인공기질 부착조류에 의한 대천의 수질평가 (Water Quality Assessment Using the Periphyton on the Artificial Substrates in Dae Stream, Busan)

  • 부산시 대천의 생물학적 수질판정을 하기 위하여 4개 정점에 인공부착기질을 설치하고 부착조류의 출현 특성을 조사하였다. 현존량의 상위 10%인 우점종은 정점 1과 2에서는 Cymbella ventricosa, Aulacoseira, granulata, Gomphonema olivaceum, Synedra ulna, Fragilaria intermedia로 나타났고, 정점 3과 4에서는 상기 4종과 더불어 Fragilaria intermedia 대신 Nitzschia palea로 나타났는데 모두 도심하천에서 주로 우점하여 출현하는 지표종이었다. 조사정점별로는 제1우점종의 현존량은 정점 1과 2에서는 Aulacoseira granulata가 각각 2,830 cells $ml^{-1}$, 2,030 cells $ml^{-1}$였고 정점 3에서는 Synedra ulna가 10,450 cells $ml^{-1}$, 정점 4에서는 Nitzschia palea가 23,470 cells $ml^{-1}$로 조사되어 오염원이 없는 지역에서는 우점종의 현존량이 거의 비슷하게 조사되었으나 오염원의 유입이 있는 지역에서는 우점종별로 현존량의 차이가 크게 나타났다. 또한 오염이 심하게 되었다고 생각되었던 지역에서는 강부수성 수역에서 출현하는 오염종인 Nitzschia palea의 현존량이 다른 종들과는 달리 두드러지게 많이 출현했음을 알 수 있었다. 오탁지수에 의한 수질은 정점 1에서는 1.49로 빈부수성 수역이었고 정점 2와 3은 각각 1.75, 2.40으로 $\beta$-중부수성 수역, 정점 4는 2.86으로 $\alpha$-중부수성 수역으로 조사되었는데 전체적으로는 2.12로 $\beta$-중부수성이었으나 부산 시민들이 많이 찾는 도시공원으로서의 기능과 역할을 생각해 볼 때 향후 도시민들의 출입이 잦아져서 오염을 가속화시킬 우려가 커서 오염방지에 많은 노력을 기울여야 할 것이다.

타액내 구강질환 원인 균의 세균배양법, SYBR green qPCR법, MRT-PCR법 간의 정량분석 (Quantitative analysis of oral disease-causing bacteria in saliva among bacterial culture, SYBRgreen qPCR and MRT-PCR method)

  • Objectives: The purpose of this study was to compare SYBR Green qPCR, TaqMan, and bacterial selective medium cultures for accurate quantitative analysis of oral microorganisms. Methods: The SYBR Green method is widely used to analyze the total amount of oral microorganisms in oral saliva. However, in this study, MTR-PCR method based on TaqMan method was performed using newly developed primers and probes. In addition, it was designed to confirm the detection agreement of bacteria among bacteria detection method. Results: As a result of MRT-PCR and SYBR Green qPCR analysis, more than 40 times (0.9-362.9 times) bacterium was detected by MRT-PCR. In addition, more bacteria were detected in saliva in the order of MRT-PCR, SYBR Green qPCR, and bacterium culture, and the results of MRB-PCR and SYBR Green qPCR showed the highest agreement. The agreement between the three methods for detecting P. intermedia was similar between 71.4 and 88.6%, but the agreement between MRT-PCR and SYBR Green qPCR was 80% for S. mutans. Among them, the number of total bacteria, P. intermedia and S. mutans bacteria in saliva was higher than that of SYBR Green qPCR method, and bacterium culture method by MRT-PCR method. P. intermedia and S. mutans in saliva were detected by MRT-PCR and MRT-PCR in 88.6% of cases, followed by the SYBR Green qPCR method (80.0%). Conclusions: The SYBR Green qPCR method is the same molecular biology method, but it can not analyze the germs at the same time. Bacterial culturing takes a lot of time if there is no selective culture medium. Therefore, the MRT-PCR method using newly developed primers and probes is considered to be the best method.

Morphological and Molecular Characterization of Apharyngostrigea pipientis Faust, 1918 (Trematoda: Diplostomidea) from Ardeacinerea jouyi, Egretta intermedia and Nycticorax nycticorax in Korea

