Mineral Carbonation of Serpentinite: Extraction, pH swing, and Carbonation (사문암(Serpentinite)을 이용한 광물탄산화: Mg 추출과 pH swing 및 탄산화)

  • Mineral carbonation by indirect method has been studied by serpentinite as cation source. Through the carbonation of $CO_2$ and alkaline earth ions (calcium and magnesium) from serpentinite, the pure carbonates including $MgCO_3$ and $CaCO_3$ were synthesized. The extraction solvent used to extract magnesium (Mg) was ammonium sulfate ($(NH_4)_2SO_4$), and the investigated experimental factors were the concentration of $(NH_4)_2SO_4$, reaction temperature, and ratio of serpentinite to the extraction solvent. From this study, the Mg extraction efficiency of approximately 80 wt% was obtained under the conditions of 2 M $(NH_4)_2SO_4$, $300^{\circ}C$, and a ratio of 5 g of serpentinite/75 mL of extraction solvent. The Mg extraction efficiency was proportional to the concentration and reaction temperature. $NH_3$ produced from the Mg extraction of serpentinite was used as a pH swing agent for carbonation to increase the pH value. About 1.78 M of $NH_3$ as the form of $NH_4{^+}$ was recovered after Mg extraction from serpentinite. And, the main step in Mg extraction process of serpentinite was estimated by geochemical modeling.

Expression and Characterization of Calcium- and Zinc-Tolerant Xylose Isomerase from Anoxybacillus kamchatkensis G10

  • The enzyme xylose isomerase (E.C. 5.3.1.5, XI) is responsible for the conversion of an aldose to ketose, especially xylose to xylulose. Owing to the ability of XI to isomerize glucose to fructose, this enzyme is used in the food industry to prepare high-fructose corn syrup. Therefore, we studied the characteristics of XI from Anoxybacillus kamchatkensis G10, a thermophilic bacterium. First, the gene coding for XI (xylA) was inserted into the pET-21a(+) expression vector and the construct was transformed into the Escherichia coli competent cell BL21 (DE3). The expression of recombinant XI was induced in the absence of isopropyl-thio-${\beta}$-galactopyranoside and purified using Ni-NTA affinity chromatography. The optimum temperature of recombinant XI was $80^{\circ}C$ and measurement of the heat stability indicated that 55% of residual activity was maintained after 2 h incubation at $60^{\circ}C$. The optimum pH was found to be 7.5 in sodium phosphate buffer. Magnesium, manganese, and cobalt ions were found to increase the enzyme activity; manganese was the most effective. Additionally, recombinant XI was resistant to the presence of $Ca^{2+}$ and $Zn^{2+}$ ions. The kinetic properties, $K_m$ and $V_{max}$, were calculated as 81.44 mM and $2.237{\mu}mol/min/mg$, respectively. Through redundancy analysis, XI of A. kamchatkensis G10 was classified into a family containing type II XIs produced by the genera Geobacillus, Bacillus, and Thermotoga. These results suggested that the thermostable nature of XI of A. kamchatkensis G10 may be advantageous in industrial applications and food processing.

Formation of Environment Friendly Electrodeposition Films by CO2 Gas Dissolved in Seawater and Their Corrosion Resistance (해수 중 CO2 기체의 유입에 의한 환경 친화적인 전착 코팅막의 형성과 그 내식특성)

  • The peculiar feature of cathodic protection in seawater has the capability to form mineral calcareous deposits such as magnesium and calcium on metal surfaces. It is assumed that $OH^-$ ions are generated close to the metal surface as a result of cathodic protection and generated $OH^-$ ions increases the pH of the metal/seawater interface outlined as the following formulae. (1) $O_2+2H_2O+4e{\rightarrow}4OH^-$, or (2) $2H_2O+2e{\rightarrow}H_2+2OH^-$. And high pH causes precipitation of $Mg(OH)_2$ and $CaCO_3$ in accordance with the following formulae. (1) $Mg^{2+}+2OH^-{\rightarrow}Mg(OH)_2$, (2) $Ca^{2+}+CO{_3}^{2-}{\rightarrow}CaCO_3$. The focus of this study was to increase the amount of $CO{_3}^{2-}$ with the injection of $CO_2$ gas to the solution for accelerating process of the following formulae. (1) $H_2O+CO_2{\rightarrow}H_2CO_3$, (2) $HCO^{3-}{\rightarrow}{H^+}+CO{_3}^{2-}$. Electrodeposit films were formed by an electro-deposition technique on steel substrates in solutions of both natural seawater and natural seawater dissolved $CO_2$ gas with different current densities, over different time periods. The contents of films were investigated by scanning electron microscopy(SEM) and X-ray diffraction(XRD). The adhesion and corrosion resistance of the coating films were evaluated by anodic polarization. From an experimental result, only $CaCO_3$ were found in solution where injected $CO_2$ gas regardless of current density. In case of injecting the $CO_2$ gas, weight gain of electrodeposits films hugely increased and it had appropriate physical properties.

