Ethanol Fermentation of Fusant between Heterologous Transformant of Saccharomyces cerevisiae and Candida tropicalis in Mini-jar Fermentor Scale (Mini-jar fermentor Scale에서의 Fusant의 Ethanol 발효)

  • The optimum conditions for ethanol fermentation and ethanol productivity of the fusant ESC-14-15 were examined in a mini-jar formentor scale (working volume : 2.5 liters) to assess the possibility of practical application. Addition of yeast extract to fermentation broth greatly enhanced the ethanol productivity and shortened the period of fermentation. The pH 4.2 was more favorable than pH 5.5 with respect to ethanol productivity and fermentation speed. The optimum concentration of liquefied potato starch for ethanol fermentation of FSC-14-15 was 15%(w/v) and the corresponding productivity was 8.7%(v/v) of ethanol with an efficiency of 80.6% to the theoretical maximum. When the fresh fermentation broth containing 20% of liquefied potato starch was inoculated with love(v/v) of inoculum, the fusant FSC-14-75 produced 11.0%(v/v) of ethanol in 4 days, which is considered comparable to that from an industrial process. From the liquefied cassava starch or the equal mixture of liquefied barley and sweet potato starch prepared according to the same method as in the industrial process except saccharification step, the fusnnt FSC-14-75 produced 8.5%(v/v) or 7.6%(v/v) of ethanol in 4 days, respectively.

A Study in Packing of Changran-Jeotgal -1. Shelf-life of a Jar Packing in Changran-Jeotgai- (창란젓갈의 포장에 관한 연구 -1. 병포장 젓갈의 품질유지기한-)

  • Quality variation of jar packing of Chanran-Jeotgal produced by improved process and conventional process were investigated during storage at 10, 20, and $30^{\circ}C$, respectively. The increasing rate of pH, volatile basic nitrogen (VBN), $CO_{2}$, and viable cell counts of Chanran-Jeotgal produced by improved process was slower than that of conventional process at all storage temperature. Also the result of sensory evaluation indicate that the production of Changran-Jeotgal by improved process may extend the shelf·life of jar packing Chanuan-Jeotgal. From above results, relationship between quality parameters was concluded. In case of Changran-Jeotgal in a jar, pressure, pH, L-value, VBN and sensory score were highly correlated. Therefore, these parameters are expected to use as shelf-life indication.

  • The great jar estimated in Proto-Three Kingdoms period was found at the site from Pyeongtaek Daechuri and the bonding traces from pieces destroyed when they were excavated are observed. Therefore this study would figure out the components of bonding material used in great jars by optical microscope, FT-IR, py-GC/MS and the kind of textile attaching to great jar by transmitted light microscope. As a result of optical microscope for the cross-section, black material is uniformly applied between a piece of jar and the part of bonding and the textile line are observed. Black bonding material is resulted in korean rhus lacquer by FT-IR and py-GC/MS and the kind of textile is identified as hemp by transmitted light microscope. Materials for repairing archaeological objects are hardly known causing little information, but this study proves that the bonding material had used lacquer for great jars before Proto-Three Kingdoms period. Therefore if ancient bonding materials are systematically analysed in the further studies, it would be significant help for studying bonding and restoration materials of ancient objects.

Effects and Batch Kinetics of Agitation and Aeration on Submerged Cultivation of Ganoderma Iucidum (영지의 액체배양에 미치는 통기.교반의 효과와 동력학적 특성)

  • The effects of agitaion and aeration on mycelial growth, exo-polysaccharide (EPS) production, and substrate consumption upon the submerged cultivation of G. lucidum were investigated, and the batch kinetics of the EPS fermentation of G. lucidum were interpreted as function of agitation speed and aeration rate. In a 2.6 L jar fermenter system, the optimum agitation speed and aeration rate for EPS production were determined to be 400 rpm and 1.0 vvm, respectively. The maximum production of EPS obtained was 15 g/L. The logistic model for mycelial growth fitted the experimental data better than that determined by the Monod and the two-thirds power models. The Luedeking-Piret equation adequately modelled the kinetic data obtained for product and substrate.

  • A survey study was carried out on the preparation methods of the Winter Kimchies in Seoul. Chung Cheong and Kang Won area by questioning 203 homes Individually. The results showed that the cabbage Kimchi was prepared more than 95o of total Kimchi a good heads of the Chinese cabbage were selected as proper quality of the cabbage. The cabbage were brined for 7∼10 hours with using sun-dried salts mostly(66∼78%) In Seoul and Chung Cheong area the quality of Chinese cabbage was regarded as the primary factor and the seasoning materials in Kang Won area for good quality of Kimchi, more than 70eA of the residents prepared Kimchi in Kimchi jar, and the correlation between the jar use kind of resident such as house or apartment was significantly high (p< 0.000)

  • The characteristics of cell growth and cellulose production by Acetobcter xylinum BRC5 were studied in shaking flasks and jar fermentors. Of the substrates tested, the highest cellulose yield was obtained from the fructose medium. Some inductive cultivation was observed and then cellulose was produced with cell growth. When glucose alone or mixture of glucose and fructose was used as the carbon source, cellulose could be biosynthesized under the glucose limitation. Corn steep liquor (CSL), as a low-cost nitrogen source, was a good substitute for yeast extract. In a jar fermentor experiment, 4.14 g/l of disintegrated cellulose was obtained from 8% CSL- medium containing 0.5% glucose and 1.5% fructose.

  • In this paper, a simple power control scheme of Class-D inverter, which is varied duty cycle of fixed frequency to desired output power. It is more suitable and acceptable for high-frequency induction heating (IH) jar applications. The proposed control scheme has the advantages of not only wide power regulation range but also ease to control output power. Also it can achieve the stable and efficient Zero-Voltage-Switching (ZVS) in whole load range. The control principles of proposed method are described in detail and its validity is verified trough simulations results on 38.5kHz IGBT for induction heating rated on 1.6kW with constant frequency variable power.

  • In this paper, a new control scheme of Class-E inverter for Induction Heating (IH) Jar applications with clamped voltage characteristics using Pulse-Frequency-Modulation (PFM) is introduced. To reduce the voltage stress of switch, the proposed PFM control scheme doesn't need any auxiliary circuit in comparison to a family of Active Clamped Class-E (ACCE) inverter. It can decrease voltage stress of switch through modulation of switching frequency. The Class-E inverter using the proposed control scheme has the advantage of not only the same output power when it is compared with a Hybrid-Active Clamped Class-E (Hybrid-ACCE) inverter but also Zero-Voltage-Switching (ZVS), which are characteristics of conventional Class-E and ACCE inverter. The control principles and analysis of proposed method are explained in detail and its validity is verified through simulation and experimental results.

  • Colistin produced from Penibacillius polymyxa was widely used as an antibiotic active against gram-negative bacteria and as feed additive. This research studied on increment of colistin productivity by mutation of P. polymyxa. As a result, several mutants were obtained from the strain by UV radiation and NTG treatment. They produced approximately 8.5${\sim}$9.0 g/L of colistin in flask and jar culture. Colistin productivity of the mutant, named Penibacillius polymyxa CBY, showed 100 times than that of wild type. When Penibacillius polymyxa CBY fermented in the optimal medium, it produced up to 18 g/L of colistin in jar fermentation.