  • An intestinal strigeiid trematode, Apharyngostrigea pipientis, was detected from Ardea cinerea jouyi, Egretta intermedia and Nycticorax nycticorax between July, 2006 and June, 2020 in Daejeon metropolitan city and Kangwon-do, Republic of Korea. It was observed with a light and scanning electron microscope (SEM), and molecularly analyzed with 18S rRNA genes. The infection rate of the strigeiid trematode was 25.9% (7/27). The body is cylindrical in whole mount, greatly elongated in outline and shows a strong dorsal curvature. The overall measurements are: length 4.1-4.8 mm; fore-body 1.00-1.20 mm by 0.8-0.9 mm; hind-body 3.2-3.6 mm by 0.43-0.46 mm. The body is covered by a relatively thick cuticle, devoid of spines. The oral sucker leads into a short esophagus that is devoid of a muscular pharynx. The holdfast organ is enormously developed. The vitelline follicles distributed dorsal wall of fore-body and hind-body. This strigeiid trematode especially differs from other species of genus Apharyngostrigea in morphology. Although these A. pipientis are several morphological distinctions from Apharyngostrigea cornu, the partial sequence of 18S rRNA showed the sequence homology of A. pipientis (99.9%) and A. cornu (99.8%), and this result revealed their conspecific relationship between A. cornu and A. pipientis. This is the first report of A. pipientis in the Republic of Korea.

  • 기존의 많은 동기화 모델 중 EFSM(Extended Finite State Machine)기반 동기화 모델을 수정 보완한 MFSM(Modified Finite State Machine)을 제안한다. MFSM은 미디어간 동기화를 위한 Intermedia Synchronization 역할을 Sync Master와 Sync Slave로 나누어 기존 논문의 단점을 보완한다. 3GPP2 환경에서 멀티미디어 데이터 송수신시에 미디어 소스간에 동기화를 위해서 MRF(Media Resource Function)가 데이터를 중계하는 MFSM기반의 동기화 모델을 제안한다.

  • Black-pigmented bacteria anaerobes have been implicated in the endodontic infections. This group of microorganisms includes Porphyromonas endodontalis, Porphyromonas gingivalis, prevotella intermedia, and Prevotella nigrescens. The organisms display a wide variety of virulence factors that may be pertinent to acute endodontic infections.(omitted)

웅천천(熊川川) 수계산(水系産) 납자루의 개체발생(個體發生) (Development of Acheilognathus lanceolatus from Ungchon river, Korea)

  • 웅천천(熊川川) 수계산(水系産) 납자루의 개체발생(個體發生)에 대(對)하여 $22{\pm}^{\circ}C$의 사육(飼育) 수온하(水溫下)에 있어서 경시적(經時的)인 관찰(觀察)을 실시(實施)하고 동시(同時)에 자어(仔魚)의 표피상돌기(表皮上突起)의 검토(檢討)도 실시(實施)하여 일본산(日本産) 납자루와의 비교(比較)도 행(行)하였다. 양국산(兩國産) 납자루는 완숙란(完熟卵)의 형태(形態)가 한국산(韓國産)이 일본산(日本産) 보다 현저(顯著)하게 가느다란 통종형(統鍾形)인 점(點)으로 구별(區別)되었으나 란발생(卵發生)과 자어(仔魚)의 발육형태(發育形態) 그리고 자어(仔魚)의 표피상돌기(表皮上突起)의 형태(形態) 등(等)에서는 차이(差異)를 볼 수 없었으며 초기발생(初期發生)에 있어서 생리적(生理的)인 격리(隔離)가 존재(存在)하지 않음이 밝혀졌다. 본(本) 종(種)과 근록종(近綠種)이라고 생각되는 칼납자루는 일본산(日本産)과 한국산(韓國産) 사이에서 개체발생상(個體發生上)의 차이(差異)나 생이적(生理的)인 격리(隔離)가 있음이 밝혀져 있는데 양국산(兩國産) 납자루 사이에서는 완숙란(完熟卵)의 형태(形態)만 차이(差異)가 있었을 뿐이다. 완숙란(完熟卵)의 형태(形態)에는 아종(亞種) 간(間)의 차이(差異)나 지리적(地理的)인 변이(變異)가 있음이 알려져 있으므로 양국산(兩國産) 납자루 사이에는 종(種) 수준(水準)의 분화(分化)라기 보다는 아종적(亞種的) 분화(分化) 단계(段階)에 있다고 생각되었다.

