Ultrastructural Study on the Cerebellar Purkinje Cell of the Head-Irradiated Rat (과량의 방사선 국소조사가 흰쥐 소뇌 Purkinje세포의 미세구조에 미치는 영향)

  • The acute irradiation effect on rat Purkinje cell was carried out. Anesthetized rats, weighing 200-250g each, were exposed their heads to the linear accelerator (ML-4MV) with the doses of 3,000 rads or 6,000 rads respectively. Irradiated rats were sacrificed by perfusion fixation under anesthesia, six hours, two days and six days following the irradiations. Rats were perfused with the fixative of 1% glutaraldehyde-1% paraformaldehyde solution (pH 7.4). Small pieces of cerebellar cortices were taken out. Tissue blocks were washed out, and were refixed in the 2% osmium tetroxide solution. After dehydration, tissues were embedded in the araldite mixture. Ultrathin sections stained with uranyl acetate-lead citrate solution, were examined with an electron microscope. The results observed were as follow; 1. Many dark Purkinje cells exhibited most severe cellular alterations on 6 hours. But after the 2 or 6 days, the cells exhibited only some alterations of cytoplasmic organelles. 2. Many granular and agranular endoplasmic reticula exhibited the fusion of cisterns. These reticular alterations were most severe on 6 hours following irradiation. But the alterations were hardly found on 6 days. 3. In the Golgi region, alterations including the adhesion of lamelliform cisterns, enlarged saccules, and increased number of vesicles, etc, were seen on 6 hours. But the Golgi complexes were almost recovered on 6 days. 4. Lysosomes were abundant on 6 hours or 2 days, but some residual bodies were found on 6 days. 5. Mitochondrial changes were also most severe at on hours, and they were recovered thereafter. From the results, it was concluded that the cerebellar Purkinje cells reacted to the high doses of irradiation by hyperactive protein synthesis, autolytic activities and energy metabolism. The reaction was most active in the early stage. It implies that motor-control function of Purkinje cells are severely disturbed in the early stage of irradiation.

  • The purpose of this study was to investigate the irradiation effects on the palatal mucosa. For this study, Sprague-Dawley strain rats were irradiated to their head and neck region with the dose of 5Gy and l0Gy by 6MV X -radiation and sacrificed on the experimental periods after irradiation. The authors observed the histological changes of the hard and soft palatal mucosae. The results were as follows: In the light microscopic examination, hydropic change on the basal cells, increased cell size of the epithelium, and decreased epithelial cell layers were observed on the 3hours, 6hours, and 12hours groups after irradiation. But, basal cell hyperplasia, increased epithelial cell layers, and elongated rate pegs were observed on the 3days group after irradiation. After then, these changes were recovered in the mucosa of the hard palate on the 7days and 14days groups, and in the mucosa of the soft palate on the 14days and 2&lays groups after irradiation. And such changes were greater in the mucosa of the soft palate than in that of the hard palate, and more prominent in l0Gy irradiated groups than in 5Gy irradiated groups.

Ultrastructural Study on the Substantia Nigra of the Head-Irradiated Rats (머리에 방사선 상해를 받은 흰쥐 흑색질의 미세구조)

  • An experimental study on the acute irradiation effects on the substantia nigra of head-irradiated rats were carried out. Rats anesthetized with sodium thiopental, were exposed only on their head areas with a single dose of 3,000 rads or 6,000 rads, respectively. Radiation was produced by Mitsubishi linear accelerator at the speed of 200 rads/min. Aminals were sacrificed on 6 hours, 2 days and 6 days following irradiations. By the perfusion fixation through the heart, rats were fixed with 1% glutaraldehyde-1% paraformaldehyde solution. Two hours later, brains were exposed and immersed in the same fixatives over night. Tissue blocks from subtantia nigra were punched out, and they were refixed in the 2% osmium tetroxide solution. Blocks were dehydrated through alcohol series, and embedded in the araldite mixture. Ultrathin sections were stained with uranyl acetate and lead citrate solutions, From the ultrastructural study, following results were made: 1. Six hours after irradiation, severe depletion of synaptic vesicles was occurred in the many axon terminals of the nigral neuropil. 2. Dramatical decrease of lysosomes and dense granules was observed. 3. Two days following irradiation, alterations of ribosomes, granular endoplasmic reticula, mitochondria, etc, were noticed. 4. Many of the malformations were seen to be repaired on the 6th day. 5. Above results were interpreted as follows. At the acute stage of heavy irradiation, neurotransmitters in the substantia nigra are released severely. But they are recovered within 6 days. It is concluded that acute head-irradiation may result severe disturbance of nigral motor control function during the first few days.

