Actions of Korean Ginseng and Benzoyl Peroxide on Inflammation Relevant to Acne (여드름과 관련된 염증에 대한 고려인삼과 벤조일퍼옥시드의 영향)

  • The intradermal injection of Propionibacterium acnes (ATCC 6919) into the ears of female Sprague-Dawley rats produced a chronic inflammation with the formation of acneiform lesions. Inflammation was characterized by more than four times of ear thickness and 2.8 times of ear weight at day 21. Histologically, massive infiltration of neutrophils, macrophage and lymphocytes, hyperplastic epidermis, comedones containing keratin mass and inflammatory materials were observed. Both ginseng saponin and extract from Korean red ginseng significantly redliced the ear thickness and their effects were similar to that of benzoyl peroxide. Ginseng samples and beneoyl peroxide modified lipid constituents of P. acnesinjected rat ear tissues. Even though no marked histological changes in inflammatory lesions were observed in ginseng-treated ear tissues, Korean red ginseng showed a possibility of reduce in the risk of acne development.

  • The Effects of Hoichunyanggyuksan on the Anti-allergic Effect, Analgesic Action, Anti-inflammatory Action and Antipyretic Action. Experimental studies were done to research the clinical effects of Hoichunyanggyuksan on the Anti-allergic effect, Analgesic action, Anti-inflammatory action and Antipyretic action. The results obtained as follows; 1. On vascular permeability responses to intradermal histamine, Hoichnyanggyuksan showed significant effect. 2. In the homologous PCA provoked by the IgE-like antibody against white egg albumin, Hoichunyanggyuksan showed the decreasing tendency, but was none significant effect. 3. In the delayed type hypersensitivity responses to Picryl chloride, Hoichunyanggyuksan was proved significant effect. 4. In the delayed type hypersensitivity resposes to SRBC, Hoichunyanggyuksan revealed significant effect. 5. In Anti-pyretic action by yeast method, Hoichunyanggyuksan showed significant effect. 6. In Anti-inflammatory action by carrageenin method, Hoichunyanggyuksan showed significant effect. 7. In analgesic action by acetic acid method, Hoichunyanggyuksan was recognized significantly.

Local Irritation of DA-125 (DA-125의 국소자극성)

  • DA-125, a new anthracycline derivative, shows significant anticancer activities. We conducted a study to examine the local irritating effect of DA-125 using mice and rabbits. In the skin test in mice, intradermal injection of 0.4 mg of DA-125, compared to a dosage of 0.2 mg of adriamycin, had weak irritating potentials to induce skin ulceration and erythematous induration. A dosage of 0.6 mg of DA-125 produced similar degree of lesions in perivascular irritation model to that of 0.2 mg of adriamycin, but the healing time was shorter in the case of mice treated with DA-125. In ocular irritation study in rabbit, the highest M.O.I.(mean ocular irritation index) of 0.5% DA-125 solution was 0.67, therefore DA-125 could be considered as a practically non-irritating anticancer agent. These results suggest that substitution of DA-125 for Adriamycin would reduces the possibility of outbreaks of local irritation and the severity of the lesions.

Prevalence of Anhidrosis in Thoroughbred Racehorses in Korea

  • The primary means of heat dissipation in horse results from the evaporation of sweat. Anhidrosis refers to a decreased ability or loss of ability to sweat in response to appropriate stimuli. This disease is fatal and causes poor performance, increased body temperature, collapse, convulsion and even death. There are some reports about this disease abroad but not in Korea. We performed intradermal epinephrine test to investigate the prevalence of anhidrosis in Thoroughbred racehorses (n=50). The prevalence was 22% and it was similar to that of other countries. There does not appear to be sex, foaling country, coat color, age or pedigree of dam predispositions. In this study, we found the presence of anhidrosis in Korea. Anhidrosis should be prevented for economical purposes and general animal welfare.

