Studies on analysis of the Korean lager beers. (I) "On the estimation of total carbohydrates, amino acids and peptides" (한국산 맥주성분에 관한 연구 (제 1 ) "Total carbohydrates, amino acids 및 peptides 의 정 에 관하여")

  • Total carbohydrates, amino acids and peptide-like substances in two kinds of Korean lager beers have been analyzed by the calorimetric method of Dreywood's anthrone reagent and Fowden's ninhydrin reagent. The samples were fractionated with column of ion-exchange resin. The experimental results are as follows; 1. Amounts of non-hydrolyzed carbohydrates in the part of column processed is 1. 82% and 1. 96 % (the value was measured by Bertrand's method). But the amounts of those measured by Dreywood's anthrone method are 5.57% and 4.25%, this values are much more than those of Bertrand's method. 2. It can be estimated the amounts of gum and dextrin are 3.75% and 3.30% in both two beers, by comparison of samples with the above mentioned two method. 3. The amounts of carbohydrates by anthrone reagent in acid-hydrolyzed beers are much increased than those of non-hydrolyzed, so it is suggested the presence of polymer carbohydrates which couldn't be detected by Bertrand's method. 4. Total amounts of amino acids are 0.015% and 0.025% (as glycine) in non-hydrolyzed beers measured by ninhydrine color reaction method, on the other hand the amount of amino acids in acid hydrolyzed beers are 0.06% and 0.056%, this is much more than those of nonhydrolysis. The different amounts means that of peptide-like substances. 5. It is necessary to determine the constituent of amino acid for the better taste of beer, and also it is desirable to check the role of carbohydrates in the course of fermentation, mashing and on lager beer for effective utilization of carbohydrate materials to eliminate the losses.

Isolation and Properties of Amino Acid Antimetabolite from Streptomyces sp. 182-27 (Streptomyces sp. 182-27 균주가 생산하는 아미노산 대사길항물질의 정제와 특성)

  • A Streptomyces strain No. 182-27, which produced amino acid antimetabolite, was isolated from soil. During the course of screening for new amino acid antimetabolites from the culture broths of Actinomycetes, we found that the strain produced a substance active against Gram-positive bacteria and its activity was reversed by L-Ieucine on the synthetic minimal agar medium in the culture broth. The morphological and cultural characteristics serve to identify the producing organism strain 182-27 as the Streptomyces, although the species of this strain should be resolved in further studies. Fermentation was carried out in the synthetic medium at $28^{\circ}C$ for 78 hours. The fermentation yield reached about 2 mg per liter of the broth. Purification was done by ion exchange resin, active carbon, silica gel column chromatography and obtained 20 mg of pure active substance from the 20 $\ell$ culture broth. The 182-27 substance was obtained as white powder, mp 18SoC. From the physicochemical characteristics of the substance, it was amino acid like substance but unknown about its chemical structure. It is active against some Gram-positive bacteria and reversed by L-Ieucine.

Determination of major and minor elements in low and medium level radioactive wastes using closed-vessel microwave acid digestion (밀폐형 극초단파 산분해법을 이용한 중${\cdot}$저준위 방사성폐기물의 성분 원소 분석)

  • The conditions are obtained for the decomposition of solid radioactive wastes, including ion exchange resin, zeolite, charcoal, and sludge from nuclear power plant. In the process of decomposing the radioactive wastes was used the microwave acid digestion method with mixed acid. The solution after acid digestion by the following method was colorless and transparent. Each solution was analyzed with ICP-AES and AAS and the recovery yield for 5 different elements added into the simulated radioactive wastes were over $94{\%}$. The elemental analysis of destructive low and medium level radioactive wastes by the proposed microwave acid digestion conditions concerning the chemical characteristics of each radioactive waste are expected to be useful basic data for development of optimal glass formulation.

Analytical Method of L-Ascorbic Acid Content in Green Tea (녹차중(綠茶中)의 L-Ascorbic Acid의 정량법(定量法)에 관한 연구(硏究))

  • Effects of interfering substances on the determination of L-ascorbic acid (AsA) in green tea and it's extracts by 2,4-dinitrophenylhydrazine (DNP) method was studied. and the removal of these interfering substances was also investigated. Under the condition prescribed for DNP method, AsA content of green tea are effected by some sugar, reductones, dicarbonyl compounds, organic acids, amino acids and others. All interfering substances except amino acids were eliminated by the chloroform extraction after adding o-phenylendiamine to sample solution. and remaining amino acids were eliminated almost completely by the treatment with ion exchange resin$(Amberlite{\;}IR-120H^{+})$. After removing the interfering substances by the above mentioned procedure, total AsA in green tea was determined by DNP method. The values obtained by this method were in good agreement with those by thin layer chromatography (TLC) method. and the method was more rapid and simple than TLC method.