Radioprotective Effect of Red Ginseng in Irradiated Mice with High and Low Dose of Radiation (고선량 및 저선량 방사선 피폭에 대한 홍삼의 방사선 방호효과)

  • Studies were performed to determine the effect of Korean red ginseng (extract powder, spray-dried), it is made of choice 6-year-old raw ginseng roots, and processed by steaming and drying, on jejunal crypt survival, endogenous spleen colony formation, and apoptosis in jejunal crypt cells of irradiated mice. Jejunal crypts were protected by pretreatment of red ginseng (1 mg/head, single I.P. at 24hours before irradiation, p<0.05). Red ginseng administration before irradiation (1 mg/head, single I.P at 24hours before irradiation) resulted in an increase of the formation of endogenous spleen colony (p<0.05). The frequency of radiation-Induced apoptosis in intestinal crypt cells was also reduced by treatment of red ginseng both pretreatment (P.O.: 2 mg/ml of drinking water for 7 days, p<0.005, I.P.: 1 mg/head, single I.P. at 24 hours before irradiation, p<0.005) and post-treatment (1 mg/head, single I.P at 30 minutes after irradiation, p<0.05). These results indicated that Korean red ginseng might be a useful radio-protector, especially since it is a relatively nontoxic natural product. Further studies are needed to characterize better the promotion nature of red ginseng and its fractions.

Hyperacute Radiation Effect on Cerebral Cortex after Local Gamma-irradiation in the Rat Brain (단일 국소방사선 조사 후 백서 대뇌 피질의 초급성기 변화에 대한 연구)

  • Objective: We investigated the morphologic changes within 24 hours after a single ${\gamma}$-irradiation in the rat brain. Methods: Forty Sprague-Dawley rats were used. After a burr hole trephination on right parietal area, cerebral hemisphere was irradiated with 2Gy and 5Gy using iridium-192($^{192}Ir$), respectively. The effect was assessed at 4, 8, 12 and 24 hours after irradiation. The histological changes were scored following the detection of edema or disarray severity. TUNEL-positive cells exhibiting apoptotic morphology were counted in irradiated region. Results: Cortical edema and disarray were initially showed at 4 or 8 hour and almost all defined at 24 hour after irradiation. And the injury was wedge shape. TUNEL-positive cells were minimal at 8 hour after irradiation as the number of positive cells were $2.6{\pm}5.27$(n=5) after 2Gy, and $0.8{\pm}0.84$(n=5) after 5Gy. But, the number of apoptotic cells were increased markedly to $60{\pm}6.24$ at 12 hour after 2Gy and to $104{\pm}19.7$ at 24 hour after 5Gy. Conclusion: There were prominent morphologic changes immediately after ${\gamma}$-irradiation. And, apoptosis was increased according to the time period. These findings implicate that brain irradiation induces rapid apoptotic change, which may play an important role in the pathogenesis of radiation-induced pathologic conditions.

ALTERATION OF RADIATION-INDUCED HEMATOTOXICITY BY BU-ZHONG-YI-QI-TANG IN MOUSE

  • The purpose of this study reported here was to investigate the ability of Bu-Zhong-Yi-Qi-Tang (BZYQT), known to elevate hematopoietic functions, to protect mice undergoing treatment with whole body single gamma-irradiation. BZYQT was given (25 mg/kg B.W.) intraperitoneally at 36 and 12 hours before irradiation and 30 minute and 24 hours after irradiation. Recovery of neutrophil and lymphocyte counts was significantly stimulated by extract of BZYQT. Stimulated recovery by the extract from the BZYQT was also observed in thrombocyte. However, the anti-radiation effect of erythrocyte, hemoglobin and hematocrit was not as significant as that of leukocyte. Further studies are needed to better characterize the protective nature of BZYQT extract and its ingredients.

