Quantitative analysis of coumarins in Artemisia keiskeana and A. stolonifera using HPLC/PDA

  • Artemisia keiskeana and A. stolonifera are plants of the genus Artemisia, distributed in various regions, especially China and Korea. They are renowned as medicinal plants with biological and pharmacological activities. Fraxidin, isofraxidin, and daphnoretin are coumarins present in Artemisia spp.; however, research on them is limited. Therefore, this study was carried out to quantify the content of these compounds in the aerial parts of A. keiskeana and A. stolonifera in different regions in Korea. High-performance liquid chromatography was performed with a photodiode array detector and a reverse-phase INNO column. A. stolonifera only contained fraxidin with the highest amount found in Yongmun commune. A. keiskeana cultivation in Soyang commune gave the highest fraxidin and daphnoretin content. However, isofraxidin was not present in all samples. The findings suggest that the concentrations of the three compounds may differ depending on the growth site and provide a foundation for future studies.

  • Chemical investigation of the aerial parts of Chloranthus japonicus Sieb. led to the isolation a new compound, 9-hydroxy heterogorgiolide (1) and $isofraxidin-7-O-{\beta}-D-glucopyranoside$ (2), the isolation of which is reported for the first time from this plant, along with the known components, ${\beta}-sitosterol,\;{\beta}-sitosterol-3-O-{\beta}-D-glucopyranoside$, palmitic acid and octacosanoic acid. The structures of compound 1 and 2 were determined on the basis of spectroscopic data including two dimensional NMR and high resolution MS.

  • Acanthopanacis Cortex (Acanthopanax sessiliflorum, Araliaceae, KP VIII), an important Korean medicinal herbal drug, has been widely used as tonic, anti-stress and immuno-enhancing drugs. To monitor the contents of active ingredients (acanthoside D[=eleutheroside E], eleutheroside B, isofraxidin, chlorogenic acid, and caffeic acid) in Acanthopanax sp., we developed the HPLC analysis method and validated. The simultaneous determination of five active ingredients was achieved in a C$\sub$18/ column with an acetonitrile water (containing 1 % phosphoric acid) (15 : 85) mobile phase. (omitted)

Changes in the Contents of Phenolic Components in the Stem of Acanthopanax koreanum Depending on Extracting Batches (섬오가피의 추출 조건에 따른 페놀성 성분 함량 변화)

  • This study compared the contents of phenolic components depending on the extracting conditions of Acanthopanax koreanum stem to provide basic information for developing Acanthopanax koreanum-based functional foods. Our findings show that the content of total phenolic component peaked at 16 hours of extraction (WAK-16, 7.22%) and when water extracted at $100^{\circ}C$. However, 11-hours water extraction (WAK-11) showed highest eleutheroside B concentration of 1.32%, a main component of A. koreanum, and the level of chlorogenic acid concentration was the highest when 1-hour water extraction (WAK-1) was conducted, being 2.12%. Moreover, highest concentration of eleutheroside E was observed in 16-hours water extraction (WAK-16) as 1.49%. With 60-hours water extraction (WAK-60), the content of syringaresinol, an active phenolic aglycon substance, concentration was the highest with the value of 0.10%. Isofraxidin showed the highest concentration of water extract(0.09%) for 20 hours (WAK-20) and 0.42% for sinapyl alcohol[16 hours (WAK-16)].
