소의 뇌로부터 Inositol Triphosphate Kinase의 정제 (Purification of Inositol Triphosphate Kinase from Bovine Brain)

  • 세포막 정보전달 과정에 참여하는 효소로 inositol triphosphate($InsP_3$)를 분해하는 $InsP_3$ kinase를 소의 뇌 조직으로 부터 새로운 정제방법을 개발하고 isozyme들의 특성을 관찰하여 다음과 같은 결과를 얻었다. 분쇄한 소의 뇌조직을 PEG로 단백침전하고 DEAE cellulose chromatography 하여 $InsP_3$ kinase I, II의 두 isozyme을 얻었으며, 각각의 isozyme 분획을 Matrix green gel 및 calmodulin-Affigel 15 column으로 chromatography하였다. Phenyl-TSK HPLC하여 정제하였으며 I은 3,103배, II는 2,310 배의 정제를 나타내었다. 정제 단계에서 specific activity를 비교해 볼 때 Matrix green gel chromatography가 DEAE cellulose에서 보다 I이 30배, II가 약 2.3배로 나타났고 정제배수도 I이 17.2%에서 62.1%로 II가 16.6%에서 38%로 나왔으나 calmodulin-Affigel 15과 비교시는 큰 차이가 없었다. 그러므로 Matrix green gel이 정제에서 매우 중요한 단계라 할 수 있다. 효소의 분자량을 알기 위하여 DEAE HPLC로 두 개의 isozyme을 분리하여 전기영동한 결과 I은 환자량 145,000, 85,500 및 69,500이 3개의 단백질을 얻었고 II는 분자량 79,000 및 57,000의 2개의 단백질을 얻었다.

제스프리 그린 키위의 주요 영양성분 및 헥산 분획물의 신경세포 보호효과 (Nutritional Components of Zespri Green Kiwi Fruit (Actinidia delicosa) and Neuronal Cell Protective Effects of the n-hexane fraction)

  • Zespri green kiwi의 무기질은 K, P, Ca 순으로 많았으며, 아미노산은 glutamic acid, aspartic acid, arginine 순으로 많이 함유되어 있었다. 지방산 함량은 a-linolenic acid, oleic acid 순으로 많이 함유되어 있었으며, 유리당은 fructose 함량이 가장 높았다. Vitamin C와 ${\beta}$-carotene 함량은 각각 29.21 mg/100 g, $1.35{\mu}g/100g$이었다. Total phenolics는 n-hexane 분획물이 11.83 mg GAE/g으로 가장 높았으며, ABTS radical 소거활성 및 지질 과산화 억제활성에서는 농도 의존적으로 in vitro 항산화 활성이 증가하는 경향을 보였다. 또한 oxidative stress에 대한 신경세포 보호효과 역시 농도의존적 경향을 보여주었다. 본 연구 결과를 종합해볼 때, 생리활성 소재로서의 phenolics를 함유한 kiwi fruit (A.deliciosa)는 in vitro 항산화 활성과 함께 신경세포 보호효과를 기초로 한 고부가 가치 건강지향식품 소재로서의 활용가치가 높다고 판단된다.

Phospholipase Cγ의 생리적 기능과 질병과 연관된 돌연변이 (Physiological Roles of Phospholipase Cγ and Its Mutations in Human Disease)

  • Phospholipase C gamma (PLCγ)는 phosphatidylinositol을 가수분해하여 신호전달 과정에 참여하는 PLC의 주요한 isotype으로 γ-specific array의 특징적인 구조를 바탕으로 receptor tyrosine kinases 및 non-receptor tyrosine kinase 신호를 주로 매개한다. PLCγ1과 PLCγ2의 두 isozyme이 존재하며 다양한 세포에서 발현하여 cell proliferation, migration 및 differentiation 등 여러 세포작용을 조절하고 있다. 최근의 연구들에서 PLCγ 돌연변이가 cancer와 immune disease 및 brain disorder 등에 연관된다는 것이 밝혀지고 있으며 genetic model을 통해 PLCγ의 생리적·병리적 기능이 제시되었다. 본 리뷰에서는 최신의 연구 결과들을 바탕으로 PLCγ의 구조와 활성 조절 기전에 대해 기술하고 나아가 여러 질병의 발병과 진행에서 보고된 PLCγ의 돌연변이와 knockout 마우스를 활용한 연구 결과를 바탕으로 생리적·병리적 관점에서 PLCγ의 역할에 대해 고찰하였다.

