Agrobacterium tumefaciens-mediated transformation(ATMT) of Flammulina velutipes was used to produce a diverse number of transformants to discover the functions of gene that is vital for its variation color, spore pattern and cellulolytic activity. Futhermore, the transformant pool will be used as a good genetic resource for studying gene functions. Agrobacterium-mediated transformation was conducted in order to generate intentional mutants of F. velutipes strain KACC42777. Then Agrobacterium tumefaciens AGL-1 harboring pBGgHg was transformed into F. velutipes. This method is use to determine the functional gene of F. velutipes. Inverse PCR was used to insert T-DNA into the tagged chromosomal DNA segments and conducting sequence analysis of the F. velutipes. But this experiment had trouble in diverse morphological mutants because of dikaryotic nature of mushroom. It needed to make monokaryotic fruiting varients which introduced genes of compatible mating types. In this study, next generation sequencing data was generated from 28 strains of Flammulina velutipes with different phenotypes using Illumina Hiseq platform. Filtered short reads were initially aligned to the reference genome (KACC42780) to construct a SNP matrix. And then we built a phylogenetic tree based on the validated SNPs. The inferred tree represented that white- and brown- fruitbody forming strains were generally separated although three brown strains, 4103, 4028, and 4195, were grouped with white ones. This topological relationship was consistently reappeared even when we used randomly selected SNPs. Group I containing 4062, 4148, and 4195 strains and group II containing 4188, 4190, and 4194 strains formed early-divergent lineages with robust nodal supports, suggesting that they are independent groups from the members in main clades. To elucidate the distinction between white-fruitbody forming strains isolated from Korea and Japan, phylogenetic analysis was performed using their SNP data with group I members as outgroup. However, no significant genetic variation was noticed in this study. A total of 28 strains of Flammulina velutipes were analyzed to identify the genomic regions responsible for producing white-fruiting body. NGS data was yielded by using Illumina Hiseq platform. Short reads were filtered by quality score and read length were mapped on the reference genome (KACC42780). Between the white- and brown fruitbody forming strains. There is a high possibility that SNPs can be detected among the white strains as homozygous because white phenotype is recessive in F. velutipes. Thus, we constructed SNP matrix within 8 white strains. SNPs discovered between mono3 and mono19, the parental monokaryotic strains of 4210 strain (white), were excluded from the candidate. If the genotypes of SNPs detected between white and brown strains were identical with those in mono3 and mono19 strains, they were included in candidate as a priority. As a result, if more than 5 candidates SNPs were localized in single gene, we regarded as they are possibly related to the white color. In F. velutipes genome, chr01, chr04, chr07,chr11 regions were identified to be associated with white fruitbody forming. White and Brown Fruitbody strains can be used as an identification marker for F. veluipes. We can develop some molecular markers to identify colored strains and discriminate national white varieties against Japanese ones.
Considering that the number of cases in which a structure foundation is located on weathered rock has been increasing recently, for adequate design bearing capacity of a foundation on weathered rock, allowable bearing capacities of such foundations in geotechnical investigation reports were studied. With reference to the study results, the allowable bearing capacity of a foundation on weathered rock was approximately 400-700 kN/m2, with a large variation, and was considered a conservative value. Because the allowable bearing capacity of the foundation ground is an important index in determining the foundation type in the early design stage, it can have a significant influence on the construction cost and period according to the initial decision. Thus, in this study, six large-scale plate-bearing tests were conducted on weathered rock, and the bearing capacity and settlement characteristics were analyzed. According to the test results, the bearing capacities from the six tests exceeded 1,500 kN/m2, and it shows that the results are similar with the one of bearing capacity formula by Pressuremeter tests when compared with the various bearing capacity formula. In addition, the elastic modulus determined by the inverse calculation of the load-settlement behavior from the large-scale plate-bearing tests was appropriate for applying the elastic modulus of the Pressuremeter tests. With consideration of the large-scale plate-bearing tests in this study and other results of plate-bearing tests on weathered rock in Korea, the allowable bearing capacity of weathered rock is evaluated to be over 1,000 kN/m2. However, because the settlement of the foundation increases as the foundation size increases, the allowable bearing capacity should be restrained by the allowable settlement criteria of an upper structure. Therefore, in this study, the anticipated foundation settlements along the foundation size and the thickness of weathered rocks have been evaluated by numerical analysis, and the foundation size and ground conditions, with an allowable bearing capacity of over 1,000 kN/m2, have been proposed as a table. These findings are considered useful in determining the foundation type in the early foundation design.
