The trend of antimicrobial resistance of Escherichia coli isolated from healthy volunteers of community and chicken butchers in Incheon (일반인 및 닭도축장 근무자에서 분리한 대장균의 항생제 내성 양상)

  • We monitored antibiotic resistance of Escherichia coli isolates from healthy volunteers of community and chicken butchers from February to July in 2006. From disc diffusion test on 473 E coli isolates from healthy volunteers of the community, the resistance rates of tetracycline, ampicillin, and ticarcillin were 44.8%, 40.8%, and 37.4%, respectively. In the disc diffusion test on 18 E coli isolates from chicken butchers, the resistance rates of tetracycline, nalidixic, streptomycin, and ampicillin were 94.4%, 61.1%, 55.6%, and 50.0%, respectively.

Effects of Water Stress on the Endophytic Fungal Communities of Pinus koraiensis Needles Infected by Cenangium ferruginosum

  • To examine the effects of water stress and Cenangium ferruginosum (CF) on the fungal endophytic community of needles of Pinus koraiensis (PK), fungal endophytes isolated from the needles of 5-year-old PK seedlings were compared before and after exposure to water stress conditions and artificial inoculation with CF ascospores. Artificial CF inoculation was successfully confirmed using PCR with CF-specific primers (CfF and CfR). For comparison of the degree of water deficit in water-stressed and control groups of PK seedlings infected with CF, the water saturation deficit and water potential were measured. Lower water potential estimates were found in the water-stressed seedlings than in the control group. The fungal endophytes isolated from the second-year needles of non-water-stressed seedlings before and after CF inoculation revealed that primary saprobes were approximately 30% and 71.7%, respectively, and the remaining endophytes were rot fungi or pathogens. Sixty days after CF inoculation, diverse fungal endophytes in the first-year needles were isolated from the water-stressed seedlings. However, some fungal endophytes isolated from the non-water-stressed seedlings were also identified. Fungal endophytes in the second-year needles of the water-stressed and non-water-stressed seedlings were approximately 8% and 71.7% of saprobes, respectively, and the remaining endophytes were rot fungi or pathogens. On the basis of the results, we conclude that water deficit and CF can have an effect on fungal endophytic communities in the needles of PK seedlings.

Molecular Genetic Characteristics of Methicillin-Resistant Staphylococcus aureus Isolated from University Campus Environment and Students

  • Staphylococcus aureus (S. aureus) is known as a bacterium that can cause skin infections, respiratory system infections, and sinusitis; however, it can exist as a normal flora rather than a pathogen. Recently, methicillin-resistant S. aureus (MRSA) infections have emerged in the community as a new variant of community-associated (CA)-MRSA. In the present study, S. aureus and MRSA were isolated and cultured by collecting samples from facilities and environments where students and educational personnel have multiple contacts on university campuses; specifically, the nostrils and hands of college students were tested from July to September of 2019. The molecular properties of the isolated MRSA were analyzed, and the one MRSA strain was isolated from the university campuses. One MRSA that was isolated and cultured on campus was the mec complex group A and staphylococcal cassette chromosome (SCC) mec type II, which is a characteristic of healthcare-associated (HA)-MRSA, and SCCmec type V, which is a characteristic of CA-MRSA. This result was similar to other studies wherein the SCCmec type II was detected in SCCmec typing analysis in CA-MRSA. To confirm whether there is a new variant of CA-MRSA in the Republic of Korea, additional follow-up studies on the analysis of virulence factors of MRSA are needed by additionally separating CA-MRSA from the body parts of university students and educational personnel.

Bacterial Diversity in the Initial Fermentation Stage of Korean and Chinese Kimchi (발효 초기 한국산 및 중국산 김치의 Bacteria 다양성 평가)

  • The purpose of this research is to draw the bacterial community difference between Korean and Chinese kimchi for future use in the confirmation of kimchi origin. Initial fermentation stage kimchi samples (above pH 5) were used for the analysis of bacterial diversity. From 26 Korean kimchi samples, 1,017 strains in the 45 genera and from 22 Chinese kimchi samples, 842 strains in the 54 genera were isolated with use of marine medium, nutrient medium, succinate minimal medium (SMM), leuconostocs selective medium (LUSM) agars. In the order of isolated numbers, Bacillus, Weissella, Leuconostoc, Pseudomonas, and Lactobacillus genera and Bacillus, Weissella, Lactobacillus, Pseudomonas, Serratia, and Enterobacter genera were predominated in Korean and Chines kimchi, respectively. Among the isolated lactic acid bacteria, Weissella spp. were isolated most dominantly owing to the biased growth of Weissella spp. on LUSM agar. Species in the genera Leuconostoc and Lactobacillus were the next frequently isolated LAB from Korean and Chinese kimchi, respectively. Weissella confusa was isolated only from Korean kimchi and W. soli and Serratia proteamculans were isolated only from Chinese kimchi. They have a possibility to be used as target bacteria to differentiate Korean kimchi from Chinese kimchi.

