Genetic Similarity and Difference between Common Carp and Israeli Carp (Cyprinus carpio) Based on Random Amplified Polymorphic DNAs Analyses

  • Common carp (Cyprinus carpio) and its aquaculture breed Israeli carp samples were obtained from two separate aquaculture facilities under the similar raising conditions during two years in the Kunsan National University, Korea. Genomic DNA was isolated from the common carp and Israeli carp for identification of genetic characteristics and genomic polymorphisms by polymerase chain reaction amplification of DNA using arbitrary primers. The arbitrary primer No.21 (ACTTCGCCAC) yielded the highest number of fragments with the average of 15.0 among the primers used in Israeli carp. A tota1 of 294 polymorphic products in common carp and 336 in Israeli carp were observed by random primers. The average number of polymorphic products generated by random RAPD primer No. 2 (GTAGAC-CCGT) showed 8.0 in Israeli carp. On average, each random RAPD primer produced 5.4 amplified polymorphic products in common carp and 6.2 in Israeli carp. An average genetic similarity (BS value) was 0.44$\pm$0.05 within the common carp and 0.32$\pm$0.04 within the Israeli carp. The degree of similarity frequency (BS) between two carps was 0.67 as generated by the primer No. 19 (GACGGATCAG). The average level of bandsharing was 0.57$\pm$0.03 between the two carps. Accordingly, the two carp populations were genetically a little distant. The electrophoretic analysis of PCR-RAPD products showed middle levels of variation between the two carp populations. This result implies that the genetic diversity among intra-population may be higher when compared with that between the two carps. The RAPD polymorphism generated by these random primers might be used as a genetic marker for populations or lines identification in important aquacultural carp.

Histological Changes of Oocytes Development and Hormone Levels in the Israeli carp, Cyprinus carpio, according to Season (이스라엘 잉어(Cyprinus carpio) 난모세포의 계절에 따른 조직학적 변화 및 호르몬 수준의 변화)

  • This study was undertaken to understand development stage of ovary and changes of hormones concentration from February to May in the Israeli carp, Cyprinus carpio. The results obtained in these experiments are as follows ; 1. Serum LH levels began to increase sharphy in March, coinciding with the onset of rapid ovarian development. 2. LH levels were well correlated with changes in gonadosomatic index. 3. Dramatic increase in gonadosomatic index occurred during the months of March. 4. Ripe stage(Stage Ⅴ) rapidly increase in March. 5. Early perinucleolus oocyte radpily develop into late perinucleolus oocytes in March. 6. The vitellogenic phase begins as these late perinucleolus oocytes become transformed into early maturing oocytes through the accumulation of yolk. 7. The cytoplasm completely fills with yolk as oocytes reach the late maturing stage. 8. Changes in the microscopic appearances of the ovaries were well correlated with changes in both gonadosomatic index and macroscopic appearance. 9. It is concluded from these observations that LH plays a major role in sexual maturation of the Israeli carp.

First report of Aeromonas veronii infection in farmed Israeli carp Cyprinus carpio in Korea

  • In May 2007, mass mortality of Israeli carp, Cyprinus carpio L., was occured on a pond farm located in Jeollabukdo Province, Korea. The mortality rates reached up to 2% of the total fish in the farm per day. Typical clinical signs were abdominal distension, reddish foci on the skin, enteritis, liver congestion and enlarged spleen and kidney. On the basis of biochemical characteristics and 16S rDNA sequence, the causative bacteria isolated from affected carp were identified as Aeromonas veronii. Histologically, degeneration of hepatocytes and congestion in sinusoids were observed in the liver. Spleen showed hemorrhage and the destruction of the sheathed tissues. In kidney, necrotized renal tubules and glomerular destructions were observed. Intestinal tissues revealed necrotized and severe hemorrhage. Mass hemorrhage was observed in muscles. This is the first report that A. veronii caused mortality in cultured Israeli carp in Korea.

