쑥, 양파 및 원지가 흰쥐의 장내환경에 미치는 영향 (Effects of Mugwort, Onion and Polygalae Radix on the Intestinal Environment of Rats)

  • 원지, 양파 및 쑥을 추출물, 즙 또는 분말로 하여 기준식이에 5 및 10% 첨가하여 흰쥐에게 14일간 먹여 대장내용물에서 주요균의 변화와 ${\beta}-glucosidase$${\beta}-glucuronidase$의 효소활성 및 인돌과 volatile basic nitrogen의 생성량을 조사하였다. 실험쥐들의 사료섭취량과 체중증가량은 쑥의 물추출물 5% 섭취구를 비롯하여 시료 첨가구들에서 대체로 높았으나, 원지물추출물 5%와 쑥의 에탄올추출물 10% 섭취구에서는 체중이 감소하였다. 쑥의 물 또는 에탄올추출물 첨가구에서 유익균인 bifidobacteria가 대조구에 비해 높았고, clostridia와 E. coli는 낮았다(p<0.05). 그외 양파즙 10% 첨가구에서도 유익균의 증가와 유해균의 감소와 같은 유리한 균총변화를 보였다. 한편 쑥분말 5% 첨가구에서 대장내용물중 ${\beta}-glucosidase$${\beta}-glucuronidase$ 활성은 대조구와 비교해 통계적인 유의성이 없으나, 인돌함랑과 pH는 대조구에 비해 낮았다(p<0.05). 그러나 원지물추출물과 양파를 10% 첨가구에서는 ${\beta}-glucosidase$ 활성이, 쑥추출물들의 첨가구에서는 ${\beta}-glucuronidase$ 활성이 높았다(p<0.05). 인돌함량은 단백질 함량이 높은 원지물추출물과 쑥에탄올추출물 첨가구에서 높았다(p<0.05). 한편 휘발성 염기태 질소화합물의 함량은 처리구들에서 유의적인 차이가 없었다.

  • This study aimed to determine the effects of dietary microalgae (Tetradesmus sp. (TO)) on intestinal immunity and microbiota of pre-starter broilers. One hundred and twenty 1-day-old birds (Ross 308) were allocated to two dietary treatment groups with six blocks in a randomized complete block design. The two experimental diets consisted of a corn-soybean meal-based basal diet and a diet with 0.5% TO powder instead of cornstarch in the basal diet. After feeding the experimental diets for ten days, all birds' body weight and feed intake were measured, and representative eight birds were selected from each treatment group. Small intestinal lamina propria cells were isolated using flow cytometry to examine the frequency of immune cells. Cecal feces were harvested for 16s rRNA gut microbiota analysis and fecal IgA levels. Here, we found that 0.5% TO supplementation increased CD3+CD4+ T cells in the small intestine, but decreased CD3+CD8+ T cells in the small intestine. Gut microbial analysis showed that TO supplementation significantly increased the alpha diversity of the gut microbiome. Taxonomic analysis showed that TO treatment increased the abundance of Firmicutes and decreased that of Bacteroidetes at the phylum level. The distribution of Enterobacteriaceae containing many harmful bacteria at the family level, was lower in the TO group. In the LEfSe analysis, the TO group had a significantly enriched abundance of Agathobaculum at the genus level. Overall, results show that Tetradesmus sp. supplementation influences intestinal T-cell immunity and induces the expansion of beneficial gut microbes in pre-starter broiler chickens.

Butyric acid and prospects for creation of new medicines based on its derivatives: a literature review

  • The widespread use of antimicrobials causes antibiotic resistance in bacteria. The use of butyric acid and its derivatives is an alternative tactic. This review summarizes the literature on the role of butyric acid in the body and provides further prospects for the clinical use of its derivatives and delivery methods to the animal body. Thus far, there is evidence confirming the vital role of butyric acid in the body and the effectiveness of its derivatives when used as animal medicines and growth stimulants. Butyric acid salts stimulate immunomodulatory activity by reducing microbial colonization of the intestine and suppressing inflammation. Extraintestinal effects occur against the background of hemoglobinopathy, hypercholesterolemia, insulin resistance, and cerebral ischemia. Butyric acid derivatives inhibit histone deacetylase. Aberrant histone deacetylase activity is associated with the development of certain types of cancer in humans. Feed additives containing butyric acid salts or tributyrin are used widely in animal husbandry. They improve the functional status of the intestine and accelerate animal growth and development. On the other hand, high concentrations of butyric acid stimulate the apoptosis of epithelial cells and disrupt the intestinal barrier function. This review highlights the biological activity and the mechanism of action of butyric acid, its salts, and esters, revealing their role in the treatment of various animal and human diseases. This paper also discussed the possibility of using butyric acid and its derivatives as surface modifiers of enterosorbents to obtain new drugs with bifunctional action.