한국 토종다래(Actinidia arguta)순의 주요 영양성분 및 in vitro 항산화 활성 (Nutritional Components of Korean Traditional Actinidia (Actinidia arguta) Sprout and in vitro Antioxidant Effect)

  • 토종다래 순의 무기질은 Ca, K, P 순으로 많았으며, 아미노산은 glutamic acid, aspartic acid가 많이 함유되어 있었다. 지방산 함량은 ${\alpha}$-linolenic acid와 linoleic acid 등의 불포화지방산 함유량이 높았으며, 유리당은 sucrose의 함량이 상대적으로 높았다. Vitamin B 그룹에서는 vitamin $B_6$가 17.99mg/100 g으로 가장 높았고, vitamin C와 ${\beta}$-carotene 함량은 각각 42.57mg/100 g, $390.69{\mu}g/100g$이었다. Total phenolics는 40% EtOH 추출물이 198.08 mg GAE/g으로 가장 높았으며, ABTS radical 소거활성 및 DPPH radical 소거 활성에서는 농도 의존적으로 radical 소거 활성을 보였다. 지질 과산화 억제활성(MDA contents)과 세포내 산화적 스트레스 생성 억제효과에서는 지질과산화를 막아주며 신경세포 손상을 일정 수준에서 억제해 주는 것으로 확인하였다. 본 연구 결과를 종합해볼 때, 상대적으로 높은 phenolics를 함유한 토종다래순은 우수한 영양학적인 구성과 in vitro antioxidant activities를 기초로 한 고부가가치 건강지향식품 소재로서의 활용가치가 높다고 판단된다.

  • 혈소판은 1차 및 2차 지혈에서 근본적인 역할을 하는 세포지만 혈소판의 과도한 활성화는 혈전증을 유발할 수 있다. 따라서 혈소판 응집의 적절한 조절은 혈전증 매개 질환을 예방하는 데 중요하다. 최근 곤충소재의 개발이 주목을 받고 있다. 다양한 곤충 자원 중 고영양 기능성 식품원으로는 쌍별귀뚜라미(Gryllus bimaculatus)와 같은 곤충류가 있다. 쌍별귀뚜라미 는 고단백 및 불포화지방산을 함유하고 있으며 2015년 9월 식품의약품안전처로부터 식품원료로 등록되었다. 본 연구에서는 쌍별귀뚜라미 에탄올 추출물(G. bimaculatus extract)이 혈소판 응집, 세포 내 Ca2+ 조절, thromboxane A2 생산 및 glycoprotein IIb/IIIa (integrin αIIb/β3) 활성화를 억제하는지 여부를 확인하고. 1, 4, 5-triphosphate receptor type I, extracellular signal-regulated kinase, cytosolic phospholipase A2, mitogen-activated protein kinases p38, vasodilator-stimulated phosphoprotein, phosphatidylinositol-3 kinase, Akt, glycogen synthase kinase-3α/β 및 SYK 같은 신호 분자를 조절할 수 있는지 여부를 조사했다. 우리는 쌍별귀뚜라미 추출물이 혈소판 관련 혈전증 및 심혈관 질환을 예방할 수 있는 잠재적인 치료 약물로 가치가 있음을 규명하였다.

양파 농축액이 고지혈증 성인 남자의 체성분, 혈장 전해질 및 지질 성분에 미치는 영향 (The Effect of Concentrated Onion Juice in a Body Composition, Serum Electrolytes and Lipids Levels on Hyperlipidemia)

  • It is known as that onion is antioxidation effect, antibiotic effect, blood pressure decreasing effect and reducing serum cholesterol levels. This research about effect that onion concentrate gets blood cholesterol levels and body composition. Subject was 17 adult men of hyperlipidemia. Age distribution of investigation subjects were average 49.4 years old by 40~56 years old, and average height and weight were 167.6cm and 75.5kg each, BMI was 26.9kg/$m^2$, and BMR was 1,460.6$\pm$87.5㎉, and AMC was 25.0$\pm$1.05cm, and BCM was 41.0$\pm$2.79cm. In the meantime, the body muscle was 53.7$\pm$3.7kg, and fat mass was 18.7$\pm$3.8kg, and intracellular fluid was 26.6$\pm$1.8kg, and extracellular fluid was 12.8$\pm$0.9kg. The % body fat was 24.6$\pm$3.8%, and fat distribution was 0.9$\pm$0.0%, and the obesity degree was 125.4$\pm$8.2%. Vegetables, seaweeds, fruits and juices increased by change of dietary life and greasy foods, instants, breads, rices etc. decreased or there was no change, fast foods and eggs were no change. Also, subject previewed that guidance about stress, smoking, drinking and beverage intake need. If compared the nutrient intake amount with before onion concentrate allowance, it was similar level almost without significant. Energy, calcium and riboflavin are lower than the RDA for koreans. After 3 months, the levels of plasma total cholesterol, triglycerides had decreased significantly : 15.0%, 31.2% respectively. And the HDL-cholesterol and LDL-cholesterol levels also showed a marked reduction of 6.8%, 8.7% respectively. Plasma lipid level change by onion concentrate supplement would can know that case of triglyceride more greatly than plasma cholesterol. The pH and Na+ level of plasma were low significant since 8 weeks after, and $K^{+}$ level increase significant. While $Ca^{++}$ level was low significant after 1 month, there was no change since 2 months after, but nC $a^{++}$ level was low significantly. Plasma $Mg^{++}$ level was no change and nM $g^{++}$ level was low significant after intake.e.e.e.e.e.

