Balb/c mouse spleen cells in vitro sensitized against ICR spleen cells were cultured in conditioned media(CM). The CM was produced by ICR spleen cells stimulated with Concanavalin-A(Con-A), and sensitized lymphoid cells were grown in CM. ICR mouse spleen cells were appeared to be a good generator of IL-2. Optimal growth was seen in growth medium containing 20% fetal calf serum. and 25% CM. When cultures were initiated at 1, 5, $10{\times}10^4\;cells/ml$, the cells were increased in numbers by about 20, 13, 5-fold, respectively, every 9 days. Such growth pattern was sustained for about 4-6 weeks and thereafter the cell growth was diminished gradually. Direct immunofluorescence indicated that 93% of the lymphoid cells grown in CM(for 10 days) expressed Thyl surface antigen. And the cells grown in CM were cytotoxic to the sesitizing ICR mouse spleen cells though cytotoxicity level was not high. According to these results, the cells grown in CM were considered to be cytotoxic T lymphocytes. The lymphoid cells grown for 20 days were nearly unresponsive to Con-A and therefore dependent only IL-2 to be used for IL-2 assay.
Aim: Historically, mineral compound herbal medicines have long been used in treatments of immune-related diseases in Korea, China and other Asian countries. In this study, we investigated the anti-inflammatory effect of egg white combined with chalcanthite (IS4) on lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. Methods: RAW 264.7 cells cultured with LPS and various concentrations of IS4 were analyzed to determine the production of pro-inflammatory cytokines and mediators by using enzyme-linked immune sorbent assays (ELISAs). Results: IS4 concentration inhibited the production of interleukin-1beta (IL-$1{\beta}$), interleukin-6 (IL-6), and granulocyte-macrophage colony-stimulating factor (GM-CSF) induced by LPS. IS4 at high concentrations (25 and 50 ${\mu}g/ml$) inhibited, in concentration-dependent manner, the expression of tumor necrosis factor-alpha (TNF-${\alpha}$) stimulated by LPS. Conclusion: IS4 has shown an anti-inflammatory effect in RAW 264.7 cells.
The purpose of this study is to investigate effects of White Ginseng-Ejung-tang water extract (EJ) on production of various cytokines such as interleukin (IL)-2, IL-5, IL-6, IL-10, IL-12p70, macrophage inflammatory protein (MIP)-2, vascular endothelial growth factor (VEGF), keratinocyte-derived chemokine(KC), tumor necrosis factor (TNF)-${\alpha}$, and granulocyte macrophage colony-stimulating factor (GM-CSF) in RAW 264.7 mouse macrophages stimulated by lipopolysaccharide (LPS). Levels of cytokines were measured by High-throughput multiplex bead array cytokine assay based on xMAP (multi-analyte profiling beads) technology. EJ significantly decreased levels of IL-2, IL-12p70, IL-5, MIP-2 for 24 h incubation at the concentrations of 25, 50, and 100 ${\mu}g/mL$ in LPS-induced RAW 264.7 (P < 0.05). EJ significantly decreased levels of IL-6 at the concentrations of 50 and 100 ${\mu}g/mL$ (P < 0.05). EJ significantly decreased levels of IL-10 and VEGF at the concentrations of 25 and 100 ${\mu}g/mL$ (P < 0.05). EJ significantly decreased levels of KC at the concentrations of 100 ${\mu}g/mL$ (P < 0.05). EJ did not show any significant effect on TNF-${\alpha}$ and GM-CSF production. These results suggest that EJ has anti-inflammtory property related with its inhibition of IL-2, IL-5, IL-6, IL-10, IL-12p70, MIP-2, VEGF, and KC production in LPS-induced macrophages.