Hydrolysis of Cellulose by Immobilized Cellulase in a Packed Bed Reactor (충진층 반응기에서 고정화 cellulase에 의한 셀룰로스 가수 분해)

  • Immobilized cellulase on weak ion exchange resin showed a typical Langmuir adsorption isotherm. Immobilized cellulase had better stability with respect to pH and temperature than free cellulase. Kinetics of thermal inactivation on free and immobilized cellulase followed first order rate, and immobilized cellulase had a longer half-life than free cellulase. The initial rate method was used to characterize the kinetic parameters of free and immobilized enzyme. The Michaelis-Menten constant $K_m$ was higher for the immobilized enzyme than it was for the free enzyme. The effect of the recirculation rate on cellulose degradation was studied in a recycling packed-bed reactor. In a continuous packed-bed reactor, the increasing flow rate of cellulose decreased the conversion efficiency of cellulose at different input lactose concentrations. Continuous operation for five days was conducted to investigate the stability of long term operation. The retained activity of the immobilized enzymes was 48% after seven days of operation.

Rapid Extraction of DNA using Ion Exchange Resin for Early Detection of Mycobacterium tuberculosis by the Polymerase Chain Reaction (결핵균 PCR에서 이온교환수지를 이용한 신속한 DNA 분리)

  • Background: The extraction methods of DNA from clinical samples are the major obstacle to use the PCR(polymerase Chain Reaction) in routine labortary for early detection of M. tuberculosis. We tried to improve the extraction method of DNA from sputum for establishment of the PCR in routine labortary by reducing the possibility of cross contamination and performing it easily and safely. Methods: We used the $InstaGene^{TM}$ DNA extraction kit(BioRad Co.) using Chelex 100 ion exchange resin for preparation of DNA. We compared InstaGene method in 100 cases of sputum from proteinase K method which is known as the most commonly used method for DNA purification(Experiment 1). And we compared InstaGene method in 98 cases of sputum from Microwave method developed by a company in Korea(Experiment 2). In experiment 1,245bps of IS6110 were amplified and then 188bps were amplified by nested PCR. In experiment 2,536bps in primary PCR and 276bps in nested PCR were amplified and analysed by agarose gel electrophoresis and EtBr staining. Results: When we chose AFB smear, culture, or AFB smear and culture as a standard test, PCR had low specificity and positive predictive value in both experiments. The InstaGene method has higher value in sensitivity and negative predictive value significantly than proteinase K method. The InstaGene method and the Microwave methods were similar in sensitivity, specificity, positive predictive value and negative predictive value. Conclusion: Even though both methods had lower possibility of cross contamination, shorter time requirement, simplicity, and economic advantages than Proteinase K method, the InstaGene method was a little simpler than the Microwave method. Therefore, in terms of usefulness in clinical application, the Instagene method seems to be the most useful method in DNA extraction for detection of M. tuberculosis using PCR. The reliability of this method will be clarified by further studies with enough clinical samples.

Purification of the Glycomacropeptide from Cheese Whey (치즈 유청으로부터 Glycomacropeptide의 분리.정제)

  • Glycomacropeptide(GMP) was purified from cheese whey which is obtaining as a byproduct in cheese producing. Cheese whey was first concentrated 10 times with a ultrafiltration aparratus, and then heated at 95$^{\circ}C$ for 5 min. The concentrated fraction was centrifuged at 20,000$\times$g for 30 min to remove fat layer. The supernatant layer enriched GMP protein was fractionated by ion exchange chromatography on DEAE-Sepharose Fast Flow column. GMP was bound to DEAE resin and eluted with 0.1~0.25 M NaCl when using a linear NaCl gradient from 0 M to 0.5 M. The purified GMP gave a single band of 24 kDa which seems to be trimer molecular weight in SDS-PAGE, and migrated to the same molecular weight with control GMP obtained commercially. Its amino acid composition were consistent with that of standard GMP. About 0.71 g of GMP was recovered from 1 L of cheese whey. These results indicate that glycomacropeptide could be simply purified from cheese whey by using ultrafiltration and DEAE column chromatography.

