위성영상과 GIS를 이용한 낙동강하구 지형변화탐지 (Change Detection at the Nakdong Estuary Delta Using Satellite Image and GIS)

  • 낙동강 하구는 세계적 철새 서식지로 부산의 관광명소인 해운대와 광안리의 모래 공급지로 다양한 역할을 하고 있다. 본 연구는 낙동강 하구(진우도, 신자도, 도요등, 다대포) 해안의 장기간 지형변화를 탐지 해석 하였다. 34년간의 위성영상 분석 결과 진우도의 전면과 후면의 중간부분이 증가하고, 신자도는 1970년 띠 형태로 양분, 80년대 이후 하나의 섬으로 형성하여 서쪽 방향으로 길게 형성된다. 1990년대 이후 백합등 전면의 소규모 섬들이 급격히 발달하여 90년대 후반 도요등을 형성하여 성장하고 있었다. 낙동강 하구역 해안지도 제작을 위해 1975년부터 2009년간 Landsat 영상 112매 중 조위가 $99{\pm}13cm$인 영상 12매를 선택하고 NDVI값을 이용해 수역과 육역을 구분하였다. 그리고 GIS의 DSAS 4.0(Digital Shoreline Analysis System)과 MATLAB을 이용해 EPR(End Point Rate), LRR(Linear Regression Rate)등 해안선 변화율을 산정하였다. 지형변화탐지결과 EPR은 진우도 전면과 후면은 -0.93~2.56m/yr로 남진하고 해안선과 면적변화는 적고 안정적이며 신자도 전면과 후면은 1~4m/yr로 북진하나 서측은 2~3m/yr로 안정되고 동측은 10m/yr이상 북진하고 있다. 도요등 전면과 후면은 18~27m/yr로 북진, 면적은 증가하고 다대포해안은 7m/yr로 서진하며 확대되었다. LRR 또한 EPR과 유사한 경향을 나타내었다. 위성영상과 GIS 분석을 통해 지형변화 탐지와 자연현상의 정량적 분석이 가능하였지만 자연현상의 지속적인 관찰과 다양한 분석 방법이 필요함을 알 수 있었다.

Production of Transgenic Micro-Pig Expressing Human Heme Oxygenase 1

  • Xenotransplantation of pig islet regarded as a good alternative to allotransplantation. However, cellular death mediated by hypoxia-reoxygenation injury after transplantation disturb success of this technique. In the present study, we produce transgenic pig expressing human heme oxygenase 1 (HO1) genes to overcome cellular death for improving efficiency of islet xenotransplantation. Particularly, Korean miniature pig breed, Micro-Pig, was used in the present study. Somatic cell nuclear transfer (SCNT) technique was used to produce the HO1 transgenic pig. Six alive transgenic piglets were produced and all the transgenic pigs were founded to have transgene in their genomic DNA and the gene was expressed in all tested organs. Also, in vitro cultured fibroblasts derived from the HO1 transgenic pig showed low reactive oxygen species level, improved cell viability and reduced apoptosis level.

Evaluation of the Antidiabetic, Islet Protective and Beta-Cell Regenerative Effects of Tinospora crispa (L.)

  • Tinospora crispa (L.) is a medicinal plant traditionally used to treat various ailments including diabetes. The stem has been widely studied for its antidiabetic properties, however the antidiabetic potential of its leaves has not been explored. This study investigates the antidiabetic properties of methanolic T. crispa stem and leaves extracts on alloxan-induced diabetic rats. The stems and leaves were extracted using Soxhlet extraction with methanol as solvent. Crude extracts were administered at 500 mg/kg body weight (BW) (high dose) and 250 mg/kg BW (low dose) via oral route to alloxan induced diabetic rats. T. crispa stem and leaves extracts was found to significantly reduce blood glucose following a twelve-week treatment period. The highest mean difference in blood glucose level was exhibited by animals in the high dose treated stem and low dose leaf extracts. Both extracts showed approximately 75% percentage recovery from hyperglycaemia. The highest regenerative capacity was observed in animals treated with the low dose leaf extract.