$M_1$$M_2$ 무스카린성 수용체에서 아미노산 Triplet Repeat의 Site-Mutagenesis가 수용체기능에 미치는 영향 (Effects of Site-Mutagenesis of an Amino Acid Triplet Repeat at $M_1$ and $M_2$ Muscarinic Receptors on Receptor Function)

  • $M_1$$M_2$ 무스카린성 수용체의 두 번째 transmembrane domain의 C-말단에는 leucine(L), tyrosine(Y), threonine(T)로 구성된 3중체(triplet)가 있다. 이 3중체는 $M_2$ 무스카린성 수용체에서는 두 번째 transmembrane domain과 첫 번째 세포외 고리사이의 연접부위에서 LYT-LYT의 반복구조로 존재하며 $M_1$ 무스카린성 수용체에서는 흥미롭게도 LYT-TYL의 역상구조로 존재한다. 본 연구에서는 site-directed mutagenesis방법을 사용하여 이와 같은 특이한 구조적차이가 두 subtype의 수용체의 기능상 차이와 관련한 역할을 가지고 있는지를 확인하고자 하였다. $M_1$ 수용체에서는 LYTTYL서열을 $M_2$ 수용체의 서열에 해당하는 LYTLYT로 mutation시켰으며 $M_2$ 수용체에서는 LYTLYT8서열을 $M_1$ 수용체의 서열에 해당하는 LYTTYL로 mutation시켰다. 이와같은 mutation은 $M_1$$M_2$ 수용체에서 효능제 carbachol의 수용체 결합친화력에 유의한 변화를 주지 않았다. 또한 $M_1$ 수용체에서의 mutation은 cyclic AMP 증가작용에 대한 coupling은 변화시키지 않고 phosphoinositides (PI) hydrolysis 촉진작용과 세포내 $Ca^{2+}$ 농도 상승을 현저히 증가시켰다. 또한 $M_2$ 수용체에서의 mutation은 adenylate cyclase 억제에 대한 coupling은 변화시키지 않고 PI hydrolysis 촉진을 약간 증가 시켰다. 이상의 결과는 $M_1$$M_2$ 수용체에서 LYTTYL/LYTLYT 아미노산 서열의 차이는 두 수용체의 PI hydrolysis에 대한 coupling을 조절하는 역할을 하지만, 두 수용체 사이에서 ligand 결합과 신호전달계의 차이를 구분하는데 중요한 역할을 하지는 않는다.

Porphyromonas endodontalis에 대한 Polyphosphate의 항균기전에 관한 연구 (MECHANISM IN ANTIBACTERIAL ACTIVITY OF POLYPHOSPHATES AGAINST PORPHYROMONAS ENDODONTALIS)

  • Poly-P has been used to prevent decomposition of foods and has been shown to have inhibitory effect on the growth of gram positive bacteria. The purpose of this study was to evaluate the effect of poly-P on the growth of Porphyromonas endodontalis, a gram negative obligate anaerobic rod, endodontopathic bacterium. P. endodontalis ATCC 35406 was in BHI broth containing hemin and vitamin K with or without poly-P. Inhibitory effect of each poly-P which was added at the beginning(lag phase) or during(exponential phase) the culture, MIC(minimum inhibitory concentration) was determined by measuring the optical density of the bacterial cell at 540nm. Viable cell counts were measured to determined whether poly-P has a bactericidal effect. Leakage of intracellular nucleotides from P. endodontalis was determined at 260nm and morphological change of P. endodontalis was observed under the TEM(transmission electron microscope). Binding of 32P-labeled poly-P to P. endodontalis was examined. SDS-polyacrylamide gel electrophoresis and zymography were performed to observe the changes in protein and enzyme profiles of P. endodontalis, respectively. The results from this study were as follows : 1. The minimal inhibitory concentration(MIC) of poly-P to P. endodontalis appeared to be 0.04~0.05%. 2. Poly-P added to the P. endodontalis culture during the exponential phase of P. endodontalis was as much effective as poly-P added at the begining of the culture, suggesting that the antibacterial effect of poly-P is not much dependent on the initial inoculum size of P. endodontalis. 3. Poly-P are bactericidal to P. endodontalis, demonstrating the decrease of the viable cell counts. 4. Intracellular nucleotide release from the P. endodontalis, was not increased in the presence of poly-P and was not reversed by the addition of divalent cations like $Ca^{2+}$ and $Mg^{2-}$. 5. Under the TEM, it was observed that fine electro-dense materials were prominent in the poly-P grown P. endodontalis, appearing locally in the cell, and the materials were more abundant and more dispersed in the cell as the incubation time with poly-P increased. In addition, highly electron dense granules accumulated in many poly-P grown cells, most of which were atypical in their shape. 6. Binding of 32P-labeled poly-P to P. endodontalis appeared to be 32.8 and 45.5 and 53.4% at 30 minutes, 1 hours and 2 hours, respectively. 7. In the presence of poly-P. the synthesis of proteins with apparent molecular masses of 25, 27, 35, 45 was lost or drastically decreased whereas expression of a protein with an apparent molecular mass of 75 was elevated. 8. Proteolytic activity of P. endodontalis was decreased by poly-P. The overall results suggest that use of poly-P may affect the growth of P. endodontalis, and the anti-bacterial activity of poly-P seems largely bactericidal. Changes in shape, protein expression, and proteolytic activity of P. endodontalis by poly-P may be directly and indirectly attributed to the antibacterial effect of poly-P. Further studies will be needed to confirm the effect of poly-P.