Optical Reactivity Modification of Titanium Oxide coatings on Ceramic filters by Nitrogen ion Implantation

  • We investigated the modification of optical response properties of titanium dioxide (TiO2) coatings on the ceramic water-purification filters by using ultraviolet-visible absorption spectroscopy and X-ray diffraction. The TiO2 coatings were prepared on ceramic substrate by e-beam evaporation method. These amorphous TiO2 were turned into anatase phase by heat treatment at $700^{\circ}C$ for 2 hours. The doping of N atoms into the TiO2 coatings was done by using 70KeV of N+ ion implantation with the dose of $1.0{\times}1017$ ions/cm2, followed by post-irradiation heat treatment at $550^{\circ}C$ for 2 hours. Methylene blue test of TiO2 coatings to solar irradiation showed that the post-evaporation heated TiO2 was photocatalytic and N-doped TiO2 reacted to the visible part of solar irradiation.

The Inhibitory UV-B Blocking Rate of Eyeglasses Lens on the Enzymes Denaturation in Cornea (각막 내 효소의 변성을 억제하는 안경 렌즈의 UV-B 차단율)

  • Purpose: To investigate the UV-B blocking rate of eyeglasses lens which can prevent enzymes denaturation in cornea. Methods: The denaturation degree of RNase A and catalase, superoxide dismutase (SOD) was determined by using Acrylamide gel electrophoresis after UV-B irradiation of 312 nm for 1, 3, 6, 24 and 96 hours. Also, the inhibitory effect of eyeglasses lens having UV-B blocking rate of 50%, 80%, 95% and 99% on the enzymes denatration was measured. Results: The denaturation of RNase A was induced by 1 hour-irradiation of UV-B. To inhibit RNase A denaturation after UV-B irradiation between 1 hour and 6 hours, UV-B blocking lens of 95% were effective. UV-B blocking lens of 99% suppressed the inhibition of RNase A denaturation after the UV-B exposure between 24 hours and 96 hours. The denaturation of catalase was not induced by 1 hourirradiation of UV-B. To inhibit enzyme denaturation after UV-B irradiation between 1 hour and 6 hours, UV-B blocking lens of 50% were effective. UV-B blocking lens of 95% suppressed the inhibition of enzyme denaturation induced by UV-B irradiation between 24 hours and 96 hours. The SOD denaturation was not induced by UV-B irradiation shorter than 6 hours exposure. The UV-B blocking lens of 50% could inhibit SOD denaturation after the UV-B irradiation for 24 hours. When SOD was exposed to UV-B for 96 hrs, SOD denaturation was inhibited by eyeglasses lens with UV blocking rate higher than 95%. Conclusions: The results demonstrated that the proper UV-B blocking rates of eyeglasses lens to inhibit the enzymes denaturatioin was different according to the types of enzymes and its inhibitory effect was effective only when eyeglasses lens had higher than certain UV-B blocking rate.

The degradation of EVA for the protection of solar cell by UV-rays irradiation (자외선 조사에 따른 태양전지 보호용 EVA의 열화)

  • We studied the degradation of EVA for the protection of solar cell by UV-rays irradiation. We investigated the reduction of electrical efficiency, photo transmmitance and degradation of EVA by UV-rays irradiation. We utilized the UV irradiation equiped with fluorescent 313nm UV lamp and radiated for 400 hours. For the chemial analysis, we used the UV-vis spectrometer, XPS and examined the degradation mechanism by UV irradiation. It is found that the discolored phenomena, the decrease of photo transmmitance and oxidation reaction is occured by UV irradiation on the EVA sample for the protection of solar cell.

Neutron irradiation of alloy N and 316L stainless steel in contact with a molten chloride salt

  • Capsules containing NaCl-MgCl2 salt with 316L stainless steel or alloy N samples were irradiated in the Ohio State University Research Reactor for 21 nonconsecutive hours. A custom irradiation vessel was designed for this purpose, and details on its design and construction are given. Stainless steel samples that were irradiated during exposure had less corrosive attack than samples exposed to the same conditions without irradiation. Alloy N samples showed no significant effect of irradiation. This work shows a method for conducting in-reactor irradiation-corrosion experiments in static molten salts and presents preliminary data showing that neutron irradiation may decelerate corrosion of alloys in molten chloride salts.
