  • In this study, we implement the potable blood gas analyzer measuring pH, $pCO_2\;and\;pO_2$ of the arterial blood. The implemented system by this study is divided into hardware and software part and also the hardware portion is parted by mechanism and electronic circuit mit. The system program is composed of operating, washing, correcting and measuring program. And to correct the system, two-point calibration method is used, one-point calibration method is also added for more accuracy, and system program is coded. For verifying the implemented system, We examine to response property of each electrode. And evaluate accuracy of the system using standard reagent and was construed as statistical.

High Accuracy Mass Measurement Approach in the Identification of Phospholipids in Lipid Extracts: 7 T Fourier-transform Mass Spectrometry and MS/MS Validation

  • In the present study, the approach of high accuracy mass measurements for phospholipid identifications was evaluated using a 7 T ESI-FTMS/linear ion trap MS/MS. Experiments were carried out for porcine brain, bovine liver, and soybean total lipid extracts in both positive and negative ion modes. In total, 59, 55, and 18 phospholipid species were characterized in the positive ion mode for porcine brain, bovine liver, and soybean lipid extracts, respectively. Assigned lipid classes were PC, PE, PEt, PS, and SM. In the negative ion mode, PG, PS, PA, PE, and PI classes were observed. In the negative ion mode, for porcine brain, bovine liver, and soybean lipid extracts, 28, 34, and 29 species were characterized, respectively. Comparison of our results with those obtained by other groups using derivatization-LC-APCI MS and nano-RP-LC-MS/MS showed that our approach can characterize PC species as effectively as those methods could. In conclusion, we demonstrated that high accuracy mass measurements of total lipid extracts using a high resolution FTMS, particularly, 7T FTMS, plus ion-trap MS/MS are very useful in profiling lipid compositions in biological samples.

Nonribosomal Peptide Synthase is Responsible for the Biosynthesis of Siderophore in Vibrio vulnificus MO6-24/O

  • Vibrio vulnificus produces siderophores, low-molecular-weight iron-chelating compounds, to obtain iron under conditions of iron deprivation. To identify genes associated with the biosynthesis of siderophore in V. vulnificus MO6-24/O, we screened clones with mini-Tn5 random insertions for those showing decreased production of siderophore. Among 6,000 clones screened, nine such clones were selected. These clones contain the transposon inserted in VV2_0830 (GenBank accession number) that is a homolog of a nonribosomal peptide synthase (NRPS). There is an another NRPS module, VV2_0831, 49-bp upstream to VV2_0830. We named these two genes vvs (Vibrio vulnificus siderophore synthase) A and B, respectively. Mutation of either vvsA or vvsB showed a decreased production of siderophore. The expression of an NRPS-lux fusion was negatively modulated by the presence of iron, and the regulation was dependent on Fur (ferric uptake regulator). However, the expression of the NRPS genes was still not fully derepressed in the iron-rich condition, even in furnull mutant cells, suggesting that some other unknown factors are involved in the regulation of the genes. We also demonstrated that the NRPS genes are important for virulence of the pathogen in a mice model.

Estimation of the Blood Pressure Using Point Variation Aspect of Dicrotic Notch on Pulsating Waveform at Each Cardiac Periods (주기별 맥동파형의 절흔점 위치변화 특성을 이용한 혈압 추정)

  • In the study, novel blood pressure estimation method was proposed to improve the accuracy of oscillometric method. The proposed algorithm estimated the blood pressure by comparing and analyzing the point variation aspect of dicrotic notch on pulsating waveform during each cardiac cycle. The waveforms of each cardiac cycle were extracted by maximum points. The extracted pulsating waveforms were applied by re-sampling, end-matching, and normalization. The systolic and diastolic blood pressures were estimated by point variation aspect of dicrotic notch. The blood pressures, which were estimated from proposed algorithm, were compared and analyzed by blood pressures from oscillometric methods and auscultation. The systolic blood pressure from oscillometric methods were +0.88 mmHg more than proposed algorithm, and 1.875 less than the diastolic blood pressures from proposed algorithm. The systolic and diastolic blood pressures from auscultation were 2.89 mmHg and 3.44 mmHg less than the blood pressures from proposed algorithm. As the errors between blood pressures from proposed algorithm, oscillometric method and auscultation were less than 5 mmHg, the proposed algorithm was effective.

IVET-based Identification of Virulence Factors in Vibrio vulnificus MO6-24/O

  • Vibrio vulnificus is an opportunistic pathogen that causes septicemia in humans. To identify the genes associated with its pathogenicity, in vivo expression technology (IVET) was used to select genes specifically expressed in a host, yet not significantly in vitro. Random lacZ-fusions in the genome of V vulnificus strain MO6-24/O were constructed using an IVET vector, pSG3, which is a suicide vector containing promoterless-aph and -lacZ as reporter genes. A total of ${\sim}18,000$ resulting library clones were then intraperitoneally injected into BALB/c mice using a colony forming unit (CFU) of $1.6{\times}10^6$. Two hours after infection, kanamycin was administered at $200{mu}g$ per gram of mouse weight. After two selection cycles, 11 genes were eventually isolated, which were expressed only in the host. Among these genes, VV20781 and VV21007 exhibiting a homology to a hemagglutinin gene and tolC, respectively, were selected based on having the highest frequency. When compared to wild-type cells, mutants with lesions in these genes showed no difference in the rate of growth rate, yet a significant decrease in cytotoxicity and the capability to form a biofilm.

A Study on the Electrochemical Synthesis of L-DOPA Using Oxidoreductase Enzymes: Optimization of an Electrochemical Process

  • Levodopa or L-3,4-dihydroxyphenylalanine (L-DOPA) is the precursor of the neurotransmitter dopamine. L-DOPA is a famous treatment for Parkinson's disease symptoms. In this study, electroenzymatic synthesis of L-DOPA was performed in a three-electrode cell, comprising a Ag/AgCl reference electrode, a platinum wire auxiliary electrode, and a glassy carbon working electrode. L-DOPA had an oxidation peak at 376 mV and a reduction peak at -550 mV. The optimum conditions of pH, temperature, and amount of free tyrosinase enzyme were pH 7, $30^{\circ}C$, and 250 IU, respectively. The kinetic constant of the free tyrosinase enzyme was found for both cresolase and catacholase activity to be 0.25 and 0.4 mM, respectively. A cyclic voltammogram was used to investigate the electron transfer rate constant. The mean heterogeneous electron transfer rate ($k_e$) was $5.8{\times}10^{-4}$ cm/s. The results suggest that the electroenzymatic method could be an alternative way to produce L-DOPA without the use of a reducing agent such as ascorbic acid.
