구속스트레스에 의한 백서 악하선의 Amylase 분비 변화 (Change of the Amylase Secretion on the Rat Submandibular Gland in the Restraint Stress Condition)

  • 현대인들에게는 다양한 증상과 징후를 나타내는 스트레스성 질환이 증가되고 있는 추세에 있다. 이러한 스트레스 시에 구강 내에는 구강건조이나 구강작열감증후군, 구강점막질환, 구취 등이 호발 된다는 것은 이미 널리 알려진 사실이다. 이때 대부분의 환자에서 공통적으로 나타나는 현상은 타액선의 기능적 및 구조적 변화에 따른 타액성분의 변화이다. 이에 저자는 스트레스가 구강 내 타액선에 어떠한 영향을 미치며, 타액선으로부터 분비되는 ${\alpha}$-amylase에 어떠한 영향을 미치는지를 알아보고자 본 실험을 시행한 결과 다음과 같은 유의성 있는 결과를 얻었다. 1. 구속 스트레스에 의하여 타액선 조직은 선포세포가 공포화되며 변성되었고, 선포간극이 이개되었으며, 도관주위조직도 변성되었다. 2. 구속 3시간 후에는 선포세포가 위축되기 시작하였고, 구속 6시간 후에는 선포 간 이개현상이 관찰되었으며, 구속 72시간 후에는 도관 주위 조직도 변성되기 시작하였고, 점차 심화되어 구속 168시간 후에는 선포와 도관 주위조직이 모두 심하게 변성되었다. 3. 면역조직화학적 검사에서 구속 3시간 후에는 amylase의 발현이 부분적으로 불규칙하게 관찰되었으며, 구속 6시간 후에는 다소 감소되었고, 구속 12시간 후에는 amylase가 부분적으로 응집되었으며, 점차 감소되고 응집되어, 구속 48시간 후에는 전 타액선에 amylase의 응집현상이 심화되었고, 구속 168시간 후에는 amylase의 발현이 매우 감소되었다. 이상의 결과를 바탕으로 정서적인 스트레스를 받으면 타액선이 변성될 수 있으며, 타액선으로부터 분비되는 중요한 소화 효소인 amylase가 변화되어 소화 장애를 일으키거나 구강건강과 관계없이 구취발생에도 영향을 미칠 수 있다고 예측할 수 있어, 향후 amylase의 분비변화에 따른 지속적인 연구가 필요할 것으로 생각된다.

한국재래산양 태아 및 신생아의 결장상피 발달에 관한 투과전자현미경적 연구 (Transmission electron microscopical study of the developing colonic epithelia in fetuses and neonates in Korean native goat (Carpus hircus))

  • The morphological development of colonic epithelia in fetuses between 60-. 90-, and 120-days gestation and neonates of Korean native goat were investigated by transmission electron microscopy. In the 60-day-old fetuses, the colonic epithelial cells contained nuclei, nucleoli, mitochondria, free ribosomes, and shoo granular and agranular endoplasmic reticula. The zonula occludens, zonula adherens, desmosomes, short microvilli, and masses of glycogen granules were also obsrved. The goblet cells contained a few secretory granules. In the 90-day-old fetuses, the cell organelles of the colonic epithelial cells were better developed than those in the 60 day old fetuses. Increased number of endoplasmic reticula, digitiform intercellular junctions, mitochondria, and Golgi complexes was observed. The goblet cells contained a lot of secretory granules. In the 120-day-old fetuses, the colonic epithelial cells contained long microvilli and well developed cell organelles. The nuclear cleft and large intercellular space were also appeared. Nunerous fibroblasts were seen in the basement membrane. The number of goblet cells was further observed. In the 120 day old fetuses, all colonic epithelial cells shape simple columnar cells. In newborns, the colonic epithelial cells were covered with extensive microvilli. There were many goblet cells with a lot of secretory granules protruding into the intestinal lumen, and some goblet cells secreted their secretory granules into the lumen. In the 60-and 90-day-old fetuses, the colonic epithelial cells appeared to be either simple columnar or stratified columnar depending on areas.

