Matrix metalloproteinase 1 (MMP-1), a calcium-dependent zinccontaining collagenase, is involved in the initial degradation of native fibrillar collagen. Tissue necrosis factor-alpha (TNFα) is a pro-inflammatory cytokine that is rapidly produced by dermal fibroblasts, monocytes/macrophages, and keratinocytes and regulates inflammation and damaged-tissue remodeling. MMP-1 is induced by TNFα and plays a critical role in tissue remodeling and skin aging processes. However, the regulation of the MMP1 gene by TNFα is not fully understood. We aimed to find additional cis-acting elements involved in the regulation of TNFα-induced MMP1 gene transcription in addition to the nuclear factor-kappa B (NF-κB) and activator protein 1 (AP1) sites. Assessments of the 5'-regulatory region of the MMP1 gene, using a series of deletion constructs, revealed the requirement of the early growth response protein 1 (EGR-1)-binding sequence (EBS) in the proximal region for proper transcription by TNFα. Ectopic expression of EGR-1, a zinc-finger transcription factor that binds to G-C rich sequences, stimulated MMP1 promoter activity. The silencing of EGR-1 by RNA interference reduced TNFα-induced MMP-1 expression. EGR-1 directly binds to the proximal region and transactivates the MMP1 gene promoter. Mutation of the EBS within the MMP1 promoter abolished EGR-1-mediated MMP-1 promoter activation. These data suggest that EGR-1 is required for TNFα-induced MMP1 transcriptional activation. In addition, we found that all three MAPKs, ERK1/2, JNK, and p38 kinase, mediate TNFα-induced MMP-1 expression via EGR-1 upregulation. These results suggest that EGR-1 may represent a good target for the development of pharmaceutical agents to reduce inflammation-induced MMP-1 expression.

In this paper, the performance of IS-95B CDMA system adopting multi-code scheme for data service on the forward link is obtained by theoretical analysis and computer simulation. The wireless channel is assumed to include AWGN, MUI(Multi-User Interference), and MPI(Multi-Path Interference), and multipath Rayleigh fading modeled by TDL(Tapped Delay Line). To obtain actual performance, IS-95B system using PN codes(M-sequence) concatenated with Walsh codes is considered. The BER performance of voice/data service satisfying QoS requirement and the maximum capacity of voice users is derived in terms of various system parameters(voice activity factor, required SNR of voice/data users, the number of multi-codes, the number of voice/data users.,etc.). As a result, under the condition that the maximum allowable number of voice user is 38 in voice only system, as the number of multi-code is decreased by 1, the maximum allowable number of voice user is increased by 5% for system with 1 data user and 22.3% for system with 4 data users. Therefore, it is possible to increase maximum allowable number of users in IS-95B CDMA system by appropriate control of the number of multi-codes and required SNR according to channel environment and traffic characteristic.

We consider the throughput-maximization problem for both the up- and downlink in a wireless network with interference channels. For this purpose, we design an iterative and distributive uplink algorithm based on Lagrangian relaxation. Using the uplink power prices and network duality, we achieve throughput-maximization in the dual downlink that has a symmetric channel and an equal power budget compared to the uplink. The network duality we prove here is a generalized version of previous research [10], [11]. Computational tests show that the performance of the up- and downlink throughput for our algorithms is close to the optimal value for the channel orthogonality factor, ${\theta}{\in}$(0.5, 1]. On the other hand, when the channels are slightly orthogonal (${\theta}{\in}$(0, 0.5]), we observe some throughput degradation in the downlink. We have extended our analysis to the real downlink that has a nonsymmetric channel and an unequal power budget compared to the uplink. It is shown that the modified duality-based approach is thoroughly applied to the real downlink. Considering the complexity of the algorithms in [6] and [18], we conclude that these results are quite encouraging in terms of both performance and practical applicability of the generalized duality theorem.