Activation Mechanism of Protease in the Germination of Mulberry Seeds (뽕나무 종자 발아시의 Protease 발현기구)

  • The activity change of mulberry seeds protease was compared during germination for 5 days at 28$^{\circ}C$ in the dark place after daily hormone injection of different concentration. The protease from germinated mulberry seeds for 4 days was partially purified and the enzyme characteristics was investigated. The protease activity of mulberry seeds treated by hormone was highest with 10 $\mu$m GA3 followed by 10 $\mu$M zeatin and 10 $\mu$M kinetin. The protease activity of mulberry seeds was increased by 14% with 10ml agar culture that control at 4th day of germination. The protease from mulberry seeds was purified 313 fold by DEAE-Toyo-pearl 650M, Butyl-Toyopearl, Hydrozylapatite and Toyopearl HW 55M. After purification, the specific activity of the enzyme was 175 units/mg. Optimum pH and temperature of protease from mulberry seeds was 5.0 and 37$^{\circ}C$, respectively. The protease was stable below 37$^{\circ}C$ and the enzyme activity was decreased by 50%, when incubated at 52$^{\circ}C$ for 10minutes. The protease activity of mulberry seeds was inhibited by metal ions such as mercury, iron, zinc, copper, but activited by magnesium, choromium, aluminium ions. The Km value of the protease was 0.89mM with azocasein as a subscribe.

Odor Reduction of Pig Wastewater Using Magnesia (in-situ test) (마그네시아를 이용한 돈분 폐수의 악취 저감(현장 시험))

  • In this study, we tried to obtain the optimal conditions to reduce odors generated from pig wastewater using magnesia (MgO) through in-situ test after producing a reactor for removing odors. For this purpose, the filling amount of magnesia, the injection amount of pig wastewater, the aeration method, the aeration amount and the aeration time were considered. The field experiment was conducted at Cheongwoon Livestock Farm, which has a pig wastewater reservoir. As the amount of magnesia added to the weight of wastewater (500 kg) increases, the amount of ammonia (NH3) and hydrogen sulfide (H2S) generated tended to gradually decrease. As a result of the test, ammonia and hydrogen sulfide in the pig wastewater decreased up to 65% and 77%, respectively, for 2 days aeration after 0.8% of magnesia was added to the reaction tank. The initial pH of the pig wastewater in the reactor was 8.2, and the pH was found to be 9.2 when magnesia was added up to 0.8%. In the light of this trend, it can be known that magnesia gradually increases the pH in the pig wastewater and makes it weakly alkaline. As the pH increases, part of the ammonia gas present in the pig wastewater vaporizes into the air and the remaining part is removed by precipitation after chemical bonding with dissolved magnesium ions and phosphate ions. In order to remove the odor of pig wastewater and turn it into compost, most of the existing livestock farms go through a six-month aeration process using microorganisms. In contrast, the current study proved the effect of removing odors from pig wastewater within 2 days through chemical reactions that do not affect microbial activity.

Highly Luminescent (Zn0.6Sr0.3Mg0.1)2Ga2S5:Eu2+ Green Phosphors for a White Light-Emitting Diode

  • Green phosphors $(Zn_{1-a-b}M_aM^{\prime}_b)_xGa_yS_{x+3y/2}:Eu^{2+}$ (M, M' = alkali earth ions) with x = 2 and y = 2-5 were prepared, starting from ZnO, MgO, $SrCO_3$, $Ga_2O_3$, $Eu_2O_3$, and S with a flux $NH_4F$ using a conventional solidstate reaction. A phosphor with the composition of $(Zn_{0.6}Sr_{0.3}Mg_{0.1})_2Ga_2S_5:Eu^{2+}$ produced the strongest luminescence at a 460-nm excitation. The observed XRD patterns indicated that the optimized phosphor consisted of two components: zinc thiogallate and zinc sulfide. The characteristic green luminescence of the $ZnS:Eu^{2+}$ component on excitation at 460 nm was attributed to the donor-acceptor ($D_{ZnGa_2S_4}-A_{ZnS}$) recombination in the hybrid boundary. The optimized green phosphor converted 17.9% of the absorbed blue light into luminescence. For the fabrication of light-emitting diode (LED), the optimized phosphor was coated with MgO using magnesium nitrate to overcome their weakness against moisture. The MgO-coated green phosphor was fabricated with a blue GaN LED, and the chromaticity index of the phosphor-cast LED (pc-LED) was investigated as a function of the wt % of the optimized phosphor. White LEDs were fabricated by pasting the optimized green (G) and the red (R) phosphors, and the commercial yellow (Y) phosphor on the blue chips. The three-band pc-WLED resulted in improved color rendering index (CRI) and corrected color temperature (CCT), compared with those of the two-band pc-WLED.