CEPA 처리(處理)가 옻나무의 칠액분비(漆液分泌) 및 수피생리(樹皮生理)에 미치는 영향(影響) (Effects of CEPA on the Secretion of Lacquer and Bark Physiology of Lactree(Rhus verniciflua Stokes))

  • 본 연구는 CEPA(2-chloroethyl phosphonic acid)처리가 옻나무(Rhus verniciflua Stokes)의 수피생리 및 해부학적 변화와 칠액의 분비촉진에 미치는 효과를 알아보기 위하여 수행되었다. 강원도 횡성군에 조성된 7년생 옻나무단지에서 크기가 비슷하고 생장이 균일한 개체 40본을 선발하여, CEPA가 각각 0%, 0.1%, 1%, 10% 함유된 lanolin pastes를 1.2m 높이에서 4방향으로 처리하고 5주 후 농도별 수피두께와 수피내 옻산함량을 측정하였다. 또한 수피조직의 해부학적 변화를 관찰하기 위하여 수피조직을 광학현미경하에서 관찰하였다. CEPA 처리시 수피의 내피두께는 현저히 증가하였으며 수피내 옻산 생산량과 밀접한 상관관계를 보였다. CEPA 0.1% 처리시 평균 내피두께는 1.65mm이고, 1% 처리구는 1.96mm였으며, 10% 처리구에서 3.59mm로 대조구의 1.43mm에 비해 약 2.5배 증가하였다. 이는 사부조직의 확대와 세포간극이 넓어진 것에 기인한다. 또한 CEPA에 의해 수피내 옻산 분비가 촉진되어 CEPA 처리목은 무처리목에 비해 더 많은 양의 옻산을 분비하였다. 수피내 단위면적당 평균 옻산함량은 CEPA 0.1% 처리구의 경우 $5.11mg/cm^2$이였고, 1% 처리구는 $6.86mg/cm^2$였으며, 10% 처리는 $12.12mg/cm^2$으로 대조구의 $4.29mg/cm^2$보다 2.8배 증가하였다. 옻나무 내피를 해부학적으로 관찰한 결과 단위면적당 칠액구 수에는 큰 차이가 없었지만 전체 칠액구 수는 유의하게 증가하였고, 칠액구의 크기가 외피쪽으로 갈수록 현저히 커졌다. 또한 CEPA를 처리한 나무의 경우 유세포가 치밀하였으며, ray층 수가 무처리구의 경우 1-2층인데 반하여 처리구에서는 2-3층으로 잘 발달되어 있었다.

산양에 있어서 분만후 자궁의 변화 II. 한국재래산양에 있어서 분만후 자궁 및 질상피세포의 전자현미경적 관찰 (Postpartum Changes in the Uterus of Goats II. Electron Microscopic Observations in the Uterine and Vaginal Epithelium of Post-partum Korean Native Goats)