This study was performed to find the problems according to interference factors (organic matter, pH, Cr, Mn, Fe, clay, and etc.) when we analyzed the hexavalent chromium [Cr(VI)] in soils using UV/VIS spectrometer (US EPA 7196A), attempted to evaluate the domestic applicability of analytical method (US EPA 7199) using IC-UV/VIS spectrometer as alternative method. The recovery rate of certified reference materials was 75.0% (US EPA 7196A) and 101.4% (US EPA 7199) by the analytical methods. As the results of performing QA/QC about US EPA 7199, method detection limit (MDL) and limit of quantification (LOQ) were 0.062 mg/kg, 0.196 mg/kg, respectively. The LOQ of US EPA 7199 was lower than that of the current soil official testing method in Korea (0.5 mg/kg). Cr(VI) contents in 23 soil samples were compared by the analytical methods of EPA 7196A and 7199. Cr(VI) was detected in 13 of 23 soil samples by EPA 7196A, while EPA 7199 was not detected in any soil samples. The Cr(VI) content in 23 soil samples by EPA 7196A was not clearly correlated with Cr, Fe, Mn and clay content in the soil samples. However, the contents of Cr(VI) and organic matter of the soil samples had the highest coefficient of determination ($R^2$) of 0.80. In order to evaluate the correlation between the recovery rates of Cr(VI) and organic matter contents in the soil samples, the recovery rates of 5 soil samples added Cr(VI) standard solution were analyzed by the analytical methods. According to the results, the higher the organic matter contents in soil samples, the lower the recovery rates of Cr(VI) by US EPA 7196, while in case of US EPA 7199, the recovery rates were stable regardless of the organic matter contents.
To study effects of cellular FLICE (FADD-like IL-$1{\beta}$-converting enzyme)-inhibitory protein (c-FLIP) inhibition by RNA interference (RNAi) on sensitivity of U2OS cells to tumor necrosis factor (TNF)-related apoptosis-inducing ligand (TRAIL)-induced apoptosis, plasmid pSUPER-c-FLIP-siRNA was constructed and then transfected into U2OS cells. A stable transfection cell clone U2OS/pSUPER-c-FLIP-siRNA was screened from the c-FLIP-siRNA transfected cells. RT-PCR and Western blotting were applied to measure the expression of c-FLIP at the levels of mRNA and protein. The results indicated that the expression of c-FLIP was significantly suppressed by the c-FLIP-siRNA in the cloned U2OS/pSUPER-c-FLIP-siRNA as compared with the control cells of U2OS/pSUPER. The cloned cell line of U2OS/pSUPER-c-FLIP-siRNA was further examined for TRAILinduced cell death and apoptosis in the presence of a pan-antagonist of inhibitor of apoptosis proteins (IAPs) AT406, with or without 4 hrs pretreatment with rocaglamide, an inhibitor of c-FLIP biosynthesis, for 24 hrs. Cell death effects and apoptosis were measured by the methods of MTT assay with 3-(4,5-dimethylthiazol-2-yl)-2,5-diphenyltetrazolium bromide and flow cytometry, respectively. The results indicated that TRAIL-induced cell death in U2OS/pSUPER-c-FLIP-siRNA was increased compared with control cells U2OS/pSUPER in the presence or absence of AT406. Flow cytometry indicated that TRAIL-induced cell death effects proceeded through cell apoptosis pathway. However, in the presence of rocaglamide, cell death or apoptotic effects of TRAIL were similar and profound in both cell lines, suggesting that the mechanism of action for both c-FLIP-siRNA and rocaglamide was identical. We conclude that the inhibition of c-FLIP by either c-FLIP-siRNA or rocaglamide can enhance the sensitivity of U2OS to TRAIL-induced apopotosis, suggesting that inhibition of c-FLIP is a good target for anti-cancer therapy.
Down syndrome (DS) results from overexpressed genes on an extra copy of human chromosome 21. Among various phenotypes seen in DS patients, mental retardation, such as learning and memory deficits, is a major factor that prevents DS individuals from leading fully independent lives. The Dyrk1A gene that plays a critical role in neurodevelopment has been isolated from chromosome 21, and transgenic mice with over-expression of Dyrk1A show severe hippocampal dependent learning and memory defects. In the present study, as an initial step to test the treatment of Dyrk1A dependent mental retardation phenotypes in model animals, we isolated human Dyrk1A specific lentiviral short hairpin RNA (shRNA) that inhibits the exogenous human Dyrk1A expression, but not the endogenous mouse expression in transgenic mice with human Dyrk1A overexpression. This limited and specific repression of exogenous human Dyrk1A will prove to be valuable information, if Dyrk1A dependent learning and memory defects in DS patients could be treated or at least ameliorated in vivo.

OFDM (Orthogonal Frequency Division Multiplexing) system is robust to frequency selective fading and narrowband interference in high-speed data communications. However, an OFDM signal consists of a number of independently modulated subcarriers and the superposition of these subcarriers causes a problem that can give a large PAPR(Peak-to-Average Power Ratio). Phase rotation method can reduce the PAPR without nonlinear distortion by multiplying phase weighting factors. But computational complexity of searching phase weighting factors is increased exponentially with the number of subblocks and considered phase factor. Therefore, a new method, which can reduce computational complexity and detect phase weighting factors efficiently, should be developed. In this paper, a modeling process is introduced, which apply metaheuristic algerian in phase rotation method and optimize in PTS (Particle Swarm Optimization) scheme. Proposed algorithm can solve the computational complexity and guarantee to reduce PAPR We analyzed the efficiency of the PAPR reduction through a simulation when we applied the proposed method to telecommunication systems.