The study examines changes in calcium volume on born by comparing two figures; one is measured by dual energy computed tomography(DECT) followed by applying variation in monochromatic energy selection(keV), material decomposition(MD), and material suppressed iodine(MSI) analysis, and the other is measured by conventional single source computed tomography(CSCT). For this study, based on CSCT images taken by using human mimicked phantom, 70, 100, 140 keV and MSI, MD material calcium weighting(MCW) and MD material iodine weighting(MIW) of DECT were applied respectively. Then calculated calcium volume was converted to Agatston score for comparison. Volume of human mimicked phantom was in inverse proportion to keV. The volume decreased while keV increased(p<0.05). Several parameters of hydraulic architecture relating to hydraulic conductance in xylem vessels were investigated in the current-year shoots of six species of deciduous oak trees. The above parameters were also investigated in the sprouts of Quercus mongolica and Q. variabilis, as well as in the seedlings of Q. mongolica and Q. acutissima. The values of specific conductivity, leaf specific conductivity and Hagen-Poiseuille's relative hydraulic conductivity relating to vessel diameter of Q. dentata were the highest in all of the species studied. The above values of most of the species studied were higher in May-June than in September-October because of increasing the vessel embolism by cavitation and so on through the growing season. The estimated values of relative hydraulic conductivity of vessel by Hagen-Poiseuille's empirical equation and the real values of hydraulic conductivity presented positive relationships in most of the species studied. Huber value and leaf specific conductity using leaf area or leaf weight generally exhibited similar patterns each other even if having some exceptions. The hydraulic conductances of sprouting shoots were much better than those of normal growing shoots in Q. rnongolica and Q. variabilis. The specific conductivity and leaf specific conductivity were rapidly decreased by the vessel embolism through cavitating just after cutting the shoots in Q. mongolica and Q. acutissima seedlings. Diurnal changes of the conductivities in the seedlings of Q. mongolica and Q. acutissima presented the possibility of their self-controlling of conductance by active moisture absorption under mild water stress. Specific conductivity and leaf specific conductivity, and so on of Q. acutissima seedlings subjected to periodical moisture stress or not have decreased through the growing season, but the influences of moisture stress to the conductance were not proved definitely because of influencing similarly and simultaneously to the development of xylem and leaf having inverse relation in the influences. The values of conductivities were higher generally in middle or upper parts of stems than root collar in the seedlings.
Background: To decide the optimal antibiotics and application of chest tube, examination of pleural fluid is fundamental in the management of empyema. Some criteria for drainage of pleural fluid have been recommended but some controversies have been suggested. Recently, newer radiologic methods including ultrasound and computed tomography scanning, have been applied to the diagnosis and management of pleural effusions. We undertook a retrospective analysis of 30 patients with pleural effusion who had CT scans of the chest in order to apply the criteria of Light et al retrospectively to patients with loculation and to correlate the radiologic appearance of pleural effusions with pleural fluid chemistry. Method: We analyzed the records of 30 out of 147 patients with pleural effusion undergoing chest CT scans. Results: 1) Six of the pleural fluid cultures yielded gram negative organisms and three anaerobic bacterias and one Staphylococcus aureus and one non-hemolytic Streptococci. No organism was cultured in ninteen cases(63.0%). 2) The reasons for taking chest CT scans were to rule out malignancy or parenchymal lung disease(46.7%), poor response to antibiotics(40.0%), hard to aspirate pleural fluid(10.0%) and to decide the site for chest tube insertion(3.3%). 3) There was no significant correlations between ATS stages and loculation but there was a tendency to loculate in stage III. 4) There was a significant inverse relationship between the level of pH and loculation(p<0.05) but there appeared to be no relationship between pleural fluid, LDH, glucose, protein, loculation and pleural thickening. 5) In 12 out of 30, therapeutic measures were changed according to the chest CT scan findings. Conclusion: We were unable to identify any correlations between the plerual fluid chemistry, ATS stages and loculations except pH, and we suggest that tube thoracotomy should be individualized according to the clinical judgement and serial observation. All patients with empyema do not need a chest CT scan but a CT scan can provide determination of loculation, guiding and assessing therapy which should decrease morbidity and hospital stay.
The wall shear stress in the vicinity of end-to end anastomoses under steady flow conditions was measured using a flush-mounted hot-film anemometer(FMHFA) probe. The experimental measurements were in good agreement with numerical results except in flow with low Reynolds numbers. The wall shear stress increased proximal to the anastomosis in flow from the Penrose tubing (simulating an artery) to the PTFE: graft. In flow from the PTFE graft to the Penrose tubing, low wall shear stress was observed distal to the anastomosis. Abnormal distributions of wall shear stress in the vicinity of the anastomosis, resulting from the compliance mismatch between the graft and the host artery, might be an important factor of ANFH formation and the graft failure.