  • Morphological changes in the uterine and vaginal epithelial cells of the Korean native goats were studied in fifteen primiparous goats slaughtered on the day of parturition and on days 1, 3, 10 and 21 postpartum. 15 uterus and vagina from goats were examined by scanning and transmission electron microscopy. The results obtained in this study were summarized as follows : 1. Transmission electron microscopically, long microvilli which sometimes ramified were found until 10 days postpartum, while short microvilli were found at 21 days. The high electron dense irregular-shaped mitochondria were found in the cytoplasm and the crystalline structure of the mitochondrial matrix was also found from 1 day to 10 days postpartum. Well-developed rough-endoplasmic reticulum (rER) with dilated cisternae which contained the proteins materials was observed at 21 days postpartum. These materials were fused each other and then large granules were found in the free surface of the cytoplasm. A few lipid droplets were generally appeared in the cytoplasm, while numerous droplets were found at 21 days postpartum. A moderate number of ribosomes, a few multivesicular bodies, vesicles, lysosomes and macrophages were found. The globule leucocytes were observed from 0 to 3 days postpartum by transmission electron microscopy. The short microvilli, high electron dense cytoplasm and severe indentation of the nuclear enbelope were found in the vaginal epithelium. Numerouos small vesicles and a few vacuoles were observed in the apical cytoplasmic portion of the epithelium. A few mitochondria were high electron dense and irregular in shape. A moderate amounts of microfilaments, loose intercellular space and dilated rER were also found at 21 days postpartum. 2. Scanning electron microscopically, the folds of the uterine mucosa were generally deep. The long microvilli of the epithelium were found until 3 days postpartum, while short microvili were found at 10 and 21 days postpartum. The distinct intercellular boundary was seen. The apporcine secretory profile of the epithelium observed at between 3 and 10 days postpartum and the cells were somewhat protruded into the lumen. The short microvilli were found on the surface of the protruded cells, while polygonal microridge profile of the epithelium and some dome-shaped epithelium were also observed at 21 days postpartum. The folds of the vaginal mucosa were deep and epithelium was polygonal in shape. The microvilli of the epithelium were long until 3 days postpartum, while they were short at 10 and 21 days. The polygonal epithelium was invaginated into the center of the cell surface until 10 days postpartum. The microridge and dome in shape of the epithelium were found at 10 days postpartum, while the polygonal and exfoliating epithelium were observed at 21 days.

바퀴의 後腸 上皮細胞들에 대한 微細構造 (Ultrastructure of the Hindgut Epithelial Cells in the Cockroach, Blattella germanica L.)

  • 가옥내에 서식하는 바퀴 (Blattella germanica Linne)의 後腸을 回腸과 直腸部로 구분하여, 그들을 구성하는 上皮細胞의 微細構造를 관찰하였다. 回腸은 扁平 혹은 立方型의 細胞들이 單層上皮를 만들며 그 內面은 cuticular intima로 덮여 있었다. 細胞들은 細胞器官이 고르게 발달하여 吸收細胞의 形態를 하였으며, 細胞質內에서 동심원적인 環狀構造를 하는 層板狀 結晶體, "spherites"가 많이 관찰됨이 특징이었다. 直腸은 圓柱狀인 圓柱狀吸收細胞의 尖端部原形質膜은 微細絨毛로 변형되었고, 側部原形質膜은 약 200$\\AA$간격으로 심하게 주름이 잡혔으며 그 사이에는 크게 발달한 mitochondria가 밀착하여 mitochondrial-scalarifrom complexes를 형성하였다. 그리고 인접 細胞間에는 많은 細胞間隙을 만들며, 그 사이 사이에는 septate junction과 desmosomes 등으로 연접된 것이 여러 곳에서 관찰되었다. 基底細胞는 납작한 杯모양으로서 細胞質의 電子密度가 높아 검게 보였으며, 그 내에는 Golgi complex, endoplasmic reticulum 그리고 mitochondria 등 細胞器官들이 발달되었고 膜의 구조 등으로 미루어 보아 역시 吸收機能을 가진 細胞임을 확인하였다. 한편 直腸의 上皮層과 hemolymph 사이에서 壁의 역할을 하는 basal sheath가 基底面을 따라 평행하게 부착되어 있고, 그 basal sheath와 근육층 사이에는 結合組織이 넓은 subepithelial space를 만들어서 그 내에는 많은 氣官小支들과 밀도가 높은 顆粒들을 갖는 軸索이 분포하고 있었다.

초등학교 교사들의 세포에 대한 개념 (Elementary School Teachers에 Conceptions on Cell)