Objective: This study aimed to investigate expression of the proto-oncogene POK erythroid myeloid ontogenic factor (Pokemon) in colorectal cancer (CRC), and assess inhibitory effects of a small interference RNA (siRNA) expression vector in SW480 and SW620 cells. Methods: Semi-quantitative reverse transcription-polymerase chain reaction (PCR) and immunohistochemistry were performed to determine mRNA and protein expression levels of Pokemon in CRC tissues. Indirect immunofluorescence staining was applied to investigate the location of Pokemon in SW480 and SW620 cells. The siRNA expression vectors that were constructed to express a short hairpin RNA against Pokemon were transfected to the SW480 and SW620 cells with a liposome. Expression levels of Pokemon mRNA and protein were examined by real-time quantitative-fluorescent PCR and western blot analysis. The effects of Pokemon silencing on proliferation of SW480 and SW620 cells were evaluated with reference to growth curves with MTT assays. Results: The mRNA expression level of Pokemon in tumor tissues ($0.845{\pm}0.344$) was significantly higher than that in adjacent tumor specimens ($0.321{\pm}0.197$). The positive expression ratio of Pokemon protein in CRC (87.0%) was significantly higher than that in the adjacent tissues (19.6%). Strong fluorescence staining of Pokemon protein was observed in the cytoplasm of the SW480 and SW620 cells. The inhibition ratios of Pokemon mRNA and protein in the SW480 cells were 83.1% and 73.5% at 48 and 72 h, respectively, compared with those of the negative control cells with the siRNA. In the SW620 cells, the inhibition ratios of Pokemon mRNA and protein were 76.3% and 68.7% at 48 and 72 h, respectively. MTT showed that Pokemon gene silencing inhibited the proliferation of SW480 and SW620 cells. Conclusion: Overexpression of Pokemon in CRC may have a function in carcinogenesis and progression. siRNA expression vectors could effectively inhibit mRNA and protein expression of Pokemon in SW480 and SW620 cells, thereby reducing malignant cell proliferation.
In this paper the performance of a closed-loop power control scheme using variable step size decision method for DS/CDMA based-low earth orbit(LEO) mobile satellite systems in which the long round trip delay is a dominant performance degradation factor is evaluated. Because there are fundamental differences in the characteristics between the LEO mobile satellite channel and terrestrial mobile channel, such as long round trip delay and different elevation angle, these factors are considered in channel modeling based on the European Space Agency(ESA) measurement data. Since the round trip delay (from the mobile terminal to the gateway station via satellite) is typically 10∼20ms in low altitude satellite channels, closed-loop power control is much less effective than it is on a terrestrial channel. Thus, the adaptive power control scheme using a variable step size control is essential for overcoming the long round trip delay and fading due to the elevation angle. It is shown that the standard deviation of signal to interference ratio(SIR) adopting a variable step size closed-loop power control scheme is much less than that of a fixed step size closed-loop power control. Furthermore, we have driven the conclusion that the measurement interval of power control commands is optimal choice when it is twice the round trip delay.

Even though extensive research results have been applied to wireless cellular systems to improve their capacity and coverage, severe performance degradation experienced in cell boundary areas still remains as a major limiting factor to prohibit further improvement of user equipment (UE) throughput. In the Long Term Evolution-Advanced (LTE-A) standard of the Third Generation Partnership Project (3GPP), Some advanced techniques have been introduced to overcome this "cell-edge problem", including coordinated multipoint transmission and reception (CoMP) and inter-cell interference coordination (ICIC). In this paper, we propose yet another strategy to improve the performance of low-tier UEs by using the concept of multiple beam direction patterns (BDPs). Such multiple BDPs can be implemented using multi-layer antenna arrays stacked vertically at base station (BS) sites to transmit signals in different main beam directions. In comparison to conventional three-sector antennas with a fixed beam pattern, the proposed methods makes signal transmission in a rotational fashion to significantly enhance the reception quality of UEs located near sector (or cell) edge areas, preventing the situation where certain UEs are marginally covered by the BS for the whole transmission time. Performance evaluation results show that the proposed scheme outperforms the conventional three-sector transmission by 171% in low 5% UEs in terms of the UE throughput.
