Quantitative Analysis of Central Nervous System Tissues (CNST) in Beef By-Products in Retail Market

  • This study was carried out to investigate the applicability of the detection of central nervous system tissues (CNST) in beef by-products in retail market. Beef by-products including large intestine, brain, spinal cord, liver, lung, spleen and heart were purchased and tested for the presence of CNST using an ELISA method. The ELISA test was evaluated and showed a high correlation coefficient by a standard curve (R value = 0.999). Based on the analytical instruction, the positive indication of the CNST contamination of brain and spinal cord was detected above 0.1% but large intestine, liver, lung, spleen, and heart was negative. Result suggests that the ELISA method is applicable to a real meat system and may provide a method to ensure confidence for consumer against bovine spongiform encephalopathy (BSE).

$^{86}Rb$ Distribution in the Lung of the Rabbit with Pneumothorax (가토 기흉에서 본 $^{86}Rb$의 분포)

  • $^{86}Rb$ uptake of some organs and tissues, ego both lungs, both renal cortices. small intestine, liver and skeletal muscle were studied in the control and the rabbit subjected to pneumothorax. $^{86}Rb$ in the form of chloride mixed with physiological saline was intravenously. injected. The doses were $100{\mu}c$ for a rabbit. The rabbits were sacrificed at intervals of 10, 20, 40, and 60 seconds after the injection of $^{86}Rb$, by the injection of saturated KCI solution. After sacrification, the organ and tissue sample were quickly removed. $^{86}Rb$ uptake in gm of the organs and tissues were measured. On the basis of uptake value, administered doses and body weight, % dose/gm tissues per 200 gm body weight was calculated. Followings were the results; 1. Pneumothorax resulted in a marked elevation in $^{86}Rb$ uptake value of collapsed lung and returned to normal level lately. 2. Contralateral lung of pnemothorax also showed marked elevation in $^{86}Rb$ uptake value and recovered to normal level. 3. Initial $^{86}Rb$ uptake value of liver, small intestine of the rabbit with pneumothorax showed some elevation as compared to control, but that of late stage were similar with control. 4. Local blood flow determination by means of $^{86}Rb$ uptake were inadequate in the collapsed lung of pneumothorax. 5. It was suggested that the mechanism for the initial elevation of $^{86}Rb$ uptake value in each organs and tissue were different from each other.

Metabolic Activation of Ester- and Amide-Type Drugs by Carboxylesterases

  • Carboxylesterase is widely distributed in the tissues of vertebrates, insects, plants and mycobacteria. Among various tissues of animals and humans, the highest esterase activity with various substrates is found in the liver. Kidney has moderate carboxylesterase activity in the proximal tubules. Considerable esterase activity is also found in the small intestine epithet elial cells and serum of mammals. Besides these tissues, carboxylesterase has been found in the lung, testis, adipose tissue, nasal mucosa and even in the central nervous system. Hepatic microsomal carboxylesterase catalyzes the hydrolysis of a wide variety of endogenous and exogenous compounds such as carboxylester, thioester and aromatic amide. Since carboxylesterases are important for metabolic activation of prodrugs and detoxification of xenobiotics, differences in substrate specificity and immunological properties of this enzyme are important in connection with choosing a suitable laboratory animal for the evaluation of biotransformation and toxicity of drugs. On the other hand, liver, kidney, intestine and serum were found to contain multiple forms of carboxylesterases in animal species and humans. In fact, we have purified more than fifteen isoforms of carboxylesterases from microsomes of liver, kidney and intestinal mucosa of nine animal species and humans. and characteristics of these isoforms were compared each other in terms of their physical and immunochemical properties. On the other hand, we have reported that hepatic microsomal carboxylesterases are induced by many exogenous compounds such as phenobarbital, polycyclic aromatic hydrocarbons, Aroclor 1254, aminopyrine and clofibrate. Later, we showed that some isoforms of hepatic carboxylesterase were induced by glucocorticoids such as dexamethasone and 16 ${\alpha}$-carbonitrile, but other isoforms were rather inhibited by these compounds. These findings indicate that involvement of carboxylesterases in the metabolism and toxicity of drugs should be explained by the isoforms involved. Since 1991, we have carried out detailed research investigating the types of carboxylesterases involved in the metabolic activation of CPT-11, a derivative of camptothecin, to the active metabolite, SN-38. The results obtained strongly suggest that some isoforms of carboxylesterase of liver microsomes and intestinal mucosal membrane are exclusively involved in CPT-11 metabolism. In this symposium, the properties of carboxylesterase isoforms purified from liver, kidney and intestine of animal species and humans are outlined. In addition, metabolism of CPT-11, a novel antitumor agent, by carboxylesterases in relation to the effectiveness will also be discussed.

Biochemical Changes in the Tissue of Mice Irradiated with LINAC (선형가속기를 이용한 방사선조사에서 생쥐조직의 생화학적 변화)

  • In this study, a linear accelerator (LINAC) through 3 Gy of radiation per body irradiated mice of the small intestine and the liver to produce in order to protect the cells after radiation exposure that caspase (caspase 3 &caspase 9) and NO (nitric oxide), and looked like to know cytokine of IL-6 and TNF-${\alpha}$, the result is as follows. First, caspase 3 & caspase 9 showed a noticeable increase in the radiation group than in the control group both small intestine and liver tissues (P <0.001). Second, NO are both intestine and liver tissue showed a marked increase in the radiation group than in the control group (P <0.001). Third, one of Cytokine IL-6 and TNF-${\alpha}$ showed a significant increase in the irradiated group than the control group both small intestine and liver tissues (P <0.001).

