Short hairpin RNA targeting of fibroblast activation protein inhibits tumor growth and improves the tumor microenvironment in a mouse model

  • Fibroblast activation protein (FAP) is a specific serine protease expressed in tumor stroma proven to be a stimulatory factor in the progression of some cancers. The purpose of this study was to investigate the effects of FAP knockdown on tumor growth and the tumor microenvironment. Mice bearing 4T1 subcutaneous tumors were treated with liposome-shRNA complexes targeting FAP. Tumor volumes and weights were monitored, and FAP, collagen, microvessel density (MVD), and apoptosis were measured. Our studies showed that shRNA targeting of FAP in murine breast cancer reduces FAP expression, inhibits tumor growth, promotes collagen accumulation (38%), and suppresses angiogenesis (71.7%), as well as promoting apoptosis (by threefold). We suggest that FAP plays a role in tumor growth and in altering the tumor microenvironment. Targeting FAP may therefore represent a supplementary therapy for breast cancer.

  • The error probability of the DS-CDMA / DPSK cellular mobile communication system with CCI canceller and MRC diversity reception technique has been analyzed in the cellular radio channel which is characterized by AWGN, Multi-User Interference(MUI) and m-distribution fading. System capacity i. e., number of user per cell has been derived and the evaluated results are shown in figures as a function of PN code sequence length, fading index, BER, number of diversity branches and $E_b/N_o$, Here, the voice activity factor is assumed to be 3/8, the number of sectors in a cell 3 and Multi-User Iner- ference is modeled as Gaussian process.

Development of Designation Criteria for Ecological Protected Areas and its Application Methodology (생태계 보호지역의 합리적 지정을 위한 평가방법의 개발과 적용방안)

  • Attempts to develope designation criteria for ecological protected areas were made for rational and scientific designation and management of 'ecosystem and landscape conservation areas', 'wetland protection areas', 'special islands protection areas', and 'wildlife protected areas' which have been designated and managed by the Ministry of Environment. Through analysis of the requisites and criteria of IUCN, UNESCO, Natura 2000, the Ramsar convention, the United States, the United Kingdom, Japan, and Germany as well as various academic researches, evaluation items of the protected areas were classified into naturalness, biodiversity, ecosystem, and scientific values. These classification titles are reflection of Korean laws concerning the 4 protected areas described above. Of these items 'naturalness' is composed of 3 factors of wilderness, geomorphology and landscape, and vegetation. 'Biodiversity' is composed of the 5 factors of species diversity, endangered species, rare species, indigenous biological resources, and habitat of wetland wild animals. 'Ecosystem' is composed of 5 factors of typicalness, diversity, rarity, restoration ability, and degree of interference. All factors are scored using a 3 point scale of high, middle, or low and are then transformed into the numerical index for designating and zoning purposes. Conclusively, it is expected that the developed methodology will be highly applicable with field verifications.

A Short-chain Dehydrogenase/reductase Gene is Required for Infection-related Development and Pathogenicity in Magnaporthe oryzae

  • The phytopathogenic fungus Magnaporthe oryzae is a major limiting factor in rice production. To understand the genetic basis of M. oryzae pathogenic development, we previously analyzed a library of T-DNA insertional mutants of M. oryzae, and identified ATMT0879A1 as one of the pathogenicity-defective mutants. Molecular analyses and database searches revealed that a single TDNA insertion in ATMT0879A1 resulted in functional interference with an annotated gene, MGG00056, which encodes a short-chain dehydrogenase/reductase (SDR). The mutant and annotated gene were designated as $MoSDR1^{T-DNA}$ and MoSDR1, respectively. Like other SDR family members, MoSDR1 possesses both a cofactor-binding motif and a catalytic site. The expression pattern of MoSDR1 suggests that the gene is associated with pathogenicity and plays an important role in M. oryzae development. To understand the roles of MoSDR1, the deletion mutant ${\Delta}Mosdr1$ for the gene was obtained via homology-dependent gene replacement. As expected, ${\Delta}Mosdr1$ was nonpathogenic; moreover, the mutant displayed pleiotropic defects in conidiation, conidial germination, appressorium formation, penetration, and growth inside host tissues. These results suggest that MoSDR1 functions as a key metabolic enzyme in the regulation of development and pathogenicity in M. oryzae.

  • Hypercholesterolemia is a main risk factor to develop cardiovascular disease, a major cause of death in Korea currently. Dietary factors which shows hypocholesterolemic effect have been reported, which includes plant proteins, unsaturated fatty acids, milk, calcium, flavonoids. Numerous animal and human studies confirmed the pronounced hypocholesterolemic effects of soluble dietary fiber, such as psyllium husk, pectin, sodium alginate, guar gum. Several hypotheses have been proposed to explain the hypocholesterolemic effect of dietary fiber, including binding of bile acids by fiber, interference of lipid absorption and reduced hepatic cholesterol synthesis by propionate, a product of soluble dietary fiber fermentation. Several other hypotheses have been proposed, and these hypotheses are not mutually exclusive.