마라도에 번식하는 섬개개비의 번식지 특성 (Breeding Site Characteristics of Styan's Grasshopper Warblers(Locustella pleskei) on Mara Islet, Jeju Province, Korea)

  • 국제적 멸종위기종이며, 동아시아에 국한하여 분포하는 섬개개비(Locustella pleskei)는 한국의 추자도, 마라도, 칠발도, 홍도 등에서 번식하는 것으로 알려져 왔으나, 구체적인 번식 정보는 아직 알려져 있지 않다. 본 연구는 마라도(N $33^{\circ}$ 06', E $126^{\circ}$ 16')에 번식하는 섬개개비의 번식 현황과 번식지 특성을 파악하기 위하여 실시되었다. 2008년 5월부터 9월까지 섬개개비 총 11쌍이 번식하는 것을 확인하였고, 번식하는 둥지는 곰솔숲의 관목층에 주로 분포하였다. 둥지는 곰솔숲의 관목층에 주로 분포하였으며, 동백나무(Camellia japonica)와 돈나무(Pittosporum tobira) 등 관목성 나무를 둥지목으로 선호하는 것으로 나타났다. 둥지목의 수고는 $2.77{\pm}1.10m$, 지면으로부터 둥지의 높이는 $1.75{\pm}0.56m$였으며, 둥지목의 수고는 다른 주변의 나무보다 낮은 것으로 나타나 마라도의 강한 해풍을 피하는데 유리한 것으로 판단되었다. 주로 억새잎이나 사초과 식물의 잎을 이용하여 밥그릇 형태의 둥지를 만들었으며, 둥지의 지름은 $11.9{\pm}0.5cm$, 높이는 $11.1{\pm}1.1cm$, 깊이는 $5.8{\pm}0.4cm$로 나타났으며, 산좌지름은 6.0cm였다. 마라도에서 섬개개비는 다른 섬지역보다 번식시기가 다소 늦었으며, 한배산란수는 3개로서 보통 다른 번식지의 4-5개보다 작았다. 이는 번식시기가 늦어질수록 한배산란수가 적어지는 일반적인 경향을 따르는 것으로 보인다. 이상의 결과로 볼 때, 마라도 곰솔숲의 솎아베기와 관목층 제거는 밀집된 관목층을 둥지로 선호하는 섬개개비의 번식에 영향을 미칠 수 있다. 따라서 섬개개비 번식 생태 및 서식지에 지속적이고 체계적인 조사가 요구되며, 숲가꾸기 수행에 있어서도 섬개개비의 보전을 위한 세심한 배려가 필요하다.

Modulation of Inflammatory Cytokines and Islet Morphology as Therapeutic Mechanisms of Basella alba in Streptozotocin-Induced Diabetic Rats

  • The mechanism of the previously reported antidiabetic effect of Basella alba is unknown. This study investigated the role of B. alba aqueous leaf extract in the modulation of inflammatory cytokines and islet morphology in streptozotocin-induced diabetic rats. Forty male Wistar rats, between 8 and 10 weeks old, were randomly divided into four groups (n = 10) and administered the following treatments: Healthy control (H-c) and Diabetic control (D-c) animals received normal saline 0.5 mL/100 g body weight daily, while Healthy Treatment (H-Ba) and Diabetic Treatment (D-Ba) rats received the plant extract 200 mg/kg body weight daily. All treatments were administered by oral gavage. Diabetes was induced in D-c and D-Ba rats by a single intraperitoneal injection of streptozotocin (55 mg/kg body). The body weight and fasting blood sugar (FBS) levels were recorded every week for 4 weeks, after which the rats were euthanized and samples collected for further analysis. After the experiment, FBS level was significantly reduced (p < 0.0001) in rats in the D-Ba group, but increased (p < 0.001) in rats in the D-c group. The absolute (H-c and H-Ba vs D-c, p < 0.05) and relative (D-Ba vs H-c, p < 0.05; D-Ba vs H-Ba, p < 0.005) weights of the pancreases were significantly higher after the experiment. The rats in the D-c group had significantly higher levels of serum interleukin-$1{\beta}$ (p < 0.001 vs H-c; p < 0.05 vs H-Ba and D-Ba) and monocyte chemotactic protein-1 (p < 0.0001), but lower levels of interleukin-10 (p < 0.05) in comparison with the other groups. Histopathological examination revealed severe interstitial congestion, reduced islet area (p < 0.0001), and increased islet cell density in the D-c group compared with those in the D-Ba group. From these findings, it was concluded that the aqueous extract of B. alba stimulates the recovery of beta-islet morphology in streptozotocininduced diabetic rats by modulating the peripheral production of inflammatory cytokines.