Feline panleukopenia virus infection in imported cats

  • The cases of feline panleukopenia virus (FPLV) infection were diagnosed in three imported cats. All cats died within one week after mild emaciation, depression and anorexia. One cat showed yellowish watery diarrhea. At necropsy, all cats had segmental hemorrhage on the serosa and mucosa of the small intestine. Histopathologically, severe diffuse necro-hemorrhagic enteritis was observed in small intestine especially in jejunum and ileum. The crypts of Lieberkuhn were dilated and contained necrotic epithelia. Severely damaged epithelia of crypts were transformed into bizarre shapes. Multifocal lympholysis and lymphoid depletion were found in Peyer's patches and other lymphoid tissues. Direct fluorescent antibody (FA) test revealed the characteristic FPLV antigen in the cytoplasms of crypt epithelial cells. Based on the clinical signs, characteristic pathologic findings and FA test, these cases were diagnosed as FPLV infection. In our best knowledge, this study is the first case report for FPLV infection in imported cats in Korea.

  • Titania nanomaterials are widely used as cosmetics and dyes, however the impacts on human health are uncertain, We investigated the biodistribution of inhaled titania nanoparticles in rats, Methods Eight weeks-old SD rats were intubated and inhaled with 3 mg titania nanoparticles, twice a week, for 2 weeks, After inhalation, the rats were sacrificed and tissues or heart, lung. intestine, brain, and liver were obtained, We investigated the tissues with optical microscope (OM), transmission electron microscope (EM), scanning EM, And to analyze titania concentration of each tissue, we lysed the tissues with radioimmunoprecipitation assay (RlPA) lysis buffer or acid. Results Granulation tissues in lung were confirmed on the optical microscope, however the other organs had no abnormalities in OM images, In EM images, the rats which inhaled titania nanoparticles showed calcium deposition at heart, brain, and intestine, Titania concentration in lung was increased on the inhaled rat sacrificed I month after last exposure. Conclusion Inhaled titania nanoparticles is thought to be deposited and make inflammatory reaction in lung, and the deposition was not efficiently cleared over a month. However inhaled titania nanoparticles may rarely pass through the alveolus-blood barrier and distribute to other organs of the bod.

  • This study investigated the distribution of scuticociliates in the intestine, spleen, kidney, testis, brain, pericondrial bone and muscle layer of the tiger puffer, Takifugu rubripes, infected with scuticociliates. Scuticociliates were infiltrated in the connective tissues of the outer layer of the intestine, spleen, kidney, testis, brain, pericondrial bone and muscle layer. In brain, membranous tissue and optic lobe cortex separation were accompanied by the infection of scuticociliates. Other internal tissues and organs did not show any lesions expect for heavy deposition of hemosiderin in spleen.

  • An endoscope is usually inserted into the human body for the inspection of the gullet, stomach, and large intestine (colon) and this may cause discomfort to patients and damage to tissues during diagnostic or therapeutic procedures. This situation necessitates a self-propelling endoscope. There are many kinds of mechanism to move in a rigid pipe. However, these methods are difficult to apply directly to the endoscope. The main reason is that human intestine cannot be considered as a uniform, straight, and rigid pipe. This paper proposes a flexible loop wheel mechanism, which is adaptable to the human intestine. This mechanism is designed and fabricated by a simple modeling, and tested by an experiment. Finally, the actuator is inserted into the pig colon.

Effect of Dietary Taurine on Free Amino Acid Concentrations in Blood and Various Tissues of Cats (식이 내의 타우린 함량이 고양이의 혈액 및 주유조직의 유리 아미노산 농도에 미치는 영향)

  • Taurine concentrations in blood and various tissues were measured in cats fed normal trurine diet(0.15% taurine) and trurine-free diet(0% taurine) for 5 weeks and 12 weeks, respectively. Cumulative body weight gain in young cats fed normal taurine diet and taurine-free diet for the initial 5 weeks were 463$\pm$43g and 383$\pm$53g respectively. Taurine concentration was one to two orders magnitude higher in most tissues than in plasma, among which the liver taurine-free diet for 12 weeks resulted in 98% reduction in plasma taurine concentration, 93% reduction in whole blood tarurine concentration, and 40~90% reduction in tissue taurine concentrations compared to the values for normal taurine diet group. Among the tissues tested, taurine depletion was most profound in heart and kidney, while liver and intestine were relatively resistant to taurine depletion. Whole blood taruined concentration appeared to be a better index for the body taurine status of cats as in more closely reflects the rate of taurine depletion in tissues. Taurine depletion in cats significantly increased the concentrations of essential and non-essential free amino acids in liver, while free amino acid concentrations in other tissues were not affected by taurine depletion.

  • Taurine (2-ethaneaminosulfonic acid, $^+{NH}_3{CH_2}{CH_2}{SO_3^{-}}$) is endogenous amino acid with functions as modulator of osmoregulation, antioxidation, detoxification, transmembrane calcium transport, and a free radical scavenger in mammalian tissues. Taurine transporter(TAUT) contains 12 transmembrane helices, which are typical of the $Na^+$- and $Cl^-$-dependent transporter gene family, and has been cloned recently from several species and tissues. To analyze the expression of TAUT mRNA, one step RT-PCR was performed from human and mouse cultured cell lines and from various mouse tissues. The primers were designed to encode highly conserved amino acid sequences at the second transmembrane domain and at the fourth and fifth intracellular domains. RT-PCR analysis showed both of the human intestine HT-29 and mouse macrophage RAW264.7 cell lines expressed mRNA of TAUT. To define the expression patterns of the TAUT mRNA in the murine organs, RT-PCR was performed to detect cDNA representing TAUT mRNA from seven different mouse tissues. The TAUT was detected in all of the mouse tissues analyzed such as heart, lung, thymus, kidney, liver, spleen and brain. A large amount of transcript was fecund from heart, liver, spleen, kidney, and brain, while lung contained a very small amount of transcript.
