Sources Identification of Anthropogenic Pb in Ulleung Basin Sediments using Stable Pb Isotope Ratios, East/Japan Sea (동해 울릉분지 시추 퇴적물에서 안정 Pb 동위원소를 이용한 Pb의 기원 추정)

  • This study investigated temporal and spatial variation of Pb and stable Pb isotopes accumulated in Ulleung Basin core sediments (4) using MC ICP/MS in order to identify the sources of anthropogenic Pb in the East/Japan Sea. Leached (1M HCl) Pb concentration and isotope ratios ($^{207}Pb/^{206}Pb\;and\;^{208}Pb/^{206}Pb$) were nearly constant during 300 yrs past than 1930, but increased up to twice in concentration and as much as 3.41% (1.70%) after 2000. On the other hand, residual Pb concentrations were nearly constant for past 400 yrs. The accumulation rates of anthropogenic Pb in the basin area were in the range of $3.1-3.5mg/m^2/yr$, which were similar levels to total atmospheric Pb deposition fluxes from 1990s to the present. In the slope area, more increase of anthropogenic Pb accumulation than the levels expected from mass accumulation rate could be found after the middle of 1990s. From the detailed evaluation for the temporal and spatial variation of accumulation rate and isotope ratios of anthropogenic Pb, we proposed probable sources and pathways of anthropogenic Pb. Pb emmision by coal burning from the China and Korea initiated the accumulation of anthropogenic Pb in the sediments of East/Japan Sea from 1930s. The accumulation of Pb increased by the addition of anti-nocking agents from both countries untill the beginning of 1990s, but from the middle of 1990s to the present, the phase-out of gasoline additives and the rapid increase of coal burning from the China maintained the atmospheric Pb levels in the Ulleung basin nearly similar to before. However, the local sources within this basin might take an important role in the rapid increase of anthropogenic Pb accumulation in slope areas from the middle of 1990s.

Differential Protein Quantitation in Mouse Neuronal Cell Lines using Amine-Reactive Isobaric Tagging Reagents with Tandem Mass Spectrometry

  • The high-throughput identification and accurate quantification of proteins are essential strategies for exploring cellular functions and processes in quantitative proteomics. Stable isotope tagging is a key technique in quantitative proteomic research, accompanied by automated tandem mass spectrometry. For the differential proteome analysis of mouse neuronal cell lines, we used a multiplexed isobaric tagging method, in which a four-plex set of amine-reactive isobaric tags are available for peptide derivatization. Using the four-plex set of isobaric tag for relative and absolute quantitation (iTRAQ) reagents, we analyzed the differential proteome in several stroke time pathways (0, 4, and 8 h) after the mouse neuronal cells have been stressed using a glutamate oxidant. In order to obtain a list of the differentially expressed proteins, we selected those proteins which had apparently changed significantly during the stress test. With 95% of the peptides showing only a small variation in quantity before and after the test, we obtained a list of eight up-regulated and four down-regulated proteins for the stroke time pathways. To validate the iTRAQ approach, we studied the use of oxidant stresses for mouse neuronal cell samples that have shown differential proteome in several stroke time pathways (0, 4, and 8 h). Results suggest that histone H1 might be the key protein in the oxidative injury caused by glutamate-induced cytotoxicity in HT22 cells.

A Geo-statistical Assessment of Heavy Metal Pollution in the Soil Around a Ship Building Yard in Busan, Korea (통계지표를 활용한 부산지역 조선소 주변 토양 내 중금속 오염조사 연구)

  • With the increase of metal usage in various industries, metal pollution and ecological toxicity in the environmental system have become a significant concern. A geo-statistical index has been widely used to determine contamination level with normalization through a background value. In this study, geo-statistical indexes such as an enrichment factor, accumulation index, and potential ecological risk index were used to assess metal pollution in soil at locations associated with shipbuilding manufacturing industries. Metal contamination, especially of Cu and Pb, was observed in some samples located closer to manufacturing sites. Enrichment factor and accumulation (IGEO) values were indicative of concerning levels of soil contamination in specific samples, and the soil contamination could be induced by anthropogenic sources. In further study, after more detailed sampling for soil and potential pollution sources, high interpretation techniques such as Pb isotope analysis and X-ray analysis will be needed to investigate source identification.