Background: Immune responses appear to be affected by anesthetics and analgesics. We investigated the effects of caudal tramadol on the postoperative immune response and pain management in pediatric patients. Methods: Sixty ASA-I pediatric patients aged 3-10 years undergoing lower abdominal surgery. Patients were randomly assigned either to a caudal bupivacaine (0.25%) group (group B), or a group that received caudal tramadol (1 mg/kg) added to the bupivacaine (0.25%) (group T). Both were diluted in a 0.9% NaCl solution to a total volume of 1ml/ kg. The systemic immune response was measured by collecting blood samples preoperatively, at the end of anesthesia, and at 24 and 72 hours postoperatively, and studied for interleukin IL-6, C-reactive proteins (CRP) cortisol levels, and leucocytes with its differential count. Postoperative pain was assessed along with sedation scales. Results: Postoperative production of IL-6 was significantly higher in group B at the end of anesthesia, than at the $24^{th}$ hour, and at the $72^{nd}$ hour in group B and group T, respectively. The immune response showed leukocytosis with increased percentages of neutrophil and monocytes, and a decreased lymphocyte response rate within both groups with no significant differences between the groups. Cortisol and CRP were significantly higher in group B. Conclusions: Adding tramadol to a caudal bupivacaine block can attenuate the pro-inflammatory cytokine response, Cortisol, and CRP in children undergoing lower abdominal surgery.
Immunoregulatory cytokines have an influence on hepatitis C virus (HCV) infection outcome. This study aimed to determine whether single nucleotide polymorphisms (SNP) in IFN- ${\gamma}$ and IL-10 genes are associated with susceptibility and/or are markers of prognosis regarding chronic hepatitis C outcomes. IFN ${\gamma}$ (+874T/A) and IL-10 (-1082G/A) genotypes were determined in 75 HCV genotype 4 patients with different disease severities (chronic hepatitis, n=25, liver cirrhosis and hepatocellular carcinoma (HCC) on top of liver cirrhosis, n=50) and 25 healthy participants using allele-specific polymerase chain reaction. No statistical differences in allele or genotype distributions of IFN ${\gamma}$ and IL-10 genes were detected between patients and controls or between patientgroups. No significant difference in the frequency of IL-10 SNP at position -1082 or IFN-${\gamma}$ at position +874T/A was found between chronic HCV genotype 4 and with progression of disease severity in liver cirrhosis or HCC. In conclusion; interferon-${\gamma}$ and interleukin-10 gene polymorphisms are not predictors of disease progression in patients with chronic hepatitis C (Genotype-4).
면역 작용에 중요한 역할을 하는 림프구의 일종인 T세포가 성장 및 증식하는 기관인 흉선의 흉선세포증식과 생존력에 미치는 인삼 saponin의 영향과, 흉선 세포의 증식에 관여하는 Interleukin2(IL2)에 미치는 인삼saponin의 영향과,IL2의 생성에 영향을 미치는 Adenosin Deaminase(ADA)에 대한 인삼 saponin의 영향 등을 알아보기 위하여 생쥐의 흉선세포를 사용하여 다음과 같은 결과를 얻었다. 여러 농도의 인삼 saponin에서 배양한 전체 흉선세포수와 살아있는 흉선세포수의 변화는 $10^{-5}$%농도에서 대조군보다 증가하였으나 $10^{-1}$%농도군에서는 감소되었다. 여러 농도의 IL2에 인삼 saponin$10^{-5}$%농도를 첨가하여 배양한 전체 흉선세포수는 1.5%,3%및 6%농도군에서 9시간 또는 12시간배양시 증가하였으며,6% 실험군은 24시간과 48시간 배양시에도 유의성있게 증가하였다. 그러나 살아있는 흉선세포수는 대조군과 별차이가 없었다. 마지막으로 흉선세포의 ADA활성도에 미치는 인삼 saponin의 영향은 $10^{-5}$%농도에서 25%의 유의성있는 활성 증가가 일어났다.