Antifungal Activity of Lactic Acid Bacteria Strains Isolated from Natural Honey against Pathogenic Candida Species

  • The role of lactic acid bacteria (LAB) in honey as antifungal activity has received little attention and their mechanism of inhibitory of fungi is not fully understood. In this study, LAB were isolated from honey samples from Malaysia, Libya, Saudi Arabia, and Yemen. Twenty-five isolates were confirmed LAB by catalase test and Gram staining, and were screened for antifungal activity. Four LAB showed inhibitory activity against Candida spp. using the dual agar overlay method. And they were identified as Lactobacillus plantarum HS isolated from Al-Seder honey, Lactobacillus curvatus HH isolated from Al-Hanon honey, Pediococcus acidilactici HC isolated from Tualang honey and Pediococcus pentosaceus HM isolated from Al-Maray honey by the 16S rDNA sequence. The growth of Candida glabrata ATCC 2001 was strongly inhibited (>15.0 mm) and (10~15 mm) by the isolates of L. curvatus HH and P. pentosaceus HM, respectively. The antifungal activity of the crude supernatant (cell free supernatant, CFS) was evaluated using well diffusion method. The CFS showed high antifungal activity against Candida spp. especially The CFS of L. curvatus HH was significantly (p < 0.05) inhibited growth of C. glabrata ATCC 2001, C. parapsilosis ATCC 2201, and C. tropicalis ATCC 750 with inhibitory zone 22.0, 15.6, and 14.7 mm, respectively. While CFS of P. pentosaceus HM was significantly (p < 0.05) effective against C. krusei, C. glabrata, and C. albicans with inhibition zone 17.2, 16.0, and 13.3 mm, respectively. The results indicated that LAB isolated from honey produced compounds which can be used to inhibit the growth of the pathogenic Candida species.

  • We monitored antibiotic resistance of Escherichia coli isolates from healthy volunteers of community and hospital patients from February to July in 2006. From disc diffusion test on 4915 E. coli isolates from healthy volunteers of the community, the resistance rates were as follows; tetracycline resistant, 46.6%; ampicillin resistant, 41.1%; ticarcillin resistant,37.9%. From disc diffusion test on 120 E. coli isolates from hospital patients, the resistance rates were as follows: ampicillin resistant, 66.9%; ticarcillin resistant, 63.8%; tetracycline resistant, 47.2%. Extended spectrum $\beta$-lactamase producing E. coli were isolated 0.6% and 4.1% from healthy volunteers and hospital patients.

Effects of a Volunteer-Run Peer Support Program on Health and Satisfaction with Social Support of Older Adults Living Alone (지역사회 노인 자원봉사자를 활용한 독거노인 건강지킴이 프로그램이 독거노인의 건강상태와 사회적 지지 만족도에 미치는 효과)

  • Purpose: The purpose of the study was to evaluate effectiveness of a peer support program conducted by older community volunteers for older adults living alone. Methods: Thirty volunteers trained as peer supporters were matched with low-income, older adults living alone in the community on gender. Visits occurred on a weekly basis over the 12 month study period, and the volunteers provided peer support for health management to solitary older adults. Data were collected, before the start of the program and again 6 and 12 months after its initialization, from intervention and control groups regarding physical health, general health, mental health, depression, social functioning, and satisfaction with social support. Repeated measures ANOVA was used to analyze data. Results: By the end of the program, socially isolated older adults in the intervention group had significantly higher scores in physical health and general health than elders in the control group. Significant interaction effects between time and group were found for depression, social functioning, and satisfaction with social support. Conclusion: The peer support program undertaken by older community volunteers was effective in improving physical health, general health, depression, social functioning, and satisfaction with social support in socially isolated, low-income, older adults.

  • Purpose The purpose of this study is to investigate the quality of life and stress level of the elderly living alone in their neighborhood volunteers. Methods Data collection was done from July 1 to October 1, 2015 and the data were collected by self-reported questionnaire at Nowon community in Seoul area in South Korea. The subjects were 146 older adults living alone agreed to participate in the study(accepted by IRB). The questionnaire was constructed with quality of life 26 items, 5-point Likert type scale(1-5), stress 15 items, 5-point Likert type. The data were analyzed by descriptive statistics. Results Socially isolated older adults had significantly higher scores ($3.7{\pm}.03$) in quality of life, moderate scores ($2.9{\pm}.03$) in stress. Conclusion The peer support undertaken by older community volunteers was effective in improving quality of life in socially isolated older adults. It is necessary to listen to the voice of elderly people living alone through the activation of volunteer groups in the community.

Diversity of Endophytic Fungi Isolated from Korean Ginseng Leaves

  • We investigated the diversity of the foliar endophytes of Korean ginseng. Endophytic fungi were isolated from healthy leaves of mountain-cultivated ginseng (MCG) and field-cultivated ginseng (FCG) at 4 sites in Chungbuk Province. A total of 24 species of fungal endophytes were identified using molecular approaches. Additionally, the diversity of these endophytic fungi was compared between MCG and FCG. The major isolated endophytes were Edenia gomezpompae and Gibberella moniliformis in the MCG and FCG samples, respectively. The results suggest that ginseng endophytes have different community structures in different environments, and this understanding may prove useful in ginseng cultivation.