Optimal Dietary Protein and Lipid Levels for Growth of Juvenile Israeli Carp Cyprinus carpio

  • A feeding trial of four dietary protein levels (20, 30, 40, and 50%) and two lipid levels (7 and 14%) with a factorial design was conducted to determine the optimal dietary protein and lipid levels for juvenile Israeli carp Cyprinus carpio. Triplicate groups of fish (average body weight, $1.3{\pm}0.02g$) were fed the experimental diets for 9 weeks. Survival of fish was not affected by either dietary protein or dietary lipid level. Weight gain and feed efficiency increased as dietary protein levels increased up to 40 and 50%, respectively. Weight gain was higher in fish fed the high-lipid diets with 20 and 40% protein content. Feeding efficiency increased as the dietary lipid level increased for the 30, 40, and 50% protein diets. Daily feed intake decreased with increasing protein level and the minimum feed consumption was observed in fish fed the 50% protein diet with 14% lipid content. Moisture and lipid contents of the whole body were affected by both dietary protein and lipid levels. The crude lipid content of fish fed the 14% lipid diet was higher than that of fish fed the 7% lipid diet at each protein level. The results of this study indicate that a diet containing 40% protein with 14% lipid content is optimal for the growth and effective protein utilization of juvenile Israeli carp.

  • We reported an outbreak of Aeromonas (A.) veronii responsible for ulcerative dermatitis in Israeli carp (Cyprinus carpio). The major clinical signs were darkening body, abdominal extension, exophthalmos and severe ulcerative necrosis in the skin. The necropsy showed yellowish ascites, necrosis in liver and enlargement of kidney and spleen in the morbid fish. In blood agar for culturing bacteria, three different colonies were identified as A. veronii, Plesimonas shigelloides and Shewanella putrefaciens by phylogenetic identification using 16S rRNA or gyrB gene sequences. A. veronii was the most dominant species among them and was resistant to ampicillin, nalidixic acid and oxytetracycline.

Growth Comparison of Israeli Carp (Cyprinus carpio) to Different Breeding Combination

  • The purpose of this study was to investigate the improvement of growth in Israeli carp (Cyprinus carpio), and the cross experiment was carried out with two strains of Israeli carp. Four combinations of Israeli carp from Jeonbuk fisheries farm and Songpu mirror carp from Heilong Jiang, China (KK; Jeonbuk ♀ ${\times}$ Jeonbuk ♂, KC; Jeonbuk ♀ ${\times}$ China ♂, CC; China ♀ ${\times}$ China ♂ and CK; China ♀ ${\times}$ Jeonbuk ♂) were developed and reared. Body length, body weight and condition factor were determined at 20, 40, 60 and 170 days post-hatch (DPH). The results showed that there were differences in growth rate of the four groups. Body length of four groups were CK > CC > KC > KK and body weight were CC > CK > KC > KK at 170 DPH. The growth perfomance of four groups were statistically significant difference (P<0.05). During the rearing, CC group had longer length and higher weight at 170 DPH compared to other three groups and also condition factor was highest in the CC group, but there was no significant difference in a survival rate. These results indicated that the growth performance mainly depended upon brooder combination but survival rate could not significantly affect brooder.

Comparison of Acute Toxicity of Pesticides between Carp(cyprinus carpio L.) and Israeli Carp(Cyprinus israeli carpio L.) (농약(農藥)에 의한 참잉어 및 이스라엘잉어의 급성독성비교(急性毒性比較))

  • This study was carried out to compare the acute toxicity(96hr) of 13 chemicals to carp (Cyprinus carpio L.) and israeli carp (Cyprinus israeli carpio L.) and the activities of acetylcholinesterase (AchE) and glutathione S-transferase (GST) in israeli carp exposed to five insecticides (diazinon, malathion, carbofuran, cartap, methomyl). $LC_{50}$ values of acute toxicity of the chemicals to israeli carp were endosulfan 0.0061ppm, captafol 0.041ppm, chlorothalonil 0.073ppm, butachlor 0.48ppm, captan 0.14ppm, carbofuran 1.13ppm, cartap 1.15ppm, diazinon 1.35ppm, nitrofen 3.72ppm, methomyl 4.39ppm, propanil 10.61ppm, malathion 11.78ppm and isoprothiolane 12.81ppm. The acute toxicity of endosulfan 0.0061ppm was 2100 times higher than that of isoprothiolane 12.81ppm. $LC_{50}$ values of acute toxicity of the chemicals to carp were endosulfan 0.0026ppm, captafol 0.062ppm, chlorothalonil 0.078ppm, captan 0.14ppm, and butachlor 0.47ppm, carbofuran 0.52ppm, nitrofen 0.58ppm, diazinon 0.81ppm, cartap 0.82ppm, methomyl 5.03ppm, propanil 10.67ppm, malathion 11.92ppm, and isoprothiolane 13.20ppm. The acute toxicity of endosulfan was 5,000 times higher than that of isoprothiolane. The toxicity of diazinon, carbofuran, cartap, endosulfan, and nitrofen to carp was approximately 2-6 times as high as that to israeli carp, but the toxicity of malathion, methomyl and captafol to israeli carp was slightly higher than that to carp. AchE activity was inhibited by 31% and 52% after 96hr’s exposure of israeli carp to diazinon and malathion respectively. GST activity in israeli carp was significantly induced by methomyl exposure for 96 hr.