Therapeutic Effects of S-Petasin on Disease Models of Asthma and Peritonitis

  • To explore the anti-allergic and anti-inflammatory effects of extracts of Petasites genus, we studied the effects of s-petasin, a major sesquiterpene from Petasites formosanus (a butterbur species) on asthma and peritonitis models. In an ovalbumin-induced mouse asthma model, s-petasin significantly inhibited the accumulations of eosinophils, macrophages, and lymphocytes in bronchoalveolar fluids. S-petasin inhibited the antigen-induced degranulation of ${\beta}$-hexosamidase but did not inhibit intracellular $Ca^{2+}$ increase in RBL-2H3 mast cells. S-petasin inhibited the LPS induction of iNOS at the RNA and protein levels in mouse peritoneal macrophages. Furthermore, s-petasin inhibited the production of NO (the product of iNOS) in a concentration-dependent manner in the macrophages. Furthermore, in an LPS-induced mouse model of peritonitis, s-petasin significantly inhibited the accumulation of polymorpho nuclear and mononuclear leukocytes in peritoneal cavity. This study shows that s-petasin in Petasites genus has therapeutic effects on allergic and inflammatory diseases, such as, asthma and peritonitis through degranulation inhibition in mast cells, suppression of iNOS induction and production of NO in macrophages, and suppression of inflammatory cell accumulation.

Regulation of L-type Calcium Channel Current by Somatostatin in Guinea-Pig Gastric Myocytes

  • To study the direct effect of somatostatin (SS) on calcium channel current ($I_{Ba}$) in guinea-pig gastric myocytes, $I_{Ba}$ was recorded by using whole-cell patch clamp technique in single smooth muscle cells. Nicardipine ($1{\mu}M$), a L-type $Ca^{2+}$ channel blocker, inhibited $I_{Ba}$ by $98{\pm}1.9$% (n=5), however $I_{Ba}$ was decreased in a reversible manner by application of SS. The peak $I_{Ba}$ at 0 mV were decreased to $95{\pm}1.5$, $92{\pm}1.9$, $82{\pm}4.0$, $66{\pm}5.8$, $10{\pm}2.9$% at $10^{-10}$, $10^{-9}$, $10^{-8}$, $10^{-7}$, $10^{-5}$ M of SS, respectively (n=3∼6; $mean{\pm}SEM$). The steady-state activation and inactivation curves of $I_{Ba}$ as a function of membrane potentials were well fitted by a Boltzmann equation. Voltage of half-activation ($V_{0.5}$) was $-12{\pm}0.5$ mV in control and $-11{\pm}1.9$ mV in SS treated groups (respectively, n=5). The same values of half-inactivation were $-35{\pm}1.4$ mV and $-35{\pm}1.9$ mV (respectively, n=5). There was no significant difference in activation and inactivation kinetics of $I_{Ba}$ by SS. Inhibitory effect of SS on $I_{Ba}$ was significantly reduced by either dialysis of intracellular solution with $GDP_{\beta}S$, a non-hydrolysable G protein inhibitor, or pretreatment with pertussis toxin (PTX). SS also decreased contraction of guinea-pig gastric antral smooth muscle. In conclusion, SS decreases voltage-dependent L-type calcium channel current ($VDCC_L$) via PTXsensitive signaling pathways in guinea-pig antral circular myocytes.