Korean red ginseng (Panax ginseng Meyer) is a well-known traditional medicine, with numerous biological functions in the body. Portulaca oleracea (P. ole) belongs to the Portulacaceae family and has bioactive potential as a traditional medicine. This study aimed to determine the anti-inflammatory effects of Korean red ginseng extract (RGE) and P. ole extract on lipopolysaccharide (LPS)-treated RAW 264.7 cells. The combination of RGE (50 ㎍/mL) and P. ole (6.25 ㎍/mL) extracts significantly suppressed LPS-induced nitric oxide synthesis. The expression of proinflammatory mediators, including inducible nitric oxide synthase (iNOS) and cyclooxygenase-2 (COX-2), and proinflammatory cytokines, including interleukin-1β, interleukin-6, and tumor necrosis factor-α, were markedly decreased by the combined treatment with RGE (50 ㎍/mL) and P. ole (6.25 ㎍/mL). Moreover, iNOS and COX-2 protein expression levels were also significantly reduced in the combined treatment compared to the LPS-stimulated group. In addition, the nuclear translocation of phosphorylated nuclear factor kappa-B was suppressed by the treatment with RGE and P. ole. Moreover, the mitogen-activated protein kinase pathway was also partially inhibited by the combination treatment with RGE and P. ole. Our results demonstrate that the treatment mixture with RGE and P. ole could be used as functional food and therapeutic herbal medicine in various inflammatory diseases.
Objective: The present research was conducted to evaluate the interactive effect of dietary concentration of calcium (Ca) and 25-hydroxy vitamin D3 (25OHD3) on the performance, blood composition and digestibility of laying hens. Methods: A total of 540 Hy-line brown laying hens aged 61 to 70 weeks were randomly allotted in a 3×3 factorial arrangement, consisting of three levels of 25OHD3 (0, 25, and 50 ㎍/kg) and three levels of Ca (3.5%, 4.0%, and 4.5%). All diets had basal concentration of 3,000 IU/kg of vitamin D3 including the 2,800 kcal/kg of metabolic energy and 16% of crude protein. Results: The results showed that interactive effect (p<0.05) between Ca and 25OHD3 was such that dietary 25OHD3 linearly increased interleukin-6 at all levels of Ca inclusion. Interaction (p<0.05) occurred with the highest parathyroid hormone in laying hens that received dietary concentration of Ca (3.5%) with 25OHD3 (50 ㎍/kg), and Ca (4.0%) with 25OHD3 (50 ㎍/kg). Egg production and egg weight significantly (p<0.05) increased in the 4.5% Ca group compared to the 3.5% to 4.0% Ca groups. Egg shell thickness and tibia bone length also increased (p<0.05) in groups fed a high-Ca diet (4.0% to 4.5%). Phosphorus digestibility significantly (p<0.05) increased along with dietary Ca level. Among the tested 25OHD3 groups, higher (p<0.05) egg production and tibia thickness were present in hens fed 50 ㎍/kg of 25OHD3. Furthermore, Ca digestibility serum Ca and 25OHD3 were significantly increased in group offered 50 ㎍/kg of 25OHD3. Conclusion: The results gathered in this study indicate that dietary concentrations of 4.0% to 4.5% Ca and 50 ㎍/kg 25OHD3 improve the performance of hens from 61 to 70 weeks of age.
1. 연구배경 최근 놀라운 과학기술의 발전에 힘입어 난치병이라고 생각되어 오던 질환들이 하나씩 정복되어 가고 있으나 평균수명의 연장, 생활수준 향상으로 인한 고지방식, 스트레스 및 과학기술발전의 부작용 등을 이유로 새로운 질환들이 나타나고 있는 실정이다. 종양도 이런 대표적인 질환중 하나로 최근 경제적, 시간적으로 많은 투자를 해왔으나 아직까지 그 발생원인이나 성장기전 등이 자세히 밝혀지지 않았다. 이에 본 연구는 효후(哮吼)를 치료하는 청상보하환(淸上補下丸)을 이용하여 흑색종의 폐전이 억제능과 이에 의한 면역증강에 관하여 연구하고자 하였다. 2. 연구결과 B-16 세포를 이용한 C57BL/6계 마우스에 있어서의 폐전이 억제능을 측정한 결과 청상보하환(淸上補下丸) 투여군이 대조군에 비해 검액처리 7일후에는 52.70%, 14일후에는 19.43%의 유의성있는 억제율을 보였으며, 청상보하환(淸上補下丸)의 B-16 세포에 대한 직접적인 항암작용을 알아 보기 위한 시험관내 세포독성 실험에서는 청상보하환(淸上補下丸)의 용량이 $5{\mu}g/well$, $2.5{\mu}g/well$, $1.25{\mu}g/well$, and $0.625{\mu}g/well$인 경우 유의성있는 효과를 얻을수 있었으며 생존율에 의한 $IC_{50}$은 $2.17{\mu}g/well$이었다. NK-activity를 측정한 결과 청상보하환(淸上補下丸) 투여군이 작동세포와 표적세포의 비율이 100:1, 50:1의 경우에 있어서 $72.63{\pm}5.17%$, $26.94{\pm}2.43%$를 보여 대조군에 비하여 각각 31.12%, 43.68% 유의성있는 증가를 보였다. IL-2의 생산능을 측정한 결과 청상보하환(淸上補下丸) 투여군은 $1392.00{\pm}22.31pg/ml$을 보여 대조군에 비해 4.04%의 유의성 있는 증가를 보였다.
