과산화아질산염과 DPPH 라디칼에 대한 해조추출물의 소거 활성 효과 (Studies on Screening of Seaweed Extracts for Peroxynitrite and DPPH Radical Scavenging Activities)

  • As a part of our search for novel antioxidants from the seaweeds, we have investigated radical scavenging effect for their crude extracts using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical, authentic peroxynitrite, and 3-morpholinsydnonimine (SIN-1), a peroxynitrite-generating species in vitro. Thirty-four seaweeds were screened for $ONOO^-$ and DPPH radical scavenging activities. A potent inhibitory effect against peroxynitrite generated by SIN-1 at $5{\mu}g/ml$ of methanol extracts was observed in order of Ishige okamurae(95.3%), Sargassum hemiphyllum(90.2%), Symphyocladia latiuscula(89.6%), Porphyra suborbiculata(86.7%), and Gelidium amamsii(85.9%), Also, a significant scavenging effect against direct authentic peroxynitrite was revekaled for methanol extracts of Ishige okamurae(66.2%) and Sargassum hemiphyllum(55.2%) and the acetone/methylene chloride(1:1) extract of Gigatina tenella (61.0%). In our measurement for evaluating the capacity to scavenge the stable free radical of DPPH, acetone/methylene chloride(1:1) extracts of Symphyocladia latiuscula, Gloiopeltis furcata, and Sargassum thunbergii and the methanol extract of Sargassum sp. showed an inhibitory potency of 85.8%, 82.8%, 74.1%, and 64.0%, respectively.

한국 남서해안 도초군도 무인도서의 하계 해조상 (Summer Algal Flora of Uninhabited Islands in Dochodo, Southwestern Coast of Korea)

  • The summer algal flora and community of 15 uninhabited islands in Dochodo, southwestern coast of Korea, were investigated from 9 June to 16 September 2007. A total 53 species (10 green, 14 brown and 29 red algae) of marine algae were identified. Among 15 uninhabited islands, the number of species observed was the highest as 33 species at Jeongdo and Gyeongchido the least as 24 species at Hugdo and Mido. The dominant species were Enteromorpha compressa, Ishige okamurae, Gloiopeltis furcata, Ulva pertusa and Sargassum thunbergii. The algal zonation of intertidal zone was figured out by Gloiopeltis furcata, Caulacanthus okamurae – Ulva pertusa, Sargassum thungergii – Gelidium amansii, Sargassum horneri from upper to lower zone. The flora investigated could be classified into six functional groups such as coarsely branched form (39.6%), filamentous form (25.8%), sheet form (14.1%), thick leathery form (9.9%), jointed calcarious form (5.6%) and crustose form algae (4.9%).

  • 한국 남해안 해남군에 위치한 무인도의 해조상과 군집조사가 1999년 6월 12일에서 29일까지 이루어졌다 조사결과 채집 동정된 해조류는 녹조류 10종, 갈조류 15종 및 흥조류 62종이었다. 우점종은 구멍갈파래와 지충이이며, 준우점종은 바위수염, 패, 톳, 불등풀가사리, 애기돌가사리이었다. 수직분포는 조간대 상부에서 하부까지 불등풀가사리, 애기가시덤불, 바위수염-참풀가사리, 애기풀가사리, 애기우뭇가사리, 구멍갈파래, 지충이-톳, 뜸부기, 진두발, 미역, 감태가 분포하였다.

Screening and identification of bioorganic light-emitting substances from marine macrophytes

  • Organic light-emitting materials in marine macrophytes from various coastal environments were identified. Twentyeight species from the solvent fractions were examined and identified as candidates for bioorganic light-emitting materials using photoluminescence (PL) spectra and gas chromatography-mass spectrometry. We selected 16 solvent fractions from a total of 1,221 prepared from Ishige okamurae, Sargassum confusum, Grateloupia elliptica, Chondracanthus intermedius, Porphyra yezoensis, Meristotheca papulosa, Gelidium amansii, and Scytosiphon lomentaria. The maximum light-emitting PL spectra appeared at various colors, mainly between blue and green, based on chromaticity coordinates, from solvent fractions of M. papulosa, G. amansii, G. elliptica, P. yezoensis, S. lomentaria, I. okamurae, and C. intermedius. These results will contribute to the development of novel organic light-emitting materials.

Antioxidant Efficacy of Extracts from a Variety of Seaweeds in a Cellular System

  • As a part of an ongoing search for antioxidants from marine sources, antioxidant activities of 24 kinds of seaweeds (4 green algae, 8 brown algae, and 12 red algae) were investigated. The seaweeds were extracted by acetone/dichloromethane and methanol, respectively. The antioxidant properties of both extracts were evaluated using four different activity tests, including degree of occurrence of intracellular reactive oxygen species (ROS), NO, lipid peroxidation, and GSH (glutathione) in mouse macrophage Raw 264.7 cells. The levels of intracellular reactive oxygen species (ROS) and GSH were measured using 2',7'-dichlorofluorescin diacetate (DCFDA) and monobromobimane as fluorescence probe, respectively. Moreover, the generation of NO and lipid peroxidation products were determined by each method based on the Griess reaction and TBARS assay. Solvent extracts from seaweeds such as Scytosiphon lomentaria, Prionitis cornea, Laruencia okamurae, Callophyllis japonica, Sargassum horneri, Dictyopteris divaricata, Lomentaria catenata, Corallina confuse, Ishige okamurae, and Ahnfeltiopsis flabelliformi exhibited high antioxidant activities in cellular oxidizing systems.