  • Eye irritation, primary skin irritation and skin sensitization tests for Aloewhite were tested in New Zealand White rabbits and Hartley guinea pig. In primary skin irritation test of male New Zealand White rabbits, body weights were not significantly changed and Primary Irritation Index (PII) was O.47, indicating Aloewhite as mildly irritating material. In ocular irritation test, any injury on iris, conjunctival membrane, and cornea in New Zealand White rabbits was not observed. No injuries of the ocular mucous membrane were also recorded. Skin sensitization was tested in guinea pig after intradermal and epicutaneous induction and graded 1 with zero % sensitization rate. These results indicate that Aloewhite was not considered to be irritant in test organs of animals.

  • Pedunculagin is an ellagitannin purified from Alnus hirsuta var. microphylla. Betulaceae. The effects of pedunculagin on immune system have been characterized to induce enhancement of NK (natural killer) cell cytotoxicities against tumor cells. Here, we report the evaluation of the effects of pedunculagin on the growth of murine Bl6-F10 melanoma in vivo. After the intradermal inoculation of Bl6-F10 melanoma, Bl6-F10 tumors grew progressively in immunocompetent syngenic C57BL/6 mice. The mice treated with pedunculagin(10 mg/kg, every 48 hrs) resulted in a significant improvement in survival. Inhibitory effects of pedunculagin on lung metastasis in C57BL/6 mice were also detected. Summarizing treatment with pedunculagin has a significant antitumor effect upon Bl6-F10 murine melanoma.

  • Although rare, antihistamines can cause adverse effects, including drug-induced eruptions or anaphylaxis. A 4-year-old child visited the pediatric department of a hospital for skin eruptions after administration of antihistamines, (e.g., ucerax [hydroxyzine] or leptizine [levocetirizine]), for cholinergic rashes; he did not have pruritus. Skin prick, intradermal, and drug provocation tests were performed to determine the relationship between the antihistamines and eruptions. Levocetirizine induced wheals in the skin prick test and a rash in the oral drug provocation test. In contrast, ketotifen induced no reaction in the skin prick test but showed a positive reaction in the oral provocation test. Our case report highlights that children can experience the same types of adverse reactions as seen in adults, and cross-reactivity between various antihistamines can occur.

Effect of Chromium Dietary Supplementation on the Immune Response and Some Blood Biochemical Parameters of Transport-stressed Lambs

  • Forty-eight Naemi lambs (avg. BW 31.7 kg) were transported by truck for a distance of 1,450 km from Al-Jouf to Riyadh, Saudi Arabia. On arrival day, the lambs were randomly allocated to four groups receiving diets supplemented with 0.0, 0.3, 0.6 and 0.9 ppm organic chromium (Cr). Each group consisted of four separately housed replicates of three lambs each. The animals were fed ad libitum on a grower diet for 84 days. Blood samples were obtained shortly before transportation, upon arrival and at weekly intervals thereafter from all lambs for analysis of plasma and serum. Plasma glucose and serum cortisol, total protein, albumin, urea-N and total cholesterol concentrations were determined. A cursory clinical examination of the lambs, along with rectal temperature, was undertaken at different intervals during the experiment. The lambs were inoculated each with 2 ml i.v. chicken red blood cells (CRBC) on days 0, 21, and 42. Serum total, IgG and IgM antibody titers were determined at weekly intervals post-immunization. An in vivo intradermal hypersensitivity test was carried out on 6 lambs from each group on days 10 and 70. Transportation of the lambs resulted in a significant (p<0.001) elevation of serum cortisol, total protein and albumin levels, as well as increased plasma glucose concentration, with corresponding decrease in total cholesterol, while blood urea-N remained largely unchanged. These constituents returned to normal levels during subsequent weeks, with no significant differences in their concentrations being observed between the Cr-supplemented groups and controls. Rise in rectal temperature after transportation was reduced to a greater extent (p<0.05) in Cr-supplemented versus control lambs. Total, IgG and IgM antibody titers against CRBC rose significantly (p<0.05) during immunizations in all groups, with significantly and linearly higher (p<0.05) total and IgG titers in Cr-supplemented versus control lambs. By contrast, no significant effect due to Cr supplementation was recorded in IgG titers, which increased equally in Cr-fed and control groups. Skin thickness in response to intradermal inoculation of phytohaemagglutinin (PHA) was also significantly (p<0.01) increased as a result of Cr supplementation. These results indicate that dietary Cr supplementation might be useful during stress especially for enhancing immune responses in transport-stressed lambs.