Relationship of Oral Bacterial Load Over One Year of Smoking Cessation

  • Background: Smoking exerts an adverse effect on the periodontal tissue by reorganizing the ecosystem of oral microorganisms and is considered to be an important factor in the development of periodontal disease. Although cross-sectional studies on smokers and non-smokers have been attempted to investigate the microbial differences in periodontal oral cavity, only few studies have been conducted to investigate the changes in oral microorganisms during smoking cessation. The purpose of this study was to investigate the changes of bacteria in saliva and gingival crevicular fluid (GCF) over a period of one year among 11 smokers trying to quit smoking. Methods: Eleven smokers trying to quit smoking visited the clinic at baseline, two weeks, two months, four months, six months, and 12 months to give saliva and GCF samples. The amounts of 16S rRNA, Porphyromonas gingivalis, Treponema denticola, Prevotella intermedia, Fusobacterium nucleatum subsp. nucleatum, Streptococcus mutans, and Streptococcus sobrinus in saliva and GCF were quantified using real-time polymerase chain reaction TaqMan probe assay. The results were analyzed by nonparametric statistical analysis using Friedman test and Spearman correlation coefficient. Results: After cessation of smoking, the amounts of 16S rRNA corresponding to P. gingivalis, F. nucleatum, P. intermedia, and T. denticola in saliva decreased and then again increased significantly. The amount of F. nucleatum 16S rRNA in GCF decreased significantly after smoking cessation. Positive correlations were observed between 16S rRNA and F. nucleatum and between F. nucleatum and T. denticola in saliva and GCF. Conclusion: Even if the number of subjects in this study was small, we suggest that smoking cessation may reduce the total bacterial amount and F. nucleatum in GCF. However, the results regarding changes in the microbial ecosystem due to smoking or smoking cessation were inconsistent. Therefore, further in-depth studies need to be carried out.

The Herbal Composition Gangjihwan from Ephedra intermedia, Lithospermum erythrorhizon and Rheum palmatum Ameliorates Hepatic Inflammation and Fibrosis in Obese C57BL/6J Mice and HepG2 Cells

  • It was demonstrated that Gangjihwan (DF), which is the herbal composition composed of Ephedra intermedia, Lithospermum erythrorhizon, and Rheum palmatum, inhibits obesity and hepatic steatosis in high fat diet (HFD)-fed obese mice. The aim of this study was to determine the effects of DF on visceral obesity, hepatic inflammation and fibrosis and the mechanism of actions involved in this process using in vivo and in vitro approaches. DF was extracted with water (DF-FW), 30% grain alcohol (DF-GA30), and 70% grain alcohol (DF-GA70). Administration of DF to HFD-fed control mice decreased visceral tissue mass and visceral adipocyte size without adverse effects. Visceral fat mass was decreased by DF-GA30 and DF-GA70, and visceral adipocyte size by all three DF extracts compared with obese control mice. Histological analysis revealed that three kinds of DF extracts reduced toluidine blue-stained mast cells and collagen accumulation in the liver, the extents of which were most eminent in DF-GA70-treated mice. DF-GA70 decreased the mRNA levels of the inflammation ($TNF{\alpha}$ and VCAM-1), fibrosis (${\alpha}-SMA$), and apoptosis (caspase 3) genes, but increasing the anti-apoptosis gene (Bcl-2) mRNA levels in the liver of obese control mice. Consistent with the in vivo data, GA-70 also altered the expression of inflammation genes ($TNF{\alpha}$ and MCP-1) in HepG2 cells. These results indicate that DF not only inhibits visceral obesity, but also ameliorates visceral obesity-induced hepatic inflammation and fibrosis and that this process may be mediated by regulating the hepatic expression of inflammatory and fibrogenic genes.

성인성 치주염환자에서 분리한 Porphyromonas gingivalis와 Prevotella intennedia의 항생제 내성에 관한 연구 (Antibiotic susceptibility of Porphyromonas gingivalis and Prevotella intennedia from the Patients with Adult Periodontitis)

  • The purpose of this study was to an in estimate the antibiotic susceptibility of P. gingivalis and P. interrnedia isolate from the subgingival plaque to adult periodontitis. Six P. gingivalis and five P. intermedia bacterial strains were tested for their susceptibility to 10 antimicrobial agents under disc diffusion method and broth dilution method. Ten patients with deep pocket(6mm) were selected for this study. They had not taken antibiotics for 6 months and no history of dental treatment for 6 months before this study. The result were as follow : 1. For antibiotic disc diffusion method, six P. gingivalis and five P. interrnedia were tested with 10 antimicrobial agents which comprised penicillin, gentamycin, clindamycin, lincomycin, ampicillin, erythromycin, tetracycline, amikacin, chloramphenicol, vancomycin. The sensitive antibiotics were tetracycline, penicillin, ampicillin, vancomycin, chloramphenicol and resisitent antibiotics were lincomycin. The other antimicrobial agents were less active. 2. From the study of determination on the minimal inhibitory concentration(MIC) by broth dilution method, the MIC of tetracycline to P. gingivalis and P. intermedia were $0.5-1.0{\mu}g/ml$, $0.5{\mu}g/ml$, that of ampicillin were $1-8{\mu}g/ml$, that of clindamycin were $1-32{\mu]g/ml$, $8-16{\mu}g/ml$, that of lincomycin were $16-32{\mu}g/ml$, $2-32{\mu}g/ml$. These data suggest that tetracycline and ampicillin may be valuable drug in the elemination of P. gingivalis and P. interrnedia from the patients with adult periodontitis.