Structure of Intertidal Macroalgal Community at the Yeonpyeongdo Islets, Korea

  • Intertidal benthic algal communities on the shores of Gujido and Daeyeonpyeongdo islets, Korea, were examined in October 2007. At both sites, 45 seaweeds including 7 green, 6 brown and 32 red algae were identified. The number of species at Gujido(38 species) was 1.5 times higher than at Daeyeonpyeongdo(25 species), but seaweed coverage was very similar with about 20%. Dominant seaweeds in terms of coverage and importance value were Hildenbrandtia sp., Caulacanthus okamurae, Ulva pertusa, and Gelidium amansii at Gujido and U. pertusa, Hildenbrandtia sp., Gelidium divaricatum at Daeyeonpyeongdo. The vertical distribution pattern of the seaweeds was G. divaricatum-U. pertusa, Hildenbrandtia sp.-U. pertusa, Hildenbrandtia sp., Ishige okamurae from upper to lower intertidal zone but seaweed zonations were not observed on the Gujido rocky shore. At both sites, coarsely-branched forms were the dominant functional group in species number and percent cover(among benthic algal species). The rocky shores of the two sites were dominated by crustose coralline and green algae, whose presence generally results in decreased seaweed biodiversity and community stability. Therefore, the shores of the Yeonpyeongdo islets are of considerable environmental concern and should be monitored for seaweed species composition and community structure.

남해안 신지도의 해조군집 (Benthic Marine Algal Communities of Shinjido, Southern Coast of Korea)

  • 남해안 신지도 주변 해조군집의 종조성과 군집 구조 분석을 통하여 이 지역 해조 군집의 특성을 구명하고자 하였다. 이 연구에서 출현한 종은 총 120종으로 녹조류 15종, 갈조류 31종, 홍조류 74종이었다. 이들중 87종이 강독에서 그리고 104종이 동고리에서 출현하였다. 조간대 식생은 크게 3그룹으로 구분되며, 대표적인 종들로는 상부에서 Enteromorpha compressa, Ulva Pertusa, Porphyra suborbiculata, Ishige okamurae, 중부에서는 Hizikra fusiformis, Sargassum thunbergii, Gigartina intermedia, Corallina pilulifera, 하부에는 Pachymeniopsis elliptica, Gelidium amansii, Sargassum homeri 등이 분포하였다. 중요도 값이 높게 나타난 종은 Ulva pertusa, Ishige ekamurae, Hizikia fuisformis, Sargassum thunbergii, Corallina pilulifera, Gelidium amansii, Gigartina tenella로 조사지점 모두에서 공통적으로 나타났다. 기능형군별 분석에서 나타난 비율은 직립분기형 $36.7\%$, 사상형 $27.5\%$, 엽상형 $15.8\%$, 다육질형 $10.0\%$, 유절산호말형 $5.0\%$, 각상형 $5.0\%$로 나타났다.

한국 남서해안 다도해 해상국립공원의 하계 해조상 (Summer Algal Flora of Dadohae National Park, Southwestern Coast of Korea)

  • The summer algal flora and community of 9 islands in Dadohae National Park, southwestern coast of Korea, were investigated from June to September 2008. A total of 81 species (13 green, 22 brown and 46 red algae) of marine algae were identified. Among 9 islands, the number of species observed was the highest as 65 species at Jindo and the least as 37 species at Hongdo. The dominant species were Enteromorpha compressa, Ulva pertusa, Sargassum thunbergii, Gelidium amansii, Caulacanthus okamurae, Gloiopeltis furcata, and Symphyocladia latiuscula. The algal zonation of intertidal zone was figured out by Gloiopeltis furcata, Ishige okamurae, Caulacanthus okamurae-Enteromorpha compressa, Sargassum thungergii-Gelidium amansii, Symphyocladia latiuscula from upper to lower zone. The flora investigated could be classified into six functional groups such as coarsely branched form (38.1%), filamentous form (24.7%), sheet form (11.3%), thick leathery form (13.9%), jointed calcarious form (6.3%) and crustose form algae (5.7%). R/P and (R+C)/P values were 2.2 and 2.8, respectively. A cluster analysis of species occurrence was suggested that the number of marine algal species was different from greatly among the sampling sites.

Evaluation of Effective MMP Inhibitors from Eight Different Brown Algae in Human Fibrosarcoma HT1080 Cells

  • Matrix metalloproteinases (MMPs) are crucial extracellular matrices degrading enzymes that have important roles in metastasis of cancer progression as well as other significant conditions such as oxidative stress and hepatic fibrosis. Marine plants are on the rise for their potential to provide natural products that exhibit remarkable health benefits. In this context, brown algae species have been of much interest in the pharmaceutical field with reported instances of isolation of bioactive compounds against tumor growth and MMP activity. In this study, eight different brown algae species were harvested, and their extracts were compared in regard to their anti-MMP effects. According to gelatin zymography results, Ecklonia cava, Ecklonia bicyclis, and Ishige okamurae showed higher inhibitory effects than the other samples on MMP-2 and -9 activity at the concentrations of 10, 50, and $100{\mu}g/mL$. However, only I. okamurae was able to regulate the MMP activity through the expression of MMP and tissue inhibitor of MMP observed by mRNA levels. Overall, brown algae species showed to be good sources for anti-MMP agents, while I. okamurae needs to be further studied for its potential to yield pharmaceutical molecules that can regulate MMP-activity through cellular pathways as well as enzymatic inhibition.

A New Antioxidant Polyphenolic Compound from Two Korean Brown Algae

  • In the course of our researches for biologically active compound from Korean algae, purification of the methanolic extracts of two brown algae (Sagassum Sagamianum and Ishige Okamurae) collected off Jeju Island afforded an antioxidant polyphenolic compound (1). The molecular formular of 1 was established as C$\sub$24/H$\sub$16/ O$\sub$13/ on the basis of the FAB mass and $\^$13/C NMR spectrum. (omitted)