INFLUENCE OF DRIED Sacillus subtilis AND LACTOBACILLI CULTURES ON INTESTINAL MICROFLORA AND PERFORMANCE IN BROILERS

  • Two hundred 10-day-lid, male Arbor Acres broiler chicks divided randomly into 4 groups of 50 chicks each were used. Different feeding treatment was carried out for each group. Chicks in treatment 1 were fed a basal diet(Starter feed)(control); treatment 2, a basal diet + 0.1% B. subtilis culture; treatment 3, a basal diet + 0.2% lactobacilli culture in the feed; and treatment 4, a basal diet + 5 g lactobacilli in the drinking water. The viable bacterial counts for each treatment were approximately $10^9cells/kg$ feed. The weight gain in chickens given feeds incorporated with B. subtilis and lactobacilli was significantly(p < 0.05) higher than those of the control. With regard to feed efficiency, there was a definite tendency towards a higher feed : gain lower(p < 0.05) feed : gain ratio. A significantly(p < 0.05) larger population of Lactobacillus was found in the small intestine of chickens fed with feed incorporated with B. subtilis at 21 and 28 days and with lactobacilli at 14, 21 and 28 days. Populations of intestinal E. coli in broilers given feed added with B. subtilis were not significantly(p < 0.05) different from those of the control, but in chickens fed lactobacilli-added feed, their populations wee significantly lower(p < 0.05) at 14 and 21 days. No significant differences were found among the treatments and the control in the occurrence of Salmonella and Campylobacter during the whole experimental period.

The effect of palm kernel meal supplementation in the diet on the growth performance and meat quality of swine, and on the level of odorous compounds and bacterial communities in swine manure

  • In the present study, we aimed to investigate the effect of dietary supplementation of palm kernel meal (PKM), as a fermentable carbohydrate source, on the growth performance, meat quality, concentration of odorous compound, and changes in bacterial community in swine manure. Swine (average initial body weight of $51.36{\pm}1.02kg$) were fed diet which included three levels of PKM (0, 2 and 4%), and their manure samples were collected from the slurry pit. Growth performance and meat quality were not affected by PKM treatments (p > 0.05). Levels of phenols and indoles were decreased in the 2 and 4% PKM treatments compared to 0% PKM (control; p < 0.05). Especially, compared to the control, the 2% PKM group showed decreased levels of phenols by 35% and indoles by 34%. Among the dominant bacterial genera, the main change in relative abundance occurred in those belonging to the Firmicutes phylum in PKM treatments. Terrisporobacter and Clostridium were decreased in the PKM groups compared to the control. However, the relative abundance of Intestinibacter, AM406061_g, Coprococcus_g2, Phascolarcotobacterium, EF401875_g, Lactobacillus, and Streptococcus were increased in the PKM group compared to control. Taken together, administration of PKM had a beneficial effect on reducing production of odorous compounds in swine manure, possibly by modulating the communities of predominantly carbohydrate-utilizing bacteria in the large intestine of swine.

Virulence factors, antimicrobial resistance patterns, and genetic characteristics of hydrogen sulfide-producing Escherichia coli isolated from swine

  • Escherichia (E.) coli is commensal bacteria found in the intestine; however, some pathogenic strains cause diseases in animals and humans. Although E. coli does not typically produce hydrogen sulfide ($H_2S$), $H_2S$-producing strains of E. coli have been identified worldwide. The relationship between virulence and $H_2S$ production has not yet been determined. Therefore, characteristics of $H_2S$-producing isolates obtained from swine feces were evaluated including antibiotic resistance patterns, virulence gene expression, and genetic relatedness. Rates of antibiotic resistance of the $H_2S$-producing E. coli varied according to antibiotic. Only the EAST1 gene was detected as a virulence gene in five $H_2S$-producing E. coli strains. Genes conferring $H_2S$ production were not transmissible although the sseA gene encoding 3-mercaptopyruvate sulfurtransferase was detected in all $H_2S$-producing E. coli strains. Sequences of the sseA gene motif CGSVTA around Cys238 were also identical in all $H_2S$- producing E. coli strains. Diverse genetic relatedness among the isolates was observed by pulsed-field gel electrophoresis analysis. These results suggested that $H_2S$-producing E. coli strains were not derived from a specific clone and $H_2S$ production in E. coli is not associated with virulence genes.