  • The purpose of this study is to investigate degree of the concept formation on cell in elementary school teachers and to clarify the patterns of their misconceptions. Data were collected by interview with 120 elementary school teachers, ranged from twenties to fifties in age, working in Taegu city. The instrument was developed by researchers and was categorized into four: cell as the basic units of life, morphology of cell, function of cell, growth of individual from the viewpoint of cell. The results are as follows: First, about 80% of teachers had two perspective outcome of cell: Cells are the basic units of life on earth. But the potato and meat, which we used to eat as food, are not constructed of cells but aggregated of nutrients. Second, most of elementary school teachers recognized that shapes of the cell in organisms were diverse, but some of them only could present several kinds of shape on cell. The 35% of teachers had misconception that shape of cell is all the same in a individual. It shows that this result is caused by lack of opportunity to observe the various kinds of cells. Third, most teachers understood tell can be seen only through a microscope. Yet in comparison with relative size, it was revealed that misconception of cell size was induced by the term of 'basic unit'. In addition, they thought that large organisms are built from large cells, and small organisms from small cells. Fourth, Elementary school teachers used to confuse the terms between chloroplast and chlorophyll, and believed that the genetic material was contained only in the reproductive cells and seeds. It was also revealed that they thought the nutrients such as starch and fat are located at intercellular space. Fifth, the 60% of teachers conceived correctly that growth of individual depends on mitosis and increase in cell number. The rest of them, however, misunderstood that it is due to the increase of tell volume.

방사선조사가 백서 협점막에 미치는 영향에 관한 전자현미경적 연구 (AN ELECTRON MICROSCOPIC STUDY ON THE EFFECTS OF IRRADIATION ON THE BUCCAL MUCOSA OF RAT)

  • The author studied the acute reaction of cobalt-60 irradiation to buccal mucosa in rats and difference of the effects of single versus fractionated exposure. 195 Sprague Dowley strain rats, weighing about 120gm, were used in this experiment. 3 rats served as controls and the remaining 192 rats were divided into six groups of 32 rats each. Experimental group Ⅰ, Ⅱ, Ⅲ were received a single dose of 15Gy, 16.5Gy, 18Gy and group Ⅳ, Ⅴ, Ⅵ were received two equal sized fractionated dose of 9Gy, 9.75Gy, 10.5Gy at 4 hour intervals, respectively. The experimental groups were irradiated with cobalt-60 teletherapy unit, Picker model 4M 60 (Field size, 12x5 cm, SSD, 50㎝, Dose rate, 222cGy/min, Depth, 1㎝). The animals were sacrificed at 1, 2, 3, 6, 12 hours, 1, 3, 7 days after irradiation and the changes of the irradiated buccal mucosa were observed by electron and light microscopy. The results were as follows: 1. A single exposure was more damaging than fractionated exposure, and as the radiation dose increased, the changes of cell organelles became faster, but the healing of radiation-induced damage in fractionated exposure was faster than in single exposure. 2. The radiation-induced changes of the basal cells were the most prominent in 18Gy-single exposure group, and the least in 18Gy-fractionated exposure group. 3. Electron-microscopically, there appeared nuclear changes, swelling of mitochondria and rough endoplasmic reticulum, decrease of free ribosome, presence of vesicles, widening of intercellular space, and loss of basal lamina. The early remarkable changes were partly loss of nuclear membrane and swelling of mitochondria. 4. Light-microscopically, derangement and pyknosis of basal cells, hydropic changes of spinous cells, enlargement of granular cells, indistinctness of basement membrane, and proliferation of epithelium were observed.

소풍도적탕(消風導赤湯)이 아토피 피부염의 항염증효과와 각질층 ceramide변화에 미치는 영향 (Changes in Ceramide in Stratum Corneum and Anti-inflamatory Effects of Sopungdojeok-tang on Atopic Dermatitis)

