Optimization of HVP-flavour formation using Candida utilis (Candida Utilis 효모를 이용한 HVP 특유의 향 생성 최적화)

  • 본 연구에서는 분리대두단백(ISP: isolated soy protein)과 탈지대두박(DSF: defatted soy flake)을 Devolase Flavourzyme 효소로 가수분해한 각각의 HVP를 발효 배지로 사용하여 Candida utilis(KCCM 50342) 효모의 ethylalcohol, 4-ethylguaiacol의 생성과 향 생성의 주요 인자인 a -galactosidase의 활성을 측정하였다. Ethylalcohol은 발효 1일째에 탈지대두박을 원료로 하고 여과후 미생물 처리한 경우에서 가장 높게 생성되었고, 4-ethylguaiacol은 일부 여과하지 않고 미생물 처리한 경우에서 낮은 농도의 생성을 보였다. a-galactosidase 효소활성은 탈지대두박 보다 ISP를 원료로 한 HVP에서 높은 활성을 보였다. 관능검사결과 탈지대두박을 사용한 HVP의 관능적 특성이 ISP를 원료로 한 HVP보다 더 우수하였다. 결과적으로 탈지대두박을 원료로 한 HVP의 향기성분 및 관능적 특성이 우수함을 확인하였다.

  • This experiment was performed to investigate the growth and acid production of lactic acid bacteria in soy yoghurts prepared from soybeans produced using various nitrogen sources and the ability of the bacteria to utilize the available carbohydrates in the medium. Various soybeans did not show significant difference in the growth and acid production of lactic acid bacteria, soybean D with newly developed complex fertilizer being the best. The acid production in soy milk by L. acidophilus was better than those in defatted soy milk and in isolated soy protein, that in isolated soy milk being significantly poor. Of lactic acid bacteria tested, L bulgaricus and L. acidophilus exhibited more population and acid production in soy yoghurts. The population and acid production of lactic acid bacteria was increased by the addition of various carbohydrates, and significantly by glucose and yeast extract.

Effects of Different Kinds of Korean Soybeans on Body Lipids and Lipid Peroxidation in Rats (다양한 콩종류가 흰쥐의 체내지질과 지질과산화에 미치는 영향)

  • This study was conducted to investigate the effect of different kinds of traditional Korean soybeans on blood and liver lipids and lipid peroxidation. SD male rats (n=48) were assigned to six different groups and provided experimental diets for 28 days. The protein source of the diet was casein, isolated soy protein (ISP), or casein plus traditional soy powders (yellow bean, huktae, jinuni or seorietae). The ISP, jinuni and seorietae diet decreased blood triglyceride compared to casein group. The ISP groups had lower hepatic total lipid and triglycerides than casein groups. The yellow bean and seorietae group had hepatic lipid lowering effects, but the effect was not found in huktae and jinuni group. The glutamate-oxaloacetate transaminase activity was significantly decreased in ISP and jinuni groups than in casein group. The glutamate-pyruvate transaminase activity was also decreased in ISP group. The results of this study suggest that jinuni, yellow bean and seorietae groups are more effective in lowering body lipids than huktae group.

Effect of Different Kinds of Protein Fed with Coffee and/or Methionine on the Protein & Lipid Metabolism of Rats (단백질의 종류를 달리한 식이에 첨가한 Coffee 와 Methionine이 흰쥐의 단백질과 지방대사에 미치는 영향)

  • This study was performed to investigate the effects of different kinds of dietary protein [plant protein ; Isolated Soy Protein(ISP), animal protein ; casein] on protein and lipid metabolism in rats fed with coffee and/or methionine in diet during four weeks of growing period after weanling. Forty male growing rats fo Sprague-Dawley strain, weighing 92.5$\pm$1.8g, were distributed into 8 groups by randomized complete block design, and fed diets containing 15% of protein by weight either as ISP or casein and 10% ofcalories as corn oil, supplemented with coffee and /or methionine for 4 weeks. Coffee were added at a concentration of 1.4% of diet as instant coffee, and methionine were added to ISP or casein diet to be 0.6% of diet as DL-methionine. Results were followed ; Body weight gain, F.E. R and P.E.R tanded to be higher in methionine added groups than non-methionine groups. The nitrogen content of feces was significantly higher in coffee groups than non-coffee groups, and tended to be higher in ISP groups than casein groups. but was not significantly different with or without methionine. thus, apparent protein digestilbity was significantly lower in coffee groups than non-coffee groups and was significantly lower in ISP groups than cesein groups, but was not significantly different with or without methionine. Total cholesterol content of serum tended to be higher in coffee groups than non-coffee groups, and tended to be lower in methionine groups than non-methionine groups.

Optimization of finely ground meat ball analogue formulations using proteins and polysaccharides from plant origin (세절형 식물성 미트볼의 저작 특성 모방을 위한 식물성 단백질과 다당류 소재의 배합비 최적화)

  • This study optimized the finely ground meat ball analogue formulation by a series of experiments. Replacing isolated soy protein (ISP) to isolated mung bean protein (IMP) in total 27% protein caused an increase in hardness while adhesiveness, springiness, and chewiness were decreased (p<0.05), and the best ISP to IMP ratio was 10:17. When protein content was changed from 19% (w/w) to 31% (w/w) with the best ISP:IMP ratio, adhesiveness was increased with decreasing protein content, but all texture profiles were greatly decreased comparing to control (p<0.05). To modify texture of plant meat ball, potato starch (PS), κ-carrageenan (KC), methyl cellulose (MC), konjac (KJ), and potato protein (PP) were applied in formulation. Finally, KJ and MC were combined with and without PP in formulation. As a result, 0.5% KJ+MC+PP and 1% KJ+MC were best formulation for plant meat ball, nevertheless, highly harder texture than control must be regulated which warranted further exploration.