Genetic Stock Identification of Common Carp (Cyprinus carpio) by Detection of Intraspecific DNA Sequence Variation in the Mitochondrial 12S rRNA Gene (미토콘드리아 12S rRNA 유전자 변이 조사를 통한 잉어(Cyprinus carpio)의 유전학적 동정)

  • Intraspecific sequence variation was detected by polymerase chain reaction (PCR) and direct sequencing of a 350-nucleotide region of the mitochondrial 12S rRNA gene of two natural populations (Han River and Nakdong River) and one hatchery stock (Jinhae Inland Fisheries Institute) of local strain common carp, one Israeli strain of common carp stock from Pukyong National University (PKU), and one hybrid between Israeli strain of common carp female and local strain common carp male from PKU stock. There is little variation in 350 bases of the mitochondrial 12S rRNA gene sequences among 2 natural and 1 hatchery local strain common carp populatins, representing abut 7 to 20 nucleotide differences (less than 6%). The sequence of specimens from Han River was more similar to that from Nakdong River (identity=98.0%) than to that from Jinhae Inland Fisheries Institute (identity=96.3%). Sequence variation between Israeli strain and wild local strain common carp was higher than the variation within natural stocks. The level of variation was ranged from 15.7 to 17.7%. The hybrid showed very similar nucleotide4 sequence of 12S rRNA gene to the sequence of Israeli strain with the identity of 98.9%.

Optimization of Vegetable Rice Porridge Preparation with Israeli Carp Cyprinus carpio (향어(Cyprinus carpio) 함유 야채죽의 제조공정 최적화)

  • This study optimized the vegetable rice porridge preparation process with Israeli carp Cyprinus carpio (VRP-IC). The optimum heating period for Israeli carp paste (IC-P) in a retort was 120 min as per yield results. Response surface methodology was performed with IC-P/[rice (glutinous rice: non-glutinous rice=6:4) (Rice, RI)+vegetable (carrot: zucchini=1:1) (vegetable, VE)] (X1) and RI/VE (X2) as independent variables; viscosity (Y1), hardness (Y2), amino nitrogen (Y3), and overall sensory acceptance (Y4) as dependent variables. Optimum rates of IC-P, RI, and VE were 47.37%, 24.49%, and 28.15%, respectively; predicted multiple response optimum values for the dependent variables were 20,150 mPa·s, 2,002.9 N/m2, 193.1 mg/100 g, and a 6.4 score for Y1, Y2, Y3, and Y4, respectively. Under optimum conditions, experimental values for Y1, Y2, Y3, and Y4 were 20,359.8±191.9 mPa·s, 2,015.0±9.6 N/m2, 188.1±4.9 mg/100 g, and a 6.5±0.2 score, respectively, which were not significantly different from the predicted values (P < 0.05). Results of F0 value, viscosity, hardness, volatile odor intensity, and browning suggested that the optimum sterilization period was 25 min. VRP-IC prepared under the optimum conditions was superior to commercial vegetable rice porridges in sensory evaluations.

Genetic Similarity Frequency and DNA Polymorphism between Common Carp and Israeli Carp Using Polymerase Chain Reaction-Random Amplified Polymorphic DNAs

  • Common carp (Cyprinus carpio) and Israeli carp(C. carpio) samples were obtained from a aquaculture facility in the Kunsan National University, Korea. Genomic DNA was isolated from the common carp and Israeli carp representing genetic characteristics and genomic polymorphisms by polymerase chain reaction amplification of DNA as arbitrary primers. There were observed a total of 90 species-specific genetic markers within Israeli carp. On average, each random RAPD primer produced amplified 7.9 products from 1 to 17 bands. An average genetic similarity within Israeli carp showed -.60$\pm$0.05. The average level of bandsharing was some 0.57$\pm$0.03 between common carp and Israeli carp. Accordingly, two carp species were genetically a little distant. The electrophoretic analysis of PCR-RAPD proudcts showed high levels of variation between two fish species. The RAPD polymorphism generated by primer may be used as a genetic marker for species or lines identification in important aquacultural carp.
