 0.1% MgSO$_{4}$,7H$_{2}$O and initial pH 7.5 when cultured with rotary shaker controlled at 25$\circ$C and 150 rpm. Under the optimal culture condition with jar fermentor, the maximum production was reached to flocculating activity of 780 units/ml after 4 days. From the results of the activity be fully maintained by periodate oxidization, it suggests that the activity is due to the protein.

  • A strain producing extracellular endo-type inulase was selected from Actinomycetes isolated from soil, and identified as Streptomyces sp. The maximum inulase production was obtained with medium containing inulin 1.0%, yeast extract 1.0%, (NH$_4$)$_2$HPO$_4$ 0.4%, NH$_4$H$_2$PO$_4$0.8%, KCl 0.05%, MgSO$_4$ㆍ7$H_2O$ 0.05%, FeSO$_4$ㆍ7$H_2O$ 0.001% at 96 hours culture in jar fermentor. The endo-type inulase was considered to be an inducible enzyme produced by inulin only.

Production of a Platelet Aggregation Inhibitor, Salmosin, by High Cell Density Fermentation of Recombinant Escherichia coli

  • Optimal conditions for a high cell density fermentation were investigated in a recombinant Escherichia coli producing salmosin, a platelet aggregation inhibitor. The optimized carbon and nitrogen sources were glycerol 10 g/l, yeast extract 30 g/l, and bacto-tryptone 10 g/l, yielding the dry cell weight (DCW) of 10.61 g/l in a 500 ml flask culture. The late-stage induction with 1% L-arabinose in a 5 l jar fermentor showed the highest DCW of 65.70 g/l after 27 h of the fed-batch fermentation. Around 2,200 mg/l of the protein was expressed as an inclusion body that was then refolded to obtain the active salmosin of 96 mg/l. We also confirmed the inhibitory activity against platelet aggregation of the active salmosin from the high cell density fermentation.

  • 키토산을 생산하기 위하여 Cunninggamella blak-esleeana IFO 4443을 자외선조사하여 생성능이 우수한 Cunningmella blakesleeana IFO 4443-10의 변이주를 분리하였다. 본 변이주인 C. blakesleeana IFO 4443-10을 3$0^{\circ}C$에서 pH 4.5에서 3일간 발효하여 mycelia 발효액을 알칼리로 처리하여 chitosan을 얻었다. 이때 chitosan의 수율은 1012mg/l로 높았으며 탈아세틸화는 84.6% 이였다.

  • A fungal strain was isolated as a pullulan-producer from plant leaves and identified as Aureobasidium pullulan GM21. With A. pullulans GM21, culture conditions were optimized for the pullulan production and the changes of the molecular weight of pullulan produced were investigated according to the culture conditions we obtained maximum conversion yield of pullulan about 58-60%(40.8-42.0g/l) from 7% sucrose at 25C, initial pH 7.5 by the batch cultivations either in Erlenmeyer flask or in jar fermentor.

Production of L-Ornithine by Citrulline Auxotrophic Mutants of Glutamate-Producing Bacteria

  • For the purpose of producing L-ornithine by microbial fermentation, mutant strains were developed from glutamate-producing bacteria by mutagenesis using N-methyl-N'-nitro-N-nitrosoguanidine (NTG) and UV irradiation. Brevibacterium ketoglutamicum BK1046, a L-citrulline auxotroph which is also resistant to arginine hydroxamate (Arghx), was isolated and selected as the best producer of L-ornithine. This strain was capable of producing L-ornithine at a concentration of 24 g/l after 69 hours of cultivation in the 21 jar fermentor. The optimum supplementary level of L-arginine, a substitute for L-citrulline, was found to be about 0.2 g/l in the shake-flask fermentation.

Lactobacillus jensenii YW-33이 생산하는 다당류성 생물응집제 및 생산조건

  • A bioflocculant producing bacterium for wastewater treatment was isolated and identified as Lactobacillus jensenii. The bioflocculant was supposed as a polysaccharide. Lactobacillus jensenii YW-33 produced the flocculant with highest activity in the medium composed of 2% sucrose, 0.05% tryptone, 0.5% yeast extract, 0.01% K$_{2}$HPO$_{4}$, 0.01% KH$_{2}$PO$_{4}$, 0.01% NaCl, 0.005% MnSO$_{4}$, 5H$_{2}$O and 0.2% Tween 80. The optimal culture pH and temperature were 7.0 and 25$\circ$C, respectively. In the time course of production with jar fermentor, the productivity of the flocculant was increased in proportion to the cell growth and the cultivation time for maximum production was 24 hrs, showing flocculating activity of 1,130 units per ml.

Effect of Aeration on Denitrification by Ochrobactrum authropi SY509

  • Aeration was found to affect the biological denitrification by Ochrobactrum authropi SY509. Although cell growth was vigorous under 1 vvm of aeration and an agitation speed of 400 rpm in a 3-L jar fermentor, almost no nitrate was removed. Yet under low agitation speeds (100, 200, and 300 rpm), denitrification occurred when the dissolved oxygen was exhausted shortly af-ter the inoculation of the microorganism. Ochrobactrum authropi SY509 was found to express highly active denitrifying enzymes under anaerobic conditions. The microorganism also synthesized denitrifying enzymes under aerobic conditions (1 vvm and 400 rpm), yet their activity was only 60% of the maximum level under anaerobic conditions and the nitrate removal efficiency was merely 15%. However, although the activities of the denitrifying enzymes were inhibited in the presence of oxygen, they were fully recovered when the conditions were switched to anaerobic conditions.