The Study on the Separation of the Subsidiary Elements in Iron and Steel by Using Ion Exchangers (II). The Separation of Anions (이온 교환수지에 의한 철 및 강의 분석에 관한 연구 (제2보). 음이온 성분의 분리)

  • The quantitative separations of a mixture containing equal amount of each anion such as Si(IV), As(V), P(V), S(VI), W(VI) and Cr(VI) are carried out by the elution through 20${\times}3.14cm^2$ column of anion exchange resin, Dowex 1${\times}$8. The eluents are a mixture of 0.07 M hydrochloric acid and 0.03 M sodium chloride (pH = 1.30) for Si(IV), As(V) and P(V) species, a mixture of 0.6 M sodium chloride and 0.3 M sodium hydroxide for S(VI), W(VI) and Cr(VI) species, and 0.1 N sodium sulfite (pH = 3.48) for P(V) and As(V) species. The subsidiary anions in a standard mixture such as Si(IV), As(V), S(VI), P(V) and W(VI) are separated together from large amount of Fe(III) by the elution through 30cm${\times}3.14cm^2$ column of the resin, Dowex${\times}$50w${\times}$12, using a mixture of 0.1 M sodium nitrate and 2 percent dimethylsulfoxide aqueous solution as an eluent. Si(IV), As(V), S(VI), P(V) and W(VI) eluted together are separated quantitatively under the same conditions as in the separations of the anion mixture. By the conditions obtained in the separations of the standard mixture, Fe(III) and all of the subsidiary anions in steel are quantitatively separated.

  • Enterotoxigenic E. coli is one of the major causative agents of the infantile diarrhea and traveler's diarrhea. The heat-stable enterotoxin (ST) is thought to be a virulence factor in the pathogenesis of the diarrhea and to be a maker for identification of the enterotoxigenic E. coli from non pathogenic E. coli. ST producing E. coli KM-7 strain was isolated from the swine and molecular cloning of ST gene of KM-7 strain. Transformant eKT-53 $(ST^+,\;LT^-)$ was selected by infant mouse assay (IMA). The culture supernatant of eKT-53 strain was performed purification by multipled steps. The culture supernatant (crude ST) was purified by sequentially applying batch adsorption chromatography on Amberlite XAD-2 resin, ion exchange chromatography on DEAE-Sephacel anion exchanger, gel filtration chromatography on Bio-Gel P-6 and preparative polyacrylamide slab gel electrophoresis. About 113-fold purification was achieved with a yield of about 11% of crude ST and the minimum effective dose(MED) of this purified ST was about 2.8ng in IMA. Homogeneity of purified ST was demonstrated by showing a single band in analytical SDS polyacrylamide disc gel electrophoresis.

Characteristics of Heavy Metal Biosorption by Pseudomonas cepacia KH410 (Pseudomonas cepacia KH410의 중금속 흡착특성)

  • An ubiquitous bacterium, Pseudomonar cepacia KH410 was isolated from fresh water plant root and identified. Adsorption of heavy metals of lead, cadmium and copper by this strain was investigated. Optimal conditions foradsorption was 1.0 dry g-biomass, at pH 4.0 and temperature of $40^{\circ}C$. Adsorption equilibrium reached max-imum after 120 min in 1000 mg/l metal solutions. The adsorption capacity (K) of lead was 5.6 times higher thancadmium and 4.0 times higher than that of copper. Adsorption of lead was applicable for Langmuir modelwhereas Freundlich model for cadmium and copper, respectively. Adsorption strength (1/n) of heavy metal ionswere in the order of lead>copper>cadmium. Uptake capacity of lead, cadmium and copper by dried cell was83.2,42.0,65.2 mg/g-biomass, respectively. Effective desorption was induced 0.1 M HCI for lead and 0.1 $HNO_3$ for cadmium and copper. Pretreatment to increase ion strength was the most effective with 0.1 M KOH.Uptake by immobilized cell was 77.8,58.5,71.2 mg/g-biomass for lead, cadmium and copper, respectively. Theimmobilized cell was more effective than ion exchange resin on removal of heavy metals in solution containinglight metals.
