Topical Application of Herbal-mixed Extract on back skin suppresses inflammation in Ovalbumin-induced Atopic Dermatitis (아토피 피부염 동물모델에서 피부에 도포한 한약 복합추출물의 염증 억제 효과)

  • Objective : This study is conducted to evaluate the efficiency of ethanol extracts of 4 mixed herbs(CP001 or CP002) on mouse model of atopic dermatitis induced by ovalbumin. Methods : Female BALB/c mice were internally sensitized with ovalbumin($20{\mu}g$) plus aluminum hydroxide hydrate(4mg) once per week. After 3 weeks, they were dermally challenged with patches containing ovalbumin ($100{\mu}g$) plus aluminum hydroxide hydrate(20mg) every other day for 3 weeks. After induction of atopic dermatitis, mice back skin were gently rubbed with CP001 or CP002(200mg/$m{\ell}$, $100{\mu}{\ell}$) for two weeks(every 2 days). Results : In CP001 or CP002 treated group, there was a remarkable reduction in infiltration of eosinophils on the skin areas and diminution of mast cells and total T cells in blood samples as compared with control group. Cutaneous expressions of interleukin-13, 17 were also decreased by CP001 or CP002. Moreover, blood immunoglobulin E level was decreased by drug administration while there was no decrease in OVA sensitization group. Conclusion : In summary, our result shows that herbal extracts(CP001 and CP002) could be potential candidates for the treatment of chronic atopic dermatitis.

Hepatitis C Virus Associations with Non Hodgkin's Lymphoma: Insights on Inflammation/Angiogenesis and CD Markers

  • We aimed to investigate any association between hepatitis C virus (HCV) infection and non-Hodgkin's lymphoma (NHL) in the view of cytokines that control inflammation/angiogenesis and their correlation with certain CD markers. NHL patients with or without HCV infection were studied. CD5, CD30, CD3, CD20 and CD45 were immunohistochemically evaluated. Plasma levels of vascular endothelial and platelet derived growth factors (VEGF, and PDGF), tumor necrosis factor (TNF-${\alpha}$), transforming growth factor (TGF-${\beta}$), interleukin-6 (IL-6), IL-8, IL-4, IL-12 and interferon gamma (IFN-${\gamma}$) were detected by enzyme-linked immunosorbent assay (ELISA). HCV+ve NHL patients showed a significant reduction in VEGF, PDGF, IFN-${\gamma}$, CD5 and CD45 and a significant increase in IL-12 and IL-8. In conclusion, there was a significant change in cytokine secretion and expression of CD markers in HCV+ve NHL patients. Based on our results, HCV infection in NHL patients requires more in-depth investigations to explore any role in lymphoma progression.

Effects of Torilis japonica Extract on DNCB-induced Contact Dermatitis in BALB/c Mice (사상자 추출물이 DNCB로 유도된 BALB/c 마우스의 접촉성 피부염에 미치는 효과)

  • 목적: 본 연구에서는 TJ 열수추출물이 아토피 피부염에 미치는 효과를 확인하고자 하였다. 방법: 2, 4-dinitrochlorobenzen (DNCB)으로 BALB/c 마우스에 1차 감작 후 3주간 2차 감작을 시행하여 아토피 유사 피부병변을 나타내는 접촉성 피부염을 유발한 뒤 동결건조 처리된 TJ 분말을 (PBS/EtOH/Cremophor=6:1:3)에 용해시켜 쥐의 등 피부에 10, 50 mg/ml 농도로 3주간 도포하였다. 결과: TJ 열수추출물은 아토피 피부 병변의 염증세포 침윤을 억제하였고 (10, 50 mg/ml ) 표피와 진피 두께를 회복시켰으며 (10, 50 mg/ml ), 혈청에서 히스타민 방출을 억제하였다 (00 mg/ml ). 또한 Th2 세포와 관련된 cytokine인 interleukin (IL)-4, IL-5, IL-13과 thymic stromal lymphopoietin (TSLP)의 mRNA 발현 양을 감소시켰고 (10, 50 mg/ml ), Th2 chemokine인 thymus- and activation-regulated chemokine (TARC or CCL17)의 mRNA의 발현 양을 감소시켰다 (10, 50 mg/ml ). 결론: TJ 열수추출물을 BALB/c 마우스에 외용하였을 때 항아토피 효과가 있음을 확인하였으며 따라서 TJ가 아토피 피부염 치료의 외용제로 활용될 수 있을 것으로 사료된다.