  • Fibrinopurulent peritonitis and abdominal abscesses associated with Streptococcus (S.) equi subsp. zooepidemicus is reported in a 1-year-old female thoroughbred horse. The horse died after showing 5-day history of the cold, severe abdominal distention, abdominal breathing, hyperthermia, anorexia, and loss of intestinal sound. At necropsy, several liters of turbid yellowish green fluid were seen in the abdominal cavity. Yellowish creamy and fibrinous or bloody materials were presented on the serosa of various abdominal organs that were intensively adhered with each other. Spleen and mesenteric lymph node were remarkably enlarged. Affected lobes of the lung showed severe congestion, hemorrhage and doughy consistency. Histologically, the lung showed hemorrhagic pneumonia with diffuse congestion and edema. Severe diffuse fibrinopurulent peritonitis with Gram-positive bacterial cocci and adjacent fibrosis were showed in the serosa of various abdominal organs such as liver, spleen, stomach, and intestine. And multifocal abscess pouches were presented in the granulation tissue of abdominal viscera. S. equi subsp. zooepidemicus was isolated from the peritoneal swab, abdominal organs, and lung. Hematogenous dissemination of bacteria from hemorrhagic pneumonia is proposed as the route of infection in this case.

음식물류폐기물의 돼지 발효사료화를 위한 종모배양액 제조 (The Manufacture of Inoculum for Fermented Pig Feed Production from Food Wastes)

  • 본 실험은 음식물류 폐기물을 돼지발효사료로 만들기 위해 공기주입과 보조용양원으로 돼지혈분의 이용에 대해 알아보았다. 발효용 종모균주로는 돼지의 내장에서 분리한 유산균인 Lactobacillus brevis와 딸기에서 분리한 효모인 Saccharomyces cerevisiae가 이용되었다. 배지원료로는 당밀과 유청이 효모엑스와 함께 사용되었다. 실험결과 공기주입은 생균수를 높여주었고 사멸기를 저지하는 효과를 보였다. 보조 영양원으로 돈혈분이 효모엑스를 대치할 경우에도 생균수의 변화는 거의 없었다. 종모발효액은 상온에서 저장할 경우 1주일간 Lactobacillus brevis와 Saccharomyces cerevisiae생균수가 모두 8 log cfu/mL대를 유지했다. 종모발효액이 음식물류 폐기물과 혼합될 경우 이틀 뒤부터는 잡균이 증가되기 시작했으나 5일 이내에는 E.coli가 검출되지 않았다.

Infection and Immune Response in the Nematode Caenorhabditis elegans Elicited by the Phytopathogen Xanthomonas

  • Xanthomonas oryzae pv. oryzae (Xoo) strains are plant pathogenic bacteria that can cause serious blight of rice, and their virulence towards plant host is complex, making it difficult to be elucidated. Caenorhabditis elegans has been used as a powerful model organism to simplify the host and pathogen system. However, whether the C. elegans is feasible for studying plant pathogens such as Xoo has not been explored. In the present work, we report that Xoo strains PXO99 and JXOIII reduce the lifespan of worms not through acute toxicity, but in an infectious manner; pathogens proliferate and persist in the intestinal lumen to cause marked anterior intestine distension. In addition, Xoo triggers (i) the p38 MAPK signal pathway to upregulate its downstream C17H12.8 expression, and (ii) the DAF-2/DAF-16 pathway to upregulate its downstream gene expressions of mtl-1 and sod-3 under the condition of daf-2 mutation. Our findings suggest that C. elegans can be used as a model to evaluate the virulence of Xoo phytopathogens to host.

Effect of Feeding Aspergillus Oryzae Culture on Fecal Microflora, Egg Qualities, and Nutrient Metabolizabilities in Laying Hens

  • This experiment examined the effects of feeding Aspergillus oryzae (AO) culture to laying hens, on fecal microbial populations, fecal pH and moisture content, egg quality, and metabolizabilities of several nutrients. Sixteen commercial 38-wk-old laying hens were randomly allotted to four diets: control; with 0.15% locally produced AO culture; with 0.3% locally produced AO culture, and; or with 0.3% imported AO. Each treatment consisted of four replicates (cages) containing one bird per cage according to a completely randomized design. After 4 wk, AO were recovered in the feces of birds fed the AO diets, indicating that AO might pass through the fore-gut alive and become active in the hind gut. The number of Lactobacillus spp. in feces was higher in all treated groups than that of the control, indicating that AO would provide a beneficial environment for the Lactobacillus spp. to proliferate in the intestine. The number of fecal E. coli was significantly reduced by the addition of AO. A similar trend was also found for aerobic bacteria. Although not significant, fecal moisture contents tended to be reduced by the addition of AO. Fecal pH was not significantly different among the treatments. The addition of AO did not affect the various economic traits of eggs. Metabolizabilities of gross energy and dry matter measured during the 5th wk were increased by the AO supplementation. It appears that AO culture alone could be used as a probiotic supplement for layers.