An Analysis of Phenolic Compounds, Carbon Isotopes, and Sugar Components of Whiskys Based on the Period of Maturation (위스키의 종류와 숙성기간에 따른 페놀성 화합물, 탄소동위원소비 및 당 성분 분석)

  • This paper analyzes phenolic compounds, carbon isotopes, and sugar components of whiskys based on the maturation period. For this, the paper considers a total of 40 whiskys(mainly imports) distributed in Korea. It is important to analyze the presence phenolic compounds(e.g., furfural, syringaldehyde, vanillin, syringic acid, and vanillic acid) because these are found only in whiskys ripened in oak. The results indicate that the total content of phenolic compounds increased with the increase in the storage period regardless of the type of whisky. In terms of vanillin/syringaldehyde(V/S), Scotch whiskys had 0.4~0.5; American whiskies, 0.30~0.34; and Canadian whiskies, 0.31~0.33. In terms of Scotch whiskys, Macallan had 0.25~0.34, making it unique among Scotch whiskys. In terms of the ratio of carbon isotopes, there were clear differences between malt Scotch whiskys, blended Scotch grain whiskys, American whiskys, and Canadian whiskys: -23.4~ -24.3, -16.8~-21.0, -11.0~-11.5 and -9.5~13.9, respectively. In addition, malt Scotch whiskys contained 40~230 $mg/{\ell}$ of fructose; blended Scotch whiskys, 20~120 $mg/{\ell}$; American whiskys, 50~70 $mg/{\ell}$; and Canadian whiskys, 20~100 $mg/{\ell}$, demonstrating that the fructose content of single-malt whiskys was twice the average fructose content. On the other hand, malt Scotch whiskys contained 30~170 $mg/{\ell}$ of glucose; blended Scotch whiskys, 20~120 $mg/{\ell}$; American whiskys, 20~30 $mg/{\ell}$; and Canadian whiskys, 10~110 $mg/{\ell}$, demonstrating that the glucose content of single-malt whiskys exceeded the average glucose content. This study's results can be used as a database of classification for whiskys based on the fermentation of raw ingredients and the period of maturation for distinguishing between different types of whiskys. In addition, the results can facilitate the verification of genuine whiskys by allowing for the identification of different types of whiskys based on the period of maturation.

Ecological Characteristics and Distribution of Fish in the Downstream Region of Gyeongan Stream (경안천 하류구간에 서식하는 어류의 분포 및 생태특성)

  • Fish field survey, especially fish distribution and their ecological characteristics, was performed in the downstream region of Gyeongan stream during the period of before (June) and after (October) the summer monsoon in 2010. Depending on the characteristics of each site, fyke net or casting net was used for fish sampling. Feeding classification was determined by the analysis of stomach contents. Total number of family and species sampled were 5 and 17, respectively. The dominant family was Cyprinidae (12 species), and relative abundance (RA) of the most dominant species, Lepomis macrochirus and Zacco platypus, was 38% and 24%, respectively. Exotic species and Korean endemic species observed were 3 (423 individuals, RA 44%) and 4 (98 individuals, RA 10%), respectively. Tolerance guild analysis as characteristics of ecological indicators revealed an undoubtedly high percentage (97%), compared to others as reflected by the identification of just one sensitive species. Analysis of trophic guilds showed that L. macrochirus dominated among insectivores (44% RA). The food of L. macrochirus composed of, aquatic insects, benthic invertebrates, zooplankton, Chironomidae, and waterweed. Thus, we determined that L. macrochirus could be classified as insectivores (partially carnivores) in this study. Conversely, Z. platypus consumed Cladocera exclusively, greater than 90% of their feed. We presume that stable isotope analysis would identify the exact position of these species in the food web.