Subtle inflammation: a possible mechanism of future cardiovascular risk in obese children

  • Purpose: The risk of cardiovascular disease (CVD) has been shown to be associated with systemic inflammation in obese adults with metabolic syndrome (MetS). The aims of this study were to evaluate the prevalence of MetS and its relation to inflammatory markers in obese Thai children. Methods: A cross-sectional study was conducted. Children with history of endogenous obesity, chronic diseases, drug ingestion, and any acute illness within 2 weeks prior to enrollment were excluded. Their fasting blood glucose (FBG) levels, oral glucose tolerance tests, insulin, lipid profiles, and selected inflammatory markers, including interleukin-6, tumor necrosis factor-alpha, and high-sensitivity C-reactive protein (hs-CRP) levels, were tested. Results: In this study, 58 obese Thai children (female, 20; male, 38) with a mean body mass index z score of $5.1{\pm}2.2$ were enrolled. The prevalence of MetS and prediabetes was 31% and 17.2%, respectively. None of the children had diabetes. FBG levels, 2-hour glucose levels, and lipid profiles were not statistically different between those with and without MetS. However, obese children with MetS had higher insulin levels and homeostasis model assessment of insulin resistance values. Elevated hs-CRP levels were found in 69% of the cases, although it was not statistically different between the 2 groups. Conclusion: We described a substantial prevalence of MetS in Thai obese children. Regardless of MetS status, two-thirds of the obese children had elevated hs-CRP level, indicating subtle ongoing inflammatory process. This chronic inflammation feasibly predisposes them to CVD in the future, even in children without MetS.

  • KCSYJT medicines controlled $CD4^+/IFN-\gamma$, and $CD4^+/CD25^+/foxp3^+$ revelation that an experiment that motive allergy immune reponse because an in vitro experiment stimulates T cells of a NC/Nga mouse same time by anti-CD40/rmIL-4, and interleukin-$1{\beta}$, IL-6, TNF-$\alpha$, and TGF-$\beta$ mRNA outturn that bear in T and B cells decreased remarkably by KCSYJT medicines. Intracellular staining of splenocytes anti-CD40/rmIL-4 plus rmIL-4 stimulated as described in a, assessed after 24 h, KCSYJT exerts a mainly immunosuppressive effect that acts at least partially through suppression of the transcription factor GATA3 expression in $CD4^+$ T cells. We found that skin lesions, which were clinically and histologically very similar to human AD, mite antigen-induced dermatitis on the face, neck, ears and dorsal skin of inbred NC/Nga mice. Result that Th1 cell and Th2 cell observe to be shifted by cytokine expression with GATA3 regulation by KCSYJT medicines could know that KCSYJT medicines can use usefully in allergy autoimmnune diease.

Effects of Caraganae Sinicae Radix on the osteoporosis in ovariectomized rats (금작근(金雀根)이 난소적출로 유발된 흰쥐의 골다공증에 미치는 영향)

  • Objective : This study was carried out to investigate the effects of the hormones and cytokines associated with bone metabolism in ovariectomized rats. Method : Twenty-four Female-Sprague-Dawley rats were divided into sham operated(normal) group, ovariectomized(control)group, ovariectomized and treated with extract Caraganae Sinicae Radix(treated) group. Each group was investigated the changes of body weight at 3,5,7weeks after treatment, and femur weight, femur/body weight, thickness of compact bone of body of femur, area of cancellous bone of distal epiphysis of femur, serum estrogen, serum calcitonin, serum parathormone, serum Tumor Necrosis $Factor-{\alpha}$, serum $lnterleukin-1{\beta}$ at 7weeks after treatment. The results were summerized as follows; 1. The treated group showed significant change in body weight compared with the control group at 5,7weeks after treatment. 2. The treated group revealed significant increases in femur/body weight compared with the control group. 3. The treated group showed a little thicker compact bone of body of femur than the control group. 4. The area of cancellous bone of distal epiphysis of femur in treated group was increased significantly compared with control group. 5. The level of serum estrogen showed no change compared with control group. 6. The level of serum calcitonin showed no change compared with control group. 7. The level of serum parathormone was decreased in treated group significantly compared with control group. 8. The level of serum $TNF-{\alpha}$ was decreased in treated group significantly compared with control group. 9. The level of serum $interleukin-1{\beta}$ showed no change compared with control group. The results indicate that Caraganae Sinicae Radix inhibits bones resorption in ovariectomized rats by it's lowering effects on serum parathormone and $TNF-{\alpha}$.

Enhancement of Immunomodulatory and Anticancer Activity of Fucoidan by Nano Encapsulation

  • The aim of the present study was to prepare nanosample of fucoidan using lecithin as encapsulated material and to investigate the anticancer and immunomodulatory activity of nanoparticle in vitro. The nanoparticles have been characterized by dynamic light scattering (DLS) and transmission electron microscopy (TEM). Confocal microscopy confirmed the internalization of the fucoidan conjugates into the immune cells. The uptake of nanoparticles was confirmed with confocal microscopy demonstrating their localization in the cells. The anticancer activity was increased over 5-10% in different cancer cells of fucoidan nanoparticle as compare with fucoidan. The human B and T cells growth and the secretion of interleukin-6 and tumor necrosis factor-a from B cell were also improved by fucoidan nanoparticle because of the rapid absorption of nanoparticle into the cells as compare to fucoidan. At 0.6 mg/mL concentrations, the fucoidan nanoparticle showed better activity than 1.0 mg/mL concentration in T cell growth because the cells reached their saturation capacity. When the fucoidan was encapsulated in lecithin, its anticancer as well as its immunomodulatory activity proved to be superior from that of itself in pure form.