  • Objective : This study was designed to identify lipid protection formation in stratum corneum and anti-inflammatory effects of Sopungdojeok-tang(SD) on atopic dermatitis(AD). Materials and Methods : In Vivo, SD extract was orally administered to BALB/c mice at $2.5m{\ell}/kg/day$ for 2 days after 5% sodium dodecyl sulfate evoked atopic dermatitis in abdominal skin. Morphological changes were observed by immunohistochemical stain using monoclonal antibodies(BrdU, ceramide, MIP-2, $NF-{\kappa}B$ p50, IL-4, and STAT6) and TUNEL method. In vitro, the alterations of IL-4 mRNA expression were detected by RT-PCT in SD extract treated EL4 cells after phorbol-12-myristate-13-acetate and 4-tert-Octylphenol induce Th2 skewed condition. Results : SD is used in Oriental Medicine for its potential curative for atopic dermatitis. In this study, we have investigated the anti-inflammatory and lipid lamella repair effects of SD were investigated. SD decreased the number of eosinophil in atopic dermatitis induced mice. In the histological properties, the hyperplasia, edema, infiltration of lymphocytes, damage of intercellular space of stratum corneum, BrdU positive reacted cells in stratum basal, and degranulated mast cells and capillaries in dermal papillae decreased in mice with SD. Treatment of SD also decreased MIP-2, STAT6 and IL-4 in dermal papillae. The IL-4 mRNA expression decreased in a dose-dependant manner in SD treated EL4 cells. In addition, decrease of $NF-{\kappa}B$ p50 and increase of apoptotic cells in dermis were observed in SD treated mice. These data suggest that SD may beneficial for atopic dermatitis. Conclusions : These data suggest that SD is beneficial in treatment of atopic dermatitis, and that SD provides lipid protection in stratum corneum and anti-inflammatory effects on atopic dermatitis.

Pseudomonas syringae pv. tomato DC3000 Improves Escherichia coli O157:H7 Survival in Tomato Plants

  • Recently, outbreaks of food-borne diseases linked to fresh produce have been an emerging public health concern worldwide. Previous research has shown that when human pathogens co-exist with plant pathogens, they have improved growth and survival rates. In this study, we have assessed whether Escherichia coli O157:H7 benefits from the existence of a phytopathogenic bacterium and the underlying mechanisms were further investigated. When Pseudomonas syringae pv. tomato DC3000 (Pst DC3000) and E. coli O157:H7 were co-inoculated by either dipping or infiltration methods, the populations of E. coli O157:H7 increased; however, no effect was observed when type three secretion system (T3SS) mutants were used instead, suggesting that E. coli O157:H7 benefits from the presence of Pst DC3000. In addition, this study confirmed that the E. coli O157:H7 populations increased when they occupied the tomato leaf intercellular space; this colonization of the interior of the leaves was possible due to the suppression of the PAMP-triggered immunity (PTI) by Pst DC3000, in particular with the AvrPto effector. In conclusion, our data support a plausible model that E. coli O157:H7 benefits from the presence of Pst DC3000 via AvrPto suppression of the PTI resistance.

대기-엽 수증기압차에 의한 산마늘과 울릉산마늘의 광합성 반응 비교 (Comparison of Photosynthetic Responses in Allium microdictyon Prokh and Allium ochotense Prokh from Atmosphere-Leaf Vapor Pressure Deficit (VPD))

  • This study was conducted to investigate the effect of atmosphere-leaf vapor pressure deficit (VPD) in Allium microdictyon Prokh. and Allium ochotense Prokh. The vapor pressure deficit (VPD) was rapidly increased with increasing temperature and decreasing relative humidity. Taken as a whole, the stomatal transpiration reaction was slightly late with increading of VPD. Maximum photosynthetic rate at high-VPD condition was 5.98 ${\mu}mol$ $CO_2{\cdot}m^{-2}{\cdot}s^{-1}$ in Allium microdictyon, which was a little lower than 6.59 ${\mu}mol$ $CO_2{\cdot}m^{-2}{\cdot}s^{-1}$ in Allium ochotense, respectively. After 2 p.m, stomatal transpiration of Allium microdictyon at the high VPD condition were rapidly decreased. Ci/Ca began to decline sharply at 8 a.m and showed the lowest value at 2 p.m, The results showed that Ci/Ca decreased with being used $CO_2$ in the mesophyll intercellular space for photosynthesis. In high VPD condition, The water potential values showed the highest at 5 a.m, and the lowest at 1 p.m in high VPD condition. The water saturation deficits (WSD) in high VPD condition showed about 1.5 times higher than in low VPD condition. The results indicated that physiological activities in Allium microdictyon is more limited from high VPD conditions.
