We compared the antimicrobial resistance and clonal relationships among the community-acquired (CA) and hospital-acquired (HA) methicillin-resistant Staphylococcus aureus (MRSA) strains that were isolated from blood cultures in a university hospital over a 4-year period. A total of 131 MRSA isolates, including 28 CA-MRSA and 103 HA-MRSA strains, were identified; antimicrobial susceptibility testing indicated that the CA-MRSA isolates were more susceptible to erythromycin (21 % vs 6% ; P=0.02), clindamycin (46% vs 12%; P<0.01), ciprofloxacin (43% vs 11%; P<0.01), and gentamicin (43% vs 6%; P<0.01) than were the HA-MRSA isolates. Pulsed-field gel electrophoresis (PFGE) typing and antimicrobial resistance profiles separated the 20 CA-MRSA isolates into 14 and 10 different patterns, respectively, and the 53 HA-MRSA isolates were separated into 24 and 7 different patterns, respectively. Twenty-one (40%) of the 53 HA-MRSA isolates belonged to two predominant PFGE types, and most of them showed multi-drug resistant patterns. Four (20%) of the 20 CA-MRSA and 10 (19%) of the 53 HA-MRSA isolates fell into two common PFGE patterns, and each of them showed the same multi-drug resistant pattern. This study suggests that, although the CA-MRSA blood isolates showed diverse PFGE and antimicrobial resistance patterns, some of these isolates may have originated from the HA-MRSA strains.
Clonal complex (CC) 398 community-associated methicillin-resistant Staphylococcus aureus (CA-MRSA) has emerged worldwide in a variety of livestock animals and humans. We report complete genome sequences of Panton-Valentine leucocidin (PVL) and immune evasion cluster (IEC) gene-positive CC398 MRSA strains isolated from patients in Korea.
Once known as a major community-genotype, sequence type (ST) 72 methicillin-resistant Staphylococcus aureus (MRSA) strains have been increasingly identified in hospital-associated infections in Korea. Here, we report the complete genome sequence of SA520 isolated from a patient in Korea.
A metagenomic fosmid library was constructed from genomic DNA isolated from the microbial community residing in hindguts of a wood-feeding higher termite (Microcerotermes sp.) collected in Thailand. The library was screened for clones expressing lignocellulolytic activities. Fourteen independent active clones (2 cellulases and 12 xylanases) were obtained by functional screening at pH 10.0. Analysis of shotgun-cloning and pyrosequencing data revealed six ORFs, which shared less than 59% identity and 73% similarity of their amino acid sequences with known cellulases and xylanases. Conserved domain analysis of these ORFs revealed a cellulase belonging to the glycoside hydrolase family 5, whereas the other five xylanases showed significant identity to diverse families including families 8, 10, and 11. Interestingly, one fosmid clone was isolated carrying three contiguous xylanase genes that may comprise a xylanosome operon. The enzymes with the highest activities at alkaline pH from the initial activity screening were characterized biochemically. These enzymes showed a broad range of enzyme activities from pH 5.0 to 10.0, with pH optimal of 8.0 retaining more than 70% of their respective activities at pH 9.0. The optimal temperatures of these enzymes ranged from $50^{\circ}C$ to $55^{\circ}C$. This study provides evidence for the diversity and function of lignocellulose-degrading enzymes in the termite gut microbial community, which could be of potential use for industrial processes such as pulp biobleaching and denim biostoning.

In an artificial pH-gradient batch culture system, the effects of acidification on the species composition of a heterotrophic bacterial community were analyzed. As a result of this study, it was found that total bacteria numbers were not affected by acidification and that the population of hetero-trophic bacteria decreased as pH became lower. The heterotrophic bacteria isolated from the entire pH gradient were 12 genera and 22 species. Among them, 64% were gram negative and 36% were gram positive bacteria. As pH decreased, the distribution rate of gram negative bacteria increased while that of gram positive bacteria decreased. The diversity of genera decreased from 13 to 5 as pH decreased from 7 to 3. The G+C content of all of the 202 isolated strains varied from 22.8 to 77.0%, and increased in interspecies of same genus as pH decreased. As a result of clustering analysis, the diversity index of species ranged from 1.13 to 2.37, and it had lower indices as pH decreased. In order to evaluate the diversity of numbers of sample of different size, a rarefaction method was used to analyze the expected number of species appearance according to pH. The statistical significance of species diversity was verified by the fact that the number decreased at lower pH.

The phylogenetic diversity of microbial communities in calcareous mats from Spearfish Creek, a freshwater stream located in the Black Hills of South Dakota, was examined using PCR-based 16S rDNA sequence analysis. In this study, two types of calcareous mats were compared: mature mats formed on the natural substrate of rock surfaces and developing mats on an artificial substrate of glass slides. Among 63 unique isolates from a clone library of 16S rRNA genes from mature mat samples, there were 8 phyla of Bacteria represented. The predominant phylum was Proteobacteria (48%), with the $\beta$ subclass being the largest group. Denaturing gradient gel electrophoresis (DGGE) analysis of 16S rRNA genes from slide samples collected at intervals for four months showed considerable diversity of the microbial community from the earliest stages of community development. Amplicons isolated from DGGE gels and sequenced indicated that community succession has occurred without increasing microbial diversity. However, light microscopic analysis revealed a significant increase in microbial cell density throughout the community development. Scanning electron microscopy of mat samples provides evidence that diatoms are also important members of calcareous mat communities.