Photodecomposition Properties of Formaldehyde Using PS Nanofiber and Photocatalyst (극세섬유와 광촉매를 이용한 포름알데히드의 광분해 특성)

  • In this study we proposed on effect of the photodecomcomposition of coated nanofiber by $Pd/TiO_2$ for the removal of formaldehyde gas as indoor air pollutant. The photocatalytic reactor was setup in the inside of rectangular box (volume 2 l), UV lamp and the coating nanofiber with $Pd/TiO_2$. This study investigated the reaction rate and the adsorption constant of Langmuir-Heinshelwood, conversion of formaldehyde gas on temperature ($40^{\circ}C{\sim}80^{\circ}C$), effect of conversion (%) under different concentration, and effect of conversion (%) with humidity level on added $SO_2$ gas. As results, the rate constant (k) and adsorption constant (ft) were 114.94ppmv/min, $0.0036ppmv^{-1}$, respectively. and the conversion (%) of formaldehyde gas on temperature ($40^{\circ}C{\sim}80^{\circ}C$) was decreased to about 24%, compare with the first conversion (%). In conversion effect of increasing humidity levels, the presence of sulfur dioxide further decreased than without sulfur dioxide. the decreasing reason of conversion with presence sulfur dioxide judged as a cause of interference factor on the decrease of contact chance with photocatalysts.

  • The most shortcoming of metalhalide lamps is what the instant restarting cannot be realized when the arc tube is in the hot condition. The discharge starting voltage of arc tube in the hot condition is much higher than in the cold condition. Therefore it takes about five minutes to restart the metalhalide lamps, that is to say, it is possible to start when the pressure and the temperature are decreased. But, if the lamp is restarted in the hot condition, we must supply the high voltage pulse with 20[kV] at the both electrode of lamp. The proposed electronic ballast is consist of a electromagnetic interference(EMI) filter, a power factor correction(PFC) circuit, a flyback converter, a half-bridge inverter, and a high voltage igniter circuit. By this composition we can start/restart the lamp with the voltage 20[kV], even if the lamp is in the hot condition.

Identification of interacting proteins of retinoid-related orphan nuclear receptor gamma in HepG2 cells

  • The retinoid-related orphan nuclear receptor gamma ($ROR{\gamma}$) plays critical roles in regulation of development, immunity and metabolism. As transcription factor usually forms a protein complex to function, thus capturing and dissecting of the $ROR{\gamma}$ protein complex will be helpful for exploring the mechanisms underlying those functions. After construction of the recombinant tandem affinity purification (TAP) plasmid, pMSCVpuro $ROR{\gamma}$-CTAP(SG), the nuclear localization of $ROR{\gamma}$-CTAP(SG) fusion protein was verified. Following isolation of $ROR{\gamma}$ protein complex by TAP strategy, seven candidate interacting proteins were identified. Finally, the heat shock protein 90 (HSP90) and receptor-interacting protein 140 (RIP140) were confirmed to interplay with $ROR{\gamma}$ by co-immunoprecipitation. Interference of HSP90 or/and RIP140 genes resulted in dramatically decreased expression of CYP2C8 gene, the $ROR{\gamma}$ target gene. Data from this study demonstrate that HSP90 and RIP140 proteins interact with $ROR{\gamma}$ protein in a complex format and function as co-activators in the $ROR{\gamma}$-mediated regulatory processes of HepG2 cells.

  • In modern warfare, securing communication channel by combatting opponents' electromagnetic attack is a crucial factor to win the war. Military satellite digital transponder is a communication payload of the next generation military satellite that maintains warfare networks operational in the presence of interfering signals by securely relaying signals between ground terminals. The transponder in this paper is classified as a partial processing transponder which performs cost effective secure relaying in satellite communication links. The control functions of transmission security achieve immunity to hostile interferences which may cause malicious effects on the link. In this paper, we present an efficient architecture for implementing the control mechanism. Two major ideas of pipelined processing in per-group control and software processing of blocked band information dramatically reduce the complexity of the hardware. A control code sequence showing its randomness with uniform distribution is exemplified and qualification test results are briefly presented.

Inhibition of mIGF-1 and mGHR Gene Expression using Tetracycline-Inducible RNAi System in Mouse Liver Cell (Tetracycline 유도적인 RNAi System을 이용한 생쥐 성장 관련 유전자의 발현 억제)

  • In this study, to further understand the mechanism of animal growth and to develop a miniature transgenic animal model, we constructed and tested tetracycline-inducible RNAi system using shRNA targeting the mRNA of mouse insulin-like growth factor (mIGF-1) or mouse growth hormone receptor (mGHR) gene. Quantitative real-time PCR analysis of mouse liver cell (Hepa1c1c7) cells transfected with these vectors showed 85% or 90% of expression inhibition effect of IGF-1 or GHR, respectively. In ELISA analysis, the protein level of IGF-1 in the cells expressing the shRNA targeting IGF-1 mRNA was reduced to 26% of non-transformed control cells. Unexpectedly, in case of using shRNA targeting GHR, the IGF-1 protein level was decreased to 75% of control cells. Further experiments are needed to explain the lower interference effect of GHR shRNA in IGF-1 protein. Accumulated knowledge of this approach could be applicable to a variety of related biological area including gene function study, gene therapy, development of miniature animals, etc.