  • The forest of kadok islet was investigated from aug. 1 1990 through oct. 30, 1990. The results obtained are summarized as follow : a floristic community. HY 항원 불일치 췌도 이식에 의한 면역 관용의 유도 (Immune Tolerance in Murine Islet Transplantation Across HY Disparity)

  • Background: Minor histocompatibility HY antigen, as a transplantation antigen, has been known to cause graft rejection in MHC (major histocompatibility complex) matched donor-recipient. The aim of our study is to investigate the role of male antigen (HY) disparity on MHC matched pancreatic islet transplantation and to examine the mechanism of the immune reaction. Methods: Pancreatic islets were isolated and purified by collagen digestion followed by Ficoll gradient. The isolated islets of male C57BL6/J were transplanted underneath the kidney capsule of syngeneic female mice rendered diabetic with streptozotocine. Blood glucose was monitored for the rejection of engrafted islets. After certain period of time, tail to flank skin transplantation was performed either on mouse transplanted with HY mismatched islets or on sham treated mouse. The rejection was monitored by scoring gross pathology of the engrafted skin. Results: HY mismatched islets survived more than 300 days in 14 out of 15 mice. The acceptance of second party graft (male B6 islets) and the rejection of third party graft (male BALB/c islets) in these mice suggested the tolerance to islets with HY disparity. B6 Skin with HY disparity was rejected on day $25{\pm}7$. However, HY mismatched skin transplanted on the mice tolerated to HY mismatched islets survived more than 240 days. Tetramer staining in these mice indicated the CTL recognizing MHC Db/Uty was not deleted or anergized. Conclusion: The islet transplantation across HY disparity induced tolerance to HY antigen in C57BL6 mouse, which in turn induced tolerance to HY mismatched skin, which otherwise would be rejected within 25 days. The MHC tetramer staining suggested the underlying mechanisms would not be clonal deletion or anergy.

Comparative Analysis of Phospholipase D2 Localization in the Pancreatic Islet of Rat and Guinea Pig

  • To examine the localization pattern of phospholipase D2 (PLD2) in the pancreatic islet (the islet of Langerhans) depending on species, we conducted a morphological experiment in the rat and guinea pig. Since individual islets display a typical topography with a central core of B cell mass and a peripheral boundary of A, D, and PP cells, double immunofluorescent staining with a panel of antibodies was performed to identify PLD2-immunoreactive cells in the islets PLD2 immunoreactivity was mainly present in A and PP cells of the rat pancreatic islets. And yet, in the guinea pig, PLD2 immunoreactivity was exclusively localized in A cells, and not in PP cells. These findings suggest a possibility that PLD2 is mainly located in A cells of rodent pancreatic islets, and that the existence of PLD2 in PP cells is not universal in all species. Based on these results, it is suggested that PLD2 may play a significant role in the function of A and/or PP cells via a PLD-mediated signaling pathway.

Effect of Lactic Acid Bacteria on the Regulation of Blood Glucose Level in Streptozotocin-induced Diabetic Rats

  • To identify the treatment effect of lactic acid bacteria for diabetes, the treatment effects of a single administration of acarbose (a diabetes treatment drug) or lactic acid bacteria, and the mixture of acarbose and lactic acid bacteria on diabetes in a type 1 diabetes animal model, were studied. In this study, streptozotocin was inoculated into a Sprague-Dawley rat to induce diabetes, and sham control (Sham), diabetic control (STZ), STZ and composition with live cell, STZ and composition with heat killed cell, STZ and composition with drugs (acarbose) were orally administered. Then the treatment effect on diabetes was observed by measuring the body weight, blood glucose, and serum lipid. For the histopathological examination of the pancreas, the Langerhans islet of the pancreas was observed using hematoxylin and eosin staining, and the renal cortex, outer medullar, and inner medullar were also observed. The induced diabetes decreased the body weight, and the fasting blood glucose level decreased in the lactic-acid-bacteria-administered group and the mixture-administered group. In addition, the probiotic resulted in the greatest decrease in the serum cholesterol level, which is closely related to diabetes. Also, the hematoxylin and eosin staining of the Langerhans islet showed that the reduction in the size of the Langerhans islet slowed in the lactic-acid-bacteria-administered group. The histopathological examination confirmed that the symptoms of diabetic nephropathy decreased in the group to which viable bacteria and acarbose were administered, unlike in the group to which dead bacteria was administered. The mixture of lactic acid bacteria and acarbose and the single administration of lactic acid bacteria or acarbose had treatment effects on the size of the Langerhans islet and of the kidney histopathology. Thus, it is believed that lactic acid bacteria have treatment effects on diabetes and can be used as supplements for the treatment of diabetes.