Otitis externa (OE) is a frequent disease in the ear canals of dogs. To identify the pathogens causing OE in dogs and to determine their antimicrobial resistances, specimens were collected from animal hospitals in Daejeon. The isolates were examined by morphological and biochemical tests, 16S rRNA analysis and antimicrobial susceptibility tests. We analyzed correlation between the isolated pathogens and external factors of dogs such as breed, age, gender, ear mite, hair in ears and experience with antibiotic therapy. Thirty three strains of bacteria were isolated from 26 of the 68 heads of dogs with OE. The most isolated bacteria were Enterococcus faecalis (E. faecalis) followed by Staphylococcus aureus (Sta. aureus), Sta. pseudointermedius, E. faecium, E. avium and Streptococcus canis (Strep. canis) in order of frequency of occurrence. Isolation frequency of Enterococcus spp. and Staphylococcus spp. were 51.5% and 45.5%, respectively. E. faecalis and E. faecium isolates showed VanB phenotype, which is resistant to vancomycin but sensitive to teicoplanin were 58% and 25%, respectively. Nine isolates among total twelve isolates of E. faecalis were isolated from the dogs treated with antibiotics. There was no methicillin-resistant Sta. aureus (MRSA), but were MR-Sta. pseudointermedius (MRSP) (57.1%) and vancomycin-resistant (VR)-Sta. pseudointermedius (14.3%) (VRSP) showing VanB phenotype. However, vanA, vanB and vanC genes were not detected in VR isolates from the dogs. Taken together, VR-Enterococcus spp. (VRE) is one of the major pathogens in domestic animals, as well as community-and hospital-acquired infection.
According as the modern form of the family becomes nuclearized, inevitable is the isolation of the family. The relationship of the neighborhood and the kinsmen are being dissolved, making the isolated and unsteady nuclear family. We need a community in the modern society, for we are facing a lot of social problems as the nuclear family. The problems are as follows ; the rearing of the children, the alienation of a woman and the woman's roles in the society. In addition to those problems, there are also such some other problems as the alienation of the old man and the social roles of them. That is to say, the community appeared as a kind of attempt to overcome the above-mentioned problems, Kibbutz may be itself called and enlarged family to function as one family even though it is not just a blood relationship. The American commune is a large family community as a group to overcome the alienation and solitude of the nuclear family. On the other hand, the urban community is the small-scaled and cooperative family to solve the problem that the modern nuclear family is a unit living solitarily and unsteadily. But in fact, even in such a community, there are much tension and conflict against its establishing purpose. In my opinion, if those communities have to surmount the tension and conflict which are derived from the process in a bid to overcome the troubles like the alienation and solitude of the nuclear family. The bond of family must be consolidated among all the members of the community. And then community may be co-existed with the traditional family as the alternative family in the future, which can supply the deficiency of the unsteady unclear family.
Wetlands exhibit intermediate characteristics of both terrestrial and aquatic ecosystems, and the biodiversity is rich in these unique biological habitats. The symbiotic relationships between plants and fungi that inhabit these wetlands play an important role in natural resource management, biodiversity, and conservation. Accordingly, the mujechi, having academic value for the study of the natural environment, was investigated in terms of genetic diversity of endophytic fungi, which inhabit the roots of wild plants. The internal transcribed spacer (ITS) region was amplified to identify fungal strains. In total, 226 strains were isolated and categorized into three phyla, seven classes, 10 orders, 22 families, and 31 genera. In plants by endophytic fungi were classified in Isachne globosa (Ig) to 19 genera, Scirpus karuisawensis (Sk) to 11 genera, Utricularia racemosa (Ur) to 19 genera, and one incertae sedis, Eriocaulon decemflorum (Ed) to 11 genera. The fungal taxa was identified the genera Acephala (19.9%), Tolypocladium (16.3%), Neopestalotiopsis (11.5%), and Perenniporia (7.1%). The fungal group isolated from Isachne globosa (Ig) grew the largest number of isolated fungal strains. After comprehensive evaluation, the endophytic fungal group from Utricularia racemosa (Ur) ranked highest in diversity analyses. From the roots of wild plant in mujechi-neup, it confirmed the distribution and diversity of endophytic fungi. This study provides the basic data to understand fungal community structure in peat wetlands.
Although much research has focused on various bioactive substances in starfish, research on microorganisms isolated from starfish is lacking as compared with other natural products. In this study, we investigated bacterial communities in the gut of red starfish (Certonardoa semiregularis) in Jeju Island. In total, 103 bacterial strains were isolated using marine agar and R2A medium. The isolated strains were analyzed and identified using the 16S rRNA gene sequence. Based on an analysis of this gene sequence, the 103 isolated bacteria were classified into four major groups: Proteobacteria (93%: Alpha-proteobacteria, 24.8%; Beta-proteobacteria, 4%; Gammaproteobacteria, 65%) Bacteroidetes (4%), Actinobacteria (2%), and Firmicutes (1%). In addition, the isolates were divided into seven classes (Actinobacteria, Flavobacteria, Bacilli, Sphingobacteria, Alphaproteobacteria, Betaproteobacteria, and Gammaproteobacteria), 15 orders, 19 families, and 24 genera. A phylogenetic analysis revealed two strains, Lysobacter sp. and Pedobacter sp., with similarity of 97.55% and 97.58%, respectively. As the similarity in the 16S rRNA gene sequence was 98% or less compared to previously identified bacteria, the two strains may possibly be classified as a new genus or species. We suggest that additional studies, including biochemical and morphological tests, should be performed to identify the new candidate strains.