Biochemical Properties of Seed Lectin from Korean Soybean Cultivars Developed for Soy Source (한국산 장류콩 종자 렉틴의 생화학적 특성)

  • Lectin was finally isolated on Sephadex G-100 from Korean soybean cultivars developed for soy source and investigated its some biochemical properties. Native PAGE pattern of this lectin revealed a molecular weight of 108 kDa as tetramer. The molecular weight of this lectin isolated as double protein band by SDS-PAGE was calculated to be 32 and 22 kDa from the relative mobilities compared with those of the standard proteins. Among the tested red blood cell, the isolated lectin agglutinated rabbit red blood cell treated with trypsin, but did not agglutinated human red blood cells (A, B, AB, O), rat, and untreated rabbit red blood cell. The optimal temperature and thermal stability of isolated lectin was at 20-$50^{\circ}C$ and 10-$60^{\circ}C$, respectively. This lectin was stable at 7.2, and showed complete loss in its activity below pH 6.2 and above pH 8.0.

Properties of a Bacteriocin Produced by Bacillus subtilis EMD4 Isolated from Ganjang (Soy Sauce)

  • A Bacillus species, EMD4, with strong antibacterial activity was isolated from ganjang (soy sauce) and identified as B. subtilis. B. subtilis EMD4 strongly inhibited the growth of B. cereus ATCC14579 and B. thuringiensis ATCC33679. The antibacterial activity was stable at pH 3-9 but inactive at pH 10 and above. The activity was fully retained after 15 min at 80℃ but reduced by 50% after 15 min at 90℃. The activity was completely destroyed by proteinase K and protease treatment, indicating its proteinaceous nature. The bacteriocin (BacEMD4) was partially purified from culture supernatant by ammonium sulfate precipitation, and Q-Sepharose and Sephadex G-50 column chromatographies. The specific activity was increased from 769.2 AU/mg protein to 8,347.8 AU/mg protein and the final yield was 12.6%. The size of BacEMD4 was determined to be 3.5 kDa by Tricine SDS-PAGE. The N-terminal amino acid sequence was similar with that of Subtilosin A. Nucleotide sequencing of the cloned gene confirmed that BacEMD4 was Subtilosin A. BacEMD4 showed bactericidal activity against B. cereus ATCC14579.

Characterization of a 27 kDa Fibrinolytic Enzyme from Bacillus amyloliquefaciens CH51 Isolated from Cheonggukjang

  • Bacillus amyloliquefancies CH51 isolated from cheonggukjang, a traditional Korean fermented soy food, has strong fibrinolytic activity and produces several fibrinolytic enzymes. Among four different growth media, tryptic soy broth was the best in terms of supporting cell growth and fibrinolytic activity of this strain. A protein with fibrinolytic activity was partially purified from the culture supernatant by CM-Sephadex and Phenyl Sepharose column chromatographies. Tandem mass spectrometric analysis showed that this protein is a homolog of AprE from B. subtilis and it was accordingly named AprE51. The optimum pH and temperature for partially purified AprE51 activity were 6.0 and $45^{\circ}C$, respectively. A gene encoding AprE51, aprE51, was cloned from B. amyloliquefaciens CH51 genomic DNA. The aprE51 gene was overexpressed in heterologous B. subtilis strains deficient in fibrinolytic activity using an E. coli-Bacillus shuttle vector, pHY300PLK.

Processing and Sensory Characteristics of Lipoxygenase-Deficient Soybeans (Lipoxygenase 결핍 콩의 가공 및 관능 특성)

  • The objective of this study was to investigate the application of lipoxygenase(LOX)-deficient soybean to food processing by evaluation of processing and sensory characteristics. LOX activity of some processed products of soybean was decreased as the order of whole soy flour, defatted flour, isolated protein. Particularly the decrement of LOX activity of Hwangkeumkong having all L-1, L-2 and L-3 was so high in defatting and isolation of protein. LOX activities of cotyledon of Jinpumkong sprouts were so high. When soy milks were prepared with LOX-deficient soybeans, its beany flavor was evaluated to be weaker than that of Hwangkeumkong. And soybean sprout of Jinpumkong 2 showed less beany flavor but hypocotyl elongation was poor. The beany flavor of ice cream prepared with defatted flour of Jinpumkong 2 didn't make a significant difference from that of ice cream made with defatted milk powder. We suggest that LOX-deficient soybean may have on relatively good oganoleptic properties of processed foods, so it takes advantage of food processing.

The Texturization Properties of Textured Extrudate made by a Mixture of Rice Flour and Isolated Soybean Protein (쌀과 분리대두단백 혼합에 따른 조직화 특성)

  • The texturization properties of extrudate from isolated soybean protein and rice flour by extrusion cooking were investigated. The addition of up to 30% rice flur to isolated soy proetin could give more tenderness to the texturized extrudate. As the rice flour content increased, die temperature, nitrogen solubility index, and integrity index were decreased slightly with lower chewiness and gumminess. The water content of final extrudate was increased as the addition of rice flour increased, while density was maintained without variation, and rehydration ratio was decreased. The distribution of pressure profile during extrusion were in the range of $15-100kg/cm^3$. As the addition of rice flour increased, scanning electron micrographs demonstrated the gelatinized surface structure of rice starch and the increased air cell size of the testurized extrudate.