Anti-inflammatory Effects of Gagamsoyosan (가감조요산(加減造遙散)의 항염(抗炎) 및 면역반응(免疫反應)에 대한 연구(硏究))

  • Purpose: It is the purpose of this study to investigate the anti-inflammatory effects of water extract from Gagamsoyosan(GGSYS) on the peritoneal macrophage. Methods: To evaluate anti-inflammatory effects of GGSYS. inflammatory cytokines were measured in lipopolysaccharide(LPS) -stimulated macrophages. Furthermore. the western blot has been done to look into the molecular mechanism. Results: GGSYS did not have any cytotoxicity in the peritoneal macrophages and suppressed production of LPS-induced NO. tumor necrosis factor-alpha (TNF-$\alpha$). interleukin(IL)-1$\beta$. IL-6. IL-12. GGSYS inhibited the activation of extracelluar signal-regulated kinase (ERK1/2). c-Jun N-terminal kinase(JNK), p38 kinase and the degradation of inhibitory kappa B-alpha($I_{k}B-\alpha$) in the LPS-stimulated peritoneal macro phages. GGSYS inhibited LPS-induced endotoxin shock and the production of TNF-$\alpha$. IL-l$\beta$. IL-6 in serum from LPS-stimulated mice. Conclusion: These results suggest that GGSYS may have the anti-inflammatory effect which can inhibit the production of NO and inflammatory cytokines. GGSYS is expected to protect against inflammatory diseases.

In Vitro Immune-Enhancing Activity of Ovotransferrin from Egg White via MAPK Signaling Pathways in RAW 264.7 Macrophages

  • Ovotransferrin (OTF) is a well-known protein of the transferrin family with strong iron chelating activity, resulting in its antimicrobial activity. Furthermore, OTF is known to have antioxidant, anticancer, and antihypertensive activities. However, there have been few studies about the immune-enhancing activity of OTF. In current study, we investigated the immune-enhancing activity of OTF using the murine macrophage cells in vitro. The effect of OTF on production of pro-inflammatory mediators and cytokines were determined using Griess assay and quantitative real-time PCR. Using Neutral Red uptake assay, we confirmed the effect of OTF on phagocytic activity of macrophages. Ovotransferrin significantly increased the production of nitric oxide (NO) and secretion of inducible nitric oxide synthase (iNOS) mRNA with no cytotoxic activity. Ovotransferrin (2 mg/mL) stimulated NO production up to $31.9{\pm}3.5{\mu}M$. Ovotransferrin significantly increased the mRNA expression levels of pro-inflammatory cytokines which are tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$), Interleukin-$1{\beta}$ (IL-$1{\beta}$), and IL-6: OTF (2 mg/mL) treatment increased the secretion of mRNA for TNF-${\alpha}$, IL-$1{\beta}$, and IL-6 by 22.20-, 37.91-, and 6.17-fold of the negative control, respectively. The phagocytic activity of macrophages was also increased by OTF treatment significantly compared with negative control. Also, OTF treatment increased phosphorylation level of MAPK signaling pathways. These results indicated that OTF has immune-enhancing activity by activating RAW 264.7 macrophages via MAPK pathways.

Unique epithelial expression of S100A calcium binding protein A7A in the endometrium at conceptus implantation in pigs

  • Objective: S100A7A, a member of the S100 protein family, is involved in various biological processes, including innate immunity, antimicrobial function, and epithelial tumorigenesis. However, the expression and function of S100A7A in the endometrium during the estrous cycle and pregnancy are not well understood in pigs. Therefore, this study determined the expression and regulation of S100A7A at the maternal-conceptus interface in pigs. Methods: We obtained endometrial tissues from pigs throughout the estrous cycle and pregnancy, conceptus tissues during early pregnancy, and chorioallantoic tissues during midto late pregnancy and analyzed the expression of S100A7A in these tissues. We also determined the effects of steroid hormones, estradiol-$17{\beta}$ ($E_2$) and progesterone, and interleukin-$1{\beta}$ (IL1B) on S100A7A expression in endometrial tissues. Results: We found that S100A7A was expressed in the endometrium during the estrous cycle and pregnancy in a pregnancy status- and stage-dependent manner and was localized to endometrial luminal epithelial (LE) and superficial glandular epithelial cells with strong intensity in LE cells on day 12 of pregnancy. Early stage conceptuses and chorioallantoic tissues from day 30 to term pregnancy also expressed S100A7A. The expression of S100A7A was increased by $E_2$ and IL1B in endometrial tissues. Conclusion: S100A7A was expressed at the maternal-conceptus interface at the initiation of implantation in response to conceptus-derived estrogen and IL1B and could be a unique endometrial epithelial marker for conceptus implantation in pigs. These findings provide an important insight into the understanding of conceptus-endometrial interactions for the successful establishment of pregnancy in pigs.
