  • This experiment was performed with the purpose of finding out the effect of simulated acid rain at various pH levels on the morphology and enzyme of Perilla frutescens var. japonica hara. The pH levels of simulated acid rain ranged from pH 2.0 to pH 6.0. The experiment showed the anion concentrations in the order of $SO_4^{2-}$, Cl^-$, $NO_3^-$, and $F^-$, $SO_4^{-2}$ was found out to be the main factor which contributed to the rainwater acidification. A general decrease of growth in Perilla frutescens var. japonica hara growth was shown with the decreas of pH concentration. As acidity increases a definite reduction in the rates of germination, heigth of plant, malate dehydrogenase, and 6-phosphogluconate dehydrogenase was ovserved, but the density of spots on the leaf apex was increased.

  • We have investigated the vegetation in the peninsula located in Ongjeong-Li Seomyeon in Youngweol-gun. Since this is a limestone area, boxtrees, Abelia mosanensis T. H. Chung ex Nakai, and Selaginella stauntoniana Spring were abundant, as these are types of vegetation characteristic of limestone areas. Pinus densiflora S. et Z. was the most dominant woody plant, but vegetational changes, such as the dominance of Quercus variabilis Blume, Quercus dentata Thunb, and Quercus mongolica Fischer, were observed in spots. In particular, the growth of nursery plants of pinus densiflora S. et Z. was not observed, and young trees like Quercus mongolica Fischer grow as low vegetation of Pinus densiflora S. et Z.. Thus, the ecological succession of Quercus mongolica Fischer is predicted. Outside the investigation area, as herbaceous plants, various vegetations were observed, including Selaginella stauntoniana Spring, Thalictrum coreanum H. Lev., Mukdenia rossii(Oliv.) Koidz., Platanthera freynii Kraenzl., Cephalanthera longibracteata Blume, Potentilla dickinsii Franch., Patrinia rupestris(Pall.) Juss., Swertia pseudochinensis H. Hara, Vicia venosa(Willd.) • This study was conducted to develop gibberellin treatment technique to enhance flower initiation in Aquilegia japonica Nakai & H. Hara. Seedlings were planted in 12cm-diameter pots on October 2016 and grown in green house. Ambient temperature in the green house was set at minimum $15^{\circ}C$ during day and night to suppress flower initiation at cold temperature condition. Two different types of gibberellin, $GA_3$ and $GA_{4+7}$, at 4 different concentration levels 100, 200, 400 and 600 mg/L, were tested in this study. Gibberellin was sprayed first at planting and secondly at 1-week after planting. Ten to fifteen ml of gibberellin was sprayed for each pot. Plant height and petiole length were elongated by both gibberellin types, flowering was more enhanced by $GA_3$ (91.7~100%) compared to of $GA_{4+7}$. However, abnormal flower was less observed in $GA_3$ treatment (0~16.7%) than $GA_{4+7}$. Number of flower stalks per plant ranged from 1.9 to 2.5. Number of flowers per plant ranged from 6.8 to 10.3. Differences in flowering characteristics between treatments were statistically significant. Optimal gibberellin treatment to enhance flower initiation in A. japonica Nakai & H. Hara substituting cold treatment was $GA_3$ at the concentration between 400 mg/L to 600 mg/L.

  • Objectives : Thymus quinquecistatus var. japonica (H.Hara) is a member of the genus Thymus of perennial aromatic herb, and it's designated as a natural monument of South Korea. It has traditionally been known to have protective or therapeutic effects on various human disease including cerebrovascular and neurological disease. Recently it was suggested that essential oil extracted from thyme has anti-fungal and anti-bacterial effect. The aim of this study is to investigate anti-inflammatory effect of Thymus quinquecistatus var. japonica in Raw 264.7 macrophage cell line. Methods : The cytotoxic effects of water and 70% ethanol extracts from Thymus quinquecistatus var. japonica, was tested using MTT assay. Inhibitory effects of the extracts to nitric oxide production and mRNA expression of inflammatory cytokines were examined by RT-PCR. Also, MitoSOX-red assay and JC-1 assay were performed to determine if the extracts can inhibit mitochondrial ROS accumulation and maintain mitochondrial membrane potential. Results : In LPS-induced inflammatory response, the extracts efficiently reduced nitric oxide NO production through inhibiting mRNA expression of iNOS enzyme. In addition, expression of the proinflammatory cytokines, IL-1𝛽 and IL-6, was also down-regulated by the extract treatments. Excessive accumulation of mitochondrial ROS induced by LPS was inhibited in the extract treated cells, which finally protected mitochondrial membrane potential. Conclusions : These results showed that water and 70% ethanol extracts from Thymus quinquecistatus var. japonica have anti-inflammatory effect through down regulation of IL-1𝛽, IL-6, and iNOS, and also have antioxidative effect against mitochondrial ROS accumulation that promote inflammatory response.

  • A simple GC method with a FID detector was developed in order to determine two main compounds (${\beta}$-sitosterol and lupenone) for Adenophorae Radix. ${\beta}$-Sitosterol and lupenone were analyzed by the gradient thermal ramping method. Nitrogen was used as the carrier gas at 108 kPa. The flow rate of gas was 2.0 mL/min; $2{\mu}L$ of filtered sample was injected at a split ratio of 1 : 80. This method was fully validated with respect to linearity, precision, accuracy and robustness. Further, this GC-FID method was applied successfully in order to quantify two compounds in an Adenophorae Radix extract. The GC analytical method for classification analysis was performed by repeated analysis of 59 reference samples in order to differentiate between Adenophora triphylla var. japonica Hara and 14 Codonopsis lanceolata. The results indicate that the GC-FID method is suitable and reliable for the quality evaluation of Adenophorae Radix.