The Effect of chemical and physical properties of Korean tales on the decomposition of Malathion in dust formulations

  • The decomposition of malathion in dust for mulations prepared from four Korean tales as carriers during storage period has been studied. Amberlite CG-120, a cation exchange resin . which has higher cation exchange capacity than tales, was also used as a carrier in hope of finding out the effect of nagative charge upon the decomposition of malathion. Besides the original talc powders obtained directly from the mines, the hydrogen ion saturated forms were also used as carriers for comparisonal study. The saturated ions for the resin were hydrogen, sodium and magnesium. As the physical properties of the tales, colloid content, water adsorption capacity, PH, specific surface, phosphate fixing capacity and exchangeable canons were determined, and these properties were correlated with the amount of the decomposition. Following results were obtained from the experiment. 1. The malathion in the talc in dust was found to decompose around 10-15% ofthe total withina month. About 50% of the decom position that took place after a month was found to occur within a week. 2. The resin which has higher cation exchange capacity than the tales was highly effective in the decomposition of malathion compared with the tales. 3. In every case the saturation of the exchange complexes with hydrogen ion greatly accelerated the decomposition of malathion. 4. The most highly correlated physical properties with the decomposition were colloid content and specific surface of the tales. 5. The water adsorption and phosphate fixing capacities of the tales were found not to correlate with the amount of malathion decomposed. From the experimental results it was concluded that the active negative spots on the colloidal tales or the resin attract the electropositive phosphorus atom in a malathion molecule thereby inducing the decomposition easier. The presence of hydrogen ion nearby might cause a catalytic effect in the decomposition of malathion.

Equilibrium Concentration of Radionuclides in Cement/Groundwater/Carbon Steel System

  • Equilibrium concentrations of major elements in an underground repository with a capacity of 100,000 drums have been simulated using the geochemical computer code (EQMOD). The simulation has been carried out at the conditions of pH 12 to 13.5, and Eh 520 and -520 mV. Solubilities of magnesium and calcium decrease with the increase of pH. The solubility of iron increases with pH at Eh -520 mV of reducing environment while it almost entirely exists as the precipitate of Fe(OH)$_3$(s) at Eh 520 mV of oxidizing environment. All of cobalt and nickel are predicted to be dissolved in the liquid phase regardless of pH since the solubility limit is greater than the total concentration. In the case of cesium and strontium, all forms of both ions are present in the liquid phase because they have negligible sorption capacity on cement and large solubility under disposal atmosphere. And thus the total concentration determines the equilibrium concentration. Adsorbed amount of iodide and carbonate are dependent on adsorption capacity and adsorption equilibrium constant. Especially, the calcite turns out to be a solubility-limiting phase on the carbonate system. In order to validate the model, the equilibrium concentrations measured for a number of systems which consist of iron, cement, synthetic groundwater and radionuclides are compared with those predicted by the model. The concentrations between the model and the experiment of nonadsorptive elements cesium, strontium, cobalt nickel and iron, are well agreed. It indicates that the assumptions and the thermodynamic data in this work are valid. Using the adsorption equilibrium constant as a free parameter, the experimental data of iodide and carbonate have been fitted to the model. The model is in a good agreement with the experimental data of the iodide system.

Heavy Metal Contamination in Surface Water Used for Irrigation: Functional Assessment of the Turag River in Bangladesh

  • The aim of the present study was to evaluate the degree of metal contamination of the Turag River water and its suitability for irrigation. Twenty water samples were analyzed for physicochemical parameters and metals viz., calcium, magnesium, potassium (K), sodium, copper (Cu), zinc (Zn), iron (Fe), manganese (Mn), lead (Pb), cadmium (Cd), chromium (Cr), and nickel (Ni). All water samples were slightly alkaline to alkaline. Regarding electrical conductivity (EC), all samples were suitable for crop in soils with moderate permeability and leaching. Water samples were medium salinity and low alkalinity hazard classes. In terms of total dissolved solids (TDS), all samples were classified as freshwater. As per sodium adsorption ratio (SAR) and soluble sodium percentage (SSP), all samples were classified as excellent. No residual sodium carbonate (RSC) was detected in any of the samples, indicating suitability for irrigation; and all samples were considered very hard. Cr and Mn contents in all samples were above FAO guideline values and, therefore, these metals were considered toxic. Zn, Cu, Pb, Cd, and Ni concentrations were below acceptable limit for irrigation and do not pose a threat to soil environment. Significant relationships were found between EC and TDS, SAR and SSP, SAR and RSC, and SSP and RSC. The combinations of ions such as K-Zn, K-Fe, K-Cu, K-Mn, K-Pb, Zn-Fe, Zn-Cu, Zn-Mn, Fe-Mn, Cu-Mn, Cu-Pb and Mn-Pb exhibited significant correlation. This study revealed that Turag River water samples are contaminated with Cr and Mn. This fact should not be ignored because water contamination by metals may pose a threat to human health through food chain.

Production of Hard Water From Seawater Using Electrodialysis (해수로부터 전기투석 장치를 이용한 고경도 수 제조)

  • There are various ions in seawater. In order to use seawater as the drinking water, some elements are to be concentrated and other elements are to be removed. To obtain these characteristics using seawater, it is necessary to adjust seawater quality. Because calcium and magnesium are especially healthful to human bodies, it is required to concentrate these elements. In this study, the technology to obtain the hard water from seawater by electerodialysis was investigated. After concentrated water was produced using nanofiltration membranes, sodium chloride was eliminated from the concentrated water by electrodialysis. The hard water production from seawater was successfully achieved using electrodialysis in this study.
