Influence of Magnetite Particles on Coagulation in Wastewater Treatment (자철광을 이용항 폐수처리시 응집효율에 미치는 영향)

  • In this study are investigated to the optimal coagulation conditions and magnetite which was used as coagulation-aids. To accomplish this study, analysis of water quality, removing tubidity and SS from wastewater were measured with jar-tester. The results were obtained that waste water requires 10g magnetite, 100mg aluminum sulfate, and pH valuse was 6~9.

  • To analyze food material and table wares in daily reception dishes of Choson Dynasty, studied historic book 'Young jeob Dogam Zabmulsek Euigwae' (1609, 1643 year) described the daily reception dishes for Chinese envoy in Choson Dynasty. The results obtained from this study are as follows. 1. Food material for reception Chinese envoy were vegetable, pickled vegetable, soup (?水), brown seaweed, kelp, green seaweed, garlic, pine mushroom, mushroom, driedfish, fish egg, shrimp, pork, pheasant, fowl, salted fish shrimp and etc, various fruits preserved in honey, green peajelly, buck wheat jelly and bean curd. 2. The table wares were rice bowl(鉢里), small bowl of porcelain(甫兒), water bowl(大貼), plate(貼是), small dish(鍾子), small earthen ware jar(東海), jar(缸), pottery(甕), table(盤), chopsticks(?), washbowl(洗面盆), earthen ware steamer(甑), kettle(釜), brazier(爐), measuring cup(升), unit of measure(合), meter(稱子), spoon(匙), chest (函), lid(蓋), large rice bowl(所羅) and bowl(椀).

Addition of Coagulants for Phosphorus Removal from Combined Sewer Overflows (CSOs) (합류식 하수관거 월류수의 인제거를 위한 응집제 투여)

  • The coagulation of combined sewer overflows ($CSO_{s}$) was investigated by jar-testing with several commercial coagulants. $CSO_{s}$ sample showed different characteristics of coagulation from secondary wastewater with three common coagulants, aluminum sulfate, ferric chloride and polyaluminum chloride (PACl). Jar-tests showed that relatively wide range of optimal SS and T-P removal yielded with alum and ferric chloride compared with cationic polymers, though efficient SS and T-P removal can be achieved with all three coagulants. The decrease of pH was caused by the increase in dosage of aluminum sulfate, ferric chloride and PACl as coagulants. The pH was changed from 7.0 to 4.7 with the dosages of ferric chloride 25 mL/L. Aluminum sulfate revealed pH of 5.0 and PACl was highest pH of 5.4 after dosing of coagulants. The optimal pH to treat $CSO_{s}$ with aluminum sulfate were 6-6.5; with PACl 6-7, and with ferric chloride higher than 7.

Effect of Glycine on L-Ornithine Production by a Citrulline Auxotroph of Brevibacterium ketoglutamicum and Stoichiometric Analysis

  • The effects of glycine on cell growth and L-omithine production were investigated in shake-flask and jar fermentor cultures of a citrulline auxotrophic mutant, Brevibacterium ketoglutamicum BK 1046. In the shake-flask culture, the optimal concentration of glycine for L-ornithine production was found to be 20 g/l. In the jar fermentor culture with the glycine at an initial concentration of 20 g/l, L-ornithine production increased by 28%, compared to that of the culture with no glycine added. 37 g/l of L-ornithine was produced when additional feeding of glycine (5 g/l) was made. This was a significant improvement in L-ornithine production compared to that (ca. 24 g/l) of the corresponding batch culture conducted without glycine. According to the stoichiometric analysis with the batch fermentation results, the experimental and theoretical L-ornithine yields based on the glucose consumption were 0.24 and 0.59, respectively. This indicates that the performance of L-ornithine fermentation can further be improved by the supplementation of glycine and the development of a mutant strain possessing a higher growth yield.

  • Bacillus polyfermenticus SCD which is commonly called as 'Bispan strain' has been appropriately used for the treatment of long-term intestinal disorders, since the live strains in the form of active endospores can successfully reach the target intestine. The goal of this study were to get the novel bispan mutant strain and to get higher spore number of bispan mutant in submerged fermentation. Bispan mutant KD21 was obtained using NTG mutagenesis of bispan strain, and bispan mutant was cultivated in 5 L jar fermenter. In this study, productivity and sporulation rate of the bispan mutant KD2l showed approximately 77% increase when compared with bispan strain.

  • The optimal temperature, pH and aeration rate for spore production by Bacillus polyfermenticus SCD in 500 ml shake flask and 5-1 jar fermenter were found to be $32^{\circ}C$, 7.0 and 1.0 vvm. respectively. When batch culture processes was performed under optimized culture conditions. viable cells were $3.3{\times}10^{10}$ CFU/ml and spore cells were $3.3{\times}10^{10}$ CFU/ml. Fed-batch culture processes were also examined with regard to higer maximum viable cell and spore production. The highe viable cells and spores were obtained in 5-1 jar fermenter at 72 h cultivation time by strategy in an intermediate feeding mode with 60% glucose solution 150 ml and 5% soybean flour solution 150 ml fed to the fermenter twice, and the productivity of spore cells was significantly increased. Finally. volumetric productivity of spore cells on fed-batch culture indicated $9.9{\times}10^8$ CFU/ml/h, which was approximately 2 times higher than batch culture. Thus, fed-batch culture show a promise as an industrial production method.

  • In the batch culture experiments, the addition of $MnSO_4$, was examined in flask culture, and then the optimal amounts of $MnSO_4$ was investigated in 2.5- L jar-fermenter. As a results, 0.005% $MnSO_4$ was found to enhance the ARA yield of 1.14- fold. Also the addition of $KH_2PO_4_4$ was investigated in 2.5-L jar-fermenter and the ARA yield was enhanced 1.20-fold. Fed batch culture study shown a relatively high productivity of cell mass (62.1 g/L) and ARA content (12.0 g/L) when 14% ammonia solution were alternatively used to control the pH and nitrogen source in the cultivation period.

  • A Saccharomyces cerevisiae isolated from a feed additive yeast product was cultivated in a Korean paper digestion wastewater in order to investigate the possibility of using it as substrate for the yeast. The yeast couldn't grow in the wastewater. It could grow in the wastewater diluted and the optimum dilution rate was 7.5 In batch cultivation with the jar fermenter the maximum total cell count was $1.34{\times}107/mL$ was obtained by the addition of undiluted digestion wastewater. By adding $(NH_4)_2S0_4 and KH_2P0_4$together with the undiluted wastewater the maximum cell concentration could be obtained faster.

  • Two culture modes, continuous and semi-continuous, of the decolorization fungus, Geotrichum candidum Dec 1, were compared to obtain a high treatment efficiency of molasses decolorization and a large productivity of peroxidase (DyP) to simultaneously decolorize dyes and molasses. The continuous culture of G. candidum Dec 1 using a 5-I jar-fermentor showed high DyP activity at a low dilution ratio of $0.005h^{-1}$, and decolorization ratio of molasses of 80% was obtained concomitantly. Therefore, a semi-continuous culture was performed by repeated refill and draw. In this mode, approximately 1.5 liters of the culture broth was replaced per cycle when the decolorization ratio of molasses was near 80%. The molasses medium (1.0 liter per day) was treated and the peroxidase productiveity in the drawn culture broth was 26.6U/day, whereas the peroxidase productiveity was 17.9U/day in the continuous culture with a dilution rate of $0.005h^{-1}$. The semi-continuous treatment system was an efficient decolorization method for the strain, G. candidum Dec 1.