이 연구의 목적은 초기 및 중등도 치주염 환자에서 전동칫솔을 사용할 경우 임상 지수의 향상 정도와 치주원인균의 정량적 감소 효과를 12주의 연구 기간 동안 평가하는 것이다. $25{\sim}55$세의 환자 80명을 대상으로 12주 동안 진행하였으며, 치태지수 0.5 이상, 치은지수 0.5 이상을 나타내는 대상에서 일반 칫솔 혹은 전동칫솔 ($Sonicare^{(R)}$ Elite, Philips Oral Healthcare Inc., Snoqualmie, Washington, USA) 사용 군을 임의로 선정하였다. 하루 2회, 매 회 2분 간 사용하고, 각 군의 칫솔 사용을 교육하였다. 임상지수는 치태지수 (PI; Silness & $L{\ddot{o}}e$), 치은지수 (GI; $L{\ddot{o}}e$ & Silness), 탐침 후 출혈 부위 (%), 치주낭 깊이 부착소실을 초진 1, 4, 12주에 측정하였다. Interleukin-1 (IL-1), MMP-8과 치은연하치태샘플에서 채취한 4 종류의 치주원인균 (Actinomyces visco년, Porphyromonas gingivalis, Streptococcus sanguis, Tannerella forsythensis)에 대한 16S rRNA test는 초진, 1주, 12주에 측정하였다. 측정 결과 전동칫솔과 일반 칫솔 모두 임상지수의 유의한 감소가 나타났으며, 치은지수는 일반칫솔에 비해 전동칫솔에서 감소효과가 통계적으로 더 우수하게 나타났다 (p<0.001). 탐침 후 출혈의 감소는 전동칫솔에서 76.73%, 일반칫솔에서 44.57% 정도로 전동칫솔 군이 더 우수하게 나타났다. 치주낭 깊이 감소는 초진에 비해 전동칫솔 군에서 18.55%, 일반칫솔 군에서 14.81% 정도로 나타났으며, 초진과 비교하였을 때 부착수준의 향상 정도는 전동칫솔 25.24%, 일반 칫솔 16.94% 정도로 두 군 모두 통계적으로 유의하게 개선되었다 (p < 0.001). 두 군 모두 IL-1 beta, MMP-8 농도의 감소가 있었으며, 치주원인균 중 A.viscosus, P.gingivalis, T.forsythensis 역시 두 군 모두에서 초진에 비해 12주에 유의한 감소를 나타내었으나, S.sanguis는 전동칫솔 군에서만 12주에 유의한 감소가 있었다. 이상의 결과에서 12주 간의 연구 기간 동안 초기 및 중등도 치주염 환자에서 소니케어 전동칫솔의 사용은 임상지수 및 치주 원인균 감소에 통계적으로 유의한 개선 효과를 나타내었다.

제 19 조 (관할 법원)
서비스 이용으로 발생한 분쟁에 대해 소송이 제기되는 경우 민사 소송법상의 관할 법원에 제기합니다.

1. (시행일) 이 약관은 2016년 9월 5일부터 적용되며, 종전 약관은 본 약관으로 대체되며, 개정된 약관의 적용일 이전 가입자도 개정된 약관의 적용을 받습니다.
